1The Association of Maternal Body Composition and Dietary Intake with the Risk of Gestational Diabetes Mellitus during the Second Trimester in a Cohort of Chinese Pregnant Women显示文摘Objective To investigate the association of maternal body composition and dietary intake with the risk of gestational diabetes mellitus(GDM). Methods A total 154 GDM subjects and 981 controls were enrolled in a prospective cohort study in 11 hospitals from May 20, 2012 to December 31, 2013. Bioelectrical impedance analysis and dietary surveys were used to determine body composition and to evaluate the intake of nutrients in subjects at 21-24 weeks' gestation(WG). Logistic regression analysis was applied to explore the relationships of maternal body composition and dietary intake with the risk of GDM morbidity. Results Age, pre-pregnant body weight(BW), and body mass index(BMI) were associated with increased risk of GDM. Fat mass(FM), fat mass percentage(FMP), extracellular water(ECW), BMI, BW, energy, protein, fat, and carbohydrates at 21-24 WG were associated with an increased risk of GDM. In contrast, fat free mass(FFM), muscular mass(MM), and intracellular water(ICW) were associated with a decreased risk of GDM. 2Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. 3The rice OsV4 encoding a novel pentatricopeptide repeat protein is required for chloroplast development during the early leaf stage under cold stress显示文摘Pentatricopeptide repeat(PPR)proteins,characterized by tandem arrays of a 35 amino acid motif,have been suggested to play central and broad roles in modulating the expression of organelle genes in plants.However,the molecular mechanisms of most rice PPR genes remains unclear.In this paper,we isolated and characterized a temperature-conditional virescent mutant,OsV4,in rice(Oryza sativa cultivar Jiahua1(WT,japonica rice variety)).The mutant displays albino phenotype and abnormal chloroplasts at the three leaf stage,which gradually turns green after the four leaf stage at a low temperature(20°C).But the mutant always develops green leaves and well-developed chloroplasts at a high temperature(32°C).Genetic and molecular analyses uncovered that OsV4 encodes a novel chloroplast-targeted PPR protein including four PPR motifs.Further investigations show that the mutant phenotype is associated with changes in chlorophyll content and chloroplast development.The OsV4transcripts only accumulate to high levels in young leaves,indicating that its expression is tissue-specific.In addition,transcript levels of some ribosomal components and plastid-encoded polymerase-dependent genes are dramatically reduced in the albino mutants grown at 20°C.These findings suggest that OsV4 plays an important role during early chloroplast development under cold stress in rice.Xiaodi Gong Qianqian Su Dongzhi Lin Quan Jiang Jianlong Xu Jianhui Zhang Sheng Teng Yanjun Dong 2014Journal of Integrative Plant Biology2014,56,4:19
4Design and fabrication of broadband ultralow reflectivity black Si surfaces by laser micro/nanoprocessing显示文摘Light collection efficiency is an important factor that affects the performance of many optical and optoelectronic devices.In these devices,the high reflectivity of interfaces can hinder efficient light collection.To minimize unwanted reflection,anti-reflection surfaces can be fabricated by micro/nanopatterning.In this paper,we investigate the fabrication of broadband anti-reflection Si surfaces by laser micro/nanoprocessing.Laser direct writing is applied to create microstructures on Si surfaces that reduce light reflection by light trapping.In addition,laser interference lithography and metal assisted chemical etching are adopted to fabricate the Si nanowire arrays.The anti-reflection performance is greatly improved by the high aspect ratio subwavelength structures,which create gradients of refractive index from the ambient air to the substrate.Furthermore,by decoration of the Si nanowires with metallic nanoparticles,surface plasmon resonance can be used to further control the broadband reflections,reducing the reflection to below 1.0%across from 300 to 1200 nm.An average reflection of 0.8%is achieved.Jing Yang Fangfang Luo Tsung Sheng Kao Xiong Li Ghim Wei Ho Jinghua Teng Xiangang Luo Minghui Hong 2014Light(Science & Applications)2014,3,1:17

6QTL analysis of rice low temperature germinability显示文摘A double haploid population, derived from anther culture of F, hybrid between a typical indica and a japonica (ZYQ8/JX17), has been used to investigate the low temperature germinability (LTG) at 15℃. The low temperature germinability of two parents was significantly different. In 6-11 d, the germination percentage of ZYQ8 was higher than that of JX17. In 12-16 d, the germination percentage of JX17 was higher than that of ZYQ8. The quantitative trait loci (QTLs) of every day for low temperature germinabilityhave been mapped based on a molecular linkage map constructed from this population. In 8-11 d, qLTG-9 was identified in C397B-RZ617B on chromosome 9, the additive effect was positive, showing that, the allele from JX17 could increase low temperature germinability. In 12-16 d, qLTG-4 was mapped between RG908 and CT563 on chromosome 4, the additive effect was negative, showing that the allele from ZYQ8 could increase low temperature germinability. 7Isolation and genetic charac-terization of a fragile plant mutant in rice (Oryza sativa L.)显示文摘The fragile rice mutant was isolated from an M2 population of indica variety Shuang Ke Zao (SKZ) treated with γ-rays, and designated as fp1 {fragile plant 1) because of its fragile leaves and culms. To map FP1 locus, an F2 mapping population was derived from a cross between the fp1 and C-bao, a polymorphic japonic variety. The primary mapping result places the FP1 locus in an interval between two molecular markers, microsatellite marker RM16 (3.1 cM proximal to FP1) and STS marker G144a (9.1 cM distal to FP1) in the centromere region of chromosome 3. A CAPS marker C524a was further developed between RM16 and G144a, with 0.4 cM genetic distances to the FP1 locus, providing a practical starting point for constructing a BAC con-tig spanning the FP1 locus and cloning the fp1 gene. 8The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 10The Chinese classification system compared with TNM staging in prognosis of patients with primary hepatic carcinoma after resection显示文摘BACKGROUND: The life expectancy of a patient with primary hepatic carcinoma (PHC) is hard to predict, and it is related to many prognostic factors. The Chinese classification system including five parameters: tumor, vascular thrombosis, lymph node metastasis, distant metastasis and Child-Pugh stage developed in 1999 was adopted by the 8th National Conference on liver Cancer of the Chinese Anti-Cancer Association in 2001. In this study, the discriminatory ability of the Chinese classification system -was compared with that of the TNM staging in patients for resection of PHC, in addition to the evaluation of prognostic value. METHODS: The data of 246 patients who had undergone resection of PHC from January 1986 to December 2000 (average age, 51 years; male/female ratio, 213/33) were retrospectively studied. Among the 246 patients, 227 were followed up for at least 3 years. RESULTS: The 1-, 3-, 5-, 7-, and 10-year tumor-free survival rates were 55% , 30% , 25% , 20% and 18% , respectively. The Chinese classification system was better than the TNM staging system in predicting survival rate of patients with PHC, as confirmed by survival curves shown by the Kaplain-Meier method. The mean survival time was 155, 70, 39, 16, and 4 months in patients with die Chinese classification stages Ⅰa, Ⅰb, Ⅱa,Ⅱb, and Ⅲ, respectively. The 1-, 3-, 5-, 7-, and 10-year tumor-free survival rates of the Chinese classification system and TNM staging were statistically significant and had a slightly positive relationship. The predictive capacity of the Chinese classification system was confirmed in any two subgroups of patients undergoing operation. COX proportional hazards regression analysis showed that the Chinese classification system was the only independent prognostic factor for survival. 11BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays.Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y. 12Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 13Association study of the single nucleotide polymorphisms in adiponectin-associated genes with type 2 diabetes in Han Chinese显示文摘Single-nucleotide polymorphisms (SNPs) of ADIPOQ, ADIPOR1, and ADIPOR2 have been associated with type 2 diabetes mellitus (T2DM), but there are many conflicting results especially in Chinese populations. To investigate the contribution of the adiponectin genes and their receptors to T2DM, a case-control study was performed and 11 SNPs of ADIPOQ, ADIPOR1, and ADIPOR2 were genotyped in 985 T2DM and 1,050 control subjects. rs16861194 (-11426 A>G) in the putative promoter of ADIPOQ was associated with T2DM (P = 0.007; OR = 1.29, 95% CI 1.08-1.55). None of the other 10 SNPs were associated with T2DM in this study, although rs2241766 and rs1501299 were reported to be associated with T2DM in previous Chinese studies. There was also no significant difference found from the ADIPOQ haplotype analysis, which contains rs16861194. In addition, we also assessed potential gene-gene interactions in three genes and no interactions were found. 15Atorvastatin attenuates surgery-induced BBB disruption and cognitive impairment partly by suppressing NF-κB pathway and NLRP3 inflammasome activation in aged mice显示文摘In clinic,perioperative neurocognitive disorder is becoming a common complication of surgery in old patients.Neuroinflammation and blood-brain barrier(BBB)disruption are important contributors for cognitive impairment.Atorvastatin,as a strong HMG-CoA reductase inhibitor,has been widely used in clinic.However,it remains unclear whether atorvastatin could prevent anesthesia and surgery-induced BBB disruption and cognitive injury by its anti-inflammatory property.In this study,aged C57BL/6J mice were used to address this question.Initially,the mice were subject to atorvastatin treatment for 7 days(10 mg/kg).After a simple laparotomy under 1.5%isoflurane anesthesia,Morris water maze was performed to assess spatial learning and memory.Western blot analysis,immunohistochemistry,and enzyme-linked immunosorbent assay were used to examine the inflammatory response,BBB integrity,and cell apoptosis.Terminal-deoxynucleotidyl transferase mediated nick end labeling assay was used to assess cell apoptosis.The fluorescein sodium and transmission electron microscopy were used to detect the permeability and structure of BBB.The results showed that anesthesia and surgery significantly injured hippocampal-dependent learning and memory,which was ameliorated by atorvastatin.Atorvastatin could also reverse the surgery-induced increase of systemic and hippocampal cytokines,including IL-1β,TNF-α,and IL-6,accompanied by inhibiting the nuclear factor kappa-B(NF-κB)pathway and Nucleotide-Binding Oligomerization Domain,or Leucine Rich Repeat and Pyrin Domain Containing 3(NLRP3)inflammasome activation,as well as hippocampal neuronal apoptosis.In addition,surgery triggered an increase of BBB permeability,paralleled by a decrease of the ZO-1,occludin,and Claudin 5 proteins in the hippocampus.However,atorvastatin treatment could protect the BBB integrity from the impact of surgery,by up-regulating the expressions of ZO-1,occludin,and Claudin 5.These findings suggest that atorvastatin exhibits neuroprotective effects on cognition in aged mice undergoing surgery.Pengfei Liu Quansheng Gao Lei Guan Yanting Hu Jingwen Jiang Teng Gao Weixuan Sheng Xinying Xue Hui Qiao Tianzuo Li 2021Acta Biochimica et Biophysica Sinica2021,53,5:6
16Genetic dissection of rice appearance quality and cooked rice elongation by genome-wide association study显示文摘Appearance and cooked rice elongation are key quality traits of rice. Although some QTL for these traits have been identified, understanding of the genetic relationship between them remains limited. In the present study, large phenotypic variation was observed in 760 accessions from the 3 K Rice Genomes Project for both appearance quality and cooked rice elongation. Most component traits of appearance quality and cooked rice elongation showed significant pairwise correlations, but a low correlation was found between appearance quality and cooked rice elongation. A genome-wide association study identified 74 QTL distributed on all 12 chromosomes for grain length, grain width, length to width ratio, degree of endosperm with chalkiness, rice elongation difference, and elongation index. Thirteen regions containing QTL stably expressed in multiple environments and/or exerting pleiotropic effects on multiple traits were detected. By gene-based association analysis and haplotype analysis, 46 candidate genes, including five cloned genes, and 49 favorable alleles were identified for these 13 QTL. The effect of the candidate gene Wx on rice elongation difference was validated by a transgenic strategy. 19Complete disassociation of adult pancreas into viable single cells through cold trypsin-EDTA digestion显示文摘The in vitro isolation and analysis of pancreatic stem/progenitor cells are necessary for understanding their properties and function; however, the preparation of high-quality single-cell suspensions from adult pancreas is prerequisite. In this study, we applied a cold trypsin-ethylenediaminetetraacetic acid (EDTA) digestion method to disassociate adult mouse pancreata into single cells. The yield of single cells and the viability of the harvested cells were much higher than those obtained via the two commonly used warm digestion methods. Flow cytometric analysis showed that the ratio of ductal or BCRP1-positive cells in cell suspensions prepared through cold digestion was consistent with that found in vivo. Cell culture tests showed that pancreatic epithelial cells prepared by cold digestion maintained proliferative capacity comparable to those derived from warm collagenase digestion. 20Identification and Genetic Mapping of a Lesion Mimic Mutant in Rice显示文摘A lesion mimic stripe mutant,designated as lms1(lesion mimic stripe 1),was obtained from the M2 progeny of a 60Co γ-radiation treated japonica rice variety Jiahua 1.The lms1 mutant displayed propagation type lesions across the whole growth and developmental stages.Physiology and histochemistry analysis showed that the mutant exhibited a phenotype of white stripe when grown under high temperature(30 ℃),and the lesion mimic caused by programmed cell death under low temperature(20 ℃).The genetic analysis indicated that this lesion-mimic phenotype is controlled by a single locus recessive nuclear gene.Furthermore,by using simple sequence repeat markers and an F2 segregating population derived from two crosses of lms1 × 93-11 and lms1 × Pei'ai 64S,the lms1 gene was mapped between markers Indel1 and MM0112-4 with a physical distance of 400 kb on chromosome 6 in rice.MA Jian-yang CHEN Sun-lu ZHANG Jian-hui DONG Yan-jun TENG Sheng 2012Rice science2012,19,1:4
