| 1 | Motility-based label-free detection of parasites in bodily fluids using holographic speckle analysis and deep learning显示文摘Parasitic infections constitute a major global public health issue.Existing screening methods that are based on manual microscopic examination often struggle to provide sufficient volumetric throughput and sensitivity to facilitate early diagnosis.Here,we demonstrate a motility-based label-free computational imaging platform to rapidly detect motile parasites in optically dense bodily fluids by utilizing the locomotion of the parasites as a specific biomarker and endogenous contrast mechanism.Based on this principle,a cost-effective and mobile instrument,which rapidly screens~3.2 mL of fluid sample in three dimensions,was built to automatically detect and count motile microorganisms using their holographic time-lapse speckle patterns.We demonstrate the capabilities of our platform by detecting trypanosomes,which are motile protozoan parasites,with various species that cause deadly diseases affecting millions of people worldwide.Using a holographic speckle analysis algorithm combined with deep learningbased classification,we demonstrate sensitive and label-free detection of trypanosomes within spiked whole blood and artificial cerebrospinal fluid(CSF)samples,achieving a limit of detection of ten trypanosomes per mL of whole blood(~five-fold better than the current state-of-the-art parasitological method)and three trypanosomes per mL of CSF.We further demonstrate that this platform can be applied to detect other motile parasites by imaging Trichomonas vaginalis,the causative agent of trichomoniasis,which affects 275 million people worldwide.With its costeffective,portable design and rapid screening time,this unique platform has the potential to be applied for sensitive and timely diagnosis of neglected tropical diseases caused by motile parasites and other parasitic infections in resource-limited regions. | Yibo Zhang Hatice Ceylan Koydemir Michelle M.Shimogawa Sener Yalcin Alexander Guziak Tairan Liu Ilker Oguz Yujia Huang Bijie Bai Yilin Luo Yi Luo Zhensong Wei Hongda Wang Vittorio Bianco Bohan Zhang Rohan Nadkarni Kent Hill Aydogan Ozcan | 2018 | Light(Science & Applications)2018,7,1: | 6 |

| 3 | Biosynthesis of rumbrins and inspiration for discovery of HIV inhibitors显示文摘Investigation on how nature produces natural compounds with chemical and biological diversity at the genetic level offers inspiration for the discovery of new natural products and even their biological targets.The polyketide rumbrin(1)is a lipid peroxide production and calcium accumulation inhibitor,which contains a chlorinated pyrrole moiety that is a rare chemical feature in fungal natural products.Here,we identify the biosynthetic gene cluster(BGC)rum of 1 and its isomer 12E-rumbrin(2)from Auxarthron umbrinum DSM3193,and elucidate their biosynthetic pathway based on heterolo-gous expression,chemical complementation,and isotopic labeling.We show that rumbrins are assembled by a highly reducing polyketide synthase(HRPKS)that uniquely incorporates a proline-derived pyrrolyl-CoA starer unit,and followed by methylation and chlorination.Sequent precursor directed biosynthesis was able to yield a group of rumbrin analogues.Remarkably,inspired by the presence of a human immunodeficiency virus(HIV)-Nef associated gene in the rum cluster,we predicted and pharmacologically demonstrated rumbrins as potent inhibitors of HIV at the nanomolar level.This work enriches the recognition of unconventional starter units of fungal PKSs and provides a new strategy for genome mining-guided drug discovery. | Beifen Zhong Jun Wan Changhui Shang Jiajia Wen Yujia Wang Jian Bai Shan Cen Youcai Hu | 2022 | Acta Pharmaceutica Sinica B2022,12,11: | 1 |
| 4 | Proanthocyanidins extracted from grape seeds inhibit the growth of hepatocellular carcinoma cells and induce apoptosis through the MAPK/AKT pathway显示文摘Proanthocyanidins naturally occur in various fruits,some vegetables and beverages.Increasing evidence has shown that dietary intake of proanthocyanidins is a promising alternative to tumor chemoprevention or chemotherapy.Grape seeds are the most abundant source of proanthocyanidins,and grape seed proanthocyanidins(GSPs)show an anticancer effect on hepatocellular carcinoma(HCC),which accounts for approximately 85-90%of all primary liver malignancies.Although an increasing number of studies have investigated the effects of GSPs on HCC and the associated mechanisms,the precise mechanism of GSP therapy in HCC remains unclear.This study aimed to understand the cytotoxic effect of GSPs on HepG2 cells by investigating morphology and growth inhibition.Cell proliferation and apoptosis were estimated by an MTT assay and flow cytometry,and the protein expression of HepG2 cells was determined through Western blotting.The results indicated that GSPs could markedly inhibit HepG2 liver cancer cells and induce apoptosis,with the effects varying by dose and treatment time.Furthermore,GSPs inhibited the phosphorylation of extracellular regulated protein kinases(ERK),c-Jun NH2-terminal kinase(JNK),p38 mitogen-activated protein kinase(p38 MAPK),IκBα,pyruvate dehydrogenase kinase 1(PDK1),glycogen synthase kinase-3 beta(GSK3β)and protein kinase B(AKT),which accounted for the inactivation of mitogen-activated protein kinase/AKT(MAPK/AKT)pathways.These results indicate that GSPs are a potential source of natural chemopreventive agents for the treatment of HCC. | Wancai Zheng a Yujing Feng Yujia Bai Zuoshan Feng Xijuan Yang Bin Dang Ming Xiao Jianmei Zhang Sheng-Qiang Han | 2022 | Food Bioscience2022,45,1: | 1 |
