| 1 | Characterization of acid-and pepsin-soluble collagens from spines and skulls of skipjack tuna(Katsuwonus pelamis)显示文摘Acid-soluble collagen(ASC) and pepsin-soluble collagen(PSC) from the spine(ASC-SP and PSC-SP) and skull(ASC-SK and PSC-SK) of the skipjack tuna, Katsuwonus pelamis, were successfully isolated and characterized. The yields of ASC-SP, PSC-SP, ASC-SK and PSC-SK were(2.47 ± 0.39)%,(5.62 ± 0.82)%,(3.57 ± 0.40)%, and(6.71 ± 0.81)%, respectively, on the basis of dry weight. The four collagens contained Gly(330.2-339.1 residues/1 000 residues) as the major amino acid, and their imino acid contents were between 168.8 and 178.2 residues/1 000 residues. Amino acid composition, SDS-PAGE, and FTIR investigations confirmed that ASC-SP and ASC-SK were mainly composed of type I collagen, and had higher contents of high-molecular weight cross-links than those of PSC-SK and PSC-SP. The FTIR investigation also certified all the collagens had triple helical structure. The denaturation temperatures of ASC-SK, PSC-SK, ASC-SP, and PSC-SP were 17.8, 16.6, 17.6, and 16.5 °C, respectively. All isolated collagens were soluble at acidic pH(1-5) and lost their solubilities when the NaCl concentration was above 2%(W/V). | 3 | Priming of NLRP3 inflammasome activation by Msn kinase MINK1 in macrophages显示文摘The nucleotide-binding domain,leucine-rich-repeat containing family,pyrin domain-containing 3(NLRP3)inflammasome is essential in inflammation and inflammatory disorders.Phosphorylation at various sites on NLRP3 differentially regulates inflammasome activation.The Ser725 phosphorylation site on NLRP3 is depicted in multiple inflammasome activation scenarios,but the importance and regulation of this site has not been clarified.The present study revealed that the phosphorylation of Ser725 was an essential step for the priming of the NLRP3 inflammasome in macrophages.We also showed that Ser725 was directly phosphorylated by misshapen(Msn)/NIK-related kinase 1(MINK1),depending on the direct interaction between MINK1 and the NLRP3 LRR domain.MINK1 deficiency reduced NLRP3 activation and suppressed inflammatory responses in mouse models of acute sepsis and peritonitis.Reactive oxygen species(ROS)upregulated the kinase activity of MINK1 and subsequently promoted inflammasome priming via NLRP3 Ser725 phosphorylation.Eliminating ROS suppressed NLRP3 activation and reduced sepsis and peritonitis symptoms in a MINK1-dependent manner.Altogether,our study reveals a direct regulation of the NLRP3 inflammasome by Msn family kinase MINK1 and suggests that modulation of MINK1 activity is a potential intervention strategy for inflammasomerelated diseases. | Kaixiang Zhu Xuexiao Jin Zhexu Chi Sheng Chen Songquan Wu Richard D.Sloan Xuai Lin Dante Neculai Di Wang Hu Hu Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,10: | 2 |

The most appropriate reaction condition is: reaction T =673 K, n (DCT)∶ n (NH 3 )∶ n (air)=1∶7∶30 and v s=250 h -1 . At this optimum reaction condition, the conversion of DCT is 97.8%; the molar yield of DCBN is 67.4%. | 17 | Systematic investigation of millimeter-wave optic modulation performance in thin-film lithium niobate显示文摘Millimeter-wave(mmWave)band(30–300 GHz)is an emerging spectrum range for wireless communication,short-range radar,and sensor applications.mmWave-optic modulators that could efficiently convert mmWave signals into the optical domain are crucial components for long-haul transmission of mmWave signals through optical networks.At these ultrahigh frequencies,however,the modulation performances are highly sensitive to the transmission line loss as well as the velocity-and impedance-matching conditions,while precise measurements and modeling of these parameters are often non-trivial.Here we present a systematic investigation of the mmWave-optic modulation performances of thin-film lithium niobate modulators through theoretical modeling,electrical verifications,and electro-optic measurements at frequencies up to 325 GHz.Based on our experimentally verified model,we demonstrate thin-film lithium niobate mmWave-optic modulators with a measured 3-dB electro-optic bandwidth of 170 GHz and a 6-dB bandwidth of 295 GHz.The device also shows a low RF half-wave voltage of 7.3 V measured at an ultrahigh modulation frequency of 250 GHz.This work provides a comprehensive guideline for the design and characterization of mmWave-optic modulators and paves the way toward future integrated mmWave photonic systems for beyond-5G communication and radar applications. | YIWEN ZHANG LINBO SHAO JINGWEI YANG ZHAOXI CHEN KE ZHANG KAM-MAN SHUM DI ZHU CHI HOU CHAN MARKO LONČAR CHENG WANG | 2022 | Photonics Research2022,10,10: | 1 |
