1PKM2 promotes reductive glutamine metabolism显示文摘Objective:Pyruvate kinases M(PKM),including the PKM1 and PKM2 isoforms,are critical factors in glucose metabolism.PKM2promotes aerobic glycolysis,a phenomenon known as'the Warburg effect'.The purpose of this study was to identify the roles of PKM2 in regulating cellular metabolism.Methods:The CRISPR/Cas9 system was used to generate the PKM-knockout cell model to evaluate the role of PKM in cellular metabolism.Lactate levels were measured by the Vitros LAC slide method on an autoanalyzer and glucose levels were measured by the autoanalyzer AU5800.The metabolism of ^(13)C_6-glucose or ^(13)C_5-glutamine was evaluated by liquid chromatography/mass spectrometry analyses.The effects of PKM on tumor growth were detected in vivo in a tumor-bearing mouse model.Results:We found that both PKM1 and PKM2 enabled aerobic glycolysis,but PKM2 converted glucose to lactate much more efficiently than PKM1.As a result,PKM2 reduced glucose levels reserved for intracellular utilization,particularly for the production of citrate,and thus increased theα-ketoglutarate/citrate ratio to promote the generation of glutamine-derived acetylcoenzyme A through the reductive pathway.Furthermore,reductive glutamine metabolism facilitated cell proliferation under hypoxia conditions,which supports in vivo tumor growth.In addition,PKM-deletion induced a reverse Warburg effect in tumorassociated stromal cells.Conclusions:PKM2 plays a critical role in promoting reductive glutamine metabolism and maintaining proton homeostasis.This study is helpful to increase the understanding of the physiological role of PKM2 in cancer cells.Miao Liu Yuanyuan Wang Yuxia Ruan Changsen Bai Li Qiu Yanfen Cui Guoguang Ying Binghui Li 2018Cancer Biology & Medicine2018,15,4:6
2Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 7Epidemiological characteristics and clinical manifestations of pediatric patients with COVID-19 in China: A multicenter retrospective study显示文摘Importance:The Coronavirus disease 2019(COVID-19)global pandemic poses a considerable challenge for pediatricians.Objective:This study aimed to identify the epidemiological characteristics and clinical features of pediatric patients with COVID-19 in China.Methods:This multicenter retrospective study included pediatric patients from 46 hospitals in China,covering 12 provinces and two municipalities.Epidemiological,demographic,clinical,laboratory,treatment,and outcome data were analyzed.Results:In total,211 pediatric patients with COVID-19 were included in this study.The median age was 7.0 years(range:22 days to 18 years).Approximately 16.3%of the patients exhibited asymptomatic infections,23.0%had upper respiratory tract infections,and 60.7%had pneumonia,including two with severe pneumonia and one with critical illness.Approximately 78.7%of the pediatric patients occurred in familial clusters.The most three common symptoms or signs at onset in children with COVID-19 were fever(54.5%),cough(49.3%),and pharyngeal congestion(20.8%).Only 17.6%of the patients presented with decreased lymphocyte count,whereas 13.6%had increased lymphocyte count.Among the patients with pneumonia who exhibited abnormal chest computed tomography findings,18.2%(23/127)of the patients had no other symptoms.Generally,the chest radiographs showed abnormalities that affected both lungs(49.6%);ground-glass opacity(47.2%)was the most common manifestation.The cure and improvement rates were 86.7%(183/211)and 13.3%(28/211),respectively.Only one patient with an underlying condition received invasive mechanical ventilation;none of the patients died.Interpretation:Similar to adults,children of all age groups are susceptible to COVID-19.Fortunately,most pediatric patients have mild symptoms or remain asymptomatic,despite the high incidence of pneumonia.Decreased proportions of white blood cells and lymphocytes are less frequent in children than in adults.Yan Bai Liwei Gao Xianfeng Wang Lili Zhong Jingfeng Li Shenggang Ding Yuejie Zheng Jun Liu Yuxia Cui Lina Wang Mingfeng Han Jun Liu Xinping Jiang Min Jiang Junhua Li Ning Chen Yunxiao Shang Hourong Zhou Yi Xu Gen Lu Xing Chen Jizhi Xu Qihong Fan Yu Tang Jiang Wu Cen Li Xiaoxiang Yang Chunxi Chen Yonghong Yang Gary Wing-Kin Wong Adong Shen Tianyou Wang Runming Jin Baoping Xu Kunling Shen 2021Pediatric Investigation2021,5,3:0
8NIR-activated nanosystems with self-modulated bacteria targeting for enhanced biofilm eradication and caries prevention显示文摘The efficacious delivery of antimicrobial drugs to intractable oral biofilms remains a challenge due to inadequate biofilm penetration and lack of pathogen targeting.Herein,we have developed a microenvironment-activated poly(ethylene glycol)(PEG)-sheddable nanoplatform to mediate targeted delivery of drugs into oral biofilms for the efficient prevention of dental caries.The PEGylated nanoplatform with enhanced biofilm penetration is capable of deshielding the PEG layer under slightly acidic conditions in a PEG chain length-dependent manner to re-expose the bacteria-targeting ligands,thereby facilitating targeted codelivery of ciprofloxacin(CIP)and IR780 to the bacteria after accumulation within biofilms.The nanoplatform tends to induce bacterial agglomeration and suffers from degradation in the acidic oral biofilm microenvironment,triggering rapid drug release on demand around bacterial cells.The self-modulating nanoplatform under near-infrared(NIR)irradiation accordingly displays greatly augmented potency in oral biofilm penetration and disruption compared with drugs alone.Topical oral treatment with nanoplatforms involving synergetic pharmacological and photothermal/photodynamic trinary therapy results in robust biofilm dispersion and efficacious suppression of severe tooth decay in rats.This versatile nanoplatform can promote local accumulation and specific drug transport into biofilms and represents a new paradigm for targeted drug delivery for the management of oral biofilm-associated infections.Yunjian Yu Yufei Zhang Yijie Cheng Yuxia Wang Zeyuan Chen Haonan Sun Xiaosong Wei Zhuang Ma Jie Li Yayun Bai Zhongming Wu Xinge Zhang 2022Bioactive Materials2022,7,7:0
9Electraesalopsis beuteli gen. & sp. nov., the first lucanid beetle from the Cretaceous Burmese amber(Coleoptera: Scarabaeoidea)显示文摘Absract The first stag beetle found in Cretaceous Myanmar amber, Electraesalopsis beuteli Bai, Zhang & Qiu gen. & sp. nov., is described and illustrated on the basis of one well-preserved specimen. This specimen provides more detailed morphological characters compared with compression fossils, e.g. the highly setose antennae and the punctured elytra. However, the systematic position of the new genus is still questionable and we provisionally place it as Lucanidae incertae sedis. In addition, the discovery of Electraesalopsis Bai, Zhang & Qiu gen. nov. also indicates that there is very high biodiversity in Burmese amber.
