| 1 | Starch grain analysis reveals function of grinding stone tools at Shangzhai site, Beijing显示文摘Prehistoric groundstone tools including slabs (metate, mo-pan) and mullers (mano, mo-bang) are often considered to be tools that were used to process cereals in ancient China. Most archaeologists believe that groundstone tools are indicators of an agrarian economy. This study employed starch grain analysis to examine a slab and a muller excavated from the Shangzhai Site (7500-7000 cal a BP), Beijing. Identifiable starch grains recovered from the stone tools were classified into 9 genera and 12 species, most of which were from acorns and foxtail millets. Remains from the oak species Q. mongolicus, Q. acuttssima, and Q. dentate were identified as well as a few starch grains from broomcorn (Panicum miliaseum) and bean (Vigna sp.). Other starch grains were from unidentifiable roots and grasses. A variety of starch grains indicates that the grinding stone tools were used to process not only cereals, but also other seeds or roots that had to be ground or husked, thus bringing into question the aforementioned cereal hypothesis. Because the numbers of starch grains from nuts and millets were so large, both gathering and cultivation might have been important economic patterns before 7000 years ago in Beijing area. | 3 | Downstaging disease in patients with hepatocellular carcinoma outside up-to-seven criteria: Strategies using degradable starch microspheres transcatheter arterial chemo-embolization显示文摘AIM: To evaluate the downstaging rates in hepatitis C virus-patients with hepatocellular carcinoma(HCC), treated with degradable starch microspheres transcatheter arterial chemoembolization(DSM-TACE), to reach new-Milan-criteria(nM C) for transplantation. METHODS: This study was approved by the Ethics Committee of our institution. From September 2013 to March 2014 eight patients(5 men and 3 women) with liver cirrhosis and multinodular HCC, that did not meet n MC at baseline, were enrolled in this study. Patients who received any other type of treatment such as termal ablation or percutaneous ethanol injection were excluded. DSM-TACE was performed in all patients using Embo Cept? S and doxorubicin. Baseline and follow-up computed tomography or magnetic resonance imaging was assessed measuring the longest enhancing axial dimension of each tumor according to the modified Response Evaluation Criteria In Solid Tumors measure-ments, and medical records were reviewed.RESULTS: DSM-TACE was successfully performed in all patients without major complication. We treated 35 lesions(mean 4.3 per patient). Six of eight patients(75%) had their HCC downstaged to meet nM C. Every patient whose disease was downstaged eventually underwent transplantation. The six patients who received transplant were still living at the time of this writing, without recurrence of HCC. Baseline age(P = 0.25), Model for End-stage Liver Disease score(P = 0. 77), and α-fetoprotein level(P = 1.00) were similar between patients with and without downstaged HCC. | 4 | Factors Affecting Sensory Quality of Cooked japonica Rice显示文摘The sensory quality of cooked rice is an important factor in determining its market price, as well as consumer acceptance and breeding efforts aimed at improvement of rice grain quality. In this study,the sensory quality and physicochemical properties of three japonica rice varieties harvested in two different growing locations(Xiangshui and Hangzhou of China) were compared to determine the most important factors affecting the sensory quality. All the three varieties had higher scores for overall sensory quality in Xiangshui than in Hangzhou, indicating that the growing location is a key factor in determining the sensory quality of cooked japonica rice. In addition to growing location, variety(genotype) also had an important effect. Longdao 18 scored the highest for overall sensory quality in the two locations, whereas Longdao 30 had the lowest score in Xiangshui, and both Longdao 20 and Longdao 30 had the lowest scores in Hangzhou. Many physicochemical properties, such as apparent amylose content, protein content, thermal properties and free amino acid contents, showed significant differences between the two locations. Correlation analysis showed that apparent amylose content and protein content had contrasting effects on all the sensory attributes. The overall sensory quality was negatively correlated with protein content(r =-0.89, P < 0.01) and positively correlated with gel hardness(r = 0.91, P < 0.01),indicating that the protein content and hardness are important physicochemical properties for predicting the sensory quality of japonica rice. | 5 | DEGREE OF SUBSTITUTION OF THE IONIZED STARCHES AND THEIR ADHESIVE CAPACITY TO POLYESTER / COTTON FIBERS显示文摘Adhesive capacity of quaternary ammonium starch and starch phosphate to the polyester / cotton fibers was studied by means roving method for the purpose of improving the sizing performance and decreasing the cost. By introducing different amount of quaternary ammonium or phosphate groups onto starch molecules, the modified starch with a series of defferent degree of substitution was prepared. Therefore the regularity of adhesive capacity, extended property and size-absorbing ability with the change in degree of substitution (DS) was obtained through the researches. The experiments demonstrate that the DS of the two modified starches show a marked influence upon the properties mentioned above. The starch phosphate is superior to quaternary ammonium starch as warp size for polyester / cotton yarns from the viewpoint of adhesive capacity and cost, although both of them are better than natural one. | 6 | Transarterial chemoembolization using degradable starch microspheres and iodized oil in the treatment of advanced hepatocellular carcinoma: evaluation of tumor response, toxicity, and survival显示文摘BACKGROUND: In a multidisciplinary conference patients with advanced non-resectable hepatocellular carcinoma (HCC) were stratified according to their clinical status and tumor extent to different regional modalities or to best supportive care. The present study evaluated all patients who were stratified to repeated transarterial chemoembolization (TACE) from 1999 until 2003 in terms of tumor response, toxicity, and survival. A moderate embolizing approach was chosen using a combination of degradable starch microspheres (DSM) and iodized oil (Lipiodol) in order to combine anti-tumoral efficiency and low toxicity. METHODS: Fourty-seven patients were followed up prospectively. TACE treatment consisted of cisplatin (50 mg/m2), doxorubicin (50 mg/m2), 450-900 mg DSM, and 5-30 ml Lipiodol. DSM and Lipiodol were administered according to tumor vascularization. Patient characteristics,toxicity, and complications were outlined. In multivariate regression analyses of pre-treatment variables from a prospective database, predictors for tumor response and survival after TACE were determined. RESULTS: 112 TACE courses were performed (2.4±1.5 courses per patient). Mean maximum tumor size was 75 (± 43) mm, in 68% there was bilobar disease. Best response to TACE treatment was: progressive disease (PD) 9%, stable disease (SD) 55%, partial remission (PR) 36%, and complete remission (CR) 0%. Multivariate regression analyses identified tumor size ≤75 mm, tumor number ≤5, and tumor hypervascularization as predictors for PR. The overall 1-, 2-, and 3-year-survival rates were 75%, 59%, and 41%, respectively, and the median survival was 26 months. Low α-fetoprotein levels (<400 ng/ml) (Odds ratio=3.3) and PR as best response to TACE (Odds ratio=6.7) were significantly associated with long term survival (>30 months, R2=36%). Grade 3 toxicity occurred in 7.1% (n=8), and grade 4 toxicity in 3.6% (n=4) of all courses in terms of reversible leukopenia and thrombocytopenia. The incidence of major complications was 5.4% (n=6). All complications were managed conservatively. The mortality within 6 weeks after TACE was 2.1% (one patient). CONCLUSIONS: DSM and Lipiodol were combined successfully in the palliative TACE treatment of advanced HCC resulting in high rates of tumor response and survival at limited toxicity. Favourable tumor response was associated with tumor extent and vascularization. | 7 | New methods and progress in research on the origins and evolution of prehistoric agriculture in China显示文摘China is one of the main global centers of origin of agriculture. Foxtail millet(Setaria italica), common millet(Panicum miliaceum), and rice(Oryza sativa) were the first crops to be domesticated in China. There remain many uncertainties and controversies in our current understanding of the chronology, locations, and plant types at the origins and the process of evolution of prehistoric millet and rice farming, and their relationships with climate change and human adaptation. This review summarizes the research progress made by Chinese scientists over the last decade on the origins and evolution of prehistoric agriculture. It highlights novel techniques and methods for identifying early crop remains, including plant macrofossils(carbonized seeds, spikelets), microfossils(phytoliths, calciphytoliths, starch, pollen), and biomarkers; new evidence on the origins, development, and spread of early agriculture; and research related to climate and environmental changes. | 9 | Study on the morphology, crystalline structure and thermal properties of yam starch acetates with different degrees of substitution显示文摘This study was carried out to understand and establish the changes in physicochemical properties of starch extracted from Chinese yam (Dioscorea opposita Thunb.) after acetylation. Yam starch acetates with different degrees of substitution (DS) were prepared by the reaction of yam starch with glacial acetic acid/acetic anhydride using sulfuric acid as the catalyst. Their formation was confirmed by the presence of the carbonyl signal around 1750 cm-1 in the Fourier transform infrared (FT-IR) spectra. The thermal behavior of the native starch and starch acetate were investigated using thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The results reveal that the starch acetates are more thermally stable than the native starch. The starch esters showed 50% weight loss at tem- peratures from 328℃ to 372 ℃ , while the native starch underwent 50% weight loss at 325℃ . The glass transition temperature (Tg) of the starch decreased from 273℃ to 226℃. The X-ray diffraction (XRD) patterns could be classified as typical of the C-type for yam starch. X-ray diffraction also showed the loss of the ordered C-type starch crystalline structure and the degree of crystallinity of starch de- creased from 36.10% to 10.96% with the increasing DS. | 10 | Starch grains from dental calculus reveal ancient plant foodstuffs at Chenqimogou site, Gansu Province显示文摘Chenqimogou site, located at the south bank of Tao River, is comprised of a large group of graves dated to ~4000 a BP. For its large scale, unique mortuary system, and abundant of typical artifacts, the site was named one of 'Top Ten Archaeological Discoveries of 2008 in China'. Many intact human skeletons have been excavated from the graves. Three teeth from two adult skeletons excavated from graves M187 and M194 were examined. Forty-eight starch grains were retrieved from dental calculus of three teeth. Six of those starch grains could not be identified because of damage. The others were classified into seven groups. Most of them were from wheat (Triticum aestivum) and/or barley (Hordeum vulgare). Remains possibly from foxtail millet (Setaria italica), buckwheat (Fagopyrum esculentum), gingkgo (Ginkgo biloba), acorn (Quercus spp.), bean, roots or tubers were identified. Ancient starch grains from dental calculus indicate that human foodstuffs primarily sourced from wheat, buckwheat, and foxtail millet. Acorn, beans, tubers or roots were supplements. [Result] For the starch content change of F1 hybrid sorghum, the effects of GCA and SCA accounted for 81.06% and 17.97%, respectively. In the present study, CMS lines 45A, 29A and restorer lines Hui 1, 44R were proved to be the excellent parent materials for preparing high starch hybrid sorghum cultivars. | 13 | Editing of Rice Isoamylase Gene ISA1 Provides Insights into Its Function in Starch Formation显示文摘Isoamylase 1(ISA1) is an isoamylase-type debranching enzyme which plays a predominant role in amylopectin synthesis. In this study, the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated endonuclease 9(CRISPR/Cas9) system was used to edit ISA1 gene in rice via Agrobacterium-mediated transformation. We identified 36 genetic edited lines from 55 T_0 transgenic events, and classified the mutation forms into 7 types. Of those, two homozygous mutants, cr-isa1-1(type 1, with an adenine insertion) and cr-isa1-2(type 3, with a cytosine deletion) were selected for further analysis. Seed sizes of both cr-isa1-1 and cr-isa1-2 were affected, and the two mutants also displayed a shrunken endosperm with significantly lower grain weight. Electron microscopy analysis showed that abnormal starch granules and amyloplasts were found in cr-isa1-1 and cr-isa1-2 endosperm cells. The contents of total starch, amylose and amylopectin in the endosperm of the cr-isa1 mutants were significantly reduced, whereas sugar content and starch gel consistency were observably increased compared to the wild-type. The gelatinization temperature and starch chain length distributions of the cr-isa1 mutants were also altered. Moreover, transcript levels of most starch synthesis-related genes were significantly lower in cr-isa1 mutants. | 14 | Warming decreases photosynthates and yield of soybean [Glycine max(L.) Merrill] in the North China Plain显示文摘Understanding the responses of field crops such as soybean to climate warming is critical for economic development and adaptive management of food security. A field warming experiment was conducted using infrared heaters to investigate the responses of soybean phenology, photosynthetic characteristics, and yield to climate warming in the North China Plain. The results showed that 0.4 °C and 0.7 °C increases in soybean canopy air and soil temperature advanced anthesis stage by 3.8 days and shortened the length of entire growth stage by 4.5 days. Warming also decreased the leaf photosynthetic rate by 6.6% and 10.3% at the anthesis and seed filling stages, respectively, but increased the leaf vapor pressure deficit by 9.4%, 15.7%, and 14.1% at the anthesis, pod setting, and seed filling stages,respectively. However, leaf soluble sugar and starch were decreased by 25.6% and 20.5%,respectively, whereas stem soluble sugar was reduced by 12.2% at the anthesis stage under experimental warming. The transportation amount of leaf soluble sugar and contribution rate of transportation amount to seed weight were reduced by 58.2% and 7.7%, respectively,under warming. As a result, warming significantly decreased 100-seed weight and soybean yield by 20.8% and 45.0%, respectively. | 15 | A Comparative Analysis of Four Kinds of Polysaccharides Purified from Furcellaria lumbricalis显示文摘Four kinds of polysaccharides (FA, FB, FC and FD) were extracted and purified from a red alga Furcellaria lumbri-calis. Their monosaccharide composition, average molecular mass (Mw) and structural feature were determined by gas chromatog-raphy (GC), high performance gel permeation chromatography (HPGPC) and Fourier transform infrared spectroscopy (FTIR), respectively. The results showed that FA and FB were composed of galactose (Gal), 3,6-anhydro-galactose (AnG) and 4-sulfated-galactose (G4S), which were structurally similar to β-carrageenan and κ-carrageenan. FC was composed of sulfated galactose and small amount of AnG, indicating that it was a hybrid-sulfated galactan. FD was composed of α-1, 4-glucose and belonged to floridean starch. The respective yields of FA, FB, FC and FD were 4.84%,33.74%,8.76% and 13.5%, and the cor-responding Mw were 537 kD, 432 kD, 513 kD and 51 kD respectively. Large panicle is a prominent factor of high yield for indica-japonica hybrid rice cultivars, and the panicle weight varies greatly among different indica-japonica hybrid rice cultivars. It is important to research on yield and grain quality of different panicle weight indica-japonica hybrid rice cultivars. In this study, two different panicle types indica-japonica hybrid cultivars were used to research on the relation of yield and grain quality. The yields of two heavy panicle weights indica-japonica hybrid cultivars were significantly higher than that of two medium panicle weight rice cultivars. The cooking and eating quality and starch properties of different panicle type cultivars were evaluated. Yongyou 6715(medium panicle) and Yongyou 1852(heavy panicle) got the relatively higher cooking and eating quality. Rice cultivars with medium panicle weight had more large starch granules and higher relative crystallinity than cultivars with heavy panicle weight. Transition temperature and retrogradation enthalpy(ΔHret) of medium panicle type cultivars were significantly higher than that of heavy panicle type cultivars. There was no significant difference in amylose content among different panicle type cultivars. Protein content of heavy panicle type cultivar was higher than that of medium panicle type cultivar, and protein content is the main factor affect cooking and eating quality in this study. The cultivar Yongyou 6715 got the highest taste value with the lowest protein content. Thus, it is suggested that the emphasis on improving rice cooking and eating quality of indica-japonica hybrid rice cultivars is how to reduce the protein content in rice grain. | 18 | Effect of nitrogen fertilizer on distribution of starch granules in different regions of wheat endosperm显示文摘This study provided visual evidence of a nitrogen effect on starch granules(SGs) in wheat endosperm. Winter wheat(Titicum aestivum L.) cultivar Xumai 30 was cultured under no nitrogen(control) and 240 kg ha-1of nitrogen applied at the booting stage. The number, morphology, and size of Aand B-type SGs in subaleurone of dorsal endosperm(SDE), center of dorsal endosperm(CDE), modified aleurone(MA), subaleurone of ventral endosperm(SVE), and center of ventral endosperm(CVE) were observed under light and electron microscopes.(1) The distribution of SGs in SDE was similar to that in SVE, the distributions of SGs in CDE and CVE were similar, but the distribution of SGs in MA was different from those in the other four endosperm regions. The number of SGs in the five endosperm regions was in the order SDE > CDE > SVE > CVE > MA.(2) Nitrogen increased the number of Aand B-type SGs in SDE and SVE. Nitrogen also increased the number of B-type SGs but decreased the number of A-type SGs in CDE and CVE. Nitrogen decreased the numbers of A-type and B-type SGs in MA. | 19 | Identification of unconditional and conditional QTL for oil, protein and starch content in maize显示文摘Oil, protein and starch are key chemical components of maize kernels. A population of 245 recombinant inbred lines(RILs) derived from a cross between a high-oil inbred line, By804, and a regular inbred line, B73, was used to dissect the genetic interrelationships among oil, starch and protein content at the individual QTL level by unconditional and conditional QTL mapping. Combined phenotypic data over two years with a genetic linkage map constructed using 236 markers, nine, five and eight unconditional QTL were detected for oil, protein and starch content, respectively. Some QTL for oil, protein and starch content were clustered in the same genomic regions and the direction of their effects was consistent with the sign of their correlation. In conditional QTL mapping, 37(29/8) unconditional QTL were not detected or showed reduced effects, four QTL demonstrated similar effects under unconditional and conditional QTL mapping, and 17 additional QTL were identified by conditional QTL mapping. These results imply that there is a strong genetic relationship among oil, protein and starch content in maize kernels. | 20 | Studies of poly-L-lysine-starch nanoparticle preparation and its application as gene carrier显示文摘Anion starch nanoparticle (StNP) with a diameter of 50 nm was prepared in wa- ter-in-oil microemulsion, with soluble starch as raw materials and POCl3 as crosslinking agent. PLL-StNP was prepared by linking poly-L-lysine (PLL) on the surface of StNP. At the same time, the size of PLL-StNP and its stability in aqueous solution were checked by AFM. The analysis of plasmid DNA binding, DNase I enzymatic degradation, toxicity and transfection were done. We discovered that PLL-StNP may be used as non-virus nanoparticle gene carrier. And we devel- oped the method of preparing PLL-StNP gene carrier and used it in cell transfection.
