3Microcalorimetric investigation of the effect of berberine alkaloids from Coptis chinensis Franch on Staphylococcus aureus growth显示文摘The inhibitory effects of three berberine alkaloids (BAs) from rhizome of Coptis chinensis Franch, a traditional Chinese medicinal (TCM) herb, on Staphylococcus aureus growth were investigated by mi- crocalorimetry. The power–time curves of S. aureus with and without BAs were acquired; meanwhile the extent and duration of inhibitory effects on the metabolism were evaluated by studying the growth rate constant (k), half inhibitory ratio (IC50), maximum heat-output power (Pmax), peak time of maximum heat-output power (tp) and total heat production (Qt). The value of k of S. aureus in the presence of the three BAs decreased with the increasing concentrations of BAs. Moreover, Pmax was reduced and the value of tp increased with increasing concentrations of the three drugs. The inhibitory activity varied with different drugs. The values of IC50 of the three BAs are respectively, 101.4 μg/mL for berberine, 241.0 μg/mL for palmatine and 792.3 μg/mL for jateorrhizine. The sequence of antimicrobial activity of the three BAs is: berberine > palmatine > jateorrhizine. 4Structure and function of interleukin-17 family cytokines显示文摘The recently identified interleukin-17(IL-17)cytokines family,which comprises six members in mammals(IL-17A–F),plays essential roles in the host immunity against infectious diseases and chronic inflammatory diseases.The three-dimensional structures containing IL-17A or IL-17F have become available and revealed the unique structural features of IL-17s as well as their receptors.Molecular modeling in this review shows that IL-17s may adopt a“cysteine knot”fold commonly seen in nerve growth factor(NGF)and other neurotrophins.Further modeling analysis unmasks a signature interaction feature of the IL-17F/IL-17RA complex,where a small loop of IL-17RA slots into the deep groove of the interface of IL-17F homodimer.This is quite different from the interaction between the best known four-helix cytokines and their cognate receptors.On the other hand,structure of IL-17A and its monoclonal antibody(CAT-2200)shows that,albeit that the antigenic epitope of IL-17A resides outside of the IL-17A homodimer interface,its physical proximity to the receptor binding groove may explain that antibody blockage would be achieved by interfering with the ligand-receptor interaction.This review is to summarize the advance in understanding the structure and function of IL-17 family cytokines,focusing mainly on IL-17A,IL-17F and IL-17E,in the hope of gaining better knowledge of immunotherapeutic strategies against various inflammatory diseases.Xiaoping Zhang Pornpimon Angkasekwinai Chen Dong Hong Tang 2011Protein & Cell2011,2,1:17
5Investigation of the Ce-Mn conversion coating on 6063 aluminium alloy显示文摘In this paper,the sustainable Ce-Mn chemical conversion coating was fabricated on 6063 aluminium alloy by means of Ce(NO3)3 and KMnO4 as the inhibitors and NaF as the accelerator.The morphologies,composition and valence state of the coating were analyzed by scanning electron microscopy(SEM),energy dispersive spectrometry(EDS) and X-ray photoelectron spectroscopy(XPS),respectively.The results indicated that the Ce-Mn conversion coating was formed.The anticorrosion of the coating was evaluated in 3.5 wt.% NaCl aqueous solution at room temperature by using polarization curve and electrochemical impedance spectroscopy(EIS).It indicated that the treated surface presented better anticorrosion behavior in chloride media than on the original material surface.The corrosion resistance of Ce-Mn conversion coating was about equal to the trivalent chromium conversion coating.ZHANG JunJun LI WenFang DU Jun HAN Dong ZHENG XiaoPing 2010Chinese Science Bulletin2010,55,29:9
6Clinical and genetic findings in a Chinese family with VDR-associated hereditary vitamin D-resistant rickets显示文摘Hereditary vitamin D-resistant rickets(HVDRR) is a rare autosomal recessive disorder characterized by severe rickets,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.This disorder is caused by homogeneous or heterogeneous mutations affecting the function of the vitamin D receptor(VDR),which lead to complete or partial target organ resistance to the action of 1,25-dihydroxy vitamin D.A non-consanguineous family of Chinese Han origin with one affected individual demonstrating HVDRR was recruited,with the proband evaluated clinically,biochemically and radiographically.To identify the presence of mutations in the VDR gene,all the exons and exon–intron junctions of the VDR gene from all family members were amplified using PCR and sequenced.The proband showed rickets,progressive alopecia,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.She also suffered from epilepsy,which is rarely seen in patients with HVDRR.Direct sequencing analysis revealed a homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene of the proband,which is located in the first zinc finger of the DNA-binding domain.Both parents had a normal phenotype and were found to be heterozygous for this mutation.We report a Chinese Han family with one individual affected with HVDRR.A homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene was found to be responsible for the patient's syndrome.In contrast to the results of treatment of HVDRR in other patients,our patient responded well to a supplement of oral calcium and a low dose of calcitriol.Qianqian Pang Xuan Qi Yan Jiang Ou Wang Mei Li Xiaoping Xing Jin Dong Weibo Xia 2016Bone Research2016,4,1:9

8The efficacy and safety of nimodipine in acute ischemic stroke patients with mild cognitive impairment: a double-blind, randomized,placebo-controlled trial显示文摘Nimodipine might be effective in subcortical vascular dementia(VaD). Its benefit in preventing further cognitive decline in patients with acute ischemic stroke(AIS) and vascular mild cognitive impairment(VaMCI) remains to be established. In this multicenter, double-blind trial, we randomly assigned 654 eligible patients to nimodipine 30 mg three times a day or placebo. The primary outcome was any cognitive decline defined by the changes on the Mini-Mental State Examination(DMMSE à3) or vascular AD assessment scale cognitive subscale(DADAS-cog ! 4) at 6 months. Secondary outcomes included any distribution shift of DADAS-cog, DMMSE or cognitive improvement defined by DADAS-cog à2, or DMMSE ! 0. The primary outcome in the nimodipine group and placebo group were similar for DMMSE à3(4.18% and 7.22%, respectively, P = 0.15) and DADAS-cog ! 4(8.36% and 8.93% respectively,P = 0.88). The distribution shift of DADAS-cog and DMMSE differed significantly between the two groups(P = 0.03 and P = 0.05 respectively). Cognitive improvement occurred in 55.4% in the nimodipine group and 43.6% in the placebo group measured by DADAS-cog à2(Odds Ratio, 1.54; 95% confidence interval[CI] 1.10–2.14, P < 0.01) or 84.0% and 74.6% respectively by DMMSE ! 0(Odds Ratio, 1.79; 95% CI 1.18–2.70, P < 0.01). Nimodipine was associated with better cognitive function in the memory domain. The adverse events rate was similar in two groups. This study is registered with ClinicalTrials.gov,NCT01220622. 12Effects of rapid quenching on structure and cycle stability of La-Mg-Ni-Co type hydrogen storage alloy显示文摘In order to improve the cycle stability of La-Mg-Ni-Co type alloy electrode, rapid quenching technology was employed. The effects of rapid quenching on the microstructure and cycle stability of the alloy were investigated. The obtained results show that the La2Mg(Ni0.85Co0.15)9M0.1 (M=B, Cr) alloy electrodes are composed of (La, Mg)Ni3 phase, LaNi5 phase and a small amount of the LaNi2 phase. A trace of the Ni2B phase exists in the as-cast MB alloy, and the Ni2B phase in the alloy nearly disappears after rapid quenching. Rapid quenching technology can slightly improve the cycling life of the alloy. When the quenching rate increases from 0 m·s-1 (As-cast is defined as quenching rate of 0 m·s-1) to 30 m·s-1, the cycle lives of the MB, MCr alloys enhance from 86 and 87 cycles to 106 and 119 cycles, respectively. 18PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. 20Electron back-scattering diffraction preliminary analysis of heterogeneous nuclei in magnesium alloy during solidification process under GPa high pressure显示文摘The effective heterogeneous nuclei in Mg-7.92 Zn-1.70 Y alloy during solidification process under high pressure was investigated by EBSD. It is found that the orientation relationship between Mg matrix and MgY phase, [111]_(MgY)//[1210]_(α-Mg) and(101)_(MgY)//(0001)_(α-Mg), exists between Mg matrix and MgY phase.The MgY phase is the potential heterogeneous nuclei substrate for a-Mg solidified under high pressure. It is calculated by Bramfitt mode that the mismatch between Mg matrix and MgY phase is greater than 12% under atmospheric pressure. When solidified under high pressure of 4 GPa, the solidification pressure can increase the number of the α-Mg nucleus by reducing the size of the nucleus, decreasing interfacial energy of MgY/a-Mg, and changing the wettability between MgY phase and α-Mg from non-wetting under atmospheric pressure to wetting under high pressure. The MgY phase becomes a powerful heterogeneous nuclei for α-Mg crystal.
