| 2 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 4 | Histological Subtypes of Lung Cancer in Chinese Males from 2000 to 2012显示文摘Objective To characterize the histological and epidemiological features of male lung cancer patients in China. Methods The demographic and histological information about male lung cancer patients identified from 2000-01-01 to 2012-12-31,was collected from the Cancer Hospital of the Chinese Academy of Medical Sciences. Relative frequencies(RF) were estimated for major histological subtypes and compared according to the years of diagnosis and birth. Results The RF of adenocarcinoma(ADC) increased from 21.96% to 43.36% and the RF of squamous cell carcinoma(SCC) decreased from 39.11% to 32.23% from 2000 to 2012 in the 15 427 male lung cancer patients included in this study(Z=17.909,P<0.0001; Z=-6.117,P<0.0001). The RF of ADC increased from 28.72% in 2000-2004,36.88% in 2005-2008 to 48.61% in 2009-2012 in patients born after 1960. The age-adjusted RF of ADC in 2007-2012 increased consistently in all the investigated areas. Conclusion The increased RF of ADC in male lung cancer patients highlights the need for further investigation of the etiologic factors of these tumors. | 5 | Preliminary results of environmental monitoring of the natural gas hydrate production test in the South China Sea显示文摘Natural gas hydrate (NGH)is considered as one of the new clean energy sources of the 21st century with the highest potential.The environmental issues of NGH production have attracted the close attention of scientists in various countries.From May 10 to July 9,2017,the first offshore NGH production test in the South China Sea (SCS)was conducted by the China Geological Survey.In addition,environmental security has also been effectively guaranteed via a comprehensive environmental monitoring system built during the NGH production test.The monitoring system considered sea-surface atmosphere methane and carbon dioxide concentrations,dissolved methane in the sea water column,and the seafloor physical oceanography and marine chemistry environment.The whole process was monitored via multiple means, in multiple layers,in all domains,and in real time.After the production test,an environmental investigation was promptly conducted to evaluate the environmental impact of the NGH production test. | 9 | Clinical profiles and outcomes of acute type A aortic dissection and intramural hematoma in the current era: lessons from the first registry of aortic dissection in China显示文摘Background::Acute type A aortic dissection(ATAAD)and acute type A intramural hematoma(ATAIMH)are life-threatening diseases with high mortality.To better understand their clinical features in the Chinese population,we analyzed the data from the first Registry of Aortic Dissection in China(Sino-RAD)to promote the understanding and management of the diseases.Methods::All patients with ATAAD and ATAIMH enrolled in Sino-RAD from January 1,2012 to December 31,2016 were involved.The data of patients’selection,history,symptoms,management,outcomes,and postoperation complications were analyzed in the study.The continuous variables were compared using the Student’s t test for normal distributions and the Mann-Whitney U test for non-normal distributions.Categorical variables were compared using the Chi-square test or Fisher exact test.Results::A total of 1582 patients with ATAAD and 130 patients with ATAIMH were included.The mean age of all patients was 48.4 years.Patients with ATAAD were significantly younger than patients with ATAIMH(48.9 years vs.55.6 years,P<0.001).For the total cohort,males were dominant,but the male ratio of patients with ATAAD was significantly higher compared to those with ATAIMH(P=0.01).The time range from the onset of symptom to hospitalization was 2.0 days.More patients of ATAIMH had hypertension than that of ATAAD(82.3%vs.67.6%,P<0.05).Chest and back pain were the most common clinical symptoms.Computerized tomography(CT)was the most common initial diagnostic imaging modality.84.7%received surgical treatment and in-hospital mortality was 5.3%.Patients with ATAAD mainly received surgical treatment(89.6%),while most patients with ATAIMH received medical treatment(39.2%)or endovascular repair(35.4%).Conclusions::Our study suggests that doctors should comprehensively use clinical examination and genetic background screening for patients with ATAAD and ATAIMH and further shorten the time range from symptoms onset to intervention,achieving early diagnosis and treatment,thereby reducing the mortality rate of patients with aortic dissection in China.We should standardize the procedures of aortic dissection treatment and improve people’s understanding.Meanwhile,the curing and transferring efficiency should also be improved. | Wei-Xun Duan Wei-Guang Wang Lin Xia Chao Xue Bo Yu Kai Ren Wei Yi Hong-Liang Liang Xiao-Chao Dong Jian Zuo Jin-Cheng Liu Shi-Qiang Yu Ding-Hua Yi | 2021 | Chinese Medical Journal2021,,8: | 15 |
| 10 | The causes and impacts for heat stress in spring maize during grain filling in the North China Plain--A review显示文摘High-temperature stress(HTS) at the grain-filling stage in spring maize(Zea mays L.) is the main obstacle to increasing productivity in the North China Plain(NCP). To solve this problem, the physiological mechanisms of HTS, and its causes and impacts, must be understood. The HTS threshold of the duration and rate in grain filling, photosynthetic characteristics(e.g., the thermal stability of thylakoid membrane, chlorophyll and electron transfer, photosynthetic carbon assimilation), water status(e.g., leaf water potential, turgor and leaf relative water content) and signal transduction in maize are reviewed. The HTS threshold for spring maize is highly desirable to be appraised to prevent damages by unfavorable temperatures during grain filling in this region. HTS has negative impacts on maize photosynthesis by damaging the stability of the thylakoid membrane structure and degrading chlorophyll, which reduces light energy absorption, transfer and photosynthetic carbon assimilation. In addition, photosynthesis can be deleteriously affected due to inhibited root growth under HTS in which plants decrease their water-absorbing capacity, leaf water potential, turgor, leaf relative water content, and stomatal conductance. Inhibited photosynthesis decrease the supply of photosynthates to the grain, leading to falling of kernel weight and even grain yield. However, maize does not respond passively to HTS. The plant transduces the abscisic acid(ABA) signal to express heat shock proteins(HSPs), which are molecular chaperones that participate in protein refolding and degradation caused by HTS. HSPs stabilize target protein configurations and indirectly improve thylakoid membrane structure stability, light energy absorption and passing, electron transport, and fixed carbon assimilation, leading to improved photosynthesis. ABA also induces stomatal closure to maintain a good water status for photosynthesis. Based on understanding of such mechanisms, strategies for alleviating HTS at the grain-filling stage in spring maize are summarized. Eight strategies have the potential to improve the ability of spring maize to avoid or tolerate HTS in this study, e.g., adjusting sowing date to avoidHTS, breeding heat-tolerance varieties, and tillage methods, optimizing irrigation, heat acclimation, regulating chemicals, nutritional management, and planting geometric design to tolerate HTS. | 11 | Trends in geographical disparities for cervical cancer mortality in China from 1973 to 2013:a subnational spatio-temporal study显示文摘Objective: This study was designed to explore the time trends in geographical variations of cervical cancer mortality in China's Mainland over the period 1973 to 2013,to provide subnational spatio-temporal patterns for targeted promotion of human papillomavirus vaccine in China.Methods: Data were extracted from three national retrospective death surveys and cancer registry.The rate ratio(RR) was estimated for the aggregated data for seven geographical regions using generalized linear models to evaluate time trends in geographical disparities of cervical cancer mortality.Results: There was a significant decrease in cervical cancer mortality in China from 1973–1975 to 2004–2005,but leveled off thereafter to 2011–2013.Compared to the period 1973–1975 the RR for the three last time periods were 0.33 [95% confidence interval(95% CI): 0.30–0.37] for 1990–1992,0.21(95% CI: 0.19–0.24) for 2004–2005 and 0.24(95% CI: 0.22–0.26) for 2011–2013.Females living in the Northwest China and Central China have a high risk of mortality from cervical cancer compared to the nationwide,with the RR being 2.09(95% CI:1.83–2.38) and 1.26(95% CI: 1.11–1.44) respectively,while the RRs for South China,Northeast China and Southwest China were below 1.00,indicating the lower death risk.Despite the mortality rate had increased slightly from 2004 to 2013,there was an encouraging sign that the geographical disparities in cervical cancer mortality had gradually narrowed over time across China.Conclusions: Although cervical cancer mortality in China has reduced to very low levels,the high risk of cervical cancer in Northwest China and Central China is still noteworthy.Public health policies including the promotion of vaccine should be targeted to further reduction of geographical disparities in cervical cancer mortality. | Changfa Xia Chao Ding Rongshou Zheng Siwei Zhang Hongmei Zeng Jinfeng Wang Yilan Liao Ningxu Zhang Zhixun Yang Wanqing Chen | 2017 | Chinese Journal of Cancer Research2017,29,6: | 13 |
| 12 | Tripartite motif.containing 3(TRIM3)inhibits tumor growth and metastasis of liver cancer显示文摘Background:Reduced expression of tripartite motif-containing 3(TRIM3) has been reported to be involved in the pathogenesis of human glioblastoma.In our previous research,we found that TRIM3 expression was markedly reduced in human primary hepatocellular carcinoma(HCC) tissues and that low TRIM3 expression was associated with short survival of HCC patients.However,the role of TRIM3 in liver cancer remains unknown.This study aimed to investigate the function of TRIM3 in liver cancer cells.Methods:The protein levels of TRIM3 in five liver cancer cell lines(SK-Hep1,Hep3 B,Huh7,HepG2,Bel-7402) and one normal liver cell line(L02) were detected with Western blotting.HepG2 and Bel-7402 cells with low TRIM3 expression were infected with recombinant lentiviruses overexpressing TRIM3(LV-TRIM3),whereas Huh7 and Hep3 B cells with high TRIM3 expression were transfected with TRIM3-targeted small interfering RNA(siTRIM3).The functions of TRIM3 in the proliferation,colony formation,cell cycle,migration,invasion,and apoptosis of the above cell lines were examined.The effect ofTRIM3 on tumor growth and metastases in nude mice was also investigated.Results:TRIM3 was overexpressed in HepG2 and Bel-7402 cells with LV-TRIM3 infection,which further reduced proliferation,colony formation,migration,and invasion of both cell lines.Cell cycle analysis showed that TRIM3 overexpression induced G_0/G_1 phase arrest in HepG2 and Bel-7402 cells.Moreover,apoptosis was not increased in HepG2 or Bel-7402 cells overexpressing TRIM3.Contrarily,silencing TRIM3 expression in Huh7 and Hep3 B cells by siTRIM3 led to significantly decreased percentages of both cells in the G_0/G_1 phase and promoted cell proliferation,colony formation,migration,and invasion.In vivo experiment results confirmed that TRIM3 overexpression suppressed tumor growth and metastasis.Conclusions:TRIM3 plays a tumor-suppressing role in the regulation of liver cancer development by reducing cell proliferation through cell cycle arrest at the G_0/G_1 phase. | Xu.Qiong Huang Xiao.Fei Zhang Jin.Hua Xia Jie Chao Qiu.Zhong Pan Jing.Jing Zhao Zi.Qi Zhou Chang.Long Chen Yan Tang De.Sheng Weng Jian.Hua Zhang Jian.Chuan Xia | 2017 | Chinese Journal of Cancer2017,36,9: | 13 |

| 14 | Down-Regulation of CD44 Contributes to the Differentiation of HL-60 Cells Induced by ATRA or HMBA显示文摘CD44 is highly expressed in human acute myeloid leukemia (AML) cells. Some experiments had shown that it was possible to reverse differentiation blockage in AML cells by CD44 ligation with specific antibodies, indicating that CD44 was closely related to the differentiation of leukemia cells. The differentiation of acute promyelocytic leukemia cell line HL-60 cells could be induced by all trans-retinoic acid (ATRA) and hexamethylene bisacetamide (HMBA), but so far the mechanism was not demonstrated clearly. In the present study, we investigated whether ATRA or HMBA induced the growth arrest of HL-60 cells by down-regulating the expression of CD44. The results indicated that the proliferation of HL-60 cells was obviously inhibited and the differentiation was induced by both ATRA and HMBA. The decreased expression of CD44 and cyclin E mRNA, and the increased expression of p27 and p21 at mRNA levels were observed. Furthermore, there was a negative correlation between the expression of CD44 and p27. | 16 | Protective effect of Shouwu Yizhi decoction against vascular dementia by promoting angiogenesis显示文摘Shouwu is a traditional Chinese medicine(TCM)with neuroprotective effect.Shouwu Yizhi decoction(SYD)was designed based on TCM theory.However,little is known about the roles of SYD in Vascular dementia(Va D).The present study aimed to evaluate the potential effects of SYD on the vascular cognitive impairment and explore the underlying mechanism by establishing focal cerebral ischemia/reperfusion(I/R)rat model to induce Va D.SYD administration(54 mg·kg^(-1))for 40 days obviously improved the vascular cognitive impairment in the middle cerebral artery occlusion(MCAO)rats as evidenced by the declined neurological deficit score and shortened escape latency via neurological deficit assessment and Morris water maze test.Moreover,SYD decreased neuron damage-induced cell death and ameliorated the ultrastructure of endothelial cells in the MCAO rats,thereby alleviating Va D.Mechanistically,SYD caused increases in the expression of vascular endothelial growth factor(VEGF),CD34 and CD31,compared with the MCAO rats in coronal hippocampus.Simultaneously,the expression level of mi R-210 was elevated significantly after SYD administration,compared with the vehicle rats(P<0.01).The expression of Notch 4 at both m RNA and protein levels was upregulated remarkably along with the notably downregulated DLL4 expression under SYD administration compared with the vehicle rats(P<0.05).Overall,the above results indicated that SYD promoted angiogenesis by upregulating VEGF-induced mi R210 expression to activate Notch pathway,and further alleviated neuron damage and ameliorated the ultrastructure of endothelial cells in the MCAO rats,ultimately enhancing the cognition and memory of MCAO rats.Therefore,our findings preliminarily identified the effect and the mechanism of action for SYD on Va D in rats.SYD could be a potential candidate in treatment of Va D. | YANG Xiao-Ni LI Chang-Sheng CHEN Chao TANG Xiao-Yong CHENG Guang-Qing LI Xia | 2017 | Chinese Journal of Natural Medicines2017,15,10: | 11 |
