| 1 | Response of the accumulation of proline in the bodies of Populus euphratica to the change of groundwater level at the lower reaches of Tarim River显示文摘The content of proline in the plant bodies is closely related to the converse-succession-resistant capability of the plants. In this paper, the relationship between the proline accumulation in the bodies of Populus euphratica and the change of groundwater level is analyzed by taking Popu-lus euphratica, the main community-building species of the desert riparian forests along the Tarim River, as the research object. The research results show that the accumulation of proline in the bodies of Populus euphratica is closely related to the change of groundwater level gradient under drought stress, it increases with the drawdown of groundwater level and the increase of moisture stress degree; the accumulation of proline in the bodies of Populus euphratica has two ex-tremely high points at the groundwater depth ranges of 3.64—5.14 m and 9.46—10.16 m. Combining the field investiga-tion and the analysis of the plots, it is considered that the groundwater level of 3.5—4.5 m is rational for the growth of Populus euphratica. | 2 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 3 | Antioxidant Defense Mechanisms of Salinity Tolerance in Rice Genotypes显示文摘In order to elucidate the role of antioxidant responses in salinity tolerance in rice genotypes under salt stress, experiments were conducted using four rice varieties, including salt-sensitive BRRI dhan 28 and three salt-tolerant varieties BRRI dhan 47, BINA dhan 8 and BINA dhan 10. Thirty-day-old rice seedlings were transplanted into pots. At the active tillering stage(35 d after transplanting), plants were exposed to different salinity levels(0, 20, 40 and 60 mmol/L NaCl). Salt stress caused a significant reduction in growth for all the rice genotypes. Growth reduction was higher in the salt-sensitive genotype than in the salt-tolerant ones, and BINA dhan 10 showed higher salt tolerance in all measured physiological parameters. The reduction in shoot and root biomass was found to be minimal in BINA dhan 10. Chlorophyll content significantly decreased under salt stress except for BINA dhan 10. Proline content significantly increased in salt-tolerant rice genotypes with increased salt concentration, and the highest proline content was obtained from BINA dhan 10 under salt stress. Catalase and ascorbate peroxidase activities significantly decreased in salt-sensitive genotype whereas significantly increased in salt-tolerant ones with increasing salt concentration. However, salt stress significantly decreased guaiacol peroxidase activity in all the rice genotypes irrespective of salt tolerance. K^+/Na^+ ratio also significantly decreased in shoots and roots of all the rice genotypes. | 4 | Physiological Responses of Contrasting Rice Genotypes to Salt Stress at Reproductive Stage显示文摘Salinity is a major abiotic stress affecting plant growth and productivity. Considerable genetic variation is present in rice in response to salt stress, with higher sensitivity during early seedling and reproductive stage. In this study, physiological changes in leaves and developing panicles of rice genotypes (IR686, Sadri, Rc222, CSR28, IR670 and Pokkali) contrasting in salt tolerance at the reproductive stage were evaluated in greenhouse experiment under salt stress. The results showed that IR670 and the tolerant-check Pokkali maintained lower Na^+/K^+ ratio, less reduction in chlorophyll concentration, lower malondialdehyde (MDA) production, higher concentrations of reduced ascorbate (reduced AsA), higher proline accumulation and lower percentage reduction in pollen viability than the salt-sensitive genotypes under salt stress. The higher concentration of reduced AsA suggests an efficient ROS-scavenging system. Physiological measurements and pollen viability analysis revealed that Sadri (moderately tolerant at the seedling stage) is sensitive to salt stress at the flowering stage. | 5 | Rice-duck co-culture benefits grain 2-acetyl-1-pyrroline accumulation and quality and yield enhancement of fragrant rice显示文摘Rice-duck co-culture is an integrated farming technology that benefits rice production, grain quality, and ecological sustainability in paddy fields. However, little is known about the effects of rice-duck co-culture on enzyme activity involved in the biosynthesis of 2-acetyl-1-pyrroline (2-AP), the volatile that gives fragrant rice its' distinctive and sought-after aroma. The present study aimed to examine the influence of rice-duck co-culture on the photosynthesis, yield, grain quality, rice aroma, and the enzymes involved in 2-acetyl-1-pyrroline biosynthesis in the cultivar Meixiangzhan 2 during the early and late rice growing seasons of 2016 in Guangzhou, China. We compared the rice grown in paddy fields with and without ducks. We found that rice-duck co-culture not only improved the yield and quality of fragrant rice grain, but also promoted the precursors of 2-AP biosynthesis formation and 2-AP accumulation in the grain. Grain 2-AP content in rice-duck co-culture was noticeably increased with 9.60% and 20.81% in early and late seasons, respectively. Proline and pyrroline-5-carboxylic acid (P5C) (precursors of 2-AP biosynthesis) and the activity of enzymes such as proline dehydrogenase (ProDH), ornithine aminotransferase (OAT) and Δ1 pyrroline-5-carboxylic acid synthetase (P5CS) were all improved by 10.15%–12.99%, 32.91%–47.75%, 17.81%–26.71%, 6.25%–21.78%, and 10.58%–38.87% under rice-duck co-culture in both seasons, respectively. | 6 | Gene Cloning and Expression of the Pyrroline-5-carboxylate Reductase Gene of Perennial Ryegrass(Lolium perenne)显示文摘Salt and drought limit the range of applications of perennial ryegrass(Lolium perenne L.), which is one of the important turf and forage grasses. Previous studies have suggested that pyrroline-5-carboxylate reductase(P5CR) might play a central role in proline accumulation in plants that are responsive to stresses. In the present study, the Lolium perenne L. pyrroline-5-carboxylate reductase(LpP 5CR) gene was cloned from leaves of the cultivar ‘Derby' using the RACE technique. The full-length LpP 5CR gene was 1 047 bp in length, which comprised an open reading frame(ORF) of 840 bp in size. Sequence alignment revealed that the putative Lp P5 CR had a 94.3% similarity to Ta P5 CR. q RT-PCR displayed that the mR NA levels of the LpP 5CR gene were almost the same as that in the roots, stems, and leaves of perennial ryegrass seedlings subjected to normal condition or NaC l treatment for 1 h. Moreover, the transcription level of LpP 5CR was up- or down- regulated with Na Cl, polyethylene glycol(PEG), cold, or abscisic acid(ABA) treatment for 3 to 48 h. In addition, confocal microscopy localized the GFP-Lp P5 CR fusion protein to the cytoplasm of onion epidermal cells. | 9 | Exogenous Application of Abscisic Acid (ABA)Enhances Chilling Tolerance in Seedlings ofNapier Grass (Pennisetum purpureum Schum.)显示文摘Napier grass, an important forage crop with potentials in multi-purpose applications, is widely grown throughout the tropics and subtropics. Low temperature severely limits its productivity and geographical distribution in temperate regions of the world. In this study, we investigated the effect of exogenous abscisic Acid(ABA) on chilling tolerance of napier grass(Pennisetum purpureum Schum.) seedlings. Seven-day-old napier grass seedlings were cultured in dd H_2O or ABA solution at different concentrations and exposed to 1 ℃ for different time durations. The chilling injury, membrane stability index(MSI) and proline content were estimated from leaf samples. The results showed that there was obvious morphological injury of leaf blighting and restrained growth for the seedlings under chilling stress, but this damage can be largely reduced(by 2/3) when the seedlings were treated by 100 μmol/L ABA in the culture solution, and that the application of exogenous ABA can help to maintain a good stability of leaf cell membrane as expressed by a high MSI value and a low level of proline in leaf cells. These results suggested that exogenous ABA can significantly alleviate chilling injury in napier grass seedlings by maintaining the stability of leaf cell membrane during chilling stress, and that the chilling tolerance was not ensured by a proline accumulation although a passive accumulation of proline was observed in the seedlings under chilling stress. Proline transporter(Pro T) plays an important role in proline distribution between plant organs. Using a candidate gene approach, we cloned a c DNA sequence for Pro T from common bean(Phaseolus vulgaris L.) and designated the gene Pv Pro T. The deduced amino acid sequence of Pv Pro T showed high similarity to Bet/Pro T proteins from other leguminous plants, and the highest similarity was observed with mothbean(Vigna aconitifolia L.) Vu Pro T.Relative quantification of the m RNA level of Pv Pro T using real-time PCR analysis showed that the Pv Pro T transcript level was higher in leaves than in stems and roots of common bean plants subjected to drought and salt stress. Under 20%(w/w) PEG-6000 treatment,drought-resistant plants expressed a higher level of Pv Pro T transcripts than droughtsensitive plants. | 16 | OsSCE1 Encoding SUMO E2-Conjugating Enzyme Involves in Drought Stress Response of Oryza sativa显示文摘Small ubiquitin-like modifier(SUMO)-conjugating enzymes are involved in post-translational regulatory processes in eukaryotes, including the conjugation of SUMO peptides to protein substrate(SUMOylation). SUMOylation plays an important role in improving plant tolerance to abiotic stress such as salt, drought, heat and cold. Herein, we reported the isolation of OsSCE1(LOC_Os10 g39120) gene encoding a SUMO-conjugating enzyme from rice(Oryza sativa cv. Nipponbare) and its functional validation in response to drought stress. The E2 enzyme, Os SCE1, is one of three key enzymes involved in the conjugation of SUMO to its target proteins. Activated SUMO is transferred to the cysteine of an E2 enzyme and then to the target lysine residue of the substrate, with or without the help of an E3 SUMO ligase. Expression of OsSCE1 was strongly induced by polyethylene glycol 6000(PEG6000) treatment, which suggested OsSCE1 may be involved in the drought stress response. Overexpression of OsSCE1(OsSCE1-OX) in Nipponbare reduced the tolerance to drought stress. Conversely, the drought tolerance was slightly improved by the knockdown of OsSCE1(OsSCE1-KD). These results were further supported by measurement of proline content in OsSCE1-OX and OsSCE1-KD transgenic lines under induced drought stress, which showed OsSCE1-KD transgenic lines accumulated higher proline content than the wild type, whereas OsSCE1-OX line had lower proline content than the wild type. | 18 | Expression of multi-domain type III antifreeze proteins from the Antarctic eelpout (Lycodichths dearborni) in transgenic tobacco plants improves cold resistance显示文摘Type III antifreeze proteins(AFPIIIs)are a group of small globular proteins found in some polar fishes to protect them against freezing damage.Transgenic expression of AFPs has been shown to confer cold tolerance to commercially important plants and animals.We have previously isolated multiple AFPIII genes in the Antarctic eelpout(Lycodichthys dearborni)that encode larger AFPIII isoforms with up to 12 of the conventional domains.Here we have introduced the fish AFPIII genes that encode for the monomer(ld1),dimer(ld2),trimer(ld3)and tetramer(ld4)AFPIII isoforms in tobacco plants.Pot-grown 4-week-old transgenic tobacco plants were exposed to cold stress at 4◦C for 30 days and the results show that ld1,ld2,ld3 and ld4 transgenic plants present relatively lower electrolyte leakage and lower content of malondialdehyde(MDA),but accumulated higher content of proline when compared to control plants.This indicates considerable improved membrane integrity under low temperature stress and improvement of the plant cold resistance.The plants transformed with the AFPIII tetramer-and trimer-domains demonstrated a higher cold-tolerant levels when compared with plants transformed with the dimer-and monomer AFPIII domains.Our study further supports that fish AFPIIIs,especially the multidomain proteins,protect cells from non-freezing hypothermic stresses,apart from there well-known function as ice inhibitors molecules at freezing temperature. | Qiao Huang Ruiqin Hu Hui zhu Changlian Peng Liangbiao Chen | 2021 | Aquaculture and Fisheries2021,6,2: | 2 |

| 20 | Drought-induced proline is mainly synthesized in leaves and transported to roots in watermelon under water deficit显示文摘Proline accumulation has been shown to occur in plants in response to various environmental stresses.Although proline metabolismrelated genes have been functionally characterized,the inter-organ transport of proline in stressed plants remains unclear.In this study,free proline was detected with significant accumulations in the roots,stems,and leaves of watermelon drought-tolerant germplasm M08 and drought-susceptible line Y34 under drought stress.Expression profiling and enzyme activity measurements revealed that ClP5CS1 gene,rather than its paralog ClP5CS2,mainly contributes to the proline synthesis in leaves via the Glu pathway.Additionally,over-expression of the ClP5CS genes significantly enhanced the drought tolerance of transgenic Arabidopsis lines.Furthermore,we confirmed that proline is mainly synthesized in leaves and transported to roots in watermelon under drought stress.Transcriptome and expression analyses revealed that the genes involved in proline metabolism exhibited different expression levels.Specifically,ClP5CS1 was upregulated in leaves and roots,while ClP5CS2 was downregulated under drought stress.Also,415 and 362 differently expressed TFs were identified in roots and leaves,respectively,with the majority upregulated in the former.Ultimately,a model for proline metabolism was proposed.The findings of this study provided new insights into the biosynthesis,transport,and regulatory mechanism of drought-induced proline in plants. | Zhongyuan Wang Yongchao Yang Vivek Yadav Wei Zhao Yaping He Xian Zhang Chunhua Wei | 2022 | Horticultural Plant Journal2022,8,5: | 2 |
