High-yield preparation of robust gold nanoshells on silica nanorattles with good biocompatiblity显示文摘Although gold nanoshells are widely considered as one of the promising photothermal nanomaterials used for biomedicine, the high cost, low yield and poor stability severely limit their potential application in clinical trials.Herein, robust gold nanoshells on silica nanorattles(GSNs)were easily prepared in a high yield by an improved seedmediated method employing polyvinylpyrrolidone(PVP) as a stabilizing and capping agent. The present method is very simple, effective and reproducible and can well control the growth process of gold nanoshells. The as-prepared GSNs have a narrow size distribution(<10 % in standard deviation). Furthermore, the utilization rate of Au in the solution used for the growth of gold nanoshells increases by 70 %than that in previous method. The resultant GSNs have a good structural stability after placing over 6 months due to the protection of PVP. More importantly, in vivo and in vitro toxic studies indicate that the GSNs have good biocompatibility. We believe that our preparation method will remarkably promote the use of gold nanoshells for biomedicine. | Changhui Fu Chaofeng He Longfei Tan Shunhao Wang Lu Shang Linlin Li Xianwei Meng Huiyu Liu | 2016 | Science Bulletin2016,61,4: | 5
Regeneration and identification of interspecific asymmetric somatic hybrids obtained by donor-recipient fusion in cotton显示文摘Asymmetric hybrids between Gossypium hirsutum (YZ-1) and G. davidsonii were obtained by donor-recipient fusion. YZ-1 was considered the recipient and was pretreated with iodoacetamide (IOA), while G. davidsonii was considered the donor and was irradiated with ultraviolet (UV) before fusion. YZ-1 protoplasts stopped growth when treated with 0.5 mmol/L IOA for 20 min, and G. davidsonii protoplasts stopped growth when irradiated with 38.7 J/cm2 UV for 30 s. Asymmetric somatic hybrids were obtained by electrofusion between the separately treated protoplasts of the 2 species. The regenerated plants were identified by morphological, cytological, and molecular analysis. Most regenerated plants derived from fused protoplasts displayed new morphology; some were intermediate between the two parents and a few displayed recipient-like morphology. Chromosome numbers in these somatic hybrids mostly ranged from 40 to 73. The hybridity was confirmed by random amplified polymorphic DNA and simple sequence repeat analysis. Organelle DNA inheritance of the YZ-1 and G. davidsonii somatic hybrid was investigated by cleaved amplified polymorphism sequence and chloroplast simple sequence repeat analysis, which indicated that recombination and rearrangements might have occurred in some regions of mitochondrial and chloroplastic DNA. This is the first report of completely asymmetric hybrid production via donor–recipient fusion between G. hirsutum and G. davidsonii, which is a novel case in hybrid production following the symmetric fusion and asymmetric fusion based on UV irradiation in cotton. | FU LiLi YANG XiYan ZHANG XianLong WANG ZhiWei FENG ChangHui LIU ChuanXiang JIANG Pei-Yong ZHANG JinLong | 2009 | Chinese Science Bulletin2009,54,17: | 3
Enhanced antibacterial activity of silica nanorattles with ZnO combination nanoparticles against methicillin-resistant Staphylococcus aureus显示文摘Silica nanorattles(SNs) with zinc oxide(ZnO) combination nanoparticles are reported to inhibit methicillin-resistant Staphylococcus aureus(MRSA) for the first time. SNs loaded with ZnO nanoparticles, which can produce free radicals, can cause severe damage to bacteria. ZnO nanoparticles not only provide free radicals in the combined nanostructures, which can inhibit the growth of bacteria, but also form nanorough surfaces with an irregular distribution of spikes on the SNs, which can enhance their adhesion to bacteria. Nanorough silica shell surfaces maintain the high activity and stability of small-sized ZnO nanoparticles and gather ZnO nanoparticles together to enhance production, which improves the efficiency of free radicals against the cytomembranes of bacterial cells. The enhanced adhesion of ZnO@SN nanoparticles to MRSA cells shortens the effective touching distance between free radicals and MRSA, which also improves antibacterial activity. As we expected, the ZnO@SN nanoparticles exhibit a better antibacterial effect than free ZnO nanoparticles against MRSA in vitro and in vivo. We also demonstrate that SNs loaded with ZnO nanoparticles can accelerate wound healing in MRSA skin inflammation models. This method of multilevel functionalization will be potentially applicable to the antibacterial field. | Qianqian Chai Qiong wu Tianlong Liu Longfei Tan Changhui Fu Xiangling Ren Yue Yang Xianwei Meng | 2017 | Science Bulletin2017,62,17: | 1
Luminescent silver nanoclusters for efficient detection of adenosine triphosphate in a wide range of pH values显示文摘In this work, polymethacrylic acid(PMAA)-templated silver nanoclusters(Ag NCs)were developed as the fluorescent probe for the efficient and sensitive detection of adenosine triphosphate(ATP)in a wide range of pH values. The fluorescence intensity of the Ag NCs could keep stable with pH values ranging from2.5 to 9.3.The detection of ATP was based on the quenching of the fluorescent Ag NCs in the presence of ATP.The fluorescence quenching of the Ag NCs with increasing ATP concentration was studied at pH 2.5,4.5,7.0 and 8.5 which involved a wide pH environment in body fluids. The limit of detection(LOD)for ATP was as low as 0.1 mmol/L in an acidic environment with pH of 2.5 and all the linear correlation coefficients were satisfactory under wide-span pH values from 2.5 to 8.5.In addition, the sensitive determination of ATP was also achieved by adding copper ions(Cu^2+).The high selectivity and rapid detection process proved that the fluorescent probe had great potential to detect ATP in biological samples under different pH conditions. | Meijia Liu Xiangling Ren Xin Liu Longfei Tan Hui Li Jing Wei Changhui Fu Qiong Wu Jun Ren Hongbo Li Xianwei Meng | 2020 | Chinese Chemical Letters2020,31,12: | 1
Nanoscale metal organic frameworks inhibition of pyruvate kinase of M2显示文摘Tumor cells usually show abnormally high glycolysis rate to maintain the dynamic balance of energy. The growth of tumor cells can be affected by inhibiting the activity of pyruvate kinase(especially M2-type isozyme,PKM2),the rate limiting enzyme of glycolysis. This is helpful to the treatment of tumor.Herein, metal organic frameworks(MOFs) were found to inhibit the activity of PKM2.Nanoscale ZIF-8 was synthesized by standing and ultrasonic method, respectively. The ZIF-8 has the performance of inhibiting PKM2.Further research showed that the inhibition ability was attributed to zinc ion in ZIF-8.Interestingly,the IC_(50) of ZIF-8 on PKM2 was one percent of that of zinc ion. This novel enzyme inhibitor is expected to be used in cancer therapy. | Xiangling Ren Xinyuan Huang Qiong Wu Longfei Tan Changhui Fu Yi Chen Xianwei Meng | 2021 | Chinese Chemical Letters2021,32,10: | 0
Zirconium metal-organic framework nanocrystal as microwave sensitizer for enhancement of tumor therapy显示文摘Although surgical resection and chemotherapy were widely applied in tumor therapy, the dysfunction of normal cells resulted in the side effects (such as anorexia, nausea). MW thermal therapy is a non-invasive anticancer strategy under the help of MW sensitizer, with the safety and higher efficacy. Zirconium metal-organic framework nanocubes (ZrMOF NCs) modified with polyethylene glycol, were prepared via one pot method and carbodiimide technique, resulting in their large specific surface area and porosity. Our results showed that non-ion-loaded ZrMOF NCs in 0.9% NaCl solution exhibited better heating effect, higher than that in pure water, due to the robust collision among the ions under MW irradiation. The in vivo experiments confirmed that ZrMOF-PEG NCs + MW group exhibited the higher temperature in the tumor region than that of only MW treatment, suggesting a better MW thermal therapeutic anticancer efficacy. This work provides a new preparation strategy of biosafety nanomaterial as MW sensitizer for enhancing MW thermal anticancer therapy. | Yuanwen Zou Wei Zhang Hongqiao Zhou Changhui Fu Longfei Tan Zhongbing Huang Xiangling Ren Jun Ren Xudong Chen Xianwei Meng | 2019 | Chinese Chemical Letters2019,30,2: | 0
Chemical chaperone delivered nanoscale metal–organic frameworks as inhibitor of endoplasmic reticulum for enhanced sensitization of thermo-chemo therapy显示文摘Thermotherapy and chemotherapy have received extensive attention to tumor treatment. However, thermal tolerance and drug resistance severely limit clinical effect of tumor therapy owing to endoplasmic reticulum(ER) stress. Reducing thermal tolerance and drug resistance of tumors is an urgent challenge to be solved. In this work, we design a nanoplatform of PBA-Dtxl@MIL-101 as an ER inhibitor. Amino functionalized Fe-metal organic framework(MIL-101) nanoparticles are synthesized as p H and microwave(MW) dual stimuli-responsive drug delivery system. Then, the chemical chaperones of 4-phenylbutyric acid(PBA) and antineoplastic drug Docetaxel(Dtxl) were successfully loaded into MIL-101 nanoparticles to form PBA-Dtxl@MIL-101 nanoparticles. Furthermore, PBA-Dtxl@MIL-101 nanoparticles exhibit inhibitor effect of ER stress through upregulating caspase 9 proteins and reduce thermal tolerance by downregulating HSP 90. It was demonstrated that the therapy sensitized by PBA-Dtxl@MIL-101 nanoparticles obviously destroyed tumor cells, showing simultaneously enhanced thermo-chemo therapy. | Xiaoyan Ma Qiong Wu Longfei Tan Changhui Fu Xiangling Ren Qijun Du Lufeng Chen Xianwei Meng | 2022 | Chinese Chemical Letters2022,33,3: | 0
Free-walking:Pedestrian inertial navigation based on dual foot-mounted IMU显示文摘The inertial navigation system(INS),which is frequently used in emergency rescue operations and other situations, has the benefits of not relying on infrastructure, high positioning frequency, and strong real-time performance. However, the intricate and unpredictable pedestrian motion patterns lead the INS localization error to significantly diverge with time. This paper aims to enhance the accuracy of zero-velocity interval(ZVI)detection and reduce the heading and altitude drift of foot-mounted INS via deep learning and equation constraint of dual feet.Aiming at the observational noise problem of low-cost inertial sensors,we utilize a denoising autoencoder to automatically eliminate the inherent noise.Aiming at the problem that inaccurate detection of the ZVI detection results in obvious displacement error,we propose a sample-level ZVI detection algorithm based on the U-Net neural network, which effectively solves the problem of mislabeling caused by sliding windows.Aiming at the problem that Zero-Velocity Update(ZUPT)cannot suppress heading and altitude error,we propose a bipedal INS method based on the equation constraint and ellipsoid constraint, which uses foot-to-foot distance as a new observation to correct heading and altitude error. We conduct extensive and well-designed experiments to evaluate the performance of the proposed method. The experimental results indicate that the position error of our proposed method did not exceed 0.83% of the total traveled distance. | Qu Wang Meixia Fu Jianquan Wang Lei Sun Rong Huang Xianda Li Zhuqing Jiang Yan Huang Changhui Jiang | 2024 | Defence Technology(防务技术)2024,33,3: | 0
Acute toxicity and oxidative damage induced by silica nanorattle in vivo显示文摘Hollow mesoporous nanomaterial is a kind of promising new drug delivery system due to their unique hollow structures. In order to evaluate the toxicity of silica nanorattle (SN) particles in vivo,40 female mice were used in this study to investigate the acute toxicity and oxidative damage.Mice were intravenously injected with SN suspension in sterile 5% glucose at 40,80 and 240 mg/kg,respectively.The control group was administrated with equal-volume 5% glucose.Weight, feed intake, hematology analysis, blood biochemical assay and histopathology diagnosis were examined. The activities of SOD,GSH and CAT were measured as well. The results demonstrated that the levels of ALT and AST in the mice treated with 240 mg/kg SN increased significantly as compared with the control group (P<0.05),whereas the contents of BUN and CREA changed unremarkably. The activity of SOD induced by SN in the liver decreased significantly (P<0.05).In summary, this study revealed that liver was the target organ of the SN.It also can be concluded that activity of SOD played an important role in liver injury caused by SN. | FU ChangHui LIU TianLong TANG FangQiong CHEN Dong LI LinLin LIU HuiYu LI XiaoMin | 2012 | Chinese Science Bulletin2012,57,20: | 0
Toxicity and bio-distribution of carbon dots after single inhalation exposure in vivo显示文摘Because of the advantages of excellent light stability, carbon dots(CDs) are considered to be a promising agent in the bio-marker application. Nevertheless, there are many unresolved issues with the toxicity of CDs in vitro and in vivo. In the study, CDs were synthesized by citric acid and ethylenediamine into deionized water, then the inhalation toxicity and bio-distribution of CDs in vivo were systematically assessed. The results showed that CDs caused animals death at higher dosages and induced injury in the lung and liver including inflammation and necrosis after single inhalation exposure at 5, 2 and 1 mg/kg dosages of the CDs. We also found that the injury increase with a dose-dependent and time-dependent manner. Fluorescent examination and TEM results showed that CDs mainly located at the lung and liver. And the fluorescent intensity increase with a time-dependent manner. This study provides a theoretical basis of the respiratory toxicity of CDs, and provides a basis for the use of CDs as a bio-marker. | Yue Yang Xiangling Ren Zhenning Sun Changhui Fu Tianlong Liu Xianwei Meng Zili Wang | 2018 | Chinese Chemical Letters2018,29,6: | 0
