1Identification of biomarkers for hepatocellular carcinoma by semiquantitative immunocytochemistry显示文摘AIM:To investigate the expression of key biomarkers in hepatoma cell lines,tumor cells from patients’blood samples,and tumor tissues.METHODS:We performed the biomarker tests in two steps.First,cells plated on coverslips were used to assess biomarkers,and fluorescence intensities were calculated using the NIH Image J software.The measured values were analyzed using the SPSS19.0 software to make comparisons among eight cell lines.Second,eighty-four individual samples were used to assess the biomarkers’expression.Negative enrichment of the blood samples was performed,and karyocytes were isolated and dropped onto pretreated glass slides for further analysis by immunofluorescence staining.Fluorescence intensities were compared among hepatocellular carcinoma(HCC)patients,chronic HBV-infected patients,and healthy controls following methods similar to those used for cell lines.The relationships between the expression of biomarkers and clinical pathological parameters were analyzed by Spearman rank correlation tests.In addition,we studied the distinct biomarkers’expression with three-dimensional laser confocal microscopy reconstructions,and Kaplan-Meier survival analysis was performed to understand the clinical significance of these biomarkers.RESULTS:Microscopic examination and fluorescence intensity calculations indicated that cytokeratin 8/18/19(CK)expression was significantly higher in six of the seven HCC cell lines examined than in the control cells,and the expression levels of asialoglycoprotein receptor(ASGPR)and glypican-3(GPC3)were higher in all seven HCC cell lines than in the control.Cells obtained from HCC patients’blood samples also displayed significantly higher expression levels of ASGPR,GPC3,and CK than cells from chronic HBV-infected patients or healthy controls;these proteins may be valuable surface biomarkers for identifying HCC circulating tumor cells isolated and enriched from the blood samples.The stem cell-like and epithelial-mesenchymal transition-related biomarkers could be detected on the karyocyte slides.ASGPR and GPC3 were expressed at high levels,and thus three-dimensional reconstructions were used to observe their expression in detail.This analysis indicated that GPC3 was localized in the cytoplasm and membrane,but that ASGPR had a polar localization.Survival analyses showed that expression of GPC3 and ASGPR is associated with a patient’s overall survival(OS).CONCLUSION:ASGPR,GPC3,and CK may be valuable HCC biomarkers for CTC detection;the expression of ASGPR and GPC3 might be helpful for understanding patients’OS.Hong Mu Kai-Xuan Lin Hong Zhao Shu Xing Cong Li Fang Liu Hai-Zhen Lu Ze Zhang Yu-Lin Sun Xi-Yun Yan Jian-Qiang Cai Xiao-Hang Zhao 2014World Journal of Gastroenterology2014,20,19:8
2Meiofauna Distribution in Intertidal Sandy Beaches Along China Shoreline(18?–40?N)显示文摘In this study,the distribution pattern of meiofauna from nine sandy beaches at six latitudinal gradients along Chinese coast between 18 and 40?N was studied on their meiofauna abundance to examine the effect of latitudinal gradients.In general,meiofauna abundance was lower in four subtropical beaches in Xiamen(24?N) and Zhoushan(29?N) than that in other beaches.Meiofauna abundance differed little between tropical and temperate beaches.The taxonomic structure of meiofauna showed a dominance of nematode in colder area.The relative composition of turbellarians and polychaetes increased in warmer area.In addition to latitudinal gradient,salinity,oxygenation,sediment grain size affect also the meiofauna latitudinal distribution.As for horizontal distribution,the highest meiofauna abundance was found in low tidal zone at tropical beaches,and in middle tidal zone at temperate beaches.The horizontal distribution of meiofauna was controlled by both physical and biotic factors including feeding and anthropogenic activities.Although meiofauna abundance exhibited a horizontal difference,the composition of meiofaunal main taxa was unanimous horizontally at all beaches at the same sampling latitude.HUA Er ZHANG Zhinan ZHOU Hong MU Fanghong LI Jia ZHANG Ting CONG Bingqing LIU Xiaoshou 2016Journal of Ocean University of China2016,15,1:7

5SC-IrO_2 NR-carbon hybrid:A catalyst with high electrochemical stability for oxygen reduction显示文摘The enhanced electrochemical stability of the synthesized hybrid catalyst has been demonstrated by the introduction of the synergistic effect between carbon powder additive and the prepared catalyst.Single crystal IrO 2 nanorod (SC-IrO 2 NR) catalyst was prepared by a sol-gel method.The structure and performance of the catalyst sample were characterized by X-ray diffraction spectroscopy (XRD),scanning electron microscope (SEM),transmission electron microscope (TEM),rotating disk electrode (RDE) and cyclic voltammetry (CV) measurements.XRD patterns and TEM images indicate that the catalyst sample has a rutile IrO 2 single crystal nanorod structure.The onset potential for oxygen reduction reaction (ORR) of the SC-IrO 2 NR-carbon hybrid catalyst specimen is 0.75 V (vs.RHE) in RDE measurement.CV and RDE test results show that the SC-IrO 2 NR-carbon hybrid catalyst has a better electrochemical stability in comparison with the commercial Pt/C catalyst,with attenuation ratios of 17.67% and 44.60% for the SC-IrO 2 NR-carbon hybrid catalyst and the commercial Pt/C catalyst after 1500 cycles,respectively.Therefore,in terms of stability,the SC-IrO 2 NR-carbon hybrid catalyst has a promising potential in the application of the proton exchange membrane fuel cell.WANG GuangJin CHENG Feng YU Yi LIANG Cong XU Tian PAN Mu 2013Science China Chemistry2013,56,1:3
6Probabilistic ecological risk assessment of cadmium in the Bohai Sea using native saltwater species显示文摘Predicted no-effect concentration(PNEC) is often used in ecological risk assessment to determine low-risk concentrations for chemicals. In the present study, the chronic data from native saltwater species were used to calculated PNEC values using four methods: log-normal distribution(ETX 2.0), log-triangle distribution(US EPA's water quality criteria procedure), burr III distribution(Burrli OZ) and traditional assessment factor(AF). The PNECs that were calculated using four methods ranged from 0.08 μg/L to 1.8 μg/L. Three of the SSD-derived PNECs range from 0.94 to 1.8 μg/L, about a factor of two apart. To demonstrate the use of SSD-based PNEC values and comprehensively estimate the regional ecological risk for cadmium in surface water of the Bohai Sea, in the Liaodong Bay, Bohai Bay, and Laizhou Bay, China, the dissolved cadmium concentrations were measured and obtained 753 valid data covering 190 stations from July 2006 to November 2007. Based on three ecological risk assessment approaches, namely hazard quotient(HQ), probabilistic risk quotient and joint probability curve(JPC), the potential ecological risk of cadmium in surface water of the Liaodong Bay, Bohai Bay, and Laizahou Bay were estimated. Overall, the ecological risk of cadmium to aquatic ecosystem in the whole Bohai Sea was at acceptable ecological risk level, the order of ecological risk was Liaodong Bay>Bohai Bay>Laizhou Bay. 10ITO/Cu multilayer electrodes for high-brightness electrochromic displays显示文摘Fabry-Perot(F-P)nanocavity-type electrochromic devices with multicolor tunability have attracted significant interest in the past two years for their potential uses in a wide variety of applications,such as electronic display,military camouflage,and dynamic decoration.However,challenges such as insufficient brightness,lengthy switching times,and poor cycling stability have yet to be overcome.Herein,we demonstrate electrochromic electrodes based on ITO/Cu as both the current collector and the reflective layer,with WO_(3 )as the electrochromic material,forming a unique three-layered structure.The constituted WO_(3 )/ITO/Cu films present a high brightness value of about 84%before coloration,and a decent brightness value of 48%after coloration at−0.8​V.Moreover,a fast switching time between different coloration states(tc​=​2.2​s;tb​=​1​s)is recorded,attributed to the extremely low sheet resistivity of the ITO/Cu current collector.The films also reveal excellent electrochemical cycling stability across 2000 cycles.Xueqing Tang Zishou Hu Zhen Wang Jian Chen Xinyang Mu Ge Song Peiyan Sun Zhengji Wen Jiaming Hao Shan Cong Zhigang Zhao 2022eScience2022,2,6:1

14Theoretical study of n-budding opening-up vesicle based on the spontaneous curvature model显示文摘N-budding lipid vesicles with single holes were analyzed theoretically by taking into account the line tension of membrane with a hole and the bending energy of membranes based on the Spontaneous-Curvature (SC) model. We solved numerically the Euler-Lagrange equation and the boundary conditions holding on the membrane edge to obtain axisymmetric vesicle shapes, which minimize the total energy. The numerical results show that the size of the opening-up vesicle is larger when the line tension is lower. 18The osteogenic niche-targeted arsenic nanoparticles prevent colonization of disseminated breast tumor cells in the bone显示文摘Up to 70% of patients with late-stage breast cancer have bone metastasis. Current treatment regimens for breast cancer bone metastasis are palliative with no therapeutic cure. Disseminated tumor cells(DTCs) colonize inside the osteogenic niches in the early stage of bone metastasis. Drug delivery into osteogenic niches to inhibit DTC colonization can prevent bone metastasis from entering its late stage and therefore cure bone metastasis. Here, we constructed a 50% DSS6 peptide conjugated nanoparticle to target the osteogenic niche. The osteogenic niche was always located at the endosteum with immature hydroxyapatite. Arsenic-manganese nanocrystals(around 14 nm) were loaded in osteogenic niche-targeted PEG-PLGA nanoparticles with an acidic environment-triggered arsenic release. Arsenic formulations greatly reduced 4 T1 cell adhesion to mesenchymal stem cells(MSCs)/preosteoblasts(pre-OBs) and osteogenic differentiation of osteoblastic cells. Arsenic formulations also prevented tumor cell colonization and dormancy via altering the direct interaction between 4 T1 cells and MSCs/pre-OBs.The chemotactic migration of 4 T1 cells toward osteogenic cells was blocked by arsenic in mimic 3 D osteogenic niche. Systemic administration of osteogenic niche-targeted arsenic nanoparticles significantly extended the survival of mice with 4 T1 syngeneic bone metastasis. 20Identification of Fusarium wilt resistance gene SiRLK1 in Sesamum indicum L.显示文摘Sesame Fusarium wilt(SFW),caused by Fusarium oxysporum f.sp.sesami(Fos),is one of the most devastating diseases affecting sesame cultivation.Deciphering the genetic control of SFW resistance is pivotal for effective disease management in sesame.An inheritance study on a cross between the highly resistant variety Yuzhi 11 and the highly susceptible accession Sp1 using a Fos pathogenicity group 1 isolate indicated that resistance was conferred by a single dominant allele.The target locus was located in a 1.24 Mb interval on chromosome 3 using a combination of cross-population association mapping and bulked segregant analysis.Fine genetic mapping further narrowed the interval between 21,350 and 21,401 kb.The locus Sindi_0812400 was identified as the SFW resistance gene and officially designated SiRLK1.This gene encodes a specific malectin/receptor-like protein kinase with three putative tandem kinase domains and is considered a kinase fusion protein.Sequence analysis revealed that a high proportion(49.44%)of variants within the locus was located within the kinase domainⅢ,and several of which were evidently associated with the diversity in SFW response,indicating the critical role of kinase domainⅢin expression of disease resistance.These findings provide valuable information for further functional analysis of SFW resistance genes and marker-assisted resistance breeding in sesame.Yinghui Duan Wenwen Qu Shuxian Chang Ming Ju Cuiying Wang Cong Mu Hengchun Cao Guiting Li Qiuzhen Tian Qin Ma Zhanyou Zhang Haiyang Zhang Hongmei Miao 2024The Crop Journal2024,12,1:0
