1Cartilage oligomeric matrix protein is an endogenous β-arrestin-2-selective allosteric modulator of AT1 receptor counteracting vascular injury显示文摘Compelling evidence has revealed that biased activation of G protein-coupled receptor(GPCR)signaling,including angiotensin II(AngII)receptor type 1(AT1)signaling,plays pivotal roles in vascular homeostasis and injury,but whether a clinically relevant endogenous biased antagonism of AT1 signaling exists under physiological and pathophysiological conditions has not been clearly elucidated.Here,we show that an extracellular matrix protein,cartilage oligomeric matrix protein(COMP),acts as an endogenous allosteric biased modulator of the AT1 receptor and its deficiency is clinically associated with abdominal aortic aneurysm(AAA)development.COMP directly interacts with the extracellular N-terminus of the AT1 via its EGF domain and inhibits AT1-β-arrestin-2 signaling,but not Gq or Gi signaling,in a selective manner through allosteric regulation of AT1 intracellular conformational states.COMP deficiency results in activation of AT1a-β-arrestin-2 signaling and subsequent exclusive AAA formation in response to AngII infusion.AAAs in COMP–/–or ApoE–/–mice are rescued by AT1a orβ-arrestin-2 deficiency,or the application of a peptidomimetic mimicking the AT1-binding motif of COMP.Explorations of the endogenous biased antagonism of AT1 receptor or other GPCRs may reveal novel therapeutic strategies for cardiovascular diseases.Yi Fu Yaqian Huang Zhao Yang Yufei Chen Jingang Zheng Chenfeng Mao Zhiqing Li Zhixin Liu Bing Yu Tuoyi Li Meili Wang Chanjuan Xu Yiwei Zhou Guizhen Zhao Yiting Jia Wei Guo Xin Jia Tao Zhang Li Li Ziyi Liu Shengchao Guo Mingliang Ma Heng Zhang Bo Liu Junbao Du Wengong Wang Chaoshu Tang Pei Gao Qingbo Xu Xian Wang Jianfeng Liu Jinpeng Sun Wei Kong 2021Cell Research2021,31,7:2
2A non-destructive method to measure the thermal properties of frozen soils during phase transition显示文摘Frozen soils cover about 40%of the land surface on the earth and are responsible for the global energy balances affecting the climate.Measurement of the thermal properties of frozen soils during phase transition is important for analyzing the thermal transport process.Due to the involvement of phase transition,the thermal properties of frozen soils are rather complex.This paper introduces the uses of a multifunctional instrument that integrates time domain re fl ectometry(TDR)sensor and thermal pulse technology(TPT)to measure the thermal properties of soil during phase transition.With this method,the extent of phase transition(freezing/thawing)was measured with the TDR module;and the corresponding thermal properties were measured with the TPT module.Therefore,the variation of thermal properties with the extent of freezing/thawing can be obtained.Wet soils were used to demonstrate the performance of this measurement method.The performance of individual modules was fi rst validated with designed experiments.The new sensor was then used to monitor the properties of soils during freezing e thawing process,from which the freezing/thawing degree and thermal properties were simultaneously measured.The results are consistent with documented trends of thermal properties variations.Bin Zhang Chanjuan Han Xiong(Bill) Yu 2015Journal of Rock Mechanics and Geotechnical Engineering2015,7,2:2
3A Safe and Reliable Routing Mechanism of LEO Satellite Based on SDN显示文摘Satellite networks have high requirements for security and data processing speed.In order to improve the reliability of the network,software-defined network(SDN)technology is introduced and a central controller is set in the network.Due to the characteristics of global perspective,control data separation,and centralized control of SDN,the idea of SDN is introduced to the design of the satellite network model.As a result,satellite nodes are only responsible for data transmission,while the maintenance of the links and the calculation of routes are implemented by the controller.For the massive LEO satellite network based on SDN,a state evaluation decision routing mechanism is proposed.The designed mechanism monitors the status of the entire network effectively and reduces the on-board load on the satellite network.The best routing decision is made under the comprehensive consideration of the current and historical status of each inter-satellite link between Low Earth Orbit(LEO)satellite network nodes.The calculation and storage requirements are controlled within a reasonable range.Based on the curve parameter transmission fuzzy encryption algorithm,a safe and reliable condition assessment decision routing mechanism(CADRM)is designed.It ensures that the personal information of the LEO satellite network can be transmitted safely and effectively.The experimental simulation results show the improvement of network throughput,the reduction of packet loss rate and the enhancing of network reliability.Chao Guo Juan Guo Chanjuan Yu Zhaobin Li Cheng Gong Abdul Waheed 2020Computers, Materials & Continua2020,,7:1

7Bioprosthetic Valve Size Selection to Optimize Aortic Valve ReplacementSurgical Outcome: A Fluid-Structure Interaction Modeling Study显示文摘Aortic valve replacement(AVR)remains a major treatment option for patients with severe aortic valve disease.Clinical outcome of AVR is strongly dependent on implanted prosthetic valve size.Fluid-structure interaction(FSI)aortic root models were constructed to investigate the effect of valve size on hemodynamics of the implanted bioprosthetic valve and optimize the outcome of AVR surgery.FSI models with 4 sizes of bioprosthetic valves(19(No.19),21(No.21),23(No.23)and 25 mm(No.25))were constructed.Left ventricle outflow track flow data from one patient was collected and used as model flow conditions.Anisotropic Mooney–Rivlin models were used to describe mechanical properties of aortic valve leaflets.Blood flow pressure,velocity,systolic valve orifice pressure gradient(SVOPG),systolic cross-valve pressure difference(SCVPD),geometric orifice area,and flow shear stresses from the four valve models were compared.Our results indicated that larger valves led to lower transvalvular pressure gradient,which is linked to better post AVR outcome.Peak SVOPG,mean SCVPD and maximum velocity for Valve No.25 were 48.17%,49.3%,and 44.60%lower than that from Valve No.19,respectively.Geometric orifice area from Valve No.25 was 52.03%higher than that from Valve No.19(1.87 cm2 vs.1.23 cm2).Implantation of larger valves can significantly reduce mean flow shear stress on valve leaflets.Our initial results suggested that larger valve size may lead to improved hemodynamic performance and valve cardiac function post AVR.More patient studies are needed to validate our findings.Caili Li Dalin Tang Jing Yao Christopher Baird Haoliang Sun Chanjuan Gong Luyao Ma Yanjuan Zhang Liang Wang Han Yu Chun Yang Yongfeng Shao 2021Computer Modeling in Engineering & Sciences2021,,4:1

9Corydalis Rhizoma as a model for herb-derived trace metabolites exploration:A cross-mapping strategy involving multiple doses and samples显示文摘Deciphering the metabolites of multiple components in herbal medicine has far-reaching significance for revealing pharmacodynamic ingredients.However,most chemical components of herbal medicine are secondary metabolites with low content whose in vivo metabolites are close to trace amounts,making it difficult to achieve comprehensive detection and identification.In this paper,an efficient strategy was proposed:herb-derived metabolites were predicted according to the structural characteristics and metabolic reactions of chemical constituents in Corydalis Rhizoma and chemical structure screening tables for metabolites were conducted.The fragmentation patterns were summarized from representative standards combining with specific cleavage behaviors to deduce structures of metabolites.Ion abundance plays an important role in compound identification,and high ion abundance can improve identification accuracy.The types of metabolites in different biological samples were very similar,but their ion abundance might be different.Therefore,for trace metabolites in biological samples,we used the following two methods to process:metabolites of high dose herbal extract were analyzed to characterize those of clinical dose herbal extracts in the same biological samples;cross-mapping of different biological samples was applied to identify trace metabolites based on the fact that a metabolite has different ion abundance in different biological samples.Compared with not using this strategy,44 more metabolites of clinical dose herbal extract were detected.This study improved the depth,breadth,and accuracy of current methods for herb-derived metabolites characterization.Chanjuan Yu Fengyun Wang Xinyue Liu Jiayan Miao Siqi Tang Qin Jiang Xudong Tang Xiaoyan Gao 2021Journal of Pharmaceutical Analysis2021,11,3:1
10Neuro-11:a new questionnaire for the assessment of somatic symptom disorder in general hospitals显示文摘Background Somatic symptom disorder(SSD)commonly presents in general hospital settings,posing challenges for healthcare professionals lacking specialised psychiatric training.The Neuro-11 Neurosis Scale(Neuro-11)offers promise in screening and evaluating psychosomatic symptoms,comprising 11 concise items across three dimensions:somatic symptoms,negative emotions and adverse events.Prior research has validated the scale’s reliability,validity and theoretical framework in somatoform disorders,indicating its potential as a valuable tool for SSD screening in general hospitals.Aims This study aimed to establish the reliability,validity and threshold of the Neuro-11 by comparing it with standard questionnaires commonly used in general hospitals for assessing SSD.Through this comparative analysis,we aimed to validate the effectiveness and precision of the Neuro-11,enhancing its utility in clinical settings.Methods Between November 2020 and December 2021,data were collected from 731 patients receiving outpatient and inpatient care at Shenzhen People’s Hospital in China for various physical discomforts.The patients completed multiple questionnaires,including the Neuro-11,Short Form 36 Health Survey,Patient Health Questionnaire 15 items,Hamilton Anxiety Scale and Hamilton Depression Scale.Psychiatry-trained clinicians conducted structured interviews and clinical examinations to establish a gold standard diagnosis of SSD.Results The Neuro-11 demonstrated strong content reliability and structural consistency,correlating significantly with internationally recognised and widely used questionnaires.Despite its brevity,the Neuro-11 exhibited significant correlations with other questionnaires.A test-retest analysis yielded a correlation coefficient of 1.00,Spearman-Brown coefficient of 0.64 and Cronbach’sαcoefficient of 0.72,indicating robust content reliability and internal consistency.Confirmatory factor analysis confirmed the validity of the three-dimensional structure(p<0.001,comparative fit index=0.94,Tucker-Lewis index=0.92,root mean square error of approximation=0.06,standardised root mean square residual=0.04).The threshold of the Neuro-11 is set at 10 points based on the maximum Youden’s index from the receiver operating characteristic curve analysis.In terms of diagnostic efficacy,the Neuro-11 has an area under the curve of 0.67.Silin Zeng Yian Yu Shan Lu Sirui Zhang Xiaolin Su Ge Dang Ying Liu Zhili Cai Siyan Chen Yitao He Xin Jiang Chanjuan Chen Lei Yuan Peng Xie Jianqing Shi Qingshan Geng Rafael H Llinas Yi Guo 2023General Psychiatry2023,36,4:0
