1Altered regional homogeneity in post-traumatic stress disorder: a resting-state functional magnetic resonance imaging study显示文摘Objective Little is known about the brain systems that contribute to vulnerability to post-traumatic stress disorder (PTSD). Comparison of the resting-state patterns of intrinsic functional synchronization, as measured by functional magnetic resonance imaging (fMRI), between groups with and without PTSD following a traumatic event can help identify the neural mechanisms of the disorder and targets for intervention. Methods Fifty-four PTSD patients and 72 matched traumatized subjects who experienced the 2008 Sichuan earthquake were imaged with blood oxygen level-dependent (BOLD) fMRI and analyzed using the measure of regional homogeneity (ReHo) during the resting state. Results PTSD patients presented enhanced ReHo in the left inferior parietal lobule and right superior frontal gyrus, and reduced ReHo in the right middle temporal gyrus and lingual gyrus, relative to traumatized individuals without PTSD. Conclusion Our findings showed that abnormal brain activity exists under resting conditions in PTSD patients who had been exposed to a major earthquake. Alterations in the local functional connectivity of cortical regions are likely to contribute to the neural mechanisms underlying PTSD.Yan Yin Changfeng Jin Lisa T. 3Aligned fibrin/functionalized self-assembling peptide interpenetrating nanofiber hydrogel presenting multi-cues promotes peripheral nerve functional recovery显示文摘Nerve guidance conduits with hollow lumen fail to regenerate critical-sized peripheral nerve defects(15 mm in rats and 25 mm in humans),which can be improved by a beneficial intraluminal microenvironment.However,individual cues provided by intraluminal filling materials are inadequate to eliminate the functional gap between regenerated nerves and normal nerves.Herein,an aligned fibrin/functionalized self-assembling peptide(AFG/fSAP)interpenetrating nanofiber hydrogel that exerting synergistic topographical and biochemical cues for peripheral nerve regeneration is constructed via electrospinning and molecular self-assembly.The hydrogel possesses an aligned structure,high water content,appropriate mechanical properties and suitable biodegradation capabilities for nerve repair,which enhances the alignment and neurotrophin secretion of primary Schwann cells(SCs)in vitro,and successfully bridges a 15-mm sciatic nerve gap in rats in vivo.The rats transplanted with the AFG/fSAP hydrogel exhibit satisfactory morphological and functional recovery in myelinated nerve fibers and innervated muscles.The motor function recovery facilitated by the AFG/fSAP hydrogel is comparable with that of autografts.Moreover,the AFG/fSAP hydrogel upregulates the regeneration-associated gene expression and activates the PI3K/Akt and MAPK signaling pathways in the regenerated nerve.Altogether,the AFG/fSAP hydrogel represents a promising approach for peripheral nerve repair through an integration of structural guidance and biochemical stimulation.Shuhui Yang Jinjin Zhu Changfeng Lu Yi Chai Zheng Cao Jiaju Lu Zhe Zhang He Zhao Yin-Yuan Huang Shenglian Yao Xiangdong Kong Peixun Zhang Xiumei Wang 2022Bioactive Materials2022,7,2:3
4Rice(Oryza sativa L.)seedlings enriched with zinc or manganese:Their impacts on cadmium accumulation and expression of related genes显示文摘Cadmium(Cd)contamination in paddy soils means that the rice produced there may be unsafe for human consumption.A hydroponic study was conducted to enrich rice seedlings with zinc(Zn)or manganese(Mn),and the uptake and transport characteristics of Cd in these Zn-and Mn-rich seedlings were subsequently investigated using a greenhouse pot trial.The results showed that hydroponic cultivation in 10-50μmol L^(-1) Zn(ZnSO_(4)·7 H_(2)O)or50-250μmol L^(-1) Mn(MnSO_(4)·H_(2)O)for 30 d had no significant impact on rice growth,while the accumulation of Zn and Mn was 7.31-18.5 and 25.4-47.7 times higher,respectively,than in the control(no Zn or Mn addition).The accumulation of Cd in the Zn-and Mn-rich rice plants was 26.3%-38.6% and 34.4%-44.5% lower than that in the control,respectively,and the translocation factors of Cd from roots to shoots also decreased by 23.3%-41.3% and 25.3%-37.0%,respectively,after transplanting to Cd-contaminated soils.Furthermore,the relative expression levels of OsIRT1(Oryza sativa iron-regulated transporter 1)were downregulated by 40.1%-59.3% and 16.0%-25.9%,respectively,in the Zn-and Mn-rich seedling roots.This downregulation may indicate a possible mechanism contributing to the reductions in Cd absorption.Field experiments confirmed that the Zn-and Mn-rich seedlings produced brown rice(unpolished rice grains)with significantly decreased concentrations of Cd(34.2%-44.4%).This study provides an innovative method for reducing the food safety risks from rice grown on slightly to moderately Cd-contaminated paddy soils.Gaoxiang HUANG Changfeng DING Yibing MA Yurong WANG Zhigao ZHOU Shun'an ZHENG Xingxiang WANG 2021Pedosphere2021,31,6:2

9Dietary zero-dimensional fullerene supplementation improves the meat quality,lipid metabolism,muscle fiber characteristics,and antioxidative status in finishing pigs显示文摘With the increasing demand for high-quality pork,more nutritional substances have been studied for the regulation of meat quality.Zero-dimensional fullerenes(C60)can modulate the biological behavior of a variety of cell lines and animals.In this study,we report the biological effects of C60 on finishing pigs at different concentrations.A total of 24 barrows(Duroc×Large White×Landrace),with an average body weight of 21.01±0.98 kg,were divided into 3 groups and each treated daily with C60(100 or 200 mg per kg feed)or a control diet until the end of the experiment.Our results showed that dietary C60 supplementation improved flesh color,marbling scores,and flavor amino acid contents of longissimus dorsi(LD)of growing-finishing pigs(P<0.05).C60 improved meat quality by regulating lipid metabolism and muscle fiber morphology by mediating the expression of genes,L-lactic dehydrogenase(LDH),myosin heavy chain(MyHC)IIa,MyHCIIb,peroxisome proliferator-activated receptor γ(PPARγ),and fatty acid transport protein 1(FATP1)(P<0.05).Moreover,C60 substantially promoted the mRNA expression of antioxidant enzyme genes(P<0.05),which also contributed to improving meat quality.These findings have important implications for the application of C60 in the livestock industry,especially for improving the meat quality of fattening pigs.Simeng Liao Guang Liu Bie Tan Ming Qi Xin Wu Jianjun Li Xiaoqing Li Changfeng Zhu Jiamei Huang Shuo Zhang Yulong Tang Yulong Yin 2022Animal Nutrition2022,,4:0

11Development and Characterization of Nanobody-Derived CD47 Theranostic Pairs in Solid Tumors显示文摘Overexpression of CD47 is frequently observed in various types of human malignancies,inhibiting myeloidmediated elimination of tumor cells and affecting the prognosis of cancer patients.By mapping biomarker expression,immuno-positron emission tomography has been increasingly used for patient screening and response monitoring.By immunization alpacas with recombinant human CD47,we prepared a CD47-targeting nanobody C2 and developed[^(68)Ga]Ga-NOTA-C2,followed by an exploration of the diagnostic value in CD47-expressing tumor models including gastric-cancer patient-derived xenograft models.By fusing C2 to an albumin binding domain(ABD),we synthesized ABDC2,which had increased in vivo half-life and improved targeting properties.We further labeled ABDC2 with^(68)Ga/^(89)Zr/177Lu to develop radionuclide theranostic pairs and evaluated the pharmacokinetics and theranostic efficacies of the agents in cell-and patient-derived models.Both C2 and ABDC2 specifically reacted with human CD47 with a high KD value of 23.50 and 84.57 pM,respectively.[^(68)Ga]Ga-NOTA-C2 was developed with high radiochemical purity(99>%,n=4)and visualized CD47 expression in the tumors.In comparison to the rapid renal clearance and short half-life of[^(68)Ga]Ga-NOTA-C2,both[^(68)Ga]Ga-NOTA-ABDC2 and[^(89)Zr]Zr-DFOABDC2 showed prolonged circulation and increased tumor uptake,with the highest uptake of[^(89)Zr]Zr-DFO-ABDC2 occurring at 72 h post-injection.Moreover,[177Lu]Lu-DOTA-ABDC2 radioimmunotherapy suppressed the tumor growth but was associated with toxicity,warranting further optimization of the treatment schedules.Taken together,we reported a series of nanobody-derived CD47-targeted agents,of which[^(68)Ga]Ga-NOTA-C2 and[^(89)Zr]Zr-DFO-ABDC2 are readily translatable.Optimization and translation of CD47-targeted theranostic pair may provide new prospects for CD47-targeted management of solid tumors.You Zhang Di Zhang Shuxian An Qiufang Liu Chenyi Liang Juan Li Ping Liu Changfeng Wu Gang Huang Weijun Wei Jianjun Liu 2023Research2023,,4:0
12A review:crystalline silicon membranes over sealed cavities for pressure sensors by using silicon migration technology显示文摘A silicon pressure sensor is one of the very first MEMS components appearing in the microsystem area.The market for the MEMS pressure sensor is rapidly growing due to consumer electronic applications in recent years. Requirements of the pressure sensors with low cost, low power consumption and high accuracy drive one to develop a novel technology. This paper first overviews the historical development of the absolute pressure sensor briefly. It then reviews the state of the art technology for fabricating crystalline silicon membranes over sealed cavities by using the silicon migration technology in detail. By using only one lithographic step, the membranes defined in lateral and vertical dimensions can be realized by the technology.
