1Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs.Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu 2020Cell Research2020,30,4:81

6Discrimination of abiogenic and biogenic alkane gases显示文摘We have combined the analytical data of the carbon isotope distribution pattern, R/Ra and CH4/3He values of abiogenic and biogenic (referring to the thermogenic and bacterial or microbial) alkane gases in China with those of alkane gases from USA, Russia, Germany, Australia and other countries. 8First mass measurement of short-lived nuclides at HIRFL-CSR显示文摘Recent commissioning of the HIRFL-CSR has demonstrated its ability to perform direct mass measurement for short-lived nuclides. Projectile fragments produced by the 78Kr ions at 481.88 MeV/u were separated with the new radioactive beam line in Lanzhou (RIBLL2), and injected into and stored in the experimental storage ring (CSRe). By operating the CSRe as an isochronous mass spectrometry, a typical mass resolution around 2.0×10?5 has been achieved. The masses for 63Ge, 65As and 67Se were measured for the first time. The measured masses are compared with theoretical predictions and the location of the proton drip-line for As isotopes is discussed. 9Sleep Deprivation Selectively Down-Regulates Astrocytic 5-HT2B Receptors and Triggers Depressive-Like Behaviors via Stimulating P2X7 Receptors in Mice显示文摘Chronic loss of sleep damages health and disturbs the quality of life.Long-lasting sleep deprivation(SD)as well as sleep abnormalities are substantial risk factors for major depressive disorder,although the underlying mechanisms are not clear.Here,we showed that chronic SD in mice promotes a gradual elevation of extracellular ATP,which activates astroglial P2X7 receptors(P2X7Rs).Activated P2X7Rs,in turn,selectively down-regulated the expression of 5-HT2B receptors(5-HT2BRs)in astrocytes.Stimulation of P2X7Rs induced by SD selectively suppressed the phosphorylation of AKT and FoxO3 a in astrocytes,but not in neurons.The overexpression of FoxO3a in astrocytes inhibited the expression of 5-HT2BRs.Down-regulation of 5-HT2BsRs instigated by SD suppressed the activation of STAT3 and relieved the inhibition of Ca2+-dependent phospholipase A2.This latter cascade promoted the release of arachidonic acid and prostaglandin E2.The depression-like behaviors induced by SD were alleviated in P2X7R-KO mice.Our study reveals the mechanism underlying chronic SD-induced depression-like behaviors and suggests 5-HT2BRs as a key target for exploring therapeutic strategies aimed at the depression evoked by sleep disorders.Maosheng Xia Zexiong Li Shuai Li Shanshan Liang Xiaowei Li Beina Chen Manman Zhang Chengyi Dong Alexei Verkhratsky Dawei Guan Baoman Li 2020Neuroscience Bulletin2020,36,11:10

11Simulated Microgravity Conditions and Carbon Ion Irradiation Induce Spermatogenic Cell Apoptosis and Sperm DNA Damage显示文摘Objective To investigate the effect of simulated microgravity and carbon ion irradiation(CIR) on spermatogenic cell apoptosis and sperm DNA damage to the testis of male Swiss Webster mice,and assess the risk associated with space environment.Methods Sperm DNA damage indicated by DNA fragmentation index(DFI) and high DNA stainability(HDS) was measured by sperm chromatin structure assay(SCSA).Apoptosis of spermatogenic cells was detected by annexin V-propidium iodide assay.Bax(the expression levels of p53) and proliferating cell nuclear antigen(PCNA) were measured by immunoblotting;p53 and PCNA were located by immunohistology.Results HDS,DFI,apoptosis index,and the expression levels of p53 and Bax were detected to be significantly higher in the experimental groups(P<0.05) compared with those in the control group;however,the PCNA expression varied to a certain degree.p53-and PCNA-positive expression were detected in each group,mainly in relation to the spermatogonic cells and spermatocytes.Conclusion The findings of the present study demonstrated that simulated microgravity and CIR can induce spermatogenic cell apoptosis and sperm DNA damage.Sperm DNA damage may be one of the underlying mechanisms behind male fertility decline under space environment.These findings may provide a scientific basis for protecting astronauts and space traveler's health and safety.LI Hong Yan ZHANG Hong MIAO Guo Ying XIE Yi SUN Chao DI Cui Xia LIU Yang LIU Yuan Yuan ZHANG Xin MA Xiao Fei XU Shuai GAN Lu ZHOU Xin 2013Biomedical and Environmental Sciences2013,26,9:10
12Acute calculous cholecystitis associated with hepatic artery pseudoaneurysm after percutaneous transhepatic gallbladder drainage in a diabetic patient显示文摘Patients with acute calculous cholecystitis are usually undertaken surgery laparoscopic cholecystectomy.However,there are controversies about the decision of operation for patients with high risk factors.Percutaneous transhepatic gallbladder drainage (PTGD) is palliative therapy to alleviate symptoms and physical signs.Since improved clinical outcome has been observed for PTGD,it was recommended as an initial therapeutic option for high risk patients who might need surgery.1 Percutaneous drainage in acute calculous cholecystitis for high risk patients is to be a safe and successful treatment option for patients less eligible for surgery.2 PTGD is relatively safe,but has certain complications.We provided a case report of acute calculous cholecystitis associated with hepatic artery pseudoaneurysm after PTGD in a diabetes mellitus patient.The liver vascular pseudoaneurysms and secondary changes of liver were documented by the imaging examination.The complex interactions among inflammation,bleeding,and diabetes mellitus were involved in this case.Tian Hu Xia Mingfeng Zhang Shuai Li Jie Liu Ju 2014Chinese Medical Journal2014,,17:9

14Large-Scale Membrane Bioreactors for Industrial Wastewater Treatment in China:Technical and Economic Features,Driving Forces,and Perspectives显示文摘Membrane bioreactors(MBRs)have been and will continue playing an important role in industrial wastewater treatment and reuse in China.The sustainable development of MBR technology in its mature-application stage requires reciprocal interactions between engineering and research participants.Thus,in this study,a total of 182 large-scale MBR projects treating industrial wastewater(with individual treatment capacities5000 m3d1)commissioned and under construction from 2003 to 2019 were analyzed comprehensively.Fast growth of the cumulative treatment capacity was observed,with extension to diverse industries,and the super large-scale was enhanced recently.The treatment processes,pollutant removal efficiencies,and actual operational parameters were summarized regarding the particularity of industrial wastewater compared to municipal wastewater.Economic features including the total investment costs of the projects,their total footprint,and their operational energy consumption were analyzed as well.A vigorous MBR market has formed in China with the fast development of membrane elements and engineering suppliers,continuously increasing official oriented projects,and responsive and innovative business modes.MBR technology has been mostly applied in specific economic zones and water-deficient areas,but its widespread use all over China is foreseeable considering the vast future market for industrial wastewater treatment and recycling.The policy–economy and market–technology driving forces revealed that MBR is consistent with the national development demand.According to the survey and analysis,prospective development in both engineering and research aspects of MBR is proposed to maintain its competitive edge.Jiao Zhang Kang Xiao Ziwei Liu Tingwei Gao Shuai Liang Xia Huang 2021Engineering2021,7,6:9
15Diffusion bonding of AlCoCrFeNi2.1 eutectic high entropy alloy to TiAl alloy显示文摘High entropy alloys have special microstructure and remarkable properties.To explore their potential engineering application in high temperature structures,the microstructure evolution of bonding interface,the elemental diffusion behavior and mechanical property of the diffusion bonded joint between AlCoCrFeNi2.1eutectic high entropy alloy(EHEA)and TiAl alloy were investigated.Four reaction layers(rodlike B2 phase,Al(Co,Ni)2Ti,τ3-Al3NiTi2+TiAl,τ3-Al3NiTi2+TiAl+Ti3Al)formed in the diffusion zone near FCC phase of EHEA,but three layers(Al(Co,Ni)2Ti,τ3-Al3Ni Ti2+Ti Al,τ3-Al3Ni Ti2+Ti Al+Ti3Al)formed near B2 phase.Al and Ni controlled the reaction diffusion of EHEA and TiAl alloy,coarsened the acicular precipitated B2 phase and turned Ti Al phase into Al(Co,Ni)2Ti andτ3-Al3NiTi2 phases.All these reaction layers grew in a parabolic manner as a function of bonding temperature.Rodlike B2 phase has the lowest growth activation energy of 125.2 kJ/mol,and the growth activation energy ofτ3-Al3Ni Ti2+TiAl layer near B2 phase is much lower than that near FCC phase.The penetration phenomenon and convex structure formed in the diffusion zone,which resulted in interlocking effect and enhanced the strength of resultant joints.The highest shear strength of 449 MPa was achieved at 950℃.And the brittle fracture generally initiated at the interface between Al(Co,Ni)2Ti andτ3-Al3NiTi2+TiAl layers.Peng Li Shuai Wang Yueqing Xia Xiaohu Hao Honggang Dong 2020Journal of Materials Science & Technology2020,42,10:8

17RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy,thus calling for the development of safe and effective vaccines.The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines.Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator,elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections.The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge.These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV.Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang 2020Signal Transduction and Targeted Therapy2020,5,1:7
18Geochemistry and genesis of authigenic pyrite from gas hydrate accumulation system in the Qilian Mountain permafrost, Qinghai, northwest China显示文摘A type of authigenic pyrites that fully fill or semi-fill the rock fractures of drillholes with gas hydrate anomalies are found in the Qilian Mountain permafrost; this type of pyrite is known as 'fracture-filling' pyrite. The occurrence of 'fracture-filling' pyrite has a certain similarity with that of the hydrate found in this region, and the pyrite is generally concentrated in the lower part of the hydrate layer or the hydrate anomaly layer. The morphology, trace elements, rare earth elements, and sulfur isotope analyses of samples from drillhole DK-6 indicate that the 'fracture-filling' pyrites are dominated by cubic ones mainly aligned in a step-like fashion along the surfaces of rock fractures and are associated with a circular structure, lower Co/Ni and Sr/Ba, lower ?REE, higher LREE, significant Eu negative anomalies, and ? 34 SCDT positive bias. In terms of the pyrites' unique crystal morphology and geochemical characteristics and their relationship with the hydrate layers or abnormal layers, they are closely related with the accumulation system of the gas hydrate in the Qilian Mountain permafrost. As climate change is an important factor in affecting the stability of the gas hydrate, formation of fracture-filling pyrites is most likely closely related to the secondary change of the metastable gas hydrate under the regional climate warming. The distribution intensity of these pyrites indicates that when the gas hydrate stability zone(GHSZ) is narrowing, the hydrate decomposition at the bottom of the GHSZ is stronger than that at the top of the GHSZ, whereas the hydrate decomposition within the GHSZ is relatively weak. 19Nanoparticle monolayer-based flexible strain gauge with ultrafast dynamic response for acoustic vibration detection显示文摘The relatively poor dynamic response of current flexible strain gauges has prevented their wide adoption in portable electronics. In this work, we present a greatly improved flexible strain gauge, where one strip of Au nanoparticle (NP) monolayer assembled on a polyethylene terephthalate film is utilized as the active unit. The proposed flexible gauge is capable of responding to applied stimuli without detectable hysteresis via electron tunneling between adjacent nanoparticles within the Au NP monolayer. Based on experimental quantification of the time and frequency domain dependence of the electrical resistance of the proposed strain gauge, acoustic vibrations in the frequency range of 1 to 20,000 Hz could be reliably detected.
