2Human adipose tissue-and umbilical cord-derived stem cells: which is a better alternative to treat spinal cord injury?显示文摘Multiple types of stem cells have been proposed for the treatment of spinal cord injury,but their comparative information remains elusive.In this study,a rat model of T10 contusion spinal cord injury was established by the impactor method.Human umbilical cord-derived mesenchymal stem cells(UCMSCs)or human adipose tissue-derived mesenchymal stem cells(ADMSCs)(2.5μL/injection site,1×10^5 cells/μL)was injected on rostral and caudal of the injury segment on the ninth day after injury.Rats injected with mesenchymal stem cell culture medium were used as controls.Our results show that although transplanted UCMSCs and ADMSCs failed to differentiate into neurons or glial cells in vivo,both significantly improved motor and sensory function.After spinal cord injury,UCMSCs and ADMSCs similarly promoted spinal neuron survival and axonal regeneration,decreased glial scar and lesion cavity formation,and reduced numbers of active macrophages.BioPlex analysis of spinal samples showed a specific increase of interleukin-10 and decrease of tumor necrosis factorαin the ADMSC group,as well as a downregulation of macrophage inflammatory protein 3αin both UCMSC and ADMSC groups at 3 days after cell transplantation.Upregulation of interleukin-10 and interleukin-13 was observed in both UCMSC and ADMSC groups at 7 days after cell transplantation.Isobaric tagging for relative and absolute quantitation proteomics analyses showed that UCMSCs and ADMSCs induced changes of multiple genes related to axonal regeneration,neurotrophy,and cell apoptosis in common and specific manners.In conclusion,UCMSC and ADMSC transplants yielded quite similar contributions to motor and sensory recovery after spinal cord injury via anti-inflammation and improved axonal growth.However,there were some differences in cytokine and gene expression induced by these two types of transplanted cells.Animal experiments were approved by the Laboratory Animal Ethics Committee at Jinan University(approval No.20180228026)on February 28,2018,and the application of human stem cells was approved by the Medical Ethics Committee of Medical College of Jinan University of China(approval No.2016041303)on April 13,2016.Ai-Mei Liu Bo-Li Chen Ling-Tai Yu Tao Liu Ling-Ling Shi Pan-Pan Yu Yi-Bo Qu Kwok-Fai So Li-Bing Zhou 2020Neural Regeneration Research2020,15,12:6
3Insulin-like growth factor 1 partially rescues early developmental defects caused by SHANK2 knockdown in human neurons显示文摘SHANK2 is a scaffold protein that serves as a protein anchor at the postsynaptic density in neurons.Genetic variants of SHANK2 are strongly associated with synaptic dysfunction and the pathophysiology of autism spectrum disorder.Recent studies indicate that early neuronal developmental defects play a role in the pathogenesis of autism spectrum disorder,and that insulin-like growth factor 1 has a positive effect on neurite development.To investigate the effects of SHANK2 knockdown on early neuronal development,we generated a sparse culture system using human induced pluripotent stem cells,which then differentiated into neural progenitor cells after 3–14 days in culture,and which were dissociated into single neurons.Neurons in the experimental group were infected with shSHANK2 lentivirus carrying a red fluorescent protein reporter(shSHANK2 group).Control neurons were infected with scrambled sh Control lentivirus carrying a red fluorescent protein reporter(sh Control group).Neuronal somata and neurites were reconstructed based on the lentiviral red fluorescent protein signal.Developmental dendritic and motility changes in VGLUT1^+glutamatergic neurons and TH+dopaminergic neurons were then evaluated in both groups.Compared with sh Control VGLUT1^+neurons,the dendritic length and arborizations of shSHANK2 VGLUT1^+neurons were shorter and fewer,while cell soma speed was higher.Furthermore,dendritic length and arborization were significantly increased after insulin-like growth factor 1 treatment of shSHANK2 neurons,while cell soma speed remained unaffected.These results suggest that insulin-like growth factor 1 can rescue morphological defects,but not the change in neuronal motility.Collectively,our findings demonstrate that SHANK2 deficiency perturbs early neuronal development,and that IGF1 can partially rescue the neuronal defects caused by SHANK2 knockdown.All experimental procedures and protocols were approved by the Laboratory Animal Ethics Committee of Jinan University,China(approval No.20170228010)on February 28,2017.Shu-Ting Chen Wan-Jing Lai Wei-Jia Zhang Qing-Pei Chen Li-Bing Zhou Kwok-Fai So Ling-Ling Shi 2020Neural Regeneration Research2020,15,12:3
4Surgical intervention combined with weight-bearing walking training improves neurological recoveries in 320 patients with clinically complete spinal cord injury:a prospective self-controlled study显示文摘Although a large number of trials in the SCI field have been conducted,few proven gains have been realized for patients.In the present study,we determined the efficacy of a novel combination treatment involving surgical intervention and long-term weight-bearing walking training in spinal cord injury(SCI)subjects clinically diagnosed as complete or American Spinal Injury Association Impairment Scale(AIS)Class A(AIS-A).A total of 320 clinically complete SCI subjects(271 male and 49 female),aged 16–60 years,received early(≤7 days,n=201)or delayed(8–30 days,n=119)surgical interventions to reduce intraspinal or intramedullary pressure.Fifteen days post-surgery,all subjects received a weight-bearing walking training with the“Kunming Locomotion Training Program(KLTP)”for a duration of 6 months.The neurological deficit and recovery were assessed using the AIS scale and a 10-point Kunming Locomotor Scale(KLS).We found that surgical intervention significantly improved AIS scores measured at 15 days post-surgery as compared to the pre-surgery baseline scores.Significant improvement of AIS scores was detected at 3 and 6 months and the KLS further showed significant improvements between all pair-wise comparisons of time points of 15 days,3 or 6 months indicating continued improvement in walking scores during the 6-month period.In conclusion,combining surgical intervention within 1 month post-injury and weight-bearing locomotor training promoted continued and statistically significant neurological recoveries in subjects with clinically complete SCI,which generally shows little clinical recovery within the first year after injury and most are permanently disabled.This study was approved by the Science and Research Committee of Kunming General Hospital of PLA and Kunming Tongren Hospital,China and registered at ClinicalTrials.gov(Identifier:NCT04034108)on July 26,2019.Yansheng Liu Jia-Xin Xie Fang Niu Zhexi Xu Pengju Tan Caihong Shen Hongkun Gao Song Liu Zhengwen Ma Kwok-Fai So Wutian Wu Chen Chen Sujuan Gao Xiao-Ming Xu Hui Zhu 2021Neural Regeneration Research2021,16,5:3
5In vivo neuronal and astrocytic activation in somatosensory cortex by acupuncture stimuli显示文摘Acupuncture is a medical treatment that has been widely pra cticed in China for over 3000 years,yet the neural mechanisms of acupuncture are not fully understood.We hypothesized that neurons and astrocytes act independently and synergistically under acupuncture stimulation.To investigate this,we used two-photon in vivo calcium reco rding to observe the effects of acupuncture stimulation at ST36(Zusanli)in mice.Acupuncture stimulation in peripheral acupoints potentiated calcium signals of pyramidal neurons and astrocytes in the somatosensory cortex and resulted in late-onset calcium transients in astrocytes.Chemogenetic inhibition of neurons augmented the astrocytic activity.These findings suggest that acupuncture activates neuronal and astrocytic activity in the somatosensory co rtex and provide evidence for the involvement of both neurons and astrocytes in acupuncture treatment.Xiao-Yue Chang Kai Chen Tong Cheng Pui To Lai Li Zhang Kwok-Fai So Edward S.Yang 2022Neural Regeneration Research2022,17,11:2
6Lutein delays photoreceptor degeneration in a mouse model of retinitis pigmentosa显示文摘Retinitis pigmentosa is a retinal disease characterized by photoreceptor degeneration.There is currently no effective treatment for retinitis pigmentosa.Although a mixture of lutein and other antioxidant agents has shown promising effects in protecting the retina from degeneration,the role of lutein alone remains unclear.In this study,we administered intragastric lutein to Pde6brd10 model mice,which display degeneration of retinal photoreceptors,on postnatal days 17(P17)to P25,when rod apoptosis reaches peak.Lutein at the optimal protective dose of 200 mg/kg promoted the survival of photoreceptors compared with vehicle control.Lutein increased rhodopsin expression in rod cells and opsin expression in cone cells,in line with an increased survival rate of photoreceptors.Functionally,lutein improved visual behavior,visual acuity,and retinal electroretinogram responses in Pde6brd10 mice.Mechanistically,lutein reduced the expression of glial fibrillary acidic protein in Müller glial cells.The results of this study confirm the ability of lutein to postpone photoreceptor degeneration by reducing reactive gliosis of Müller cells in the retina and exerting anti-inflammatory effects.This study was approved by the Laboratory Animal Ethics Committee of Jinan University(approval No.LACUC-20181217-02)on December 17,2018.Hui-Jun Zhang Xiao-Bin Liu Xiong-Min Chen Qi-Hang Kong Yu-Sang Liu Kwok-Fai So Jian-Su Chen Ying Xu Xue-Song Mi Shi-Bo Tang 2022Neural Regeneration Research2022,17,7:2
