| 1 | Blumgart's technique of pancreaticojejunostomy: Analysis of safety and outcomes显示文摘Background: Blumgart's pancreaticojejunostomy(PJ) has been described with low pancreatic leak rates. This study aimed to evaluate our experience with this technique regarding the pancreatic leak and other perioperative outcomes. Methods: We performed a single-center retrospective analysis of a cohort of 81 patients who underwent pancreaticoduodenectomy in our department from January 2011 to February 2018. The primary endpoint was the occurrence of a clinically relevant postoperative pancreatic fistula(CR-POPF) and analysis of its risk factors. Results: The CR-POPF rate was 12.3%. Fistula risk score(FRS) was the only significant risk factor for the occurrence of overall POPF in multivariate analysis. However, none of the other factors including FRS was found to be significantly associated with CR-POPF risk. A strong positive correlation was found between the CR-POPF and the incidence of delayed gastric emptying, post-pancreatectomy hemorrhage and increased length of hospital stay. Conclusion: Blumgart's technique is a safe technique of pancreatico-enteric anastomosis with low rates of CR-POPF. CR-POPF with this technique is independent of most of the preoperative and intraoperative factors. | 2 | Lipid-polymer hybrid nanoparticles: Development & statistical optimization of norfloxacin for topical drug delivery system显示文摘Poly lactic acid is a biodegradable, biocompatible, and non-toxic polymer, widely used in many pharmaceuticalpreparations such as controlled release formulations, parenteral preparations, surgicaltreatment applications, and tissue engineering. In this study, we prepared lipid-polymer hybrid nanoparticlesfor topical and site targeting delivery of Norfloxacin by emulsification solvent evaporationmethod (ESE). The design of experiment (DOE) was done by using software to optimize the result, andthen a surface plot was generated to compare with the practical results. The surface morphology, particlesize, zeta potential and composition of the lipid-polymer hybrid nanoparticles were characterized bySEM, TEM, AFM, and FTIR. The thermal behavior of the lipid-polymer hybrid nanoparticles was characterizedby DSC and TGA. The prepared lipid-polymer hybrid nanoparticles of Norfloxacin exhibited anaverage particle size from 178.6 ± 3.7 nm to 220.8 ± 2.3 nm, and showed very narrow distribution withpolydispersity index ranging from 0.206 ± 0.36 to 0.383 ± 0.66. The surface charge on the lipid-polymerhybrid nanoparticles were confirmed by zeta potential, showed the value from t23.4 ± 1.5 mVto t41.5 ± 3.4 mV. An Antimicrobial study was done against Staphylococcus aureus and Pseudomonasaeruginosa, and the lipid-polymer hybrid nanoparticles showed potential activity against these two.Lipid-polymer hybrid nanoparticles of Norfloxacin showed the %cumulative drug release of 89.72% in24 h. A stability study of the optimized formulation showed the suitable condition for the storage oflipid-polymer hybrid nanoparticles was at 4 ± 2 C/60 ± 5% RH. | 3 | Drought-induced proline is mainly synthesized in leaves and transported to roots in watermelon under water deficit显示文摘Proline accumulation has been shown to occur in plants in response to various environmental stresses.Although proline metabolismrelated genes have been functionally characterized,the inter-organ transport of proline in stressed plants remains unclear.In this study,free proline was detected with significant accumulations in the roots,stems,and leaves of watermelon drought-tolerant germplasm M08 and drought-susceptible line Y34 under drought stress.Expression profiling and enzyme activity measurements revealed that ClP5CS1 gene,rather than its paralog ClP5CS2,mainly contributes to the proline synthesis in leaves via the Glu pathway.Additionally,over-expression of the ClP5CS genes significantly enhanced the drought tolerance of transgenic Arabidopsis lines.Furthermore,we confirmed that proline is mainly synthesized in leaves and transported to roots in watermelon under drought stress.Transcriptome and expression analyses revealed that the genes involved in proline metabolism exhibited different expression levels.Specifically,ClP5CS1 was upregulated in leaves and roots,while ClP5CS2 was downregulated under drought stress.Also,415 and 362 differently expressed TFs were identified in roots and leaves,respectively,with the majority upregulated in the former.Ultimately,a model for proline metabolism was proposed.The findings of this study provided new insights into the biosynthesis,transport,and regulatory mechanism of drought-induced proline in plants. | Zhongyuan Wang Yongchao Yang Vivek Yadav Wei Zhao Yaping He Xian Zhang Chunhua Wei | 2022 | Horticultural Plant Journal2022,8,5: | 2 |

| 11 | Outcome of application of primary versus secondary Illizarov's fixator in open tibial shaft fractures显示文摘BACKGROUND: The present study aimed to compare outcome of primary and secondary Illizarov's fi xator application as a treatment method for type III open tibial fractures in terms of nonunion and wound infection.METHODS: This prospective study was done in a tertiary care center. Forty-eight type III tibial fractures were treated with Illizarov's apparatus between 2008 and 2011. The patients were divided into two groups depending on the treatment protocol, timing of wound closure and Illizarov's application, primary(n=28) and secondary(n=20).RESULTS: In the primary group, healing was achieved in all 28 patients. The median time to recovery was 24 weeks, and the median number of operations was 3. There were 6 patients with a bone defect. In the secondary group, complete recovery was achieved in 18 out of 20 patients. The median time to recovery was 30 weeks, and the median number of operations 5. There were 9 patients with a bone defect. The median time to recovery and the number of operations were signifi cantly smaller in patients undergoing primary operation. Union was 100% in the primary group and more than 95% in the secondary group. | 12 | Design and evaluation of aceclofenac ocular inserts with special reference to cataracts and conjunctivitis显示文摘The eyes present unique opportunities and challenges when it comes to the delivery of pharmaceuticals.While absorption by this route is bungling,there are a few side effects with conventional dosage forms.Ocular inserts were prepared with prolonged release of drug and minimum swelling within cul-de-sac using aceclofenac.The work focused on treatment of conjunctivitis and cataracts by formulating ocular inserts of different polymeric combination of aceclofenac using hydroxypropyl methyl cellulose(HPMC,3% to 5%),chitosan(3% to 5%),poly vinyle alcohol(PVA,3% to 5%),methyl cellulose(MC,3% to 5%) as drug reservoir and ethyl cellulose(EC) polymer as rate controlling membrane by solvent casting technique with the objective of increasing contact time,achieving controlled release and greater therapeutic efficiency.The prepared ocular insert were then evaluated for physical appearances tensile strength,elongation at break(%),weight variation,uniformity of thickness,moisture absorption(%),pH,folding endurance,Fourier Transform Infrared spectroscopy,differential scanning calorimetry.Physicochemical characterization and in vitro transcorneal permeation studies reveals that,the prepared ocular insert formulations F2 and F8 containing HPMC and PVA had released their drug content,98.54% and 96.24%,respectively,over an extended period of 24 h.Hence these formulations were selected as best optimized formulations.It can be concluded that hydroxy propyl methyl cellulose is a good film forming hydrophilic polymer which shows potential agent for ocular drug delivery system.Incorporation of polyethylene glycol enhances the permeability of aceclofenac ocular insert and has perfect zero order release,proving a promising controlled release delivery system. | Vivek Dave Sachdev Yadav Sarvesh Paliwal Swapnil Sharma Dhirendra Kumar | 2013 | Journal of Chinese Pharmaceutical Sciences2013,22,5: | 1 |
| 13 | Determination of mycophenolic acid in human plasma by ultra performance liquid chromatography tandem mass spectrometry显示文摘A simple, sensitive and high throughput ultra performance liquid chromatography tandem mass spectrometry method has been developed for the determination of mycophenolic acid in human plasma. The method involved simple protein precipitation of MPA along with its deuterated analog as an internal standard(IS) from 50 mL of human plasma. The chromatographic analysis was done on Acquity UPLC C18(100 mm 2.1 mm, 1.7 mm) column under isocratic conditions using acetonitrile and 10 mM ammonium formate, pH 3.00(75:25, v/v) as the mobile phase. A triple quadrupole mass spectrometer operating in the positive ionization mode was used for quantitation. In-source conversion of mycophenolic glucuronide metabolite to the parent drug was selectively controlled by suitable optimization of cone voltage, cone gas flow and desolvation temperature. The method was validated over a wide concentration range of 15–15000 ng/mL. The mean extraction recovery for the analyte and IS was 495%. Matrix effect expressed as matrix factors ranged from 0.97 to 1.02. | 16 | Ectopic expression of VvFUS3,B3-domain transcription factor,in tomato influences seed development via affecting endoreduplication and hormones显示文摘FUSCA3(FUS3)is a member of B3-domain transcription factor family and master regulator of seed development.It has potential roles in hormone biosynthesis and signaling pathways and therefore plays diverse roles in plant life cycle,especially in seed germination,dormancy,embryo formation,seed and fruit development,and maturation.However,there is limited information about its functions in seed and fruit development of grapevine.In this study,we expressed VvFUS3 in tomato for its functional characterization.Overexpression of VvFUS3 in tomato led to a reduction in seed number and seed weight without affecting the fruit size.Histological analysis found that both cell expansion and cell division in transgenic seed and fruit pericarp have been affected.However,there were no obvious differences in pollen size,shape,and viability,suggesting that VvFUS3 affects seed development but not the pollen grains.Moreover,the expression of several genes with presumed roles in seed development and hormone signaling pathways was also influenced by VvFUS3.These results suggest that VvFUS3 is involved in hormonal signaling pathways that regulate seed number and size.In conclusion,our study provides novel preliminary information about the pivotal roles of VvFUS3 in seed and fruit development and these findings can potentially serve as a reference for molecular breeding of seedless grapes. | Bilal Ahmad Songlin Zhang Jin Yao Shengyue Chai Vivek Yadav Habib-ur-Rehman Athar Mati Ur Rahman Li Wang Xiping Wang | 2022 | Horticultural Plant Journal2022,8,3: | 0 |
