| 1 | Microcalorimetric investigation of the effect of berberine alkaloids from Coptis chinensis Franch on Staphylococcus aureus growth显示文摘The inhibitory effects of three berberine alkaloids (BAs) from rhizome of Coptis chinensis Franch, a traditional Chinese medicinal (TCM) herb, on Staphylococcus aureus growth were investigated by mi- crocalorimetry. The power–time curves of S. aureus with and without BAs were acquired; meanwhile the extent and duration of inhibitory effects on the metabolism were evaluated by studying the growth rate constant (k), half inhibitory ratio (IC50), maximum heat-output power (Pmax), peak time of maximum heat-output power (tp) and total heat production (Qt). The value of k of S. aureus in the presence of the three BAs decreased with the increasing concentrations of BAs. Moreover, Pmax was reduced and the value of tp increased with increasing concentrations of the three drugs. The inhibitory activity varied with different drugs. The values of IC50 of the three BAs are respectively, 101.4 μg/mL for berberine, 241.0 μg/mL for palmatine and 792.3 μg/mL for jateorrhizine. The sequence of antimicrobial activity of the three BAs is: berberine > palmatine > jateorrhizine. | 3 | Synergistic activity of lawsone methyl ether in combination with some antibiotics and artocarpin against methicillin-resistant Staphylococcus aureus, Candida albicans, and Trychophyton rubrum显示文摘Objective: One appealing strategy to overcome and prevent resistant problem is the use of combined two or more antibacterial substances. Lawsone methyl ether(LME) is the naphthoquinone found in the leaves of Impatiens balsamina. The objective of this study is to determine the interaction of LME with some antibiotics(ampicillin, tetracycline, norfloxacin, and clotrimazole) and a natural compound, artocarpin against methicillin-resistant Staphylococcus aureus(MRSA), Candida albicans, and Trychophyton rubrum.Methods: A broth microdilution method was used to determine the minimum inhibition concentration(MIC). Synergistic effects were evaluated at their own MIC using the checkerboard method and time-kill assay.Results: LME showed moderate antibacterial activity against MRSA with MIC value of 15.6 μg/mL, and exhibited strong antifungal activities against T. rubrum and C. albicans with MIC values of 7.8 and 3.9 μg/m L,respectively. The interaction of LME with the natural compound artocarpin against MRSA produced a synergy with fractional inhibitory concentration index(FICI) value of 0.31, while the combination of LME and clotrimazole exhibited synergy against C. albicans and T. rubrum with FICI values of 0.38 and 0.24, respectively. The time-kill assays confirmed that the compounds in combination enhanced their antimicrobial activities against the resistant microorganisms with different degrees.Conclusion: LME in combination with clotrimazole exhibited synergy effect against C. albicans and T.rubrum. | 5 | Prevalence and antibiogram study of Salmonella and Staphylococcus aureus in poultry meat显示文摘Objective:To evaluate the presence and antibiogram pattern of Salmonella and Staphylococcus aureus(S.aureus)in retail poultry meat products.Methods:Foodborne pathogens(Salmonella and S.aureus)were isolated from poultry meat and confirmed with the help of biochemical and immunological test.Antibiogram of the isolates were examined by following CLS1 methods.Results:A total number of 209 poultry meat samples were collected and studied in this study.Out of which,5.26%were found contaminated with Salmonella while 18.18%were found contaminated with S.aureus.All the Salmonella and S.aureus isolates were found resistant to at least one antibiotic.About 72.72%of the Salmonella isolates showed resistance to tetracycline,while S.aureus isolates were also found highly resistant to tetracycline equal to 44.73%.One of the Salmonella isolates showed multi-drug resistance to almost six antibiotics out of nine antibiotics used in the study.Multidrug resistant S.aureus isolates were also found in the study.Conclusions:The study confirmed the presence of Salmonella and S.aureus in retail poultry meat.It is a potential threat to consumer health.To reduce the risk of contamination,good hygiene practices are necessary from processing to storage. | Ali Akbar Anil Kumar Anal | 2013 | Asian Pacific Journal of Tropical Biomedicine2013,3,2: | 8 |
| 6 | Antibacterial activity of eight Iranian plant extracts against methicillin and cefixime restistant Staphylococcous aureus strains显示文摘Objective:To assess the antibacterial activity of eight Iranian plant extracts against Staphylococcus aureus(S.aureus) strains which were isolated from infected patients.Methods: The studied strains were isolated from urine,stool,blood and wound of infected patients and identified by biochemical tests.In further,the antibacterial activity of 8 ethanolic local plant extracts including Querqus brantii(Q.brantii),Ziziphus spina-christi(Z.spina-christi),Peganum harmala(P.harmala),Oliveira decumbens(O.decumbens),Galium tricornutum(G.tricornutum), Vitex pseudo negundo(Vi.pseudo negundo),Salvia officinalis(S.officinalis),Vaccaria pyramidata (V.pyramidata) were then evaluated using agar disc diffusion method.Results:A total of 9 isolates were isolated and identified as S.aureus.Antibacterial profile of the strains showed that the strains were resistant to methicillin and cefixime.The highest antibacterial activity against the studied strains were belong to Q.brantii,P.harmala,Z.spina-christi and O.decumbens vent extracts with 11-40 mm,15-28 mm,8-26 mm and 10-20 mm of diameters,respectively. Intermediate antibacterial activity was exhibited by G.tricornutum and Vi.pseudo negundo against some of the studied strains with 7-20 mm and 7-13 mm of diameters,respectively. | 8 | Costunolide alleviates HKSA-induced acute lung injury via inhibition of macrophage activation显示文摘Staphylococcus aureus (S. aureus) infection leads to a severe inflammatory response and causes acute lung injury (ALI), eventually threatening human life. Therefore, it is of importance to find an agent to inhibit infiammation and reduce ALI. Here, we found that costunolide, a sesquiterpene lactone, displays anti-infiammatory effects and ameliorates heat-killed S. aureus (HKSA)-induced pneumonia. Costunolide treatment attenuated HKSA-induced murine ALI in which pulmonary neutrophil infltration was inhibited, lung edema was decreased, and the production of pro-inflammatory cytokines was significantly reduced. In addition, costunolide dose-dependently inhibited the generation of IL-6, TNF-α, IL-1β, and keratinocyte-derived cytokine (KC), as well as the expression of iNOS, in HKSA-induced macrophages. Furthermore, costunolide attenuated the phosphorylation of p38 MAPK and cAMP response element-binding protein (CREB). | 14 | Microbiological assessment of indoor air of a teaching hospital in Nigeria显示文摘Objective:To investigate the quality of indoor air of different wards and units of Olabisi Onabanjo University Teaching Hospital, Sagamu, to ascertain their contribution to infection rate in the hospital.Methods:The microbial quality of indoor air of nine wards/units of Olabisi Onabanjo University Teaching Hospital, Sagamu, Nigeria was conducted. Sedimentation technique using open Petri-dishes containing different culture media was employed and samplings were done twice daily, one in the morning shortly after cleaning and before influx of people/patients into the wards/units and the other in the evening when a lot of activities would have taken place in these wards. Isolates were identified according to standard methods.Results:Results showed that there was a statistically significant difference(氈2= 6.0167) in the bacteria population of the different sampling time whereas it was not so for fungi population(氈2= 0.2857). Male medical ward(MMW) and male surgical general(MSG) recorded the highest bacterial and fungal growth while the operating theatre(OT) was almost free of microbial burden. The bacteria isolates were Staphylococcus aureus, Klebsiellasp., Bacillus cereus, Bacillus subtilis, Streptococcus pyogenes andSerratia marscenceswhile the fungi isolates includedAspergillus flavus, Penicilliumsp.,Fusariumsp.,Candida albicansandAlternariasp.Staphylococcus aureuswas the predominantly isolated bacterium whilePenicilliumsp. was the most isolated fungus.Conclusions:Though most of the microbial isolates were potential and or opportunistic pathogens, there was no correlation between the isolates in this study and the surveillance report of nosocomial infection during the period of study, hence the contribution of the indoor air cannot be established. Methods:Thirty post operative pathogenic isolated S.aureus strains were used in this study. Bacterial culture was done in Mueller-Hinton broth at 37 ℃.Characters of these strains were determined by traditional biochemical tests such as hydrolysis test of gelatin,urea,galactose, starch and protein,and fermentation of lactose and sucrose.Antibiotic susceptibility were carried out by minimum inhibilory concentration test,minium bactericidal concentration test,disc agar diffusion test and brain heart infusion oxacillin screening agar.Results:Prom this study,it was observed that 100%S.aureus isolates showed positive results in gelatin,urea and galactose hydrolysis test.50%isolates were positive in starch hydrolysis test,35%in protein hydrolysis test. 100%isolates in lactose fermenting test,but no isolate was positive in sucrose fermenting test. | 20 | Mastitis Pathogens Isolated from Samples of Milk in Dairy Cows and Their Resistance against Antimicrobial Agents显示文摘The objective of this study was to evaluate the effectiveness of different antibiotics against mastitis causing microorganisms during first phase of lactation in herd of 320 Holstein cows in east of Slovakia. Milk samples from quarters were cultured and identified bacteria were subjected to antimicrobial susceptibility test by disc diffusion method to a large number of antibiotics. The results revealed higher sensitivity against tetradelta (100% of Strep. Agalactiae and Strep. uberis, Coagulase Negative Staphylococci), (94.4% of Stap. aureus) with highest number of bacterial isolates, followed by cefalexin + kanamycin amoxicillin plus clavulanat acid and ceftiotur (100% of Strep. agalactiae and Strep. uberis).
