2Efficacy and Safety of Tofacitinib in Chinese Patients with Rheumatoid Arthritis显示文摘Background:Tofacitinib is an oral Janus kinase inhibitor for the treatment of rheumatoid arthritis (RA).This study assessed the efficacy and safety of tofacitinib in Chinese patients with RA enrolled in Phase 3and long-term extension (LTE)studies. Methods:ORAL Sync was a 1-year,randomized,placebo-controlled,Phase 3trial.Patients received tofacitinib 5 or 10 mg twice daily (BID)or placebo advanced to tofacitinib 5 or 10 mg BID at 3 or 6 months.All patients remained on >1background conventional synthetic disease-modifying antirheumatic drug.ORAL Sequel is an open-label LTE study (data-cut:March 2015;data collection and analyses were ongoing,and study database was not locked at the time of analysis;study was closed in 2017).Efficacy outcomes:American College of Rheumatology (ACR)20/50/70response rates and Disease Activity Score in 28 joints using erythrocyte sedimentation rate (DAS28-4[ESR]).Patient-and physician-reported outcomes:HealthAssessment Questionnaire-Disability Index (HAQ-DI),Patient and Physician Global Assessment of Arthritis,and pain (visual analog scale).Safety was assessed throughout. Results:ORAL Sync included 218 patients;192 were subsequently enrolled into ORAL Sequel.In ORAL Sync,more patients achieved ACR20(tofacitinib 5 mg BID,67.4%;10 mg BID,70.6%;placebo,34.1%)and DAS28-4(ESR)<2.6(tofacitinib 5 mg BID,7.1%; 10mg BID,13.1%;placebo,2,3%)with tofacitinib versus placebo at Month 6.Mean changes from baseline in HAQ-DI were greater with tofacitinib versus placebo at Month 6.In ORAL Sequel,efficacy was consistent to Month 48.Incidence rates for adverse events of special interest in tofacitinib-treated patients were similar to the global population. 3Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4
4Evidence for charge and spin density waves in single crystals of La_(3)Ni_(2)O_(7) and La_(3)Ni_(2)O_(6)显示文摘Charge and spin orders are intimately related to superconductivity in copper oxide superconductors.Elucidation of the competing orders in various nickel oxide compounds is crucial,given the fact that superconductivity has been discovered in Nd_(0.8)Sr_(0.2)NiO_(2)films.Herein,we report structural,electronic transport,magnetic,and thermodynamic characterizations of single crystals of La_(3)Ni_(2)O_(7)and La_(3)Ni_(2)O_(6).La_(3)Ni_(2)O_(7)is metallic with mixed Ni^(2+)and Ni^(3+)valent states.Resistivity measurements yield two transition-like kinks at~110 and 153 K.The kink at 153 K is further revealed from magnetization and specific heat measurements,indicative of the formation of charge and spin density waves.La_(3)Ni_(2)O_(6)single crystals obtained from the topochemical reduction of La_(3)Ni_(2)O_(7)are insulating and show an anomaly at~176 K on magnetic susceptibility.The transition-like behaviors of La_(3)Ni_(2)O_(7)and La_(3)Ni_(2)O_(6)are analogous to those observed in La_(4)Ni_(3)O_(10) and La_(4)Ni_(3)O_(8),suggesting that charge and spin density waves are a common feature in the ternary La-Ni-O system with mixed-valent states of nickel.Zengjia Liu Hualei Sun Mengwu Huo Xiaoyan Ma Yi Ji Enkui Yi Lisi Li Hui Liu Jia Yu Ziyou Zhang Zhiqiang Chen Feixiang Liang Hongliang Dong Hanjie Guo Dingyong Zhong Bing Shen Shiliang Li Meng Wang 2023Science China(Physics,Mechanics & Astronomy)2023,66,1:2

11Assembly dynamics of East Asian subtropical evergreen broadleaved forests: New insights from the dominant Fagaceae trees显示文摘The dominant species of a biome can be regarded as its genuine indicator.Evergreen broadleaved forests(EBLFs) in subtropical East Asia harbor high levels of species biodiversity and endemism and are vital to regional carbon storage and cycling.However,the historical assembly of this unique biome is still controversial.Fagaceae is the most essential family in East Asian subtropical EBLFs and its dominant species are vital for the existence of this biome.Here,we used the dominant Fagaceae species to shed light on the dynamic process of East Asian subtropical EBLFs overtime.Our results indicate high precipitation in summer and low temperature in winter are the most influential climatic factors for the distribution of East Asian subtropical EBLFs.Modern East Asian subtropical EBLFs did not begin to appear until 23 Ma,subsequently experienced a longlasting development in the Miocene and markedly deteriorated at about 4 Ma,driven jointly by orogenesis and paleoclimate.We also document that there is a lag time between when one clade invaded the region and when its members become dominant species within the region.This study may improve our ability to predict and mitigate the threats to biodiversity of East Asian subtropical EBLFs and points to a new path for future studies involving multidisciplinary methods to explore the assembly of regional biomes.Lisi Hai Xiao-Qian Li Jing-Bo Zhang Xiao-Guo Xiang Rui-Qi Li Florian Jabbour Rosa del COrtiz An-Ming Lu Zhi-Duan Chen Wei Wang 2022Journal of Integrative Plant Biology2022,64,11:1

17Biochar pyrolyzed at low temperature enhanced acidophilous plant growth by promoting rhizospheric microbes in a slightly alkaline urban soil显示文摘Biochar has been considered as a potential way to enhance acidophilous plant growth in alkaline soils.However,whether such enhancements are closely linked with biochar pyrolysis temperature and its improvements in rhizospheric soil fertility and microbial activities remains largely unknown.We performed a pot experiment to investigate changes in azalea(Rhodo-dendron)morphology and physiology,as well as its rhizosphere soil chemical and biological properties in a slightly alkaline urban soil under the amendment of biochars that pyrolyzed at three temperatures(i.e.,350,550 and 700°C).Our results showed that the effects of biochars on plant growth and soil properties depended on pyrolysis temperature.In comparison with the non-amended control,350 and 550°C biochars showed significant promotions on the azalea growth in terms of photosynthetic characteristics,biomass,root morphology,and N and P uptake.Whereas,700°C biochar showed an inhibi-tion effect on them.350°C biochar decreased soil pH and increased soil available P and K contents and the activities ofα-glucosidase,N-β-glucosaminidase,phosphatase and sulfatase.In addition,350°C biochar significantly enhanced bacterial 16S rRNA and fungal 18S rRNA gene abundances in the rhizosphere soil of azalea and mycorrhizal infection.Correlation analysis indicated that soil pH,available nutrients and fungal abundance had positive associations with the enhanced plant growth parameters.Therefore,our findings suggest that biochar produced at low temperature could be a feasible strategy for enhancing acidophilous azalea growth and improving urban soil quality.Huimin Zhou Xiaoying Li Chang Li Lisi He Xiaoqing Liu Zheng Xiao Xiaobo Sun Gaoling Shi Jiale Su Junhui Chen 2021Biochar2021,3,4:1
18The development of an iridium(Ⅲ) complex functionalized G-quadruplex probe for the stability of G-quadruplex and lifetime image in cytoplasm显示文摘G-quadruplex(G4) is widely known as a non-classical secondary structure of nucleic acid. With the indepth study of G4, it is an urgent need for a phosphorescent probe with a high G4 binding ability to evaluate the level of G4 in the cytoplasm. Thus, this study designed and synthesized Ir-PDP where an Ir(Ⅲ)complex was used as a phosphorescent emitter. Meanwhile, two installed PDPs(pyridostatin derivatives)were used to improve the combination ability with G4 and reduced the cytotoxicity of the Ir(Ⅲ) complex.Compared with other nucleic acid secondary structures, Ir-PDP produced a higher phosphorescence lifetime after interacting with G4. Ir-PDP was distributed in the cytoplasm of living cells, and two-photon phosphorescence lifetime imaging can detect the binding events of the probe in the cytoplasm. The addition of G4 binder PDS significantly regulated cytoplasmic phosphorescence lifetime. 19Luminescent gold(Ⅲ)exciplexes enable efficient multicolor electroluminescence显示文摘The control of excited states and related emissive properties of gold(Ⅲ)complexes mainly depends on the modulation of intramolecular electronic interactions among gold(Ⅲ)metal center,chelating ligands and/or peripheral groups.However,luminescent gold(Ⅲ)systems based on intermolecular electronic interactions have never been explored.Here we report a series of proof-of-concept gold(Ⅲ)exciplexes using a simple gold(Ⅲ)complex,AuDPPy,as an electron acceptor.The emissive properties of gold(Ⅲ)exciplexes can be regulated by combining AuDPPy with different donors.Inspiringly,these gold(Ⅲ)exciplexes have donor-dependent emission mechanisms:dominant phosphorescence or dual radiative channels of thermally activated delayed fluorescence(TADF)and phosphorescence.Consequently,these gold(Ⅲ)exciplexes deliver green-to-red electroluminescence with external quantum efficiencies(EQEs)of up to 10.1%.More importantly,using these gold(Ⅲ)exciplexes to host multi-resonance TADF emitters results in narrowband yellow,orange,and deep-red electroluminescence with high EQEs of 23.5%,24.4%,and 27.4%,respectively,competitive to the highest values for gold(Ⅲ)OLEDs in similar color gamut.Lisi Zhan Tianhao Chen Cheng Zhong Xiaosong Cao Yuewei Zhang Yang Zou Zhengyang Bin Jingsong You Dongdong Zhang Lian Duan Chuluo Yang Shaolong Gong 2023Science China Chemistry2023,66,11:0
20miR-103-3p regulates the differentiation of bone marrow mesenchymal stem cells in myelodysplastic syndrome显示文摘The pathogenesis of myelodysplastic syndrome(MDS)may be related to the abnormal expression of microRNAs(miRNAs),which could influence the differentiation capacity of mesenchymal stem cells(MSCs)towards adipogenic and osteogenic lineages.In this study,exosomes from bone marrow plasma were successfully extracted and identified.Assessment of miR-103-3p expression in exosomes isolated from BM in 34 MDS patients and 10 controls revealed its 0.52-fold downregulation in patients with MDS compared with controls(NOR)and was downregulated 0.55-fold in MDS-MSCs compared with NOR-MSCs.Transfection of MDS-MSCs with the miR-103-3p mimic improved osteogenic differentiation and decreased adipogenic differentiation in vitro,while inhibition of miR-103-3p showed the opposite results in NOR-MSCs.Thus,the expression of miR-103-3p decreases in MDS BM plasma and MDS-MSCs,significantly impacting MDS-MSCs differentiation.The miR-103-3p mimics may boost MDS-MSCs osteogenic differentiation while weakening lipid differentiation,thereby providing possible target for the treatment of MDS pathogenesis.NINGYU LI XIAOFANG CHEN SUXIA GENG PEILONG LAI LISI HUANG MINMING LI XIN HUANG CHENGXIN DENG YULIAN WANG JIANYU WENG XIN DU 2023BIOCELL2023,47,1:0
