3Chronic niacin overload may be involved in the increased prevalence of obesity in US children显示文摘AIM: To investigate nicotinamide’s action on glucose metabolism, and the association between niacin consumption and obesity prevalence. METHODS: Dynamic nicotinamide’s effect on plasma hydrogen peroxide and glucose metabolism was in- vestigated using oral glucose tolerance tests with or without nicotinamide in the same five healthy subjects.Lag-regression analysis was used to examine the association between the niacin consumption and the obesity prevalence among US children using the data from the Economic Research Service of the US Department of Agriculture and from US Centers for Disease Control and Prevention, respectively. RESULTS: Compared with the control oral glucose tolerance test, the 1-h plasma hydrogen peroxide (1.4 ± 0.1 μmol/L vs 1.6 ± 0.1 μmol/L, P = 0.016) and insulin levels (247.1 ± 129.0 pmol/L vs 452.6 ± 181.8 pmol/L, P = 0.028) were signif icantly higher, and the 3-h blood glucose was signif icantly lower (5.8 ± 1.2 mmol/L vs 4.5 ± 1.1 mmol/L, P = 0.002) after coadministration of glucose and 300 mg nicotinamide. The obesity prevalence among American children increased with the increasing per capita niacin consumption, the increasing grain contribution to niacin due to niacin-fortification, and the increasing niacin-fortified ready-to-eat cereal consumption, with a 10-year lag. The regression analyses showed that the obesity prevalence in the US children of all age groups was determined by niacin consumption (R2 = 0.814, 0.961 and 0.94 for 2-5 years, 6-11 years and 12-19 years age groups, respectively). CONCLUSION: The appetite-stimulating effect of nicotinamide appears to involve oxidative stress. 4Nicotinamide overload may play a role in the development of type 2 diabetes显示文摘AIM: To investigate whether nicotinamide overload plays a role in type 2 diabetes. METHODS: Nicotinamide metabolic patterns of 14diabetic and 14 non-diabetic subjects were compared using HPLC. Cumulative effects of nicotinamide and N1-methylnicotinamide on glucose metabolism, plasma H2O2 levels and tissue nicotinamide adenine dinucleotide (NAD) contents of adult Sprague-Dawley rats were observed. The role of human sweat glands and rat skin in nicotinamide metabolism was investigated using sauna and burn injury, respectively. RESULTS: Diabetic subjects had significantly higher plasma N 1-methylnicotinamide levels 5 h after a 100-mg nicotinamide load than the non-diabetic subjects (0.89 ± 0.13 μmol/L vs 0.6 ± 0.13 μmol/L, P < 0.001). Cumulative doses of nicotinamide (2 g/kg) significantly increased rat plasma N 1-methy lnico-tinamide concentrations associated with severe insulin resistance, which was mimicked by N1-methy-lnicotinamide. Moreover, cumulative exposure to N1-methylnicotinamide (2 g/kg) markedly reduced rat muscle and liver NAD contents and erythrocyte NAD/NADH ratio, and increased plasma H2O2 levels. Decrease in NAD/NADH ratio and increase in H2O2 generation were also observed in human erythrocytes after exposure to N1-methylnicotinamide in vitro. Sweating eliminated excessive nicotinamide (5.3-fold increase in sweat nicotinamide concentration 1 h after a 100-mg nicotinamide load). Skin damage or aldehyde oxidase inhibition with tamoxifen or olanzapine, both being notorious for impairing glucose tolerance, delayed N1-methylnicotinamide clearance. 5A specific objective supplemental factor in evaluating acute mountain sickness: ΔHR in combination with SaO_2显示文摘Background: So far, there have been no measurements confirmed useful in diagnosing acute mountain sickness(AMS). The aim of this study was to determine the role of heart rate(HR) difference(ΔHR) and oxygen saturation(Sa O2) as objective risk factors in aiding the diagnosis of AMS.Methods: A total of 1,019 participants were assigned to either the acute exposure group(AEG): from 500 m to 3,700 m by flight within 2.5 hours(n=752); or the pre-acclimatization group(PAG): ascended to 4,400 m from 3,650 m within three hours by car after adapting 33 days at 3,650m(n=267). The questionnaires or measurements of resting Sa O2 and HR were completed between 18 and 24 hours before departure and after arrival.Results: Incidence of AMS was 61.3%(461) in AEG, with 46.1%(347) mild cases and 15.2%(114) severe cases. In PAG, the incidence was 38.9%(104), with 30.7%(82) mild cases and 8.2%(22) severe cases. The AMS subjects showed a significant increase in HR and a decrease in Sa O2 levels compared with the non-AMS subjects in both groups. ΔHR and post-exposure Sa O2 were significantly correlated with the Lake Louise Score(LLS) in both groups. Stepwise logistic regression analysis revealed the ΔHR >25 and Sa O2 <88% in AEG as well as ΔHR >15 and Sa O2 <86% in PAG to be independent risk factors of AMS. Combining these two measurements could specifically indicate participants with AMS, which showed a positive predictive value of 89% and specificity of 97% in AEG as well as 85% and 98% in PAG.Conclusion:ΔHR or Sa O2, as objective measurements, correlate with AMS. 7PECS: Towards Personalized Edge Caching for Future Service-Centric Networks显示文摘Mobile operators face the challenge of how to best design a service-centric network that can effectively process the rapidly increasing number of bandwidth-intensive user requests while providing a higher quality of experience(QoE). Existing content distribution networks(CDN) and mobile content distribution networks(mCDN) have both latency and throughput limitations due to being multiple network hops away from end-users. Here, we first propose a new Personalized Edge Caching System(PECS) architecture that employs big data analytics and mobile edge caching to provide personalized service access at the edge of the mobile network. Based on the proposed system architecture, the edge caching strategy based on user behavior and trajectory is analyzed. Employing our proposed PECS strategies, we use data mining algorithms to analyze the personalized trajectory and service usage patterns. Our findings provide guidance on how key technologies of PECS can be employed for current and future networks. 8Interleukin-34 deficiency aggravates development of colitis and colitis-associated cancer in mice显示文摘BACKGROUND Although expression of interleukin(IL)-34 is upregulated in active ulcerative colitis(UC),the molecular function and underlying mechanism are largely unclear.AIM To investigate the function of IL-34 in acute colitis,in a wound healing model and in colitis-associated cancer in IL-34-deficient mice.METHODS Colitis was induced by administration of dextran sodium sulfate(DSS),and carcinogenesis was induced by azoxymethane(AOM).Whether the impact of IL-34 on colitis was dependent on macrophages was validated by depletion of macrophages in a murine model.The association between IL-34 expression and epithelial proliferation was studied in patients with active UC.RESULTS IL-34 deficiency aggravated murine colitis in acute colitis and in wound healing phase.The effect of IL-34 on experimental colitis was not dependent on macrophage differentiation and polarization.IL-34-deficient mice developed more tumors than wild-type mice following administration of AOM and DSS.No significant difference was shown in degree of cellular differentiation in tumors between wild-type and IL-34-deficient mice.IL-34 was dramatically increased in the active UC patients as previously reported.More importantly,expression of IL-34 was positively correlated with epithelial cell proliferation in patients with UC.CONCLUSION IL-34 deficiency exacerbates colonic inflammation and accelerates colitis-associated carcinogenesis in mice.It might be served as a potential therapeutic target in UC.Zhao-Xiu Liu Wei-Jie Chen Yang Wang Bing-Qian Chen Yi-Cun Liu Tiao-Chun Cheng Lei-Lei Luo Lin Chen Lin-Ling Ju Yuan Liu Ming Li Nan Feng Jian-Guo Shao Zhao-Lian Bian 2022World Journal of Gastroenterology2022,28,47:3
9Enrichment factors of movable hydrocarbons in lacustrine shale oil and exploration potential of shale oil in Gulong Sag,Songliao Basin,NE China显示文摘The geological characteristics and production practices of the major middle-and high-maturity shale oil exploration areas in China are analyzed.Combined with laboratory results,it is clear that three essential conditions,i.e.economic initial production,commercial cumulative oil production of single well,and large-scale recoverable reserves confirmed by the testing production,determine whether the continental shale oil can be put into large-scale commercial development.The quantity and quality of movable hydrocarbons are confirmed to be crucial to economic development of shale oil,and focuses in evaluation of shale oil enrichment area/interval.The evaluation indexes of movable hydrocarbon enrichment include:(1)the material basis for forming retained hydrocarbon,including TOC>2%(preferentially 3%-4%),and typeⅠ-Ⅱkerogens;(2)the mobility of retained hydrocarbon,which is closely related to the hydrocarbon composition and flow behaviors of light/heavy components,and can be evaluated from the perspectives of thermal maturity(Ro),gas-oil ratio(GOR),crude oil density,quality of hydrocarbon components,preservation conditions;and(3)the reservoir characteristics associated with the engineering reconstruction,including the main pore throat distribution zone,reservoir physical properties(including fractures),lamellation feature and diagenetic stage,etc.Accordingly,13 evaluation indexes in three categories and their reference values are established.The evaluation indicates that the light shale oil zones in the Gulong Sag of Songliao Basin have the most favorable enrichment conditions of movable hydrocarbons,followed by light oil and black oil zones,containing 20.8×10^(8) t light oil resources in reservoirs with R_(0)>1.2%,pressure coefficient greater than 1.4,effective porosity greater than 6%,crude oil density less than 0.82 g/cm^(3),and GOR>100 m/m^(3).The shale oil in the Gulong Sag can be explored and developed separately by the categories(resource sweet spot,engineering sweet spot,and tight oil sweet spot)depending on shale oil flowability.The Gulong Sag is the most promising area to achieve large-scale breakthrough and production of continental shale oil in China.ZHAO Wenzhi BIAN Congsheng LI Yongxin ZHANG Jinyou HE Kun LIU Wei ZHANG Bin LEI Zhengdong LIU Chang ZHANG Jingya GUAN Ming LIU Shijul 2023Petroleum Exploration and Development2023,50,3:2
10Predicting the degradation behavior of magnesium alloys with a diffusion-based theoretical model and in vitro corrosion testing显示文摘Magnesium alloys have shown great potential for their use in the medical device field, due to the promising biodegradability. However, it remains a challenge to characterize the degradation behavior of the Mg alloys in a quantitative manner. As such, controlling the degradation rate of the Mg alloys as per our needs is still hard, which greatly limits the practical application of the Mg alloys as a degradable biomaterial.This paper discussed a numerical model developed based on the diffusion theory, which can capture the experimental degradation behavior of the Mg alloys precisely. The numerical model is then implemented into a finite element scheme, where the model is calibrated with the data from our previous studies on the corrosion of the as-cast Mg-1 Ca and the as-rolled Mg-3 Ge binary alloys. The degradation behavior of a pin implant is predicted using the calibrated model to demonstrate the model’s capability. A standard flow is provided in a practical framework for obtaining the degradation behavior of any biomedical Mg alloys. This methodology was further verified via the comparison with enormous available experimental results. 11Global occurrence,bioaccumulation factors and toxic effects of polychlorinated biphenyls in tuna:A review显示文摘Polychlorinated biphenyls(PCBs)tend to accumulate in adipose tissue and induce toxicity due to their high lipophilicity and persistence.Tuna play an important role in the ocean food web and human diet.PCBs can damage the growth of tuna directly and have some adverse effects on human indirectly.This review aims to provide geographic variations,bioaccumulation factors and the toxic effects of PCBs in tuna.PCB levels found were varied significantly among the different regions and it showed a trend of offshore greater than pelagic,with the northern hemisphere higher than the southern hemisphere.The highest PCB concentration of tuna was in FAO area 37.Nearly all of the specimens presented similar domain configuration,while their concentrations varied significantly.PCB congeners and concentrations in tuna were affected by a vast of factors,such as geographical distribution,tissue types,trophic level and growth stage,gender,tuna species and physical and chemical properties of PCBs.They can damage reproductive,neurologic and development systems.This review gives a systematic insight into PCBs in tuna and highlights the more attention should be paid to the long-term changes of PCBs in tuna and oceans.There is also a need to strengthen the assessment of PCB impacts on tuna ecology.Jingqian Xie Zhihe Bian Tian Lin Ling Tao Qiang Wu Ming Chu 2020Emerging Contaminants2020,6,1:2
12Numerical analysis of heat transfer enhancement on steam condensation in the presence of air outside the tube显示文摘In loss-of-coolant accidents,a passive containment heat removal system protects the integrity of the containment by condensing steam.As a large amount of air exists in the containment,the steam condensation heat transfer can be significantly reduced.Based on previous research,traditional methods for enhancing pure steam condensation may not be applicable to steam–air condensation.In the present study,new methods of enhancing condensation heat transfer were adopted and several potentially enhanced heat transfer tubes,including corrugated tubes,spiral fin tubes,and ring fin tubes were designed.STAR-CCM+was used to determine the effect of enhanced heat transfer tubes on the steam condensation heat transfer.According to the calculations,the gas pressure ranged from 0.2 to 1.6 MPa,and air mass fraction ranged from 0.1 to 0.9.The effective perturbation of the high-concentration air layer was identified as the key factor for enhancing steam–air condensation heat transfer.Further,the designed corrugated tube performed well at atmospheric pressure,with a maximum enhancement of 27.4%,and performed poorly at high pressures.In the design of spiral fin tubes,special attention should be paid to the locations that may accumulate high-concentration air.Nonetheless,the ring-fin tubes generally displayed good performance under all conditions of interest,with a maximum enhancement of 24.2%.Wen-Tao Li Xian-Ke Meng Hao-Zhi Bian Ming Ding 2022Nuclear Science and Techniques2022,33,8:2
13Benefit of everolimus in treatment of an intrahepatic cholangiocarcinoma patient with a PIK3CA mutation显示文摘Intrahepatic cholangiocarcinoma(ICC) is a relatively rare form of liver cancer with a poor prognosis. The therapeutic options for patients with advanced ICC are limited and usually ineffective. There is currently no approved targeted therapy for ICC, although accumulating evidence supports inhibition of the PI3K/Akt/m TOR signaling pathway as a promising therapeutic strategy in the treatment of ICC. Here, we report a patient with stage IV ICC harboring a PIK3 CA mutation who responded well to the m TOR inhibitor everolimus. Computed tomography and magnetic resonance imaging demonstrated shrinkage of the tumor and maintenance of a partial response for 6.5 mo after everolimus treatment as the best response.
