2The Serum Exosome Derived MicroRNA-135a,-193b, and-384 Were Potential Alzheimer's Disease Biomarkers显示文摘Objective MicroR NAs(mi Rs) are attractive molecules to be considered as one of the blood-based biomarkers for neurodegenerative disorders such as Alzheimer's disease(AD). The goal of this study was to explore their potential value as biomarkers for the diagnosis of AD. Methods The expression levels of exosomal miR-135 a,-193 b, and-384 in the serum from mild cognitive impairment(MCI), dementia of Alzheimer-type(DAT), Parkinson's disease with dementia(PDD), and vascular dementia(VaD) patients were measured with a real-time quantitative reverse transcriptase PCR(qR T-PCR) method. Results Both serum exosome mi R-135 a and miR-384 were up-regulated while miR-193 b was down-regulated in serum of AD patients compared with that of normal controls. Exosome miR-384 was the best among the three miR s to discriminate AD, VaD, and PDD. Using the cut-off value could better interpret these laboratory test results than reference intervals in the AD diagnosis. ROC curve showed that the combination of mi R-135 a,-193 b, and-384 was proved to be better than a particular one for early AD diagnosis. 3LA-ICP-MS zircon U-Pb dating from granitoids in southern basement of Songliao basin:Constraints on ages of the basin basement显示文摘Seven LA-ICP-MS zircon U-Pb datings from granitoids in the southern basement of the Songliao basin were done in order to constrain the ages of the basin basement. The cathodoluminescence (CL) images of the zircons from seven granitoids indicate that they are euhedral-subhedral ones with striped ab-sorption and obvious oscillatory zoning rims. The dating results show that a weighted mean 206Pb/238U age is 236±3 Ma for quartz diorite (sample No.T6-1) located in the western slope of the basin,that weighted mean 206Pb/238U ages are 319±1 Ma (2126 m) and 361±2 Ma (1994 m) for diorite (sample No.YC1-1) and granite (sample No.YC1-2) located in northern part of southeastern uplift of the basin,respectively,and that weighted mean 206Pb/238U ages are 161±5 Ma,165±2 Ma,165±1 Ma and 161±4 Ma for samples Q2-1,SN121,SN122,and SN72 granitoids located in southern part of southeastern uplift of the basin,respectively. The statistical results of ages suggest that the middle Jurassic granitoids con-stitute the main part of basement granitoids,and that the Hercynian and Indo-Sino magmatisms also occur in the basin basement. 6Effects of plant species coexistence on soil enzyme activities and soil microbial community structure under Cd and Pb combined pollution显示文摘The relationship between plant species coexistence and soil microbial communities under heavy metal pollution has attracted much attention in ecology.However,whether plant species coexistence could offset the impacts of heavy metal combined pollution on soil microbial community structure and soil enzymes activities is not well studied.The modified ecological dose model and PCR-RAPD method were used to assess the effects of two plant species coexistence on soil microbial community and enzymes activities subjected to Cd and Pb combined stress.The results indicated that monoculture and mixed culture would increased microbe populations under Cd and Pb combined stress,and the order of sensitivity of microbial community responding to heavy metal stress was:actinomycetes > bacteria > fungi.The respirations were significantly higher in planted soil than that in unplanted soil.The plant species coexistence could enhance soil enzyme activities under Cd and Pb combined.Furthermore,planted soil would be helpful to enhance soil genetic polymorphisms,but Cd and Pb pollution would cause a decrease on soil genetic polymorphisms.Mixed culture would increase the ecological dose 50%(ED50) values,and the ED50 values for soil enzyme activities decreased with increasing culture time.The dehydrogenase was most sensitive to metal addition and easily loses activity under low dose of heavy metal.However,it was difficult to fully inhibit the phoshpatase activity,and urease responded similarly with phosphatase.Yang Gao Pei Zhou Liang Mao Yueer Zhi Chunhua Zhang Wanjun Shi 2010Journal of Environmental Sciences2010,22,7:19
7Bone biomaterials and interactions with stem cells显示文摘Bone biomaterials play a vital role in bone repair by providing the necessary substrate for cell adhesion,proliferation, and differentiation and by modulating cell activity and function. In past decades, extensive efforts have been devoted to developing bone biomaterials with a focus on the following issues:(1)developing ideal biomaterials with a combination of suitable biological and mechanical properties;(2)constructing a cell microenvironment with pores ranging in size from nanoscale to submicro-and microscale;and(3) inducing the oriented differentiation of stem cells for artificial-to-biological transformation. Here we present a comprehensive review of the state of the art of bone biomaterials and their interactions with stem cells. Typical bone biomaterials that have been developed, including bioactive ceramics, biodegradable polymers, and biodegradable metals, are reviewed, with an emphasis on their characteristics and applications. The necessary porous structure of bone biomaterials for the cell microenvironment is discussed,along with the corresponding fabrication methods. Additionally, the promising seed stem cells for bone repair are summarized, and their interaction mechanisms with bone biomaterials are discussed in detail.Special attention has been paid to the signaling pathways involved in the focal adhesion and osteogenic differentiation of stem cells on bone biomaterials. 8Ex vivo-expanded bone marrow stem cells home to the liver and ameliorate functional recovery in a mouse model of acute hepatic injury显示文摘BACKGROUND:Stem cell transplantation provides a theoretical approach for liver regeneration medicine;it may promote liver regeneration and self-repair.However,the transplantation of bone marrow-mesenchymal stem cells expanded ex vivo as a therapy for liver disease has rarely been investigated.This study aimed to explore whether bone marrow stem cells expanded ex vivo home to the liver and foster hepatic recovery after CCl 4 injury.METHODS:Bone marrow cells from BALB/c mice were expanded ex vivo by multiple-passage cultivation,characterized by cytoflow immunofluorescence,and pre-labeled with PKH26 before intravenous infusion into animals treated with CCl 4.The integration of bone marrow cells into the liver was examined microscopically,and plasma hepatic enzymes were determined biochemically.RESULTS:Cultured bone marrow cells exhibited antigenic profiles comparable to those of primary medullary stem cells.Double immunofluorescence showed colocalization of these cells with proliferative activity and albumin expression in the liver of CCl 4 -treated mice.Densitometry showed increased in situ cell proliferation (50±14 vs 20±3 cells/high-power field,P<0.05) and albumin expression (149±25 vs 20±5 cells/high-power field,P<0.05) in the liver,as well as reduced serum aminotransferase levels (P<0.05) and better survival rates (P<0.05) in animals receiving cultured bone marrow cells relative to controls.CONCLUSIONS:Ex vivo-expanded bone marrow cells are capable of relocating to and proliferating in the chemically- injured liver.Transplantation of these pluripotent stem cells appears to improve serum indices of liver function and survival rate in mice after CCl4-induced hepatic damage.Shi-Zhu Jin,Bing-Rong Liu,Jun Xu,Fu-Lai Gao,Zong-Jing Hu,Xin-Hong Wang,Feng-Hua Pei,Yu Hong,Hong-Yan Hu and Ming-Zi Han Department of Gastroenterology and Hepatology,and Department of Science Research Management,Second Affiliated Hospital,Harbin Medical University,Harbin 150080,China Department of Gastroenterology and Hepatology,Fourth Affiliated Hospital,Harbin Medical University,Harbin 150001,China 2012Hepatobiliary & Pancreatic Diseases International2012,11,1:16
9Oral berberine improves brain dopa/dopamine levels to ameliorate Parkinson’s disease by regulating gut microbiota显示文摘The phenylalanine-tyrosine-dopa-dopamine pathway provides dopamine to the brain.Iin this process,tyrosine hydroxylase(TH)isthe rate-limiting enzyme that hydroxylates tyrosine and generates levodopa(L-dopa)with tetranydrobiopterin(BH_(4))as a coenzyme.Here,we show that oral berberine(BBR)might supply H^(·) through dihydroberberine(reduced BBR produced by bacterial nitroreductase)and promote the production of BHl from dihydrobiopterin;the increased BH,enhances TH activity,which accelerates the production of L-dopa by the gut bacteria.Oral BBR acts in a way similar to vitamins.The L-dopa produced by theintestinal bacteria enters the brain through the circulation and is transformed to dopamine.To verify the gut-brain dialog activatedby BBR's effect,Enterococcus foecalis or Enterococcus faecium was transplanted into Parkinson's disease(PD)mice.The bacteriasignificantly increased brain dopamine and ameliorated PD manifestation in mice;additionally,combination of BBR with bacteriashowed better therapeutic effect than that with bacteria alone.Moreover,2,4,6-trimethy-pyranylium tetrafluoroborate(TMP-TFB)-derivatized matrix-assisted laser desorption mass spectrometry(MALDI-MS)imaging of dopamine identihed elevated striataldopamine levels in mouse brains with oral Enterococcus,and BBR strengthened the imaging intensity of brain dopamine.Theseresults demonstrated that BBR was an agonist of TH in Enterococcus and could lead to the production of L-dopa in the gut.Furthermore,a study of 28 patients with hyperlipidemia conhrmed that oral BBR increased bloodfecal L-dopa by the intestinalbacteria.Hence,BBR might improve the brain function by upregulating the biosynthesis of-dopa in the gut microbiota through avitamin-like effect.Yan Wang Qian Tong Shu-Rong Ma Zhen-Xiong Zhao Li-Bin Pan Lin Cong Pei Han Ran Peng Hang Yu Yuan Lin Tian-Le Gao Jia-Wen Shou Xiao-Yang Li Xian-Feng Zhang Zheng-Wei Zhang Jie Fu Bao-Ying Wen Jin-Bo Yu Xuetao Cao Jian-Dong Jiang 2021Signal Transduction and Targeted Therapy2021,6,3:16
10Effects of Honghua preserved amniotic membrane on scar healing in experimental glaucoma surgery显示文摘AIM:To investigate the efficacy and safety of Honghua preserved amniotic membrane(AM)for preventing scar formation of the filtering bleb in a rabbit model of glaucoma trabeculectomy surgery.METHODS:Totally 36 rabbits(36 eyes)were randomly divided into 3 groups:the experimental group(ocular trabeculectomy in combination with Honghua preserved AM transplantation),the control group(ocular trabeculectomy surgery in combination with AM implantation),and the blank group(single trabeculectomy).Clinical observations[including intraocular pressure(IOP),filtering blebs and complications],MassonTrichrome staining,real-time quantitative reverse transcription-polymerase chain reaction(real-time PCR),Western blot were performed on different time points(D1,D7,D14,D21 and D56)after the surgery.RESULTS:After operated for 14d,there were statistically significant differences in the filtering blebs compared to the situation before operation(P<0.05),whereas no statistically difference on that among three groups(P>0.05).After 21d,the IOP of experimental group was lowest(P<0.05).There was significant difference between control group and blank group(P<0.05).On postoperative D14,the mean number of fibroblasts in the experimental group was significantlylower(40.6±10.2)compared to those in the control group(54.4±10.8)and blank group(68.2±11.6)(P<0.05,respectively).The mean numbers of the macrophage in the experimental and control groups were respcitively significantly lower versus the blank group(P<0.05,P<0.05,respectively).Compared to that in blank group,the level of transforming growth factor-β(TGF-β1)expression in sclera and conjunctival areas was reduced in the experimental and control groups on protein and mRNA level(P<0.05),but not significant difference between these two groups(P>0.05).CONCLUSION:The trabeculectory surgery with Honghua preserved AM can control IOP,sustain the functional filtration bleb,inhibit the proliferation of fibroblasts and open the filtrating pathway on the rabbit glaucoma models.Yi Shao Yao Yu Qiu-Ping Liu Jing-Ming Li Fei Dong Xin Huang Chong-Gang Pei Ping Tu Hen-Hui Li Gui-Ping Gao 2014International Journal of Ophthalmology(English edition)2014,7,2:16
11Evaluation of novel decellularizing corneal stroma for cornea tissue engineering applications显示文摘·AIM:To develop a new decellularization method depended upon the natural corneal structure and to harvest an ideal scaffold with good biocompatibilities for corneal reconstruction.METHODS:The acellular cornea matrix (ACM) were prepared from de-epithelium fresh porcine corneas (DFPCs) by incubation with 100% fresh human sera and additional electrophoresis at 4℃. Human corneal epithelial cells (HCEs) were used for the cytotoxicity tests of ACM. ACM were implanted into the Enhanced Green Fluorecence Protein (eGFP) transgenic mouse anterior chamber for evaluation of histocompatibility.RESULTS:HE and GSIB4 results showed fresh porcine cornea matrix with 100% human sera and electrophoresis could entirely decellularize stromal cell without reducing its transparency. ACM has no cytotoxic effect ex vivo. 12Craniocerebral injury promotes the repair of peripheral nerve injury显示文摘The increase in neurotrophic factors after craniocerebral injury has been shown to promote fracture healing. Moreover, neurotrophic factors play a key role in the regeneration and repair of peripheral nerve. However, whether craniocerebral injury alters the repair of peripheral nerve injuries remains poorly understood. Rat injury models were established by transecting the left sciatic nerve and using a free-fall device to induce craniocerebral injury. Compared with sciatic nerve injury alone after 6–12 weeks, rats with combined sciatic and craniocerebral injuries showed decreased sciatic functional index, increased recovery of gastrocnemius muscle wet weight, recovery of sciatic nerve ganglia and corresponding spinal cord segment neuron morphologies, and increased numbers of horseradish peroxidase-labeled cells. 14Plant diversity reduces the effect of multiple heavy metal pollution on soil enzyme activities and microbial community structure显示文摘It is unclear whether certain plant species and plant diversity could reduce the impacts of multiple heavy metal pollution on soil microbial structure and soil enzyme activities. Random amplified polymorphic DNA (RAPD) was used to analyze the genetic diversity and microbial similarity in planted and unplanted soil under combined cadmium (Cd) and lead (Pb) pollution. A metal hyper- accumulator, Brassica juncea, and a common plant, Festuca arundinacea Schreb, were used in this research. The results showed that microorganism quantity in planted soil significantly increased, compared with that in unplanted soil with Cd and Pb pollution. The order of microbial community sensitivity in response to Cd and Pb stress was as follows: actinomycetes 〉 bacteria 〉 fungi. Respiration, phosphatase, urease and dehydrogenase activity were significantly inhibited due to Cd and Pb stress. Compared with unplanted soil, planted soils have frequently been reported to have higher rates of microbial activity due to the presence of additional surfaces for microbial colonization and organic compounds released by the plant roots. Two coexisting plants could increase microbe population and the activity of phosphatases, dehydrogenases and, in particular, ureases. Soil enzyme activity was higher in B. juncea phytoremediated soil than in F. arundinacea planted soil in this study. Heavy metal pollution decreased the richness of the soil microbial community, but plant diversity increased DNA sequence diversity and maintained DNA sequence diversity at highlevels. 15VdPKS1 is required for melanin formation and virulence in a cotton wilt pathogen Verticillium dahliae显示文摘Verticillium dahliae is a soil-borne phytopathogenic fungus that causes vascular wilt disease in a broad range of hosts.This pathogen survives for many years in soil in the form of melanized microsclerotia.To investigate the melanin synthesis in V.dahliae,we identified a polyketide synthase gene in V.dahliae,namely VdPKS1.PKS1 is known to involve in the dihydroxynaphthalene melanin synthesis pathway in many fungi.We found that VdPKS1 was required for melanin formation but not for microsclerotial production in V dahliae.The VdPKS1 gene-disruption mutant (vdpksl) formed melanin-deficient albino microsclerotia,which did not affect the fungal colonization in host tissues but significantly reduced the disease severity.Gene transcription analysis in the wild-type and the vdpksl strains suggested that VdPKS1 gene-disruption influenced the expression of a series of genes involved in ethylene biosynthesis,microsclerotial formation and pathogenesis.Our results suggest that the VdPKS1-mediated melanin synthesis is important for virulence and developmental traits of V.dahliae.Ting Zhang Bosen Zhang Chenlei Hua Pei Meng Sheng Wang Zhirong Chen Yejuan Du Feng Gao Jiafeng Huang 2017Science China(Life Sciences)2017,60,8:14
16Progress in the stratigraphy and geochronology of the Shuidonggou site,Ningxia,North China显示文摘In the past years we carried out further stratigraphy division in field and it is found that rich stone artifacts can be found in fluvial-shallow lake-alluvial sediments on the terrace Ⅱ of Biangou River, in Shuidonggou site, Ningxia and they are SDG1, 2 and 7. More luminescence and AMS 14C dating in laboratory show that Paleolithic culture develops during the Upper Paleolithic period with ages of 35―20 ka. The Paleolithic culture of SDG 1 is a little earlier than that of SDG 2 similar to SDG 7. The sandy sediments on terrace Ⅱ of Biangou River deposits in the past 72―18 ka, corresponding to Last Glacial. 17Analysis of sedimentary-geomorphologic variation and the living environment of hominids at the Shuidonggou Paleolithic site显示文摘Shuidonggou is one of the most important Upper Paleolithic sites in North China. Due to the presence of rich human remains,animal fossils,abundant sporopollen and unique geological sequence,it is the type site for Late Pleistocene to Holocene human occupation and environmental change in the Ningxia-Inner Mongolia region. Many scholars suggest that the site should be named the 'Shuidong-gou Formation' of Late Pleistocene in North China. Dating results indicate that ancient human activities at the site took place 30―24 ka (Marine Isotope Stage MIS 3). The climate at that time was warmer and moister than present day,and adequate precipitation led to the formation of water pack depressions where broad-leaf trees and sparse forest vegetations,as well as herbivorous animals flourished,mak-ing the area suitable for early human hunting,gathering and survival. The Neolithic human occupation happened 9―5 ka at the site,while similar environmental conditions with MIS3 occurred. 18Microstructure, biodegradation, antibacterial and mechanical properties of ZK60-Cu alloys prepared by selective laser melting technique显示文摘Magnesium(Mg) alloys are receiving increasing attention for body implants owing to their good biocompatibility and biodegradability. However, they often suffer from bacterial infections on account of their insufficient antibacterial ability. In this study, ZK60-x Cu(x = 0, 0.2, 0.4, 0.6 and 0.8 wt%) alloys were prepared by selective laser melting(SLM) with alloying copper(Cu) to enhance their antibacterial ability.Results showed that ZK60-Cu alloys exhibited strong antibacterial ability due to combination of release of Cu ions and alkaline environment which could kill bacteria by destroying cellular membrane structure,denaturing enzymes and inhibiting deoxyribonucleic acid(DNA) replication. In addition, their compressive strength increased due to grain refinement and uniformly dispersing of short-bar shaped MgZnCu phases. Moreover, ZK60-Cu alloys also exhibited good cytocompatibility. 19Biochemical Mechanism of Chlorantraniliprole Resistance in the Diamondback Moth, Plutella xylostella Linnaeus显示文摘The insecticide chlorantraniliprole exhibits good efficacy and plays an important role in controlling the diamondback moth, Plutella xylostella Linnaeus. However, resistance to chlorantraniliprole has been observed recently in some field populations. At present study, diamondback moths with resistance to chlorantraniliprole(resistant ratio(RR) was 82.18) for biochemical assays were selected. The assays were performed to determine potential resistance mechanisms. The results showed that the selected resistant moths(GDLZ-R) and susceptible moth could be synergized by known metabolic inhibitors such as piperonyl butoxide(PBO), triphenyl phosphate(TPP) and diethyl-maleate(DEM) at different levels(1.68-5.50-fold and 2.20-2.89-fold, respectively), and DEM showed the maximum synergism in both strains. In enzymes assays, a high level of glutathione-S-transferase(GST) was observed in the resistant moth, in contrast, moths that are susceptible to the insecticide had only 1/3 the GST activity of the resistant moths. The analysis of short-term exposure of chlorantraniliprole on biochemical response in the resistant strain also showed that GST activity was significantly elevated after exposure to a sub-lethal concentration of chlorantraniliprole(about 1/3 LC50, 12 mg L-1) 12 and 24 h, respectively. 20Morinda officinalis oligosaccharides increase serotonin in the brain and ameliorate depression via promoting 5-hydroxytryptophan production in the gut microbiota显示文摘Morinda officinalis oligosaccharides(MOO) are an oral drug approved in China for the treatment of depression in China. However, MOO is hardly absorbed so that their anti-depressant mechanism has not been elucidated. Here, we show that oral MOO acted on tryptophan → 5-hydroxytryptophan(5-HTP) → serotonin(5-HT) metabolic pathway in the gut microbiota. MOO could increase tryptophan hydroxylase levels in the gut microbiota which accelerated 5-HTP production from tryptophan;meanwhile, MOO inhibited 5-hydroxytryptophan decarboxylase activity, thus reduced 5-HT generation,and accumulated 5-HTP. The raised 5-HTP from the gut microbiota was absorbed to the blood, and then passed across the blood-brain barrier to improve 5-HT levels in the brain. Additionally, pentasaccharide,as one of the main components in MOO, exerted the significant anti-depressant effect through a mechanism identical to that of MOO.
