| 1 | An EGLN1 mutation may regulate hypoxic response in cyanotic congenital heart disease through the PHD2/HIF-1A pathway显示文摘Cyanotic congenital heart disease(CCHD),a term describing the most severe congenital heart diseases are characterized by the anatomic malformation of a right to left shunt.Although the incidence of CCHD are far less than the that of congenital heart diseases(CHD),patients with CCHD always present severe clinical features such as hypoxia,dyspnea,and heart failure.Chronic hypoxia induces hypoxemia that significantly contributes to poor prognosis in CCHD.Current studies have demonstrated that the prolyl-4-hydroxylase2(PHD2,encoded by EGLN1)/hypoxia-inducible factor-1A(HIF-1A)pathway is a key regulator of hypoxic response.Thus,we aim to assess the associations of single polymorphisms(SNPs)of the EGLN1 gene and hypoxic response in CCHD.A missense variant of EGLN1 c.380G>C(rs1209790)was found in 46 patients(46/126),with lower hypoxia incidence and higher rate of collateral vessel formation,compared with the wild type(P<0.05).In vitro experiments,during hypoxia,EGLN1 mutation reduced EGLN1 expression compared with the wild type,with higher HIF-1A,VEGF and EPO expression levels in the mutant.No difference in HK1 expression was observed between the mutant and wild type.CCHD patients with c.380G>C showed improved response to hypoxia compared with the wild-type counterparts.The EGLN1 c.380G>C mutation improves hypoxic response through the PHD2/HIF-1A pathway,which may provide a molecular mechanism for hypoxic response in CCHD.The effects of the EGLN1 c.380G>C mutation on CCHD prognosis deserve further investigation. | Yuanlin Zhou Na Ouyang Lingjuan Liu Jie Tian Xupei Huang Tiewei Lu | 2019 | Genes & Diseases2019,6,1: | 7 |

| 3 | Genetic analysis and fine mapping of an enclosed panicle mutant esp2 in rice (Oryza sativa L.)显示文摘The phenomenon of panicle enclosure in rice is mainly caused by the shortening of uppermost internode.Elucidating the molecular mechanism of panicle enclosure will be helpful for solving the problem of panicle enclosure in male sterile lines and creating new germplasms in rice.We acquired a monogenic recessive enclosed panicle mutant,named as esp2 (enclosed shorter panicle 2),from the tissue culture progeny of indica rice cultivar Minghui-86.In the mutant,panicles were entirely enclosed by flag leaf sheaths and the uppermost internode was almost completely degenerated,but the other internodes did not have obvious changes in length.Genetic analysis indicated that the mutant phenotype was controlled by a recessive gene,which could be steadily inherited and was not affected by genetic background.Apparently,ESP2 is a key gene for the development of uppermost internode in rice.Using an F 2 population of a cross between esp2 and a japonica rice cultivar Xiushui-13 as well as SSR and InDel markers,we fine mapped ESP2 to a 14-kb region on the end of the short arm of chromosome 1.According to the rice genome sequence annotation,only one intact gene exists in this region,namely,a putative phosphatidylserine synthase gene.Sequencing analysis on the mutant and the wild type indicated that this gene was inserted by a 5287-bp retrotransposon sequence.Hence,we took this gene as a candidate of ESP2.The results of this study will facilitate the cloning and functional analysis of ESP2 gene. | GUAN HuaZhong DUAN YuanLin LIU HuaQing CHEN ZhiWei ZHUO Ming ZHUANG LiJun QI WenMing PAN RunSen MAO DaMei ZHOU YuanChang WANG Feng WU WeiRen | 2011 | Chinese Science Bulletin2011,56,14: | 6 |
| 4 | Analysis of Review Helpfulness Based on Consumer Perspective显示文摘When consumers make purchase decisions, they generally refer to the reviews generated by other consumers who have already purchased similar products in order to get more information. Online transaction platforms provide a highly convenient channel for consumers to generate and retrieve product reviews. In addition,consumers can also vote reviews perceived to be helpful in making their decision. However, due to diverse characteristics, consumers can have different preferences on products and reviews. Their voting behavior can be influenced by reviews and existing review votes. To explore the influence mechanism of the reviewer, the review,and the existing votes on review helpfulness, we propose three hypotheses based on the consumer perspective and perform statistical tests to verify these hypotheses with real review data from Amazon. Our empirical study indicates that review helpfulness has significant correlation and trend with reviewers, review valance, and review votes. | 5 | Extranodal involvement in young patients with diffuse large B-cell lymphoma: distribution, prognostic value and treatment options显示文摘Objective: Extranodal involvement represents a peculiar presentation of diffuse large B-cell lymphoma(DLBCL). Previous studies have suggested that older patients are more prone to extranodal involvement. This study retrospectively addressed the distribution, prognostic value and treatment options of extranodal involvement in young patients with DLBCL.Methods: A total of 329 patients were enrolled according to the inclusion requirements. The effects of gender,extranodal involvement, age-adjusted international prognostic index(aa IPI), rituximab infusion and radiotherapy on patient outcomes were evaluated.Results: Among these patients, 59% presented extranodal involvement in 16 anatomic sites. More than one instance was linked to many poorer clinical characteristics and poorer survival compared with either nodal disease or one instance. In patients with one extranodal lesion, multivariate analysis revealed that the site of extranodal involvement, but not the aa IPI or rituximab infusion, was independently related to the outcome, and radiotherapy had a negative influence on survival.Conclusions: Extranodal involvement is common in younger patients and exhibits a ubiquitous distribution.The site of extranodal involvement is of strong prognostic significance. | 6 | Genome-wide identification and cold stress-induced expression analysis of the CBF gene family in Liriodendron chinense显示文摘Cold-resistance pathways that operate in model plants such as Arabidopsis thaliana and Oryza sativa have been studied extensively.It has been found that CBF genes play an important role in plant cold resistance.Liriodendron chinense,a tree known for its graceful tree shape and widely spread in south China,has weak cold tolerance.However,little is known about its response to cold.To further study the function of L.chinense CBF gene family,we started by characterizing all members of this gene family in the L.chinense genome and their expression profiling.Phylogenetic analysis found that 14 CBF genes in L.chinense are more closely related to their homologues in woody plants and A.thaliana than those in O.sativa.Cis-acting elements and GO analysis showed that some LcCBF genes participated in the biological process of cold stress response.The transcriptomic and RT-qPCR data showed that most of LcCBF genes displayed an initially increasing and subsequently decreasing trend during cold stress course and the expression profile of each member was different.Some LcCBF genes exhibited a different abundance in callus,root,stem and leaf tissues.The structure and expression characteristics of LcCBF genes imply that they may have similar and different functions in response to cold stress conditions.The identification and analysis of LcCBF gene family have laid the foundation for future studies into L.chinense cold stress mechanisms and for the cultivation of cold-resistance cultivars. | Yuanlin Guan Siqin Liu Weihuang Wu Kaiyue Hong Rongxue Li Liming Zhu Yang Liu Ye Lu Jinhui Chen Liming Yang Jisen Shi | 2021 | Journal of Forestry Research2021,32,6: | 3 |
