2CHH methylation of genes associated with fatty acid and jasmonate biosynthesis contributes to cold tolerance in autotetraploids of Poncirus trifoliata显示文摘Polyploids have elevated stress tolerance,but the underlying mechanisms remain largely elusive.In this study,we showed that naturally occurring tetraploid plants of trifoliate orange(Poncirus trifoliata(L.) Raf.) exhibited enhanced cold tolerance relative to their diploid progenitors.Transcriptome analysis revealed that whole-genome duplication was associated with higher expression levels of a range of well-characterized cold stress-responsive genes.Global DNA methylation profiling demonstrated that the tetraploids underwent more extensive DNA demethylation in comparison with the diploids under cold stress.CHH methylation in the promoters was associated with up-regulation of related genes,whereas CG,CHG,and CHH methylation in the 3’-regions was relevant to gene down-regulation.Of note,genes involved in unsaturated fatty acids(UFAs) and jasmonate(JA)biosynthesis in the tetraploids displayed different CHH methylation in the gene flanking regions and were prominently up-regulated,consistent with greater accumulation of UFAs and JA when exposed to the cold stress.Collectively,our findings explored the difference in cold stress response between diploids and tetraploids at both transcriptional and epigenetic levels,and gained new insight into the molecular mechanisms underlying enhanced cold tolerance of the tetraploid.These results contribute to uncovering a novel regulatory role of DNA methylation in better cold tolerance of polyploids.Yue Wang Lanlan Zuo Tonglu Wei Yu Zhang Yang Zhang Ruhong Ming Dahro Bachar Wei Xiao Khan Madiha Chuanwu Chen Qijun Fan Chunlong Li Ji-Hong Liu 2022Journal of Integrative Plant Biology2022,64,12:2
3Dietary citrus pectin drives more ileal microbial protein metabolism and stronger fecal carbohydrate fermentation over fructooligosaccharide in growing pigs显示文摘Fructo-oligosaccharide(FOS)and pectin are known soluble dietary fibers and can influence gut microbiota and consequently modulate gut health.To understand the differential impact patterns of pectin vs.FOS in modulating gut microbiota in the small and large intestine,an ileal-cannulated pig model was adopted to compare the temporal and spatial effects of FOS and citrus pectin(CP)on the gut microbiota.Sixteen terminal ileal-cannulated pigs were randomly divided into 2 groups and fed with a standard diet supplemented with either 3% FOS or 3% CP for 28 d.The CP group and FOS group showed different microbial composition,especially in the feces,with time and location as major factors affecting microbiota in the CP group,and with only location contribution in the FOS group.In the feces,relative to the FOS group,the CP group showed higher abundance of Christensenellaceae R-7 group and Ruminococcaceae UCG-010 and lower abundance of Mitsuokella and Olsenella(adjusted P<0.05),a higher level of shortchain fatty acids and a lower level of lactate at both d 14 and 25(P<0.05),and more copy numbers of genes encoding key enzymes related to propionate(mmd A)and butyrate(BCo AT)production and lactate utilization(Lcd A)(P<0.05),indicating a greater degree of microbial carbohydrate fermentation.In the ileum,as compared with FOS,CP increased the bacteria with high capability of fermenting amino acids,including Escherichia-Shigella and Klebsiella(adjusted P<0.05),and the expression of enzymes responsible for amino acid fermentation(i.e.lysine decarboxylase),as well as the amino acid fermentation products(cadaverine and tyramine)(P<0.05),indicating a greater degree of amino acid fermentation.Overall,our results highlight a differential dynamic impact of dietary CP vs.FOS on microbial composition and metabolism in the gut.The dietary CP has a stronger ability to promote microbial amino acid fermentation in the ileum and carbohydrate fermentation in the feces than FOS.These findings provide a new insight into the role of different fibers in gut nutrition and guidelines for the choice of fibers in manipulating gut health.Yanan Zhang Chunlong Mu Shuai Liu Weiyun Zhu 2022Animal Nutrition2022,,4:2

6On the measurements of individual particle properties via compression and crushing显示文摘An experimental study is presented to measure the elastic,yielding,and crushing properties of individual particles under compression using substrates made of aluminum alloy,stainless steel,and sapphire.Carefully selected,highly spherical individual Ottawa sand particles of 0.75e1.1 mm in nominal diameter were compressed between two smooth substrates,and the loadedeformation curves were analyzed by Hertz elastic contact theory to derive their reduced modulus and Young’s modulus as well as yielding and crushing strengths,which vary significantly with the type of substrate materials.Further analysis of the yielding and plastic deformation at the particle-substrate contact shows that the yield strength or hardness of the substrate materials dominates the local contact behavior and hence affects the measured apparent yielding and crushing strengths.The two softer substrates(aluminum alloy and stainless steel)actually lead to underestimated apparent shear yield strengths of quartz particles by 60.4%and 54.2%,respectively,which are actually the yielding of substrates,while the true particle yielding occurs in the sapphire-particle contact.Moreover,the two softer substrates cause much overestimated crushing strengths of the quartz particles by 50.4%and 36.4%,respectively.Selection of inappropriate substrate materials and inappropriate interpretation of the particle-substrate contact can lead to significant errors in the measured yielding and crushing strengths.It is recommended that single particle compression testing uses substrates with yield strength greater than that of the tested particles and result interpretation also considers the elastic and yielding behaviors of the substrates.Chunlong Liu Fengyin Liu Jinliang Song Fuli Ma Dongfang Wang Guoping Zhang 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,2:1

11Gulong shale oil enrichment mechanism and orderly distribution of conventional–unconventional oils in the Cretaceous Qingshankou Formation,Songliao Basin,NE China显示文摘Through the study of organic matter enrichment,hydrocarbon generation and accumulation process of black shale of the Cretaceous Qingshankou Formation in the Songliao Basin,the enrichment mechanism of Gulong shale oil and the distribution of conventional–unconventional oil are revealed.The Songliao Basin is a huge interior lake basin formed in the Early Cretaceous under the control of the subduction and retreat of the western Pacific plate and the massive horizontal displacement of the Tanlu Fault Zone in Northeast China.During the deposition of the Qingshankou Formation,strong terrestrial hydrological cycle led to the lake level rise of the ancient Songliao Basin and the input of a large amount of nutrients,resulting in planktonic bacteria and algae flourish.Intermittent seawater intrusion events promoted the formation of salinization stratification and anoxic environment in the lake,which were beneficial to the enrichment of organic matters.Biomarkers analysis confirms that the biogenic organic matter of planktonic bacteria and algae modified by microorganisms plays an important role in the formation of high-quality source rocks with high oil generation capability.There are four favorable conditions for the enrichment of light shale oil in the Qingshankou Formation of the Gulong Sag,Songliao Basin:the moderate organic matter abundance and high oil potential provide sufficient material basis for oil enrichment;high degree of thermal evolution makes shale oil have high GOR and good mobility;low hydrocarbon expulsion efficiency leads to a high content of retained hydrocarbons in the source rock;and the confinement effect of intra-layer cement in the high maturity stage induces the efficient accumulation of light shale oil.The restoration of hydrocarbon accumulation process suggests that liquid hydrocarbons generated in the early(low–medium maturity)stage of the Qingshankou Formation source rocks accumulated in placanticline and slope after long-distance secondary migration,forming high-quality conventional and tight oil reservoirs.Light oil generated in the late(medium–high maturity)stage accumulated in situ,forming about 15 billion tons of Gulong shale oil resources,which finally enabled the orderly distribution of conventional–unconventional oils that are contiguous horizontally and superposed vertically within the basin,showing a complete pattern of“whole petroleum system”with conventional oil,tight oil and shale oil in sequence.ZHANG Shuichang ZHANG Bin WANG Xiaomei FENG Zihui HE Kun WANG Huajian FU Xiuli LIU Yuke YANG Chunlong 2023Petroleum Exploration and Development2023,50,5:0

14Human 8-cell embryos enable efficient induction of disease-preventive mutations without off-target effect by cytosine base editor显示文摘Approximately 140 million people worldwide are homozygous carriers of APOE4(ε4),a strong genetic risk factor for late onset familial and sporadic Alzheimer’s disease(AD),91%of whom will develop AD at earlier age than heterozygous carriers and noncarriers.Susceptibility to AD could be reduced by targeted editing of APOE4,but a technical basis for controlling the off-target effects of base editors is necessary to develop low-risk personalized gene therapies.Here,we first screened eight cytosine base editor variants at four injection stages(from 1-to 8-cell stage),and found that FNLS-YE1 variant in 8-cell embryos achieved the comparable base conversion rate(up to 100%)with the lowest bystander effects.In particular,80%of AD-susceptibleε4 allele copies were converted to the AD-neutralε3 allele in humanε4-carrying embryos.Stringent control measures combined with targeted deep sequencing,whole genome sequencing,and RNA sequencing showed no DNA or RNA off-target events in FNLS-YE1-treated human embryos or their derived stem cells.Furthermore,base editing with FNLS-YE1 showed no effects on embryo development to the blastocyst stage.Finally,we also demonstrated FNLS-YE1 could introduce known protective variants in human embryos to potentially reduce human susceptivity to systemic lupus erythematosus and familial hypercholesterolemia.Our study therefore suggests that base editing with FNLS-YE1 can efficiently and safely introduce known preventive variants in 8-cell human embryos,a potential approach for reducing human susceptibility to AD or other genetic diseases.Yinghui Wei Meiling Zhang Jing Hu Yingsi Zhou Mingxing Xue Jianhang Yin Yuanhua Liu Hu Feng Ling Zhou Zhifang Li Dongshuang Wang Zhiguo Zhang Yin Zhou Hongbin Liu Ning Yao Erwei Zuo Jiazhi Hu Yanzhi Du Wen Li Chunlong Xu Hui Yang 2023Protein & Cell2023,14,6:0
15Regulation of serotonin production by specific microbes from piglet gut显示文摘Background Serotonin is an important signaling molecule that regulates secretory and sensory functions in the gut.Gut microbiota has been demonstrated to affect serotonin synthesis in rodent models.However,how gut microbes regulate intestinal serotonin production in piglets remains vague.To investigate the relationship between microbiota and serotonin specifically in the colon,microbial composition and serotonin concentration were analyzed in ileum-cannulated piglets subjected to antibiotic infusion from the ileum when comparing with saline infusion.Microbes that correlated positively with serotonin production were isolated from piglet colon and were further used to investi-gate the regulation mechanisms on serotonin production in IPEC-J2 and a putative enterochromaffin cell line RIN-14B cells.Results Antibiotic infusion increased quantities of Lactobacillus amylovorus(LA)that positively correlated with increased serotonin concentrations in the colon,while no effects observed for Limosilactobacillus reuteri(LR).To understand how microbes regulate serotonin,representative strains of LA,LR,and Streptococcus alactolyticus(SA,enriched in feces from prior observation)were selected for cell culture studies.Compared to the control group,LA,LR and SA supernatants significantly up-regulated tryptophan hydroxylase 1(TPH1)expression and promoted serotonin production in IPEC-J2 cells,while in RIN-14B cells only LA exerted similar action.To investigate potential mechanisms mediated by microbe-derived molecules,microbial metabolites including lactate,acetate,glutamine,andγ-aminobutyric acid were selected for cell treatment based on computational and metabolite profiling in bacte-rial supernatant.Among these metabolites,acetate upregulated the expression of free fatty acid receptor 3 and TPH1 while downregulated indoleamine 2,3-dioxygenase 1.Similar effects were also recapitulated when treating the cells with AR420626,an agonist targeting free fatty acid receptor 3.Conclusions Overall,these results suggest that Lactobacillus amylovorus showed a positive correlation with sero-tonin production in the pig gut and exhibited a remarkable ability to regulate serotonin production in cell cultures.These findings provide evidence that microbial metabolites mediate the dialogue between microbes and host,which reveals a potential approach using microbial manipulation to regulate intestinal serotonin biosynthesis.Ziyu Liu Yidan Ling Yu Peng Shuibing Han Yuting Ren Yujia Jing Wenlu Fan Yong Su Chunlong Mu Weiyun Zhu 2023Journal of Animal Science and Biotechnology2023,14,6:0
16On fluorescence imaging:The diffusion equation model and recovery of the absorption coefficient of fluorophores显示文摘To quantify fluorescence imaging of biological tissues,we need to solve an inverse problem for the coupled radiative transfer equations which describe the excitation and emission fields in biological tissues.We begin by giving a concise mathematical argument to derive coupled diffusion equations with the Robin boundary condition as an approximation of the radiative transfer system.Then by using this coupled system of equations as a model for the fluorescence imaging,we have a nonlinear inverse problem to identify the absorption coefficient in this system.The associated linearized inverse problem is to ignore the absorbing effect on the excitation field.We firstly establish the estimates of errors on the excitation field and the solution to the inverse problem,which ensures the reasonability of the model approximation quantitatively.Some numerical verification is presented to show the validity of such a linearizing process quantitatively.Then,based on the analytic expressions of excitation and emission fields,the identifiability of the absorption coefficient from the linearized inverse problem is rigorously analyzed for the absorption coefficient in the special form,revealing the physical difficulty of the3-dimensional imaging model by the back scattering diffusive system.Jijun Liu Manabu Machida Gen Nakamura Goro Nishimura Chunlong Sun 2022Science China Mathematics2022,65,6:0

18Statistical characteristics of spray autoignition of transient kerosene jet in cross flow显示文摘To study statistical characteristics of the random spray autoignition,aviation kerosene was injected transiently into non-vitiated air crossflow in a flow reactor with optical accesses.The operating conditions were relevant to gas turbine combustor:the air crossflow pressure and temperature were in the range of 1.4-1.7 MPa and 830-947 K,respectively,and the jet-tocrossflow momentum flux ratios were 20,50 and 80.Statistical distributions of random ignition delay times with adequate convergence were estimated based on histograms.The dependences of the distributions on reactor pressure,temperature,and jet-to-crossflow momentum flux ratio were studied.The results show that the resulting distributions appear more concentrated with the increase of air temperature or jet-to-crossflow momentum flux ratio.And then the correlations for the mean and standard deviation of the ignition delay time sample data were developed based on the present results.Compared with the correlations of ignition delay time of homogeneous premixed gas-phase kerosene/air mixture reported in the literature,the results show a greater significance pressure dependence and lower temperature sensitivity of the ignition delay time of nonpremixed kerosene spray.Chi ZHANG Chunlong HE Xin XUE Yuzhen LIN Jibao LI Chongchong LIU 2021Chinese Journal of Aeronautics2021,34,12:0
19Investigation of solution chemistry to enable efficient lithium recovery from low-concentration lithium-containing wastewater显示文摘In the production of lithium-ion batteries(LIBs)and recycling of spent LIBs,a large amount of low-concentration lithium-containing wastewater(LCW)is generated.The recovery of Li from this medium has attracted significant global attention from both the environmental and economic perspectives.To achieve effective Li recycling,the features of impurity removal and the interactions among different ions must be understood.However,it is generally dificult to ensure highly efficient removal of impurity ions while retaining Li in the solution for further recovery.In this study,the removal of typical impurity ions from LCW and the interactions between these species were systematically investigated from the thermodynamic and kinetics aspects.It was found that the main impurities(e.g.,Fe^+,AIP^+,Ca^2+,and Mg^2+)could be efficiently removed with high Li recovery by control-ling the ionic strength of the solution.The mechanisms of Fe^3+,Al^+,Ca^2+,and Mg^2+removal were investigated to identify the controlling steps and reaction kinetics.It was found that the precipitates are formed by a zero-order reaction,and the activation energies tend to be low with a sequence of fast chemical reactions that reach equilibrium very quickly.Moreover,this study focused on Li loss during removal of the impurities,and the corresponding removal rates of Fe^+,Al^+,Ca^2+,and Mg^2+were found to be 99.8%,99.5%,99%,and 99.7%,respectively.Conse-quently,high-purity LisPO4 was obtained via one-step precipitation.Thus,this research demonstrates a potential route for the effective recovery of Li from low-concentra-tion LCW and for the appropriate treatment of acidic LCW.Chunlong Zhao Mingming He Hongbin Cao Xiaohong Zheng Wenfang Gao Yong Sun He Zhao Dalong Liu Yanling Zhang Zhi Sun 2020Frontiers of Chemical Science and Engineering2020,14,4:0
