Cloning and functional characterization of Rht8,a“Green Revolution”replacement gene in wheat显示文摘Dear Editor,Wheat(Triticum aestivum)is a major staple crop in the world and provides~20%of the food calories for human consumption(Appels et al.,2018).During the 1960s and 1970s,the introduction of two semi-dwarfing genes Rht-B1b and Rht-D1b in wheat varieties strikingly improved lodging resistance and harvest index, and therefore substantially increased grain yield, resulting in the well-known'Green Revolution'(Peng et al.,1999).However,plants carrying Rht-B1b and Rht-D1b suffer from several drawbacks. | Hongchun Xiong Chunyun Zhou Meiyu Fu Huijun Guo Yongdun Xie Linshu Zhao Jiayu Gu Shirong Zhao Yuping Ding Yuting Li Jiazi Zhang Ke Wang Xuejun Li Luxiang Liu | 2022 | Molecular Plant2022,15,3: | 7
A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults. Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors, built and examined the operating characteristics of a point-based algorithm for HCC risk prediction. Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells, concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. At 4.5 points, only 20.86% of participants in training cohort and 23.73% in validation cohort were recognized at high risk, with positive prediction value of 22.85% and 12.35%, respectively. Conclusions: Male-ABCD algorithm identified individual's risk for HCC occurrence within short term for their HCC precision surveillance. | Yuting Wang Minjie Wang He Li Kun Chen Hongmei Zeng Xinyu Bi Zheng Zhu Yuchen Jiao Yong Wang Jian Zhu Hui Zhao Xiang Liu Chunyun Dai Chunsun Fan Can Zhao Deyin Guo Hong Zhao Jianguo Zhou Dongmei Wang Zhiyuan Wu Xinming Zhao Wei Cui Xuehong Zhang Jianqiang Cai Wanqing Chen Chunfeng Qu | 2021 | Chinese Journal of Cancer Research2021,33,3: | 3
Effect of roughness on wettability and floatability:Based on wetting film drainage between bubbles and solid surfaces显示文摘Wetting film thinning measurement was introduced to clarify the wettability and floatability of solid surfaces with varying roughness. The wettability was quantified using the contact angle measurement combined with the dynamic force microbalance test between solid surfaces and water droplets, while the floatability was investigated by the bubble-solid surface dynamic attachment observation and the induction time measurement. The results show that the water contact angles reduce(14.53°, 12.74°, and 6.71°)with the increase of glass surface roughness, while the water droplet-glass adhesion forces intensify(11.1, 19.1 and 19.2 μN) owing to the stable wetting film. The distortion of the contact surface and the Wenzel state are the causes. In contrast, the hydrophobized surfaces have the growing apparent contact angles(38.08°, 69.81°, and 81.01°), declining adhesion strength and shortening induction time(863, 352and 12 ms) along with the increasing surface roughness. The weak wettability and fine floatability on the rough hydrophobized surface is reflected in the fast wetting film drainage dynamics and three-phase contact formation, which may be attributed to the wetting film with short diameter on tiny rough nubs and the entrapped air in the grooves as a bridge between the bulk bubble and the solid surface. | Ming Li Yaowen Xing Chunyun Zhu Qinshan Liu Zili Yang Rui Zhang Youfei Zhang Yangchao Xia Xiahui Gui | 2022 | International Journal of Mining Science and Technology2022,32,6: | 1

The roles and regulation of Yes-associated protein 1 in stem cells显示文摘Yes-associated protein 1(YAP1)is a downstream effector of the Hippo signaling pathway, and it is involved in tumorigenesis, tissue repair, growth, and development. In this review, the biological roles and the mechanisms of YAP1 in mediating stem cell fate decisions are discussed, including cell proliferation, differentiation, and apoptosis. In general,YAP1 promotes the proliferation and differentiation of stem cells, including embryonic stem cells and adult stem cells. It inhibits apoptosis by binding to the transcription factors,e.g.,transcriptional enhanced associate domain(TEAD),Smad,runt-related transcription factor 1/2,p73,p63,and Erb84,to maintain tissue homeostasis. The translocalization of YAP1 in cellular nuclei and the phosphorylation in the cytoplasm work as important and unusual events for the activation of YAP1.Moreover,YAP1 serves as the crosstalk for the Hippo pathway and other signaling pathways, including the Wnt and Notch pathways. It is highlighted in this review that YAP1 is an essential regulator for stem cells that have significant applications in regenerative medicine and reproductive medicine. | QING YIN CAIHONG LIU WENYING JIANG HAIHUI GONG CHUNYUN LI ZUPING HE | 2023 | BIOCELL2023,47,1: | 0
Synthesis and Luminescence Properties of SBA-15 Functionalized with Covalently Bonded Terbium-Benzoic Acid Complex显示文摘Novel hybrid materials containing covalently bonded Terbium-benzoic acid complex in mesoporous silicaSBA-15 (denoted as Tb-SBA-15 ) were prepared via co-condensation of tetrethoxysilane (TEOS) and N-(4-benzoicacid-yl), N'-(propyltriethoxysilyl) urea (denoted as PABI).XRD, FTIR and luminescence spectroscopy were employed to characterize Tb-SBA-15.When monitored by the ligand absorption wavelength (270 nm), Tb-SBA-15 displays the emission of Tb3+ (5D4→7Fj (j = 6, 5, 4, 3 ) transition) due to the energy transfer from the ligands to Tb3+. | Peng Chunyun Zhang Hongjie Liu Fengwei Meng Qinguo Fu Lianshe Sun Lining Guo Junfang Yu Jiangbo | 2004 | Journal of Rare Earths2004,22,z1: | 0
Recent advances of intermolecular Diels–Alder reaction in bio-inspired synthesis of natural products显示文摘The Diels–Alder(D–A)reaction is one of the most powerful reactions in organic synthesis. The intermolecular D–A reaction attracts much less attentions than its intramolecular counterpart in natural product synthesis, possibly due to the issues of reactivity and selectivity. In the past decade, the intermolecular D–A reaction has been increasingly utilized in the total synthesis of structurally complex natural products. In this article,we present a few examples for the elegant applications of the intermolecular D–A reaction that are inspired by biosynthetic hypotheses of the target natural products. These examples demonstrate that D–A reaction is not only useful for preparing building blocks but also powerful for coupling structurally complicated and sterically demanding segments in a highly chemo-and stereo-controlled fashion, which may inspire further developments of intermolecular D–A reaction from both strategy and methodology perspectives. | WAN ChunYun DENG Jun LIU Hua BIAN Ming LI Ang | 2014 | Science China Chemistry2014,57,7: | 0
Promotion effect of niobium on ceria catalyst for selective catalytic reduction of NO with NH3显示文摘The CeO_(2),Ce-Nb-O_(x) and Nb_(2)O_(5) catalysts were synthesized by citric acid method and the promotion effect of Nb on ceria for selective catalytic reduction(SCR)of NO with NH_(3) was investigated. The catalytic activity measurements indicate that the mixed oxide Ce-Nb-O_(x) presents a higher SCR activity than the single oxide CeO_(2) or Nb_(2)O_(5) catalyst. In addition, the Ce-Nb-O_(x) catalyst shows high resistance towards H_(2)O and SO_(2) at 280℃.The Raman,X-ray photoelectron spectra and temperature programmed reduction with H_(2) results indicate that the incorporation of Nb provides abundant oxygen vacancies for capturing more surface adsorbed oxygen, which provides a superior redox capability and accelerates the renewal of active sites.Furthermore, the Fourier transform infrared spectra and temperature programmed desorption of NH_(3) results suggest that niobium pentoxide shows high surface acidity, which is partly retained in the Ce-Nb-O_(x) catalyst possessing a high content of Lewis and Br?nsted acid sites. Therefore, the incorporation of Nb improves both the redox and acidic capacities of Ce-Nb-O_(x) catalyst for the SCR reaction. Here, the redox behavior is primarily taken on Ce and the acidity is well improved by Nb,so the synergistic effect should exist between Ce and Nb.In terms of the reaction mechanism,in situ DRIFT experiments suggest that both NH_(3) on Lewis acid sites and NH_(4)^(+) on Bronsted acid sites can react with NO species, and adsorbed NO and NO_(2) species can both be reduced by NH_(3).In the SCR process,O_(2) primarily acts as the accelerant to improve the redox and acid cycles and plays an important role. This work proves that the combination of redox and acidic properties of different constituents can be feasible for catalyst design to obtain a superior SCR performance. | Bolin Zhang Lifeng Deng Michael Liebau Yingjie Ren Chunyun Luo Bo Liu Shengen Zhang Roger Glaser | 2022 | Journal of Rare Earths2022,40,10: | 0
Fasudil-modified macrophages reduce inflammation and regulate the immune response in experimental autoimmune encephalomyelitis显示文摘Multiple sclerosis is characterized by demyelination and neuronal loss caused by inflammatory cell activation and infiltration into the central nervous system.Macrophage polarization plays an important role in the pathogenesis of experimental autoimmune encephalomyelitis,a traditional experimental model of multiple sclerosis. This study investigated the effect of Fasudil on macrophages and examined the therapeutic potential of Fasudil-modified macrophages in experimental autoimmune encephalomyelitis. We found that Fasudil induced the conversion of macrophages from the pro-inflammatory M1 type to the anti-inflammatory M2 type,as shown by reduced expression of inducible nitric oxide synthase/nitric oxide,interleukin-12,and CD16/32 and increased expression of arginase-1,interleukin-10,CD14,and CD206,which was linked to inhibition of Rho kinase activity, decreased expression of toll-like receptors, nuclear factor-κB,and components of the mitogen-activated protein kinase signaling pathway, and generation of the pro-inflammatory cytokines tumor necrosis factor-α,interleukin-1β,and interleukin-6.Crucially,Fasudil-modified macrophages effectively decreased the impact of experimental autoimmune encephalomyelitis, resulting in later onset of disease, lower symptom scores, less weight loss, and reduced demyelination compared with unmodified macrophages. In addition,Fasudil-modified macrophages decreased interleukin-17 expression on CD4^(+)T cells and CD16/32,inducible nitric oxide synthase, and interleukin-12 expression on F4/80^(+)macrophages,as well as increasing interleukin-10 expression on CD4^(+)T cells and arginase-1,CD206,and interleukin-10 expression on F4/80^(+)macrophages,which improved immune regulation and reduced inflammation. These findings suggest that Fasudil-modified macrophages may help treat experimental autoimmune encephalomyelitis by inducing M2 macrophage polarization and inhibiting the inflammatory response, thereby providing new insight into cell immunotherapy for multiple sclerosis. | Chunyun Liu Shangde Guo Rong Liu Minfang Guo Qing Wang Zhi Chai Baoguo Xiao Cungen Ma | 2024 | Neural Regeneration Research2024,19,3: | 0
Origin and evolution of fatty acid desaturase genes in oil crop Brassica napus显示文摘Fatty acid(FA)desaturases,as the key enzymes in lipid metabolism, are responsible for biosynthesis of the unsaturated fatty FAs,which play important roles in maintaining cell membrane integrity and multiple stress responses.Although attention has been drawn to some plant FA desaturase genes, their global landscape in oil crops is still lacking. Here,we performed systematic characterization and phylogenomic synteny network analyses of the FA desaturase gene family in polyploid oil crop B.napus and other 54 species covering major streptophyte lineages.A total of 1653 FA desaturase genes were identified from these plant genomes.Based on the broad-scale family phylogeny and functional domains,we proposed a unified eight-group classification system for angiosperm FA desaturases, and found that the origin of genes responsible for FA desaturation evolved early and some genes were absent in different species.Phylogenomic analyses revealed deeply conserved syntenic relationships within each of the eight FA desaturase groups.B.napus contains up to 93 FA desaturase genes from the eight groups.Recurrent duplication events in Brassicaceae contributed to the expansion of FA desaturase genes in B.napus, leading to further functional diversification. These FA desaturase genes exhibited spatio-temporal specific expression patterns in different tissues of B.napus, and a set of FA desaturase genes seem to be orchestrated by key transcriptional factors during seed development, such as zf-HD,B3,GATA3,PEI1,NFYA7,YAB1 and YAB2.Altogether,our data have inferred the evolutionary trajectory of this important gene family across distinct plant lineages, providing theoretical basis for future manipulation of FA desaturase genes to improve the seed oil quality of B.napus. | Wei Zhao Lunwen Qian Mei Guan Jun Liu Chunyun Guan | 2022 | Oil Crop Science2022,7,4: | 0
