Schistosoma japonicum infection induces macrophage polarization显示文摘The role of macrophages(Mφ)as the first line of host defense is well accepted. These cells play a central role in orchestrating crucial functions during schistosomal infection. Thus, understanding the functional diversity of these cells in the process of infection as well as the mechanisms underlying these events is crucial for developing disease control strategies. In this study,we adopted a Mφpolarization recognition system.Ml macrophage was characterized by expressing CD16/32,IL-12 and iNOS.M2 macrophage was characterized by expressing CD206,IL-10 and arg-1.In vivo(mouse peritoneal macrophages of different infection stages were obtained)and in vitro(different 5.japonicum antigens were used to stimulate RAW264.7)were characterized by using the above mentioned system.NCA and ACA stimulated RAW264.7 express significantly higher levels of IL-12 while significantly higher levels of IL-10 were detected after soluble egg antigen(SEA)stimulation. The results showed that dramatic changes of antigen in the microenvironment before and after egg production led to macrophage polarization.Furthermore, through TLR blocking experiments, the TLR4 signaling pathway was found to play a role in the process of macrophage polarization toward Ml.Our data suggest that macrophage polarization during S.japonicum infection had significant effects on host immune responses to S.japonicum. | Jingwei Xu Hao Zhang Lin Chen Donghui Zhang Minjun Ji Haiwei Wu Guanling Wu | 2014 | The Journal of Biomedical Research2014,28,4: | 20

Structural and functional insights into the TEAD-YAP complex in the Hippo signaling pathway显示文摘The control of organ size growth is one of the most fundamental aspects of life.In the past two decades,a highly conserved Hippo signaling pathway has been identified as a key molecular mechanism for governing organ size regulation. In the middle of this pathway is a kinase cascade that negatively regulates the downstream component Yes-associated protein(YAP)/transcriptional coactivator with PDZ-binding motif(TAZ)/Yorkie through phosphorylation.Phosphorylation of YAP/TAZ/Yorkie promotes its cytoplasmic localization, leads to cell apoptosis and restricts organ size overgrowth. When the Hippo pathway is inactivated,YAP/TAZ/Yorkie translocates into the nucleus to bind to the transcription enhancer factor(TEAD/TEF)family of transcriptional factors to promote cell growth and proliferation. In this review,we will focus on the structural and functional studies on the downstream transcription factor TEAD and its coactivator YAP. | Liming Chen Portia Gloria Loh Haiwei Song | 2010 | Protein & Cell2010,1,12: | 6

The impact of sample processing on the rapid antigen detection test for SARS-CoV-2: Virus inactivation, VTM selection, and sample preservation显示文摘Many factors have been identified as having the ability to affect the sensitivity of rapid antigen detection(RAD)tests for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).This study aimed to identify the impact of sample processing on the sensitivity of the RAD tests. We explored the effect of different inactivation methods, viral transport media(VTM)solutions, and sample preservation on the sensitivity of four RAD kits based on two SARS-CoV-2 strains.Compared with non-inactivation,heat inactivation significantly impacted the sensitivity of most RAD kits;however,β-propiolactone inactivation only had a minor effect. Some of the VTM solutions(VTM2,MANTACC)had a significant influence on the sensitivity of the RAD kits, especially for low viral-loads samples. The detection value of RAD kits was slightly decreased, while most of them were still in the detection range with the extension of preservation time and the increase of freeze–thaw cycles. Our results showed that selecting the appropriate inactivation methods and VTM solutions is necessary during reagent development, performance evaluation, and clinical application。 | Haiwei Zhou Conghui Wang Jian Rao Lan Chen Tingting Ma Donglai Liu Lili Ren Sihong Xu | 2021 | Biosafety and Health2021,3,5: | 3
Hypersonic flutter and flutter suppression system of a wind tunnel model显示文摘The aeroelastic behavior of a thin flat rudder model was numerically simulated and experimentally investigated in a hypersonic wind tunnel. In particular, a flutter suppression system taking advantage of collision within small gaps was proposed and a novel system for the flutter simulation of the whole nonlinear aeroelastic system including the flutter suppression system was developed. First, the critical flutter dynamic pressure of the rudder without the flutter suppression system was calculated with different methods. Then, the whole nonlinear aeroelastic system, including theflutter suppression system, was simulated to design the gap size. Finally, the flutter suppression system was experimentally validated in a hypersonic wind tunnel operating at Mach number 5. The typical phenomenon of Limit Cycle Oscillation(LCO) was observed, avoiding the structural failure of the model and the consistency between numerical and experimental results was demonstrated. The proposed suppression system can improve the design and reusability of test models of hypersonic flutter experiments. | Chuanwei XUAN Jinglong HAN Bing ZHANG Haiwei YUN Xiaomao CHEN | 2019 | Chinese Journal of Aeronautics2019,32,9: | 3

Enhanced hydrogen evolution reaction in Sr doped BiFeO_(3) by achieving the coexistence of ferroelectricity and ferromagnetism at room temperature显示文摘The perovskite transition metal oxide(TMO) has been considered in electrocatalysis for the modern clean energy technologies as its high electrochemical activity and low cost. The atomic scale engineering to the local stoichiometry of single crystal TMO provides a clue of the relation between electronic structure and catalytic performance. Here we report a hydrogen evolution reaction(HER) activity enhancement ~ 1761% of Bi_(0.85)Sr_(0.15)FeO_3 compared to the pure BiFeO_3. By the systemic investigation of the Sr doping level of Bi_(1-x)Sr_xFeO_3(BSFO), it is found that the HER enhancement originates from the improvement of ferromagnetism of BSFO without obvious scarification of the ferroelectricity at the room temperature. The multiple ferroic orderings in BSFO are beneficial for HER activity, which offers the strengthen of hybridization of Fe 3d and O2 p orbitals from the view of ferromagnetism, and the assistance of electron drift by spontaneous electric polarization. Our study not only affords the strategy of developing multiple ferroic orderings in TMO, but also facilitates the atomic scale understanding of the improved HER activity. | Ji Qi Huan Liu Ming Feng Hang Xu Haiwei Liu Chen Wang Aopei Wang Weiming Lu | 2021 | Journal of Energy Chemistry2021,30,2: | 2

Dynamics of the SARS-CoV-2 antibody response up to 10 months after infection显示文摘COVID-19 caused by SARS-CoV-2 infection has caused substantial morbidity and mortality worldwide and paralyzed the international economy.Understanding the magnitude and duration of the antibody response to SARS-CoV-2 is important to achieve a balance between curbing the pandemic and minimizing adverse effects on society.1 Although the antibody response to SARS-CoV-2 within 9 months has been extensively studied,2,3,4,5,6 little is known about the magnitude and kinetics of antibody responses for over 9 months. Moreover, with limited observations over 9 months(n<100),2,7,8 several studies have produced inconsistent conclusions about antibody dynamics, suggesting different rates of antiviral antibody positivity at the last follow-up.2,7,8 These studies have been limited by a lack of measurement of neutralizing antibodies(NAbs),7 of inclusion of mild or asymptomatic cases,2,8 and of further exploration of potential predisposing factors for antibody dynamics.2,7 Considering the individual heterogeneity(such as disease severity)8 and time-dependent nature1 of the immune response,in-depth characterization of SARS-CoV-2 antibody kinetics across disease severity groups over a long period is urgently needed. Therefore,we repeatedly tested IgM,IgG,viral spike protein receptor-binding dom(anti-RBD)IgG,and NAb titers in COVID-19 patients during a follow-up period of up to 10 months and explored potential predisposing factors of antibody titers during follow-up. | Hao Wang Yu Yuan Mingzhong Xiao Li Chen Youyun Zhao Haiwei Zhang Pinpin Long Yana Zhou Xi Xu Yanshou Lei Bihao Wu Tingyue Diao Hao Cai Li Liu Zuoyu Shao Jingzhi Wang Yansen Bai Kai Wang Miao Peng Linlin Liu Shi Han Fanghua Mei Kun Cai Yake Lei An Pan Chaolong Wang Rui Gong Xiaodong Li Tangchun Wu | 2021 | Cellular & Molecular Immunology2021,18,7: | 1

Two-Stage Lesion Detection Approach Based on Dimension-Decomposition and 3D Context显示文摘Lesion detection in Computed Tomography(CT) images is a challenging task in the field of computer-aided diagnosis. An important issue is to locate the area of lesion accurately. As a branch of Convolutional Neural Networks(CNNs),3D Context-Enhanced(3DCE) frameworks are designed to detect lesions on CT scans. The False Positives(FPs) detected in 3DCE frameworks are usually caused by inaccurate region proposals, which slow down the inference time.To solve the above problems,a new method is proposed,a dimension-decomposition region proposal network is integrated into 3DCE framework to improve the location accuracy in lesion detection.Without the restriction of 'anchors' on ratios and scales, anchors are decomposed to independent 'anchor strings'.Anchor segments are dynamically combined in accordance with probability, and anchor strings with different lengths dynamically compose bounding boxes.Experiments show that the accurate region proposals generated by our model promote the sensitivity of FPs and spend less inference time compared with the current methods. | Jiacheng Jiao Haiwei Pan Chunling Chen Tao Jin Yang Dong Jingyi Chen | 2022 | Tsinghua Science and Technology2022,27,1: | 1
Phase characterisation of metalenses显示文摘Metalenses have emerged as a new optical element or system in recent years, showing superior performance and abundant applications. However, the phase distribution of a metalens has not been measured directly up to now, hindering further quantitative evaluation of its performance. We have developed an interferometric imaging phase measurement system to measure the phase distribution of a metalens by taking only one photo of the interference pattern.Based on the measured phase distribution,we analyse the negative chromatic aberration effect of monochromatic metalenses and propose a feature size of metalenses.Different sensitivities of the phase response to wavelength between the Pancharatnam-Berry phase-based metalens and propagation phase-reliant metalens are directly observed in the experiment.Furthermore, through phase distribution analysis,it is found that the distance between the measured metalens and the brightest spot of focusing will deviate from the focal length when the metalens has a low nominal numerical aperture, even though the metalens is ideal without any fabrication error. We also use the measured phase distribution to quantitatively characterise the imaging performance of the metalens. Our phase measurement system will help not only designers optimise the designs of metalenses but also fabricants distinguish defects to improve the fabrication process, which will pave the way for metalenses in industrial applications. | Maoxiong Zhao Mu Ku Chen Ze-Peng Zhuang Yiwen Zhang Ang Chen Qinmiao Chen Wenzhe Liu Jiajun Wang Ze-Ming Chen Bo Wang Xiaohan Liu Haiwei Yin Shumin Xiao Lei Shi Jian-Wen Dong Jian Zi Din Ping Tsai | 2021 | Light(Science & Applications)2021,10,4: | 1

Correction method of airfoil thickness effect in hinge moment calculation of a folding wing显示文摘The influences of airfoil thickness on the aerodynamic loading distribution and the hinge moments of folding wing aircraft are presented in this work. The traditional panel method shows deficiencies in the calculation of folding wing's hinge moments. Thus, a thickness correction strategy for the aerodynamic model with CFD results is proposed, and an aeroelastic flight simulation platform is constructed based on the secondary development of ADAMS.Based on the platform, the developed aerodynamic model is verified, then the flight-folding process of the folding wing aircraft is simulated, and the influences of airfoil thickness on the results are investigated. Results show that the developed aerodynamic model can effectively describe the thickness effect of the folding wing.Airfoil thickness, which cannot be considered by the panel method, has a great influence on the hinge moments during the folding process, and the thickness correction has great significance in the calculation of folding wing's hinge moments. | Hao XU Jinglong HAN Haiwei YUN Xiaomao CHEN | 2020 | Chinese Journal of Aeronautics2020,33,3: | 1
