| 1 | GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://gffzzdbc7b6aaae734bddhkvxnn699b9bo6vow.ffgz.tsg.suse.edu.cn/gsa or http://gffzz4eaf941a184140e3hkvxnn699b9bo6vow.ffgz.tsg.suse.edu.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 2 | Responses of gross primary productivity to different sizes of precipitation events in a temperate grassland ecosystem in Inner Mongolia, China显示文摘Changes in the sizes of precipitation events in the context of global climate change may have profound impacts on ecosystem productivity in arid and semiarid grasslands. However, we still have little knowledge about to what extent grassland productivity will respond to an individual precipitation event. In this study, we quantified the duration, the maximum, and the time-integrated amount of the response of daily gross primary productivity(GPP) to an individual precipitation event and their variations with different sizes of precipitation events in a typical temperate steppe in Inner Mongolia, China. Results showed that the duration of GPP-response(τR) and the maximum absolute GPP-response(GPPmax) increased linearly with the sizes of precipitation events(Pes), driving a corresponding increase in time-integrated amount of the GPP-response(GPPtotal) because variations of GPPtotal were largely explained by τR and GPPmax. The relative contributions of these two parameters to GPPtotal were strongly Pes-dependent. The GPPmax contributed more to the variations of GPPtotal when Pes was relatively small(<20 mm), whereas τR was the main driver to the variations of GPPtotal when Pes was relatively large. In addition, a threshold size of at least 5 mm of precipitation was required to induce a GPP-response for the temperate steppe in this study. | 3 | Structural variation during dog domestication: insights from gray wolf and dhole genomes显示文摘Several processes like phenotypic evolution, disease susceptibility and environmental adaptations, which fashion the domestication of animals, are largely attributable to structural variations(SVs) in the genome.Here, we present high-quality draft genomes of the gray wolf(Canis lupus) and dhole(Cuon alpinus) with scaffold N50 of 6.04 Mb and 3.96 Mb, respectively. Sequence alignment comprising genomes of three canid species reveals SVs specific to the dog, particularly 16 315 insertions, 2565 deletions, 443 repeats, 16 inversions and 15 translocations. Functional annotation of the dog SVs associated with genes indicates their enrichments in energy metabolisms, neurological processes and immune systems. Interestingly, we identify and verify at population level an insertion fully covering a copy of the AKR1 B1(Aldo-Keto Reductase Family 1 Member B) transcript. Transcriptome analysis reveals a high level of expression of the new AKR1 B1 copy in the small intestine and liver, implying an increase in de novo fatty acid synthesis and antioxidant ability in dog compared to gray wolf, likely in response to dietary shifts during the agricultural revolution. For the first time, we report a comprehensive analysis of the evolutionary dynamics of SVs during the domestication step of dogs. | 10 | Alternative Copper-Based Single-Atom Nanozyme with Superior Multienzyme Activities and NIR-II Responsiveness to Fight against Deep Tissue Infections显示文摘Nanozymes are considered to represent a new era of antibacterial agents,while their antibacterial efficiency is limited by the increasing tissue depth of infection.To address this issue,here,we report a copper and silk fibroin(Cu-SF)complex strategy to synthesize alternative copper single-atom nanozymes(SAzymes)with atomically dispersed copper sites anchored on ultrathin 2D porous N-doped carbon nanosheets(CuN_(x)-CNS)and tunable N coordination numbers in the CuN_(x) sites(x=2 or 4).The CuN_(x)-CNS SAzymes inherently possess triple peroxidase(POD)-,catalase(CAT)-,and oxidase(OXD)-like activities,facilitating the conversion of H_(2)O_(2)and O_(2)into reactive oxygen species(ROS)through parallel POD-and OXD-like or cascaded CAT-and OXD-like reactions.Compared to CuN_(2)-CNS,tailoring the N coordination number from 2 to 4 endows the SAzyme(CuN_(4)-CNS)with higher multienzyme activities due to its superior electron structure and lower energy barrier.Meanwhile,CuN_(x)-CNS display strong absorption in the second near-infrared(NIR-II)biowindow with deeper tissue penetration,offering NIR-II-responsive enhanced ROS generation and photothermal treatment in deep tissues.The in vitro and in vivo results demonstrate that the optimal CuN_(4)-CNS can effectively inhibit multidrug-resistant bacteria and eliminate stubborn biofilms,thus exhibiting high therapeutic efficacy in both superficial skin wound and deep implant-related biofilm infections. | Jiaxiang Bai Yonghai Feng Wenming Li Zerui Cheng Jessica MRosenholm Huilin Yang Guoqing Pan Hongbo Zhang Dechun Geng | 2023 | Research2023,,3: | 0 |
