1MiR-19a-3p regulates the Forkhead box F2-mediated Wnt/β-catenin signaling pathway and affects the biological functions of colorectal cancer cells显示文摘BACKGROUND Colorectal cancer(CRC)is one of the most common malignancies worldwide.AIM To explore the expression of microRNA miR-19a-3p and Forkhead box F2(FOXF2)in patients with CRC and the relevant mechanisms.METHODS Sixty-two CRC patients admitted to the hospital were enrolled into the study group,and sixty healthy people from the same period were assigned to the control group.Elbow venous blood was sampled from the patients and healthy individuals,and blood serum was saved for later analysis.MiR-19a-3p mimics,miR-19a-3p inhibitor,miR-negative control,small interfering-FOXF2,and short hairpin-FOXF2 were transfected into HT29 and HCT116 cells.Then quantitative polymerase chain reaction was performed to quantify the expression of miR-19a-3p and FOXF2 in HT29 and HCT116 cells,and western blot(WB)analysis was conducted to evaluate the levels of FOXF2,glycogen synthase kinase 3 beta(GSK-3β),phosphorylated GSK-3β(p-GSK-3β),β-catenin,p-β-catenin,α-catenin,Ncadherin,E-cadherin,and vimentin.The MTT,Transwell,and wound healing assays were applied to analyze cell proliferation,invasion,and migration,respectively,and the dual luciferase reporter assay was used to determine the correlation of miR-19a-3p with FOXF2.RESULTS The patients showed high serum levels of miR-19a-3p and low levels of FOXF2,and the area under the curves of miR-19a-3p and FOXF2 were larger than 0.8.MiR-19a-3p and FOXF2 were related to sex,tumor size,age,tumor-nodemetastasis staging,lymph node metastasis,and differentiation of CRC patients.Silencing of miR-19a-3p and overexpression of FOXF2 suppressed the epithelialmesenchymal transition,invasion,migration,and proliferation of cells.WB analysis revealed that silencing of miR-19a-3p and FOXF2 overexpression significantly suppressed the expression of p-GSK-3β,β-catenin,N-cadherin,and vimentin;and increased the levels of GSK-3β,p-β-catenin,α-catenin,and Ecadherin.The dual luciferase reporter assay confirmed that there was a targeted correlation of miR-19a-3p with FOXF2.In addition,a rescue experiment revealed that there were no differences in cell proliferation,invasion,and migration in HT29 and HCT116 cells co-transfected with miR-19a-3p-mimics+sh-FOXF2 and miR-19a-3p-inhibitor+si-FOXF2 compared to the miR-negative control group.CONCLUSION Inhibiting miR-19a-3p expression can upregulate the FOXF2-mediated Wnt/β-catenin signaling pathway,thereby affecting the epithelial-mesenchymal transition,proliferation,invasion,and migration of cells.Thus,miR-19a-3p is likely to be a therapeutic target in CRC.Fu-Bing Yu Juan Sheng Jia-Man Yu Jing-Hua Liu Xiang-Xin Qin Bo Mou 2020World Journal of Gastroenterology2020,26,6:7
2Estimating abundance and density of Amur tigers along the Sino- Russian border显示文摘As an apex predator the Amur tiger(Panthera tigris altaica)could play a pivotal role in maintaining the integ­rity of forest ecosystems in Northeast Asia.Due to habitat loss and harvest over the past century,tigers rapidly declined in China and are now restricted to the Russian Far East and bordering habitat in nearby China.To fa­cilitate restoration of the tiger in its historical range,reliable estimates of population size are essential to assess effectiveness of conservation interventions.Here we used camera trap data collected in Hunchun National Na­ture Reserve from April to June 2013 and 2014 to estimate tiger density and abundance using both maximum likelihood and Bayesian spatially explicit capture-recapture(SECR)methods.A minimum of 8 individuals were detected in both sample periods and the documentation of marking behavior and reproduction suggests the presence of a resident population.Using Bayesian SECR modeling within the 11400 km2 state space,den­sity estimates were 0.33 and 0.40 individuals/100 km^(2) in 2013 and 2014,respectively,corresponding to an es­timated abundance of 38 and 45 animals for this transboundary Sino-Russian population.In a maximum likeli­hood framework,we estimated densities of 0.30 and 0.24 individuals/100 km^(2) corresponding to abundances of 34 and 27,in 2013 and 2014,respectively.These density estimates are comparable to other published estimates for resident Amur tiger populations in the Russian Far East.This study reveals promising signs of tiger recovery in Northeast China,and demonstrates the importance of connectivity between the Russian and Chinese popula­tions for recovering tigers in Northeast China.Wenhong XIAO Limin FENG Pu MOU Dale GMIQUELLE Mark HEBBLEWHITE Joshua FGOLDBERG Hugh SROBINSON Xiaodan ZHAO Bo ZHOU Tianming WANG Jianping GE 2016Integrative Zoology2016,11,4:4
3Safety and efficacy of Yupingfeng granules in children with recurrent respiratory tract infection:A randomized clinical trial显示文摘Importance:Recurrent respiratory tract infection(RRTI)is common in children.Inappropriate RRTI treatment will lead to asthma and other diseases,thereby seriously affecting the growth and physical health of children.Immune function modulation can prevent and alleviate childhood RRTI.Yupingfeng(YPF),a patented traditional Chinese medicine(TCM),has immunomodulatory effects and is widely used in China to treat children with RRTI.Objective:To evaluate the safety and efficacy of YPF monotherapy in treating children with RRTI.Methods:This multicenter,randomized,double-blind,double-simulation,noninferiority clinical trial was conducted from January 2015 to August 2017,with an 8-week treatment period and 52-week follow-up after the drug withdrawal.Children aged 2–6 years with RRTI meeting the inclusion and exclusion criteria were enrolled in 13 hospitals in China and divided randomly into three groups(2:2:1 ratio)to receive YPF,pidotimod,or placebo.The primary outcome was the proportion of RRTI returning to normal standard level during the follow-up.The secondary outcomes were reduction in the number of RRTI recurrences,effect on clinical symptoms(in accord with TCM practice),effect per symptom,and safety.The trial was registered at the Chinese Clinical Trials Registry(www.chictr.org.cn)under the unique identifier ChiCTR-IPR-15006847.Results:Three hundred and fifty-one children were enrolled and randomly assigned to 3 groups;124,125,and 61 children in the YPF,pidotimod,and placebo groups,respectively,had completed the trial.During the follow-up,the proportion of RRTI returning to normal standard level was 73.13%,67.15%,and 38.81%with YPF,pidotimod,and placebo,respectively(P<0.0001).The proportion of cases who returned to normal standard level in the YPF group was 34.32%higher than that in the placebo group.The safety profile did not significantly differ among the groups.Interpretation:YPF granules were noninferior to the active control drug pidotimod oral solution for the treatment of RRTI in children,and were superior to placebo,with a high safety profile.Baoping Xu Xinmin Li Siyuan Hu Yixiao Bao Fengmei Chen Zhimin Chen Yonggang Du Enmei Liu Yufeng Liu Qinghui Mou Baoling Su Bo Wang Jianwen Xu Guiping Xu Qiaozhi Yang Liwei Gao Xiaohui Liu Lei Li Rong Ma Kunling Shen 2022Pediatric Investigation2022,6,2:3
4Unique sandwich design of high-efficiency heat-conducting phosphor-in-glass film for high-quality laser-driven white lighting显示文摘Multi-color phosphor-in-glass(PiG)film has been considered as a promising color converter in high-quality laser lighting owing to its outstanding merits of phosphor versatility,tunable luminescence,and simple preparation.However,the opto-thermal properties of PiG film are severely affected by the photon reabsorption and backward scattering of phosphor structure and the heat conduction of substrate.Herein,a unique sandwich design of phosphor structure was introduced in the multi-color PiG film for high-quality laser lighting.By elaborately synthesizing the borosilicate glass with low glass transition temperature(T_(g)),similar expansion coefficient,and high refractive index(RI),the sandwiched PiGs were prepared by sintering(~600℃)broadband green and red phosphor glass films on the double sides of sapphire.The green and red PiG films were tightly coated on the sapphire with no delamination and maintained higher luminescence intensity than raw phosphors at high temperatures.By simultaneously coupling photon reabsorption and backward scattering,the sandwiched green PiG film-sapphire-red PiG film(G-S-R PiG)yields a high-quality white light with a high luminous efficacy of 163 lm/W and an excellent color rendering index(CRI)of 85.4 under a laser power of 2.4 W,which are the best comprehensive results yet reported.Benefiting from the ingenious sandwich design with heat-conducting sapphire and thin PiG films,the G-S-R PiG displays low working temperatures(<200℃)under high-power laser excitation.This work reveals the role of sandwiched phosphor structure in photon loss and heat dissipation,which provides a new strategy to design PiG films for high-quality laser lighting.Yang PENG Zikang YU Jiuzhou ZHAO Qing WANG Jiaxin LIU Bo SUN Yun MOU Mingxiang CHEN 2022Journal of Advanced Ceramics2022,11,12:2

6The development of a rapid high-quality universal nucleic acid extraction kit based on magnetic separation显示文摘Nucleic acid extraction is one of the bases in molecular biology and it is the indispensable process for nucleic acid detection. Due to its excellent characteristics, the method based on magnetic beads has become one of the main methods for nucleic acid extraction.In this research, we have developed a rapid high-quality universal nucleic acid extraction kit based on magnetic beads(MBs) and named it MB-100. Some factors affecting nucleic acid extraction were optimized, such as particle size of MBs, surface groups of MBs and lysis time. Results demonstrated that micron-grade MBs@SiO_2 particles were much more appropriate for nucleic acid extraction and lysis time was 5 min. Simultaneously, the performance of MB-100 was explored. 8Early results and factors affecting arterial switch operation in provincial medical center显示文摘Transposition of the great arteries(TGA)is a serious and complicated congenital heart disease,accounting for 5%to 7%of all congenital heart malformations.Most fetuses with TGA can survive stably to delivery.After birth,their survival depends on the associated ventricular septal defect(VSD),atrial septal defect(ASD)or patent ductus arteriosus(PDA).Without treatment,approximately 30%of these infants die in the first week after birth and 90%die within the first year.[1]Arterial switch operation is now recognized worldwide as an effective treatment for TGA and good results have been achieved with this approach.ASO was performed late at our center,with a few patients and high mortality rates in the early stage.In recent years,both the number of patients and the survival rate have increased with the gradual improvement of the skills of the surgical team.More patients have been diagnosed prenatally with the development and promotion of fetal echocardiography.From pregnancy to delivery to surgery,children with TGA require monitoring with close cooperation among medical staff from multiple disciplines.In this study,the data of 87 patients who underwent ASO from January 2014 to December 2019 at our center were retrospectively summarized to analyze the major nonsur-gical factors affecting patient prognosis[Table 1].The study was approved by the Ethics Committee of Shandong Provincial Hospital.Hui-Jun Mou Mei Zhu An-Biao Wang Bo Han Chun-Yang Xu 2021Chinese Medical Journal2021,,22:0
9Expression and functional identification of the hypothetical adhesin P32 from Mycoplasma genitalium显示文摘Mycoplasma genitalium is the main causative agent for non-gonococcal and non-chlamydial urethritis. P32 is the putative surface-exposed membrane protein of M. genitalium and it has substaintial identity in amino acid sequence with adhesin protein P30 from M. pnewnoniae. Since M. pneumoniae mutants lacking P30 protein is defective in cytadherence, P32 protein has been proposed to be an essential adhesin implicated in the adherence of M. genitalium to host cells. The prokaryotic expression vector pET-30 ( + )/p32 was constructed in the present study, and the recombinant protein was expressed in E. coli and purified under denaturing condition. As demonstrated by the immunoblotting analysis, the recombinant protein could react with rabbit antisera against M. genitalium, and adherence inhibition assays were petformed with antisera against this recombinant protein. 14Multiple genotyping based on multiplex PCR and microarray显示文摘The genetic variability has obtained more and more attention in the process of diagnosis and treatment of tumors.Herein,we have described a multiple genotyping method based on magnetic enrichmentmultiplex PCR(MEM-PCR) and microarray technology.Monodisperse magnetic beads were fabricated and modified with streptavidin.Four loci on two genes(M235T and A-6G loci on AGT gene,A1298 C and C677 T loci on MTHFR gene) were selected to study single nucleotide polymorphisms(SNP).Target sequences of these SNP loci were amplified using Cy3-labeled primers through multiplex PCR in one tube after the templates were enriched and purified by functional magnetic beads(MB).Four pairs of NH_2-labeled probes,corresponding to each locus,were fixed on CHO-modified glass slide by covalent binding.Hybridization between target sequences and probes was performed under suitable conditions.The spotting locations on microarray and the ratio of fluorescence intensity,produced by different loci,were used to distinguish the SNP genotypes.Finally,three of gastric cancer samples were collected and genotyping analysis for these four SNP loci was carried out successfully simultaneously by this method.Xian-Bo Mou Zeeshan Ali Bo Li Tao-Tao Li Huan Yi Hong-Ming Dong Nong-Yue He Yan Deng Xin Zeng 2016Chinese Chemical Letters2016,27,11:0
15Machine learning-based radiomics score improves prognostic prediction accuracy of stage II/III gastric cancer: A multi-cohort study显示文摘BACKGROUND Although accurately evaluating the overall survival(OS)of gastric cancer patients remains difficult,radiomics is considered an important option for studying pro-gnosis.AIM To develop a robust and unbiased biomarker for predicting OS using machine learning and computed tomography(CT)image radiomics.METHODS This study included 181 stage II/III gastric cancer patients,141 from Lichuan People's Hospital,and 40 from the Cancer Imaging Archive(TCIA).Primary tumors in the preoperative unenhanced CT images were outlined as regions of interest(ROI),and approximately 1700 radiomics features were extracted from each ROI.The skeletal muscle index(SMI)and skeletal muscle density(SMD)were measured using CT images from the lower margin of the third lumbar vertebra.Using the least absolute shrinkage and selection operator regression with 5-fold cross-validation,36 radiomics features were identified as important predictors,and the OS-associated CT image radiomics score(OACRS)was cal-culated for each patient using these important predictors.RESULTS Patients with a high OACRS had a poorer prognosis than those with a low OACRS score(P<0.05)and those in the TCIA cohort.Univariate and multivariate analyses revealed that OACRS was a risk factor[RR=3.023(1.896-4.365),P<0.001]independent of SMI,SMD,and pathological features.Moreover,OACRS outperformed SMI and SMD and could improve OS prediction(P<0.05).CONCLUSION A novel biomarker based on machine learning and radiomics was developed that exhibited exceptional OS discrimination potential.Ying-Hao Xiang Huan Mou Bo Qu Hui-Rong Sun 2024World Journal of Gastrointestinal Surgery2024,16,2:0
