| 1 | Genetic diversity and construction of core collection in Chinese wheat genetic resources显示文摘Genetic diversity among 5029 accessions representing a proposed Chinese wheat core collection was analyzed using 78 pairs of fluorescent microsatellite (SSR) primers mapped to 21 chromosomes. A stepwise hierarchical sampling strategy with priority based on 4×105 SSR data-points was used to construct a core collection from the 23090 initial collections. The core collection consisted of 1160 accessions, including 762 landraces, 348 modern varieties and 50 introduced varieties. The core ac-counts for 23.1% of the 5029 candidate core accessions and 5% of the 23090 initial collections, but retains 94.9% of alleles from the candidate collections and captures 91.5% of the genetic variation in the initial collections. These data indicate that it is possible to maintain genetic diversity in a core collection while retaining fewer accessions than the accepted standard, i.e., 10% of the initial collections captured more than 70% of their genetic diversity. Estimated genetic representation of the core con-structed by preferred sampling (91.5%) is much higher than that by random sampling (79.8%). Both mean genetic richness and genetic diversity indices of the landraces were higher than those of the modern varieties in the core. Structure and principal coordinate analysis revealed that the landraces and the modern varieties were two relatively independent subpopulations. Strong genetic differentiation associated with ecological environments has occurred in the landraces, but was relatively weak in the modern cultivars. In addition, a mini-core collection was constructed, which consisted of 231 accessions with an estimated 70% representation of the genetic variation from the initial collections. | 2 | Genetic diversity in Chinese modern wheat varieties revealed by microsatellite markers显示文摘Genetic diversity of 1680 modern varieties in Chinese candidate core collections was analyzed at 78 SSR loci by fluorescence detection system. A total of 1336 alleles were detected, of which 1253 alleles could be annotated into 71 loci. For these 71 loci, the alleles ranged from 4 to 44 with an average of 17.6, and the PIC values changed from 0.19 to 0.89 with an average of 0.69. (1) In the three genomes of wheat, the average genetic richness was B>A>D, and the genetic diversity indexes were B>D>A. (2) Among the seven homoeologous groups, the average genetic richness was 2=7>3>4>6>5>1, and the genetic diversity indexes were 7>3>2>4>6>5>1. As a whole, group 7 possessed the highest genetic diversity, while groups 1 and 5 were the lowest. (3) In the 21 wheat chromosomes, 7A, 3B and 2D possessed much higher genetic diversity, while 2A, 1B, 4D, 5D and 1D were the lowest. (4) The highest average genetic diversity index existed in varieties bred in the 1950s, and then it declined continually. However, the change tendency of genetic diversity among decades was not greatly sharp. This was further illustrated by changes of the average genetic distance between varieties. In the 1950s it was the largest (0.731). Since the 1960s, it has decreased gradually (0.711, 0.706, 0.696, 0.695). The genetic base of modern varieties is becoming narrower and narrower. | 3 | Fast integration and accumulation of beneficial breeding alleles through an AB–NAMIC strategy in wheat显示文摘Wheat(Triticum aestivum)is among the most important staple crops for safeguarding the food security of the growing world population.To bridge the gap between genebank diversity and breeding programs,we developed an advanced backcross-nested association mapping plus inter-crossed population(AB-NAMIC)by crossing three popular wheat cultivars as recurrent founders to 20 germplasm lines from a mini core collection.Selective backcrossing combined with selection against undesirable traits and extensive crossing within and between sub-populations created new opportunities to detect unknown genes and increase the frequency of beneficial alleles in the AB-NAMIC population.We performed phenotyping of 590 AB-NAMIC lines and a natural panel of 476 cultivars for six consecutive growing seasons and genotyped these 1066 lines with a 660K SNP array.Genome-wide association studies of both panels for plant development and yield traits demonstrated improved power to detect rare alleles and loci with medium genetic effects in AB-NAMIC.Notably,genome-wide association studies in AB-NAMIC detected the candidate gene TaSWEET6-7B(TraesCS7B03G1216700),which has high homology to the rice SWEET6b gene and exerts strong effects on adaptation and yield traits.The commercial release of two derived AB-NAMIC lines attests to its direct applicability in wheat improvement.Valuable information on genome-wide association studymapping,candidate genes,and their haplotypes for breeding traits are available through WheatGAB.Our research provides an excellent framework for fast-tracking exploration and accumulation of beneficial alleles stored in genebanks. | Chengzhi Jiao Chenyang Hao Tian Li Abhishek Bohra Lanfen Wang Jian Hou Hongxia Liu Hong Liu Jing Zhao Yamei Wang Yunchuan Liu Zhiwei Wang Xin Jing Xiue Wang Rajeev K.Varshney Junjie Fu Xueyong Zhang | 2023 | Plant Communications2023,4,3: | 3 |

In recent years,considerable progress has been made in genetic studies of SCZ. A number of common variants with small effects and rare variants with relatively larger effects have been identifi ed. These variants include risk loci identifi ed by genome-wide association studies,rare copy-number variants identifi ed by comparative genomic analyses,and de novo mutations identified by high-throughput DNA sequencing. Collectively,they contribute to the heterogeneity of the disease. | 6 | Molecular cloning and characterization of a gene encoding the proline transporter protein in common bean(Phaseolus vulgaris L.)显示文摘As a typical compatible solute, proline is accumulated in plants under environmental stresses. Proline transporter(Pro T) plays an important role in proline distribution between plant organs. Using a candidate gene approach, we cloned a c DNA sequence for Pro T from common bean(Phaseolus vulgaris L.) and designated the gene Pv Pro T. The deduced amino acid sequence of Pv Pro T showed high similarity to Bet/Pro T proteins from other leguminous plants, and the highest similarity was observed with mothbean(Vigna aconitifolia L.) Vu Pro T.Relative quantification of the m RNA level of Pv Pro T using real-time PCR analysis showed that the Pv Pro T transcript level was higher in leaves than in stems and roots of common bean plants subjected to drought and salt stress. Under 20%(w/w) PEG-6000 treatment,drought-resistant plants expressed a higher level of Pv Pro T transcripts than droughtsensitive plants. Although heterologous expression of Pv Pro T in the Escherichia coli mutant mkh13 showed that Pv Pro T exhibited uptake activities for proline and betaine, no betaine content was detected in the common bean. | 13 | QTL and candidate genes associated with common bacterial blight resistance in the common bean cultivar Longyundou 5 from China显示文摘Common bacterial blight(CBB), caused by Xanthomonas axonopodis pv. phaseoli and Xanthomonas fuscans subsp. fuscans(Xff), is a worldwide disease of common bean(Phaseolus vulgaris L.).Longyundou 5, a Chinese cultivar in the Mesoamerican gene pool of common bean, displays resistance to the Xff strain XSC3-1. To identify the genetic mechanisms behind this resistance,we crossed Long 5 with a susceptible genotype to develop a mapping population of F2 plants.Plant resistance to CBB was identified at 14 and 21 days after inoculation with Xff strain XSC3-1.A major QTL at 14 and 21 days after inoculation was mapped on chromosome Pv10 with LOD scores of 6.41 and 5.35, respectively. This locus was associated with SAP6, a previouslyidentified and much-used dominant marker, but in a 4.2 cM interval between new codominant markers BMp10s174 and BMp10s244. Ten candidate genes were found between markers BMp10s174 and BMp10s244 on chromosome Pv10 and could encode defense response proteins responding to CBB pathogens. Four pairs each of epistatic QTL for CBB resistance were detected at 14 and 21 days after inoculation. Phenotypic variation explained by the epistatic QTL ranged from 7.19% to 12.15% and 7.72% to 8.80% at 14 and 21 days after inoculation, respectively. These results confirmed the importance of epistasis in CBB resistance in common bean. | 16 | Mendelian randomization study to investigate the causal relationship between plasma homocysteine and chronic obstructive pulmonary disease显示文摘BACKGROUND:Several observational studies have shown an association between homocysteine(Hcy)levels and chronic obstructive pulmonary disease(COPD),but causal relationships are not clear.Our study aimed to explore the causal relationship between plasma Hcy and COPD by two-sample Mendelian randomization(MR).METHODS:A two-sample MR study was performed to infer the causal link.Genetically predicted plasma Hcy was selected as an instrumental variable(Ⅳ)from published genome-wide association study(GWAS)meta-analyses.COPD with different etiologies was extracted as outcome variables from other GWAS meta-analyses.The main MR analysis was performed using the inversevariance weighted(IVW)method.Additional analyses were further performed using Cochran’s Q-test and MR-Egger regression to evaluate the heterogeneity or horizontal pleiotropy of our findings.RESULTS:MR analysis showed no significant association between plasma Hcy and COPD.The results of the groups were consistent with the sensitivity analysis and repeated analysis,without heterogeneity or horizontal pleiotropy.The IVW results showed COPD hospital admissions(odds ratio[OR]1.06,95%confidence interval[CI]0.91-1.24,P=0.42),asthma/COPD(OR 0.97,95%CI0.89-1.06,P=0.55),COPD-related chronic infection(OR 1.50,95%CI 0.57-3.99,P=0.41),COPDI asthma/interstitial lung disease(ILD)-related pneumonia or pneumonia-derived septicemia(OR 0.93,95%CI 0.86-1.02,P=0.13),and COPD-related respiratory insufficiency(OR 1.00,95%CI 0.7-1.44,P=0.99).CONCLUSION:There is no direct causal relationship between plasma Hcy and COPD in our study.As Hcy is known to have deleterious effects on endothelial function and vascular homeostasis,further studies are needed to investigate whether additional factors mediate the association between Hcy and COPD. | Yanlan Hu Ping Tan Juntao Wang Jun Zeng Quan Li Shijiao Yan Wenjie Hao Lanfen He Xingyue Song Caihong Zhang Chuanzhu Lyu | 2023 | World Journal of Emergency Medicine2023,14,5: | 0 |
| 17 | Efficient triplet pair separation from intramolecular singlet fission in dibenzopentalene derivatives显示文摘In intramolecular SF(iSF), the strong coupling nature and confinement of diffusional separation of ^1(TT) limits the extraction and harnessing of triplet energy. In order to investigate the possible ^1(TT) separation and the role of molecular parameters on it, a series of iSF-capable dibenzopentalene derivatives(DBPs) have been synthesized and their photoinduced dynamics are monitored. iSF takes place in DBPs, accompanied by consecutive ^1(TT) separation in polycrystalline film with almost ^100% yield. It is suggested the strong intermolecular coupling provided by the closely packing configuration in the film facilitates the disentanglement of correlated ^1(TT). | 18 | Genetic dissection of yield-related traits in response to drought stress in common bean显示文摘Drought stress severely impairs common bean production.For facilitating drought-resistance breeding in common bean,molecular markers were identified in a genome-wide level marker–trait association study.A panel of 210 common bean accessions showed large variation in 11 agronomic traits at the adult stage(plant height,pod number per plant,seed number per pod,seed number per plant,seed yield per plant,pod length,harvest index,pod harvest index,days to maturity,hundred-seed weight,and seed yield)under two water conditions.The coefficient of variation ranged from 6.21% for pod harvest index to51.00% for seed number per plant under well-watered conditions,and from 4.05% for days to maturity to 40.72% for seed number per plant under drought stress.In a genome-wide association study,119quantitative-trait loci were associated with drought resistance,including 41 adjacent to known loci.Among these loci,12 were found to be associated with at least two traits.Three major loci were identified at Pv01 and Pv02.A set of candidate genes were found that encode MYBs,AREBs,WKRYs,and protein kinases.These results reveal promising alleles that control drought resistance,shedding light on the genetic basis of drought resistance and accelerating future efforts for drought resistance improvement in common bean. | Lei Wu Yujie Chang Lanfen Wang Liang Ji Lin Peng Shumin Wang Jing Wu | 2023 | The Crop Journal2023,11,4: | 0 |
