1Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
2Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
3Wetting transition energy curves for a droplet on a square-post patterned surface显示文摘Due to the property of water repellence, biomimetic superhydrophobic surfaces have been widely applied to green technologies, in turn inducing wider and deeper investigations on superhydrophobic surfaces. Theoretical, experimental and numerical studies on wetting transitions have been carried out by researchers, but the mechanism of wetting transitions between Cassie-Baxter state and Wenzel state,which is crucial to develop a stable superhydrophobic surface, is still not fully understood. In this paper,the free energy curves based on the transition processes are presented and discussed in detail. The existence of energy barriers with or without consideration of the gravity effect, and the irreversibility of wetting transition are discussed based on the presented energy curves. The energy curves show that different routes of the Cassie-to-Wenzel transition and the reverse transition are the main reason for the irreversibility. 4Takagi-Sugeno fuzzy model identification for turbofan aero-engines with guaranteed stability显示文摘This paper is concerned with identifying a Takagi-Sugeno(TS) fuzzy model for turbofan aero-engines working under the maximum power status(non-afterburning). To establish the fuzzy system, theoretical contributions are made as follows. First, by fixing antecedent parameters, the estimation of consequent parameters in state-space representations is formulated as minimizing a quadratic cost function. Second, to avoid obtaining unstable identified models, a new theorem is proposed to transform the prior-knowledge of stability into constraints. Then based on the aforementioned work, the identification problem is synthesized as a constrained quadratic optimization.By solving the constrained optimization, a TS fuzzy system is identified with guaranteed stability.Finally, the proposed method is applied to the turbofan aero-engine using simulation data generated from an aerothermodynamics component-level model. 5Sorption of chlorophenols onto fruit cuticles and potato periderm显示文摘To better understand the interaction mechanisms of plant surfaces with polar organic compounds,sorption of 4-chlorophenol,2,4dichlorophenol,and 2,4,6-trichlorophenol by fruit cuticles(i.e.,tomato,apple,and pepper),and potato tuber periderm were investigated.The roles of cuticular components(waxes,cutin,cutan and sugar) on sorption of chlorophenols are quantitatively compared.Cutin and waxes govern the sorption capacity of bulk apple cuticle by hydrophobic interactions.Potato periderm with highest sugar content exhibits the lowest sorption capability for the chlorophenols.With the increase of hydrophobicity(i.e.,K ow) of sorbate,the relative contribution of lipophilic components(wax,cutin and cutan) on total sorption increases,however,the ratios of K oc to K ow decreases due to increasing ionization degree of sorbates.Yungui Li Yingqing Deng Baoliang Chen 2012Journal of Environmental Sciences2012,24,4:2

9Shadow geometry maps for alias-free shadows显示文摘Shadow maps sample scene visibility in the light source space and offer an efficient solution to generate hard shadows.However,they suffer from aliasing artifacts because of discretization errors,inadequate resolution and projection distortion.In this paper,we propose the shadow geometry map method,where a shadow depth map is augmented by storing geometry information about scenes.This leads to a new shadowrendering algorithm that combines a supersampling filter,a geometry-aware reconstruction kernel and an irregular sampling filter.Our method produces high quality alias-free and subpixel supersampling shadow rendering and retains the simplicity and the efficiency of shadow maps.We show that the algorithm pipeline is efficiently parallelized using current programmable graphics hardware and that our method is capable of generating high quality hard shadows.WANG Rui WU YingQing PAN MingHao CHEN Wei HUA Wei 2013Science China(Information Sciences)2013,56,6:1

11Compounding effects of human activities and climatic changes on coexistence of oasis-desert ecosystems:Prioritizing resilient decision-making for a riskier world显示文摘Water-salt balance is critical for the stable coexistence of salt-affected and groundwater-fed oasis-desert ecosystems. Yet, a comprehensive investigation of how soil salinization and groundwater degradation threaten the coexistence of oasis-desert ecosystems is still scarce, especially under the compounding effects of human activities and climatic changes. Here, we assessed the impacts of irrigated agriculture on hydrological regimes in oasisdesert systems, investigated the spatio-temporal variations of soil salinization in irrigated cropland, and evaluated the implications of the interplays of soil salinization and groundwater degradation on the coexistence of oasis-desert ecosystems in northwestern China, based on meaningful modelling approaches and comprehensive measurements over 1995–2020. The results showed that the irrigation return flow coefficient decreased sharply from 0.21 ± 0.09 in the traditional irrigation period to 0.09 ± 0.01 in the water-saving irrigation period. The continuous drop in groundwater tables and significant degradation of groundwater quality are occurring throughout this watershed. The eco-environmental flows are reaching to their limit with watershed closures(i.e.,the drainage from the oasis region into the desert region is being weakened or even eliminated), although these progressions were largely hidden by regional precipitation and streamflow variability. The process of salt migration and accumulation across different landscapes in oasis-desert system is being reshaped, and soil salinization in water-saving agricultural irrigated lands is accelerating with a regional average annual growth rate of18%. The vegetation in this watershed is degrading, and anthropogenic disturbance accelerates this trend. Our results highlight that environmental stress adaptation strategies must account for resilience maintenance to avoid accelerating catastrophic transitions in oasis-desert ecosystems. 12Spectroscopic Analysis of the Interaction between Bovine Serum Protein and Lead Ion显示文摘In this paper, the spectral characteristics of the interaction between lead and bovine serum albumin (BSA) were analyzed by fluorescence spectroscopy, ultraviolet spectroscopy and infrared spectroscopy. The effects of pH, bovine serum albumin concentration, lead ion concentration and ionic strength on the spectra of lead ion-bovine serum albumin system were investigated, and the best testing condition is to determine the interaction between lead and BSA by testing the peak changes or displacement in the UV spectrum analysis. Fluorescence spectroscopy showed that lead ion could induce the fluorescence quenching of bovine serum albumin. The influence of lead ion on the secondary structure of protein was analyzed by infrared spectroscopy. 13Targeting a cryptic allosteric site of SIRT6 with small-molecule inhibitors that inhibit the migration of pancreatic cancer cells显示文摘SIRT6 belongs to the conserved NAD^(+)-dependent deacetylase superfamily and mediates multiple biological and pathological processes.Targeting SIRT6 by allosteric modulators represents a novel direction for therapeutics,which can overcome the selectivity problem caused by the structural similarity of orthosteric sites among deacetylases.Here,developing a reversed allosteric strategy Allo Reverse,we identified a cryptic allosteric site,Pocket Z,which was only induced by the bi-directional allosteric signal triggered upon orthosteric binding of NAD^(+).Based on Pocket Z,we discovered an SIRT6 allosteric inhibitor named JYQ-42.JYQ-42 selectively targets SIRT6 among other histone deacetylases and effectively inhibits SIRT6 deacetylation,with an IC50 of 2.33μmol/L.JYQ-42 significantly suppresses SIRT6-mediated cancer cell migration and pro-inflammatory cytokine production.JYQ-42,to our knowledge,is the most potent and selective allosteric SIRT6 inhibitor.This study provides a novel strategy for allosteric drug design and will help in the challenging development of therapeutic agents that can selectively bind SIRT6.Qiufen Zhang Yingyi Chen Duan Ni Zhimin Huang Jiacheng Wei Li Feng Jun-Cheng Su Yingqing Wei Shaobo Ning Xiuyan Yang Mingzhu Zhao Yuran Qiu Kun Song Zhengtian Yu Jianrong Xu Xinyi Li Houwen Lin Shaoyong Lu Jian Zhang 2022Acta Pharmaceutica Sinica B2022,12,2:0
