2Influence of hydraulic retention time on behavior of antibiotics and antibiotic resistance genes in aerobic granular reactor treating biogas slurry显示文摘The behavior of antibiotics and the corresponding resistance genes in aerobic granular reactors for treating biogas slurry under different hydraulic retention times (10.7 h, Rl;8 h, R2) was investigated in this study. The results indicated that the hydraulic retention time could affect the effluent concentrations and removal efficiencies of sulfonamides. The average removal rates of tetracyclines, fluoroquinolones, and sulfonamides were 63%, 46%, and 90% in Rl, and 62%, 46%, and 86% in R2, respectively. Although the removal efficiencies of tetracyclines and fluoroquinolones were similar in both reactors, the respective accumulated concentrations of tetracyclines and fluoroquinolones in R1 were 7.00 and 11.15μg/g SS, which were lower than those in R2 (8.92 and 13.37μg/g SS, respectively). The difference in the relative abundance of target antibiotic resistance genes between both reactors was not significant, yet the average relative abundances of all target resistance genes in R1 were higher than those in R2 after 45 days of operation. 3Pilot-scale studies of domestic wastewater treatment by typical constructed wetlands and their greenhouse gas emissions显示文摘Three typical constructed wetlands(CWs)including Vertical Flow(VF),Free Water Surface(FWS),and Subsurface Flow(SF),and combined VF-SF-FWS constructed wetlands were investigated for the treatment of domestic wastewater with low C/N ratio.The performance of nutrient removal and the characteristics of greenhouse gas emissions,such as CH4 and N2O,from these CWs were compared.The results indicated that the four types of CWs had high removal efficiencies for organic matter and suspended solid(SS).The combined wetland also showed a comparatively good performance for nitrogen and phosphorus removal,and the removal efficiencies for total nitrogen(TN)and total phosphorus(TP)were 81.3%and 84.5%,respectively.The combined CWs had a comparative lower global warming potential.The FWS CW had the highest tendency to emit CH4 and led to a higher global warming potential among the four types of CWs,which was about 586 mg CO_(2)/m^(2)·h.Chaoxiang LIU Kaiqin XU Ryuhei INAMORI Yoshitaka EBIE Jie LIAO Yuhei INAMORI 2009Frontiers of Environmental Science & Engineering2009,3,4:2

8Fermentative hydrogen production from beet sugar factory wastewater treatment in a continuous stirred tank reactor using anaerobic mixed consortia显示文摘A low pH, ethanol-type fermentation process was evaluated for wastewater treatment and bio-hydrogen production from acidic beet sugar factory wastewater in a continuous stirred tank reactor (CSTR) with an effective volume of 9.6 L by anaerobic mixed cultures in this present study. After inoculating with aerobic activated sludge and operating at organic loading rate (OLR) of 12 kgCOD·m-3·d-1, HRT of 8h, and temperature of 35℃ for 28 days, the CSTR achieved stable ethanol-type fermentation. When OLR was further increased to 18 kgCOD·m-3·d-1, on the 53rd day, ethanol-type fermentation dominant microflora was enhanced. The liquid fermentation products, including volatile fatty acids (VFAs) and ethanol, stabilized at 1493mg·L-1 in the bioreactor. Effluent pH, oxidation-reduction potential (ORP), and alkalinity ranged at 4.1-4.5, -250-(-290) mV, and 230-260mgCaCO3·L-1. The specific hydrogen production rate of anaerobic activated sludge was 0.1 L'gMLVSS-1· d-1 and the COD removal efficiency was 45%. 9Hypoxia-induced RBBP7 promotes esophagus cancer progression by inducing CDK4 expression显示文摘Hypoxia-induced epigenetic regulation calls for more effective therapeutic targets for esophageal cancer.We used GEPIA and UALCAN databases to screen survival-related and cancer stage-associated genes.Eca109 and KYSE450 esophageal cancer cell lines were cultured under normoxia,hypoxia,or CoCl2-induced hypoxia conditions,which were further transfected with plasmids expressing RB binding protein 7(RBBP7),hypoxia-inducible factor 1(HIF1)-α,or RBBP7 shRNA.Colony formation and MTT assays were used to detect cell proliferation.Tumor sphere formation and stemness marker detection were applied to assess cell stemness.RT-PCR and western blot analysis were used to detect the relative mRNA level and protein expression,respectively.Luciferase assay was utilized to detect the direct interaction between HIF1αand RBBP7.Up-regulated RBBP7 was identified as one of the most prominent survival-related genes,which is negatively correlated with the overall survival(OS),disease recurrencefree survival(DFS),and tumor stages.Hypoxia-induced HIF1αup-regulates RBBP7 expression,which promotes esophagus cancer cell viability,proliferation,and stemness with increased cyclin-dependent kinase 4(CDK4)expression.Luciferase reporter assay verified that HIF1αtranscriptionally regulates the expression of RBBP7.We conclude that hypoxia induces high expression of RBBP7 which is at least partially mediated by HIF1α,up-regulates the expression of downstream CDK4,and thereby promotes tumor progression in esophageal cancer cells.Renfeng Wang Zhiliang Huang Zhenyang Lin Borong Chen Xuguang Pang Chaoxiang Du Hong Fan 2022Acta Biochimica et Biophysica Sinica2022,54,2:1

16Taurolidine improved protection against highly pathogenetic avian influenza H5N1 virus lethal-infection in mouse model by regulating the NF-κB signaling pathway显示文摘Taurolidine(TRD),a derivative of taurine,has anti-bacterial and anti-tumor effects by chemically reacting with cell-walls,endotoxins and exotoxins to inhibit the adhesion of microorganisms.However,its application in antiviral therapy is seldom reported.Here,we reported that TRD significantly inhibited the replication of influenza virus H5N1 in MDCK cells with the half-maximal inhibitory concentration(EC_(50))of 34.45μg/mL.Furthermore,the drug inhibited the amplification of the cytokine storm effect and improved the survival rate of mice lethal challenged with H5N1(protection rate was 86%).Moreover,TRD attenuated virus-induced lung damage and reduced virus titers in mice lungs.Administration of TRD reduced the number of neutrophils and increased the number of lymphocytes in the blood of H5N1 virus-infected mice.Importantly,the drug regulated the NF-κB signaling pathway by inhibiting the separation of NF-κB and IκBa,thereby reducing the expression of inflammatory factors.In conclusion,our findings suggested that TRD could act as a potential anti-influenza drug candidate in further clinical studies.Chaoxiang Lv Yuanguo Li Tiecheng Wang Qiqi Zhang Jing Qi Mingwei Sima Entao Li Tian Qin Zhuangzhuang Shi Fangxu Li Xuefeng Wang Weiyang Sun Na Feng Songtao Yang Xianzhu Xia Ningyi Jin Yifa Zhou Yuwei Gao 2023Virologica Sinica2023,38,1:0
17Whole-genome and genome-wide association studies improve key agricultural traits of safflower for industrial and medicinal use显示文摘Safflower(Carthamus tinctorius)is widely cultivated around the world for its seeds and flowers.The presence of linoleic acid(LA)in its seeds and hydroxysafflor yellow A(HSYA)in its flowers are the crucial traits that enable safflower to be used for industrial and medicinal purposes.Understanding the genetic control of these traits is essential for optimizing the quality of safflower and its breeding.To further this research,we present a chromosome-scale assembly of the genome of the safflower variety‘Chuanhonghua 1’,which was achieved using an integrated strategy combining Illumina,Oxford Nanopore,and Hi-C sequencing.We obtained a 1.17-Gb assembly with a contig N50 of 1.08 Mb,and all assembled sequences were assigned to 12 pseudochromosomes.Safflower’s evolution involved the core eudicotγ-triplication event and a whole-genome duplication event,which led to large-scale genomic rearrangements.Extensive genomic shuffling has occurred since the divergence of the ancestor of dicotyledons.We conducted metabolite and transcriptome profiles with time-and part-dependent changes and screened candidate genes that significantly contribute to seed lipid biosynthesis.We also analyzed key gene families that participate in LA and HSYA biosynthesis.Additionally,we re-sequenced 220 safflower lines and carried out a genome-wide association study using high-quality SNP data for eight agronomic traits.We identified SNPs related to important traits in safflower.Besides,the candidate gene HH_034464(CtCGT1)was shown to be involved in the biosynthesis of HSYA.Overall,we provide a high-quality reference genome and elucidate the genetic basis of LA and HSYA biosynthesis in safflower.This vast amount of data will benefit further research for functional gene mining and breeding in safflower.Jiang Chen Shuai Guo Xueli Hu Rui Wang Donghai Jia Qiang Li Xianmei Yin Xuejiao Liao Zunhong Hu Peiqi Wang Chaoxiang Ren Shuai Dong Chao Chen Shilin Chen Jiang Xu Jin Pei 2023Horticulture Research2023,10,11:0
18Characterization of lipid-based nanomedicines at the single-particle level显示文摘Lipid-based nanomedicines(LBNMs),including liposomes,lipid nanoparticles(LNPs)and extracellular vesicles(EVs),are recognized as one of the most clinically acceptable nano-formulations.However,the bench-to-bedside translation efficiency is far from satisfactory,mainly due to the lack of in-depth understanding of their physical and biochemical attributes at the single-particle level.In this review,we first give a brief introduction of LBNMs,highlighting some milestones and related scientific and clinical achievements in the past several decades,as well as the grand challenges in the characterization of LBNMs.Next,we present an overview of each category of LB-NMs as well as the core properties that largely dictate their biological characteristics and clinical performance,such as size distribution,particle concentration,morphology,drug encapsulation and surface properties.Then,the recent applications of several analytical techniques including electron microscopy,atomic force microscopy,fluorescence microscopy,Raman microscopy,nanoparticle tracking analysis,tunable resistive pulse sensing and flow cytometry on the single-particle characterization of LBNMs are thoroughly discussed.Particularly,the com-parative advantages of the newly developed nano-flow cytometry that enables quantitative analysis of both the physical and biochemical characteristics of LBNMs smaller than 40 nm with high throughput and statistical ro-bustness are emphasized.The overall aim of this review article is to illustrate the importance,challenges and achievements associated with single-particle characterization of LBNMs.Chaoxiang Chen Chen Chen Yurou Li Ruilan Gu Xiaomei Yan 2023Fundamental Research2023,3,4:0
19Cloning,expression and activity analysises of chalcone synthase genes in Carthamus tinctorius显示文摘Objective:Flavonoids are the bioactive compounds in safflower(Carthamus tinctorius),in which chalcone synthase(CHS)is the first limiting enzyme.However,it is unclear that which chalcone synthase genes(CHSs)are participated in flavonoids biosynthesis in C.tinctorius.In this study,the CHSs in the molecular characterization and enzyme activities were investigated.Methods:Putative chalcone biosynthase genes were screened by the full-length transcriptome sequences data in C.tinctorius.Chalcone biosynthase genes in C.tinctorius(CtCHSs)were cloned from cDNA of flowers of C.tinctorius.The cloned gene sequences were analyzed by bioinformatics,and their expression patterns were analyzed by real-time PCR(RT-PCR).The protein of CtCHS in the development of flowers was detected by polyclonal antibody Western blot.A recombinant vector of CtCHS was constructed.The CtCHS recombinant protein was induced and purified to detect the enzyme reaction(catalyzing the reaction of p-coumaryl-CoA and malonyl-CoA to produce naringin chalcone).The reaction product was detected by HPLC and LC-MS.Results:Two full-length CtCHS genes were successfully cloned from the flowers of safflower(CtCHS1 and CtCHS3),with gene lengths of 1525 bp and 1358 bp,respectively.RT-PCR analysis showed that both genes were highly expressed in the flowers,but the expression of CtCHS1 was higher than that of CtCHS3 at each developmental stage of the flowers.WB analysis showed that only CtCHS1 protein could be detected at each developmental stage of the flowers.HPLC and LC-MS analyses showed that CtCHS1 could catalyze the conversion of p-coumaryl-CoA and malonyl-CoA substrates to naringin chalcone.Conclusion:CtCHS1 is involved in the biosynthesis of naringin chalcone in safflower.Xiaohui Tang Chaoxiang Ren Jing Hu Jiang Chen Jie Wang Rui Wang Qinghua Wu Wan Liao Jin Pei 2023Chinese Herbal Medicines2023,15,2:0
20Building multipurpose nano-toolkit by rationally decorating NIR-Ⅱ fluorophore to meet the needs of tumor diagnosis and treatment显示文摘Phototheranostics have attracted tremendous attention in cancer diagnosis and treatment because of the noninvasiveness and promising effectiveness.Developing advanced phototheranostic agents with long emission wavelength,excellent biocompatibility,great tumor-targeting capability,and efficient therapeutic effect is highly desirable.However,the mutual constraint between imaging and therapeutic functions usually hinders their wide applications in biomedical field.To balance this contradiction,we herein rationally designed and synthesized three novel tumor-targeted NIR-Ⅱ probes(QR-2PEG_(321),QR-2PEG_(1000),and QR-2PEG_(5000)) by conjugating three different chain lengths of PEG onto an integrin α_(v)β_(3)-targeted NIR-Ⅱ heptamethine cyanine fluorophore,respectively.In virtue of the essential amphiphilic characteristics of PEG polymers,these probes display various degree of aggregation in aqueous buffer accompanying with differential NIR-Ⅱ imaging and photothermal(PTT) therapeutic performance.Both in vitro and in vivo results have demonstrated that probe QR-2PEG_(5000) has the best NIR-Ⅱ imaging performance with prominent renal clearance,whereas QR-2PEG_(321)possesses excellent photoacoustic signal as well as PTT effect,which undoubtedly provides a promising toolbox for tumor diagnosis and therapy.We thus envision that these synthesized probes have great potential to be explored as a toolkit for precise diagnosis and treatment of malignant tumors.Chaoxiang Cui Jiachen Li Jing Fang Yan Zhao Yuqi Zhang Shuyue Ye Anna Wang Yali Feng Qiulian Mao Hongni Qin Haibin Shi 2022Chinese Chemical Letters2022,33,7:0
