2Terrestrial carbon sinks in China and around the world and their contribution to carbon neutrality显示文摘Enhancing the terrestrial ecosystem carbon sink(referred to as terrestrial C sink) is an important way to slow down the continuous increase in atmospheric carbon dioxide(CO_(2)) concentration and to achieve carbon neutrality target.To better understand the characteristics of terrestrial C sinks and their contribution to carbon neutrality,this review summarizes major progress in terrestrial C budget researches during the past decades,clarifies spatial patterns and drivers of terrestrial C sources and sinks in China and around the world,and examines the role of terrestrial C sinks in achieving carbon neutrality target.According to recent studies,the global terrestrial C sink has been increasing from a source of (-0.2±0.9) Pg C yr^(-1)(1 Pg=1015g)in the 1960s to a sink of (1.9±1.1) Pg C yr^(-1) in the 2010s.By synthesizing the published data,we estimate terrestrial C sink of 0.20–0.25 Pg C yr^(-1) in China during the past decades,and predict it to be 0.15–0.52 Pg C yr^(-1) by 2060.The terrestrial C sinks are mainly located in the mid-and high latitudes of the Northern Hemisphere,while tropical regions act as a weak C sink or source.The C balance differs much among ecosystem types:forest is the major C sink;shrubland,wetland and farmland soil act as C sinks;and whether the grassland functions as C sink or source remains unclear.Desert might be a C sink,but the magnitude and the associated mechanisms are still controversial.Elevated atmospheric CO_(2) concentration,nitrogen deposition,climate change,and land cover change are the main drivers of terrestrial C sinks,while other factors such as fires and aerosols would also affect ecosystem C balance.The driving factors of terrestrial C sink differ among regions.Elevated CO_(2) concentration and climate change are major drivers of the C sinks in North America and Europe,while afforestation and ecological restoration are additionally important forcing factors of terrestrial C sinks in China.For future studies,we recommend the necessity for intensive and long-term ecosystem C monitoring over broad geographic scale to improve terrestrial biosphere models for accurately evaluating terrestrial C budget and its dynamics under various climate change and policy scenarios.Yuanhe Yang Yue Shi Wenjuan Sun Jinfeng Chang Jianxiao Zhu Leiyi Chen Xin Wang Yanpei Guo Hongtu Zhang Lingfei Yu Shuqing Zhao Kang Xu Jiangling Zhu Haihua Shen Yuanyuan Wang Yunfeng Peng Xia Zhao Xiangping Wang Huifeng Hu Shiping Chen Mei Huang Xuefa Wen Shaopeng Wang Biao Zhu Shuli Niu Zhiyao Tang Lingli Liu Jingyun Fang 2022Science China(Life Sciences)2022,65,5:32

5Land Use Effects on Soil Organic Carbon, Microbial Biomass and Microbial Activity in Changbai Mountains of Northeast China显示文摘Land use changes are known to alter soil organic carbon(SOC) and microbial properties, however, information about how conversion of natural forest to agricultural land use as well as plantations affects SOC and microbial properties in the Changbai Mountains of Northeast China is meager. Soil carbon content, microbial biomass carbon(MBC), basal respiration and soil carbon mineralization were studied in five selected types of land use: natural old-growth broad-leaved Korean pine mixed forest(NF); spruce plantation(SP) established following clear-cutting of NF; cropland(CL); ginseng farmland(GF) previously under NF; and a five-year Mongolian oak young forest(YF) reforested on an abandoned GF, in the Changbai Mountains of Northeast China in 2011. Results showed that SOC content was significantly lower in SP, CL, GF, and YF than in NF. MBC ranged from 304.4 mg/kg in CL to 1350.3 mg/kg in NF, which was significantly higher in the soil of NF than any soil of the other four land use types. The SOC and MBC contents were higher in SP soil than in CL, GF, and YF soils, yielding a significant difference between SP and CL. The value of basal respiration was also higher in NF than in SP, CL, GF, and YF. Simultaneously, higher values of the metabolic quotient were detected in CL, GF, and YF soils, indicating low substrate utilization of the soil microbial community compared with that in NF and SP soil. The values of cumulative mineralized carbon and potentially mineralized carbon(C0) in NF were significantly higher than those in CL and GF, while no significant difference was observed between NF and SP. In addition, YF had higher values of C0 and C mineralization rate compared with GF. The results indicate that conversion from NF into agricultural land(CL and GF) uses and plantation may lead to a reduction in soil nutrients(SOC and MBC) and substrate utilization efficiency of the microbial community. By contrast, soils below SP were more conducive to the preservation of soil organic matter, which was reflected in the comparison of microbial indicators among CL, GF, and YF land uses. 6Application of thyristor controlled phase shifting transformer excitation impedance switching control to suppress short-circuit fault current level显示文摘Short-circuit fault current suppression is a very important issue in modern large-interconnected power networks. Conventional short-circuit current limiters, such as superconducting fault current limiters, have to increase additional equipment investments. Fast power electronics controlled flexible AC transmission system(FACTS)devices have opened a new way for suppressing the fault current levels, while maintaining their normal functionalities for steady-state and transient power system operation and control. Thyristor controlled phase shifting transformer(TCPST) is a beneficial FACTS device in modern power systems, which is capable of regulating regional powerflow. The mathematical model for TCPST under different operation modes is firstly investigated in this study. Intuitively, the phase shifting angle control can adjust the equivalent impedance of TCPST, but the effect has been demonstrated to be weak. Therefore, a novel transformer excitation impedance switching(EIS) control method, is proposed for fault current suppressing, according to the impedance characteristics of TCPST. Simulation results on IEEE 14-bus system have shown considerable current limiting characteristic of the EIS control under various fault types. 7No association between elevated homocysteine levels and carotid atherosclerosis in a rural population in China显示文摘Objectives:We wish to determine if homocysteine(Hcy)is an independent risk factor for carotid atherosclerosis in a rural Chinese population.Methods:2291 individuals(1016 men and 1275 women),aged 64.6±7.4 years,from Lvliang,China participated in this study.Tests performed included carotid artery ultrasound scan and blood analysis,to measure Hcy levels and other blood components.Results:The mean serum Hcy level was 24.7±18.0μmol/L.The overall detection rate of carotid atherosclerotic lesions was 76.3%and the detection rate of plaque was 48.6%.Participants were divided into 4 groups based on their Hcy levels,<14.49,14.49–19.43,19.44–28.30,and≥28.30μmol/L.The relative risk of carotid atherosclerosis for each quartile as compared with the risk for the lowest quartile was estimated as the OR derived from the logistic regression coefficients.After adjusting for age and gender,the OR of carotid atherosclerosis in the third and fourth quartiles of Hcy were 1.219(95%CI 0.922 to 1.612)and 1.156(95%CI 0.859 to 1.555;p>0.05),respectively.After controlling for demographic variables(age,gender,current smoker,fasting blood glucose,low-density lipoprotein and systolic blood pressure)the OR of carotid plaque(s)in the third and fourth quartiles were 1.246(95%CI 0.967 to 1.606)and 1.259(95%CI 0.963 to 1.646;p>0.05).Conclusions:The mean value of Hcy among participants in this study was much higher than that previously reported.However,no significant correlation between elevated Hcy and carotid atherosclerosis was found.Yang Li Li Wang Wei Zhang Yalan Fang Xiaoyuan Niu 2016Stroke & Vascular Neurology2016,1,4:13
8Effects of Exposure to Aluminum on Long-term Potentiation and AMPA Receptor Subunits in Rats in vivo显示文摘Objective To explore the effects of exposure to aluminum(Al) on long-term potentiation(LTP) and AMPA receptor subunits in rats in vivo. Methods Different dosages of aluminum-maltolate complex [Al(mal)3] were given to rats via acute intracerebroventricular(i.c.v.) injection and subchronic intraperitoneal(i.p.) injection. Following Al exposure, the hippocampal LTP were recorded by field potentiation technique in vivo and the expression of AMPAR subunit proteins(GluR1 and GluR2) in both total and membrane-enriched extracts from the CA1 area of rat hippocampus were detected by Western blot assay. Results Acute Al treatment produced dose-dependent suppression of LTP in the rat hippocampus and dose-dependent decreases of GluR1 and GluR2 in membrane extracts; however, no similar changes were found in the total cell extracts, which suggests decreased trafficking of AMPA receptor subunits from intracellular pools to synaptic sites in the hippocampus. The dose-dependent suppressive effects on LTP and the expression of AMPA receptor subunits both in the membrane and in total extracts were found after subchronic Al treatment, indicating a decrease in AMPA receptor subunit trafficking from intracellular pools to synaptic sites and an additional reduction in the expression of the subunits. Conclusion Al(mal)3 obviously and dose-dependently suppressed LTP in the rat hippocampal CA1 region in vivo, and this suppression may be related to both trafficking and decreases in the expression of AMPA receptor subunit proteins. 10Generation of a Hutchinson-Gilford progeria syndrome monkey model by base editing显示文摘Many human genetic diseases,including Hutchinson-Gilford progeria syndrome(HGPS),are caused by single point mutations.HGPS is a rare disorder that causes premature aging and is usually caused by a de novo point mutation in the LMNA gene.Base editors(BEs)composed of a cytidine deaminase fused to CRISPR/Cas9 nickase are highly efficient at inducing C to T base conversions in a programmable manner and can be used to generate animal disease models with single amino-acid substitutions.Here,we generated the first HGPS monkey model by delivering a BE mRNA and guide RNA(gRNA)targeting the LMNA gene via microinjection into monkey zygotes.Five out of six newborn monkeys carried the mutation specifically at the target site.HGPS monkeys expressed the toxic form of lamin A,progerin,and recapitulated the typical HGPS phenotypes including growth retardation,bone alterations,and vascular abnormalities.Thus,this monkey model genetically and clinically mimics HGPS in humans,demonstrating that the BE system can efficiently and accurately generate patient-specific disease models in non-human primates.Fang Wang Weiqi Zhang Qiaoyan Yang Yu Kang Yanling Fan Jingkuan Wei Zunpeng Liu Shaoxing Dai Hao Li Zifan Li Lizhu Xu Chu Chu Jing Qu Chenyang Si Weizhi Ji Guang-Hui Liu Chengzu Long Yuyu Niu 2020Protein & Cell2020,11,11:11
11The lncRNA Malat1 functions as a ceRNA to contribute to berberine-mediated inhibition of HMGB1 by sponging miR-181c-5p in poststroke inflammation显示文摘Long non-coding RNAs(lncRNAs)have been identified as essential mediators in neurological dysfunction.Our previous study shows that berberine(BBR)hampers the nuclear-to-cytosolic translocation of high-mobility group box 1(HMGB1)in the process of poststroke inflammation.In this study,we explored the role of lncRNA metastasis‐associated lung adenocarcinoma transcript 1(Malat1)in the process of BBR-induced inhibition of HMGB1 in ischemic brain.Before the 60-min MCAO surgery,the mice were pretreated with BBR(50 mg·kg−1 per day,ig)for 14 days or ICV injected with specific lentiviral vector or shRNA.We showed that MCAO caused marked increase in the expression Malat1 and HMGB1 in the ipsilateral cortex,which was significantly attenuated by pretreatment with BBR.Knockdown of Malat1 attenuated the inflammatory injury after brain ischemia,whereas overexpression of Malat1 exacerbated ischemic brain inflammation.Overexpression of Malat1 also reversed BBR-induced reduction of HMGB1 and proinflammatory cytokines.The above results suggested a potential correlation between Malat1 and stroke inflammation.Based on informatics analysis we predicted that HMGB1 was a direct downstream target of miR-181c-5p,whereas Malat1 acted as a competitive endogenous RNA(ceRNA)for miR-181c-5p targeted the 3′-UTR of HMGB1 to promote inflammation after ischemic stroke.Knockdown of Malat1 significantly decreased HMGB1 level,which could be abrogated by transfection with miR-181c-5p inhibitors.Taken together,our results demonstrate for the first time that Malat1/miR-181c-5p/HMGB1 axis may be a key pathway of BBR-induced antiinflammation effects in stroke,and they may provide a novel avenue for targeted therapy.Ding-wen Cao Man-man Liu Rui Duan Yi-fu Tao Jun-shan Zhou Wei-rong Fang Jun-rong Zhu Li Niu Jian-guo Sun 2020Acta Pharmacologica Sinica2020,41,1:11
12Highly expressed BMP9/GDF2 in postnatal mouse liver and lungs may account for its pleiotropic effects on stem cell differentiation,angiogenesis,tumor growth and metabolism显示文摘Bone morphogenetic protein 9(BMP9)(or GDF2)was originally identified from fetal mouse liver cDNA libraries.Emerging evidence indicates BMP9 exerts diverse and pleiotropic functions during postnatal development and in maintaining tissue homeostasis.However,the expression landscape of BMP9 signaling during development and/or in adult tissues remains to be analyzed.Here,we conducted a comprehensive analysis of the expression landscape of BMP9 and its signaling mediators in postnatal mice.By analyzing mouse ENCODE transcriptome datasets we found Bmp9 was highly expressed in the liver and detectable in embryonic brain,adult lungs and adult placenta.We next conducted a comprehensive qPCR analysis of RNAs isolated from major mouse tissues/organs at various ages.We found that Bmp9 was highly expressed in the liver and lung tissues of young adult mice,but decreased in older mice.Interestingly,Bmp9 was only expressed at low to modest levels in developing bones.BMP9-associated TGFβ/BMPR type I receptor Alk1 was highly expressed in the adult lungs.Furthermore,the feedback inhibitor Smads Smad6 and Smad7 were widely expressed in mouse postnatal tissues.However,the BMP signaling antagonist noggin was highly expressed in fat and heart in the older age groups,as well as in kidney,liver and lungs in a biphasic fashion.Thus,our findings indicate that the circulating BMP9 produced in liver and lungs may account for its pleiotropic effects on postnatal tissues/organs although possible roles of BMP9 signaling in liver and lungs remain to be fully understood.Wei Liu Zhongliang Deng Zongyue Zeng Jiaming Fan Yixiao Feng Xi Wang Daigui Cao Bo Zhang Lijuan Yang Bin Liu Mikhail Pakvasa William Wagstaff Xiaoxing Wu Huaxiu Luo Jing Zhang Meng Zhang Fang He Yukun Mao Huiming Ding Yongtao Zhang Changchun Niu Rex C.Haydon Hue H.Luu Jennifer Moriatis Wolf Michael J.Lee Wei Huang Tong-Chuan He Yulong Zou 2020Genes & Diseases2020,7,2:9
13Endothelial Nitric Oxide Synthase Gene Polymorphisms Associated with Susceptibility to High Altitude Pulmonary Edema in Chinese Railway Construction Workers at Qinghai-Tibet over 4500 Meters above Sea Level显示文摘Objective To examine whether the polymorphisms of endothelial nitric oxide synthase (eNOS) gene are associated with the susceptibility to high altitude pulmonary edema (HAPE) in Chinese railway construction workers at Qinghai-Tibet where the altitude is over 4 500 m above sea level. Methods A case-control study was conducted including 149 HAPE patients in the construction workers and 160 healthy controls randomly recruited from their co-workers, matching the patients in ethnicity, age, sex, lifestyle, and working conditions. Three polymorphisms of eNOS gene, T-786C in promoter, 894G/T in exon 7, and 27bp variable number tandem repeat (VNTR) in intron 4, were genotyped using polymerase chain reaction (PCR) and confirmed with DNA sequencing. Results The frequencies of 894T allele and heterozygous G/T of the 894G/T variant were significantly higher in HAPE patients group than in the control group (P=0.0028 and P=0.0047, respectively). However, the frequencies of the T-786C in promoter and the 27bp VNTR in intron 4 were not significantly different between the two groups. Haplotypic analysis revealed that the frequencies of two haplotypes (H3,T-T-b, b indicates 5 repeats of 27 bp VNTR; H6, C-G-a, a indicates 4 repeats of 27 bp VNTR) were significantly higher in HAPE patients (both P<0.0001). On the contrary, the frequencies of H1 (T-G-b) and H2 (T-G-a) were lower in HAPE patients than in healthy controls (both P<0.001). Conclusions Two haplotypes (T-T-b and C-G-a) may be strongly associated with susceptibility to HAPE. 14The Relationship Between Neuroimmunity and Bipolar Disorder:Mechanism and Translational Application显示文摘Neuroimmune system may be involved in the pathological process of bipolar disorder(BD),but the essential association is not fully understood.Accumulating evidence has shown that BD involves the activation of immune cells and the release of inflammatory substances in the central nerve system(CNS).Meanwhile,neuroimmune responses also interact with other hypothesis of the etiology of BD that are widely recognized,such as neurotransmitter systems,neuroendocrine systems,neurotrophic factors,and oxidative stress.Simultaneously,related genes and immune changes in peripheral blood vary with it.Overall,neuroimmunity may play an important role in the pathogenesis of BD,and the inflammatory cytokines,especially interleukin-6 and tumor necrosis factor-alpha,have potential value for the clinical diagnosis and prognosis of BD,as well as predicting the therapeutic effects of drugs.Large-scale studies are needed to extend the evidence on neuroimmunity in BD,and to examine its clinical value for applications such as early prediction and treatment.Zhiang Niu Lu Yang Xiaohui Wu Yuncheng Zhu Jun Chen Yiru Fang 2019Neuroscience Bulletin2019,35,4:8

16Human pharmacokinetics of ginkgo terpene lactones and impact of carboxylation in blood on their platelet-activating factor antagonistic activity显示文摘Terpene lactones are a class of bioactive constituents of standardized preparations of Ginkgo biloba leaf extract, extensively used as add-on therapies in patients with ischemic cardiovascular and cerebrovascular diseases. This investigation evaluated human pharmacokinetics of ginkgo terpene lactones and impact of their carboxylation in blood. Human subjects received oral YinXing- TongZhi tablet or intravenous ShuXueNing, two standardized ginkgo preparations. Their plasma protein-binding and plateletactivating factor antagonistic activity were assessed in vitro. Their carboxylation was assessed in phosphate-buffered saline (pH 7.4) and in human plasma. After dosing YinXing-TongZhi tablet, ginkgolides A and B and bilobalide exhibited significantly higher systemic exposure levels than ginkgolides C and J; after dosing ShuXueNing, ginkgolides A, B, C, and J exhibited high exposure levels. The compounds’ unbound fractions in plasma were 45–92%. Apparent oral bioavailability of ginkgolides A and B was mostly >100%, while that of ginkgolides C and J was 6–15%. Bilobalide’s bioavailability was probably high but lower than that of ginkgolides A/B. Terminal half-lives of ginkgolides A, B, and C (4–7 h) after dosing ShuXueNing were shorter than their respective values (6–13 h) after dosing YinXing-TongZhi tablet. Half-life of bilobalide after dosing the tablet was around 5 h. Terpene lactones were roughly evenly distributed in various body fluids and tissues; glomerular-filtration-based renal excretion was the predominant elimination route for the ginkgolides and a major route for bilobalide. Terpene lactones circulated as trilactones and monocarboxylates. Carboxylation reduced platelet-activating factor antagonistic activity of ginkgolides A, B, and C. Ginkgolide J, bilobalide, and ginkgo flavonoids exhibited no such bioactivity. Collectively, differences in terpene lactones’ exposure between the two preparations and influence of their carboxylation in blood should be considered in investigating the relative contributions of terpene lactones to ginkgo preparations’ therapeutic effects. 17Effects of climate change on phenology and primary productivity in the desert steppe of Inner Mongolia显示文摘Variations in temperature and precipitation affect local ecosystems. Considerable spatial and temporal heterogeneity exists in arid ecosystems such as desert steppes. In this study, we analyzed the spatiotemporal dynamics of climate and vegetation phenology in the desert steppe of Inner Mongolia, China using meteorological data(1961–2010) from 11 stations and phenology data(2004–2012) from 6 ecological observation stations. We also estimated the gross primary production for the period of 1982–2009 and found that the annual mean temperature increased at a rate of 0.47oC/decade during 1961–2010, with the last 10 years being consistently warmer than the 50 years as an average. The most significant warming occurred in winters. Annual precipitation slightly decreased during the 50-year period, with summer precipitation experiencing the highest drop in the last 10 years, and spring precipitation, a rise. Spatially, annual precipitation increased significantly in the northeastern and eastern central areas next to the typical steppe. From 2004 to 2012, vegetation green-up and senescence date advanced in the study area, shortening the growing season. Consequently, the primary productivity of the desert steppe decreased along the precipitation gradient from southeast to northwest. Temporally, productivity increased during the period of 1982–1999 and significantly decreased after 2000. Overall, the last decade witnessed the most dramatic climatic changes that were likely to negatively affect the desert steppe ecosystem. 19Pulsed radiofrequency alleviated neuropathic pain by down-regulating the expression of substance P in chronic constriction injury rat model显示文摘Background:Pulsed radiofrequency(PRF),as a non-invasive treatment of neuropathic pain(NP),has been widely administered clinically.Previous studies have shown that PRF has the potential to improve hyperalgesia in animal models of NP.However,there have been few reports to clarify whether the mechanism of PRF treatment of NP involves intervention in the expression of substance P(SP).Therefore,this study administered PRF treatment to chronic constriction injury(CCI)model rats and observed the sciatic nerve mechanical pain threshold and SP expression in the spinal cord to explore the mechanism of PRF treatment.Methods:A total of 96 Sprague-Dawley rats were randomly divided into the sham-surgery-sham-treatment group(S-S group),the sham-surgery-PRF group(S-P group),the CCI-sham-treatment group(C-S group),and the CCI-PRF group(C-P group).The C-S group and the C-P group underwent sciatic nerve CCI,while the other groups received a sham operation.At 14 days after the operation,the C-P group and the S-P group were treated with PRF for 300 s.We recorded the hindpaw withdrawal threshold(HWT)and the thermal withdrawal latency(TWL)of rats in the various groups at baseline,before treatment(0 days),and at 1,7,14,and 28 days after treatment.L4 to L6 spinal cord tissues were taken before treatment(0 days)and 1,7,14,and 28 days after treatment.The transcription and translation of SP were measured by quantitative polymerase chain reaction and Western blotting,respectively.Results:The HWT and the TWL in the C-P group 28 days after PRF treatment were significantly higher than those in the C-S group(95%confidence interval[CI]:5.84–19.50,P<0.01;95%CI:2.58–8.69,P=0.01).The expression of SP in the C-P group 28 days after PRF treatment was significantly lower than that in the C-S group(95%CI:1.17–2.48,P<0.01).Conclusions:PRF may alleviate CCI-induced NP by down-regulating the expression of SP in the spinal cord of CCI model rats.Jing-Ai Wang Shao-Ning Niu Fang Luo 2020Chinese Medical Journal2020,,2:7
20Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd.Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu 2020Horticulture Research2020,7,1:7
