| 1 | Assessing environmental factors associated with regional schistosomiasis prevalence in Anhui Province,Peoples’Republic of China using a geographical detector method显示文摘Background:Schistosomiasis is a water-borne disease caused by trematode worms belonging to genus Schistosoma,which is prevalent most of the developing world.Transmission of the disease is usually associated with multiple biological characteristics and social factors but also factors can play a role.Few studies have assessed the exact and interactive influence of each factor promoting schistosomiasis transmission.Methods:We used a series of different detectors(i.e.,specific detector,risk detector,ecological detector and interaction detector)to evaluate separate and interactive effects of the environmental factors on schistosomiasis prevalence.Specifically,(i)specific detector quantifies the impact of a risk factor on an observed spatial disease pattern,which were ranked statistically by a value of Power of Determinate(PD)calculation;(ii)risk detector detects high risk areas of a disease on the condition that the study area is stratified by a potential risk factor;(iii)ecological detector explores whether a risk factor is more significant than another in controlling the spatial pattern of a disease;(iv)interaction detector probes whether two risk factors when taken together weaken or enhance one another,or whether they are independent in developing a disease.Infection data of schistosomiasis based on conventional surveys were obtained at the county level from the health authorities in Anhui Province,China and used in combination with information from Chinese weather stations and internationally available environmental data.Results:The specific detector identified various factors of potential importance as follows:Proximity to Yangtze River(0.322)>Land cover(0.285)>sunshine hours(0.256)>population density(0.109)>altitude(0.090)>the normalized different vegetation index(NDVI)(0.077)>land surface temperature at daytime(LST_(day))(0.007).The risk detector indicated that areas of schistosomiasis high risk were located within a buffer distance of 50 km from Yangtze River.The ecological detector disclosed that the factors investigated have significantly different effects.The interaction detector revealed that interaction between the factors enhanced their main effects in most cases.Conclusion:Proximity to Yangtze River had the strongest effect on schistosomiasis prevalence followed by land cover and sunshine hours,while the remaining factors had only weak influence.Interaction between factors played an even more important role in influencing schistosomiasis prevalence than each factor on its own.High risk regions influenced by strong interactions need to be targeted for disease control intervention. | Yi Hu Congcong Xia Shizhu Li Michael PWard Can Luo Fenghua Gao Qizhi Wang Shiqing Zhang Zhijie Zhang | 2017 | Infectious Diseases of Poverty2017,6,1: | 9 |
| 2 | Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART. | Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu | 2021 | Cell Research2021,31,8: | 2 |
| 3 | Analysis of QTLs for erucic acid and oil content in seeds on A8 chromosome and the linkage drag between the alleles for the two traits in Brassica napus显示文摘The history of canola breeding began with the discovery of germplasm with low erucic acid content in seeds of spring forage cultivar in the 1950's.FAE1 mutations led to a dramatic decrease of the seed erucic acid content in Arabidopsis thaliana.The products of the two FAE1 loci,BnA8.FAE1 and BnC3.FAE1,showed additive effects to the level of erucic acid content in oilseed rape.Previous research believed that the pleiotropy of FAE1 was responsible for the decrease in seed oil content along with the reduction of seed erucic acid content in the modern cultivars.TN DH population was developed from a canola cultivar Tapidor and a Chinese traditional cultivar Ningyou7.The population had been tested in 10 and 11 environments to map QTLs for the erucic acid content and oil content in seeds.As the map resolution increased,a novel QTL for seed erucic acid content was revealed,after Meta-analysis,7 cM away from the most significant seed erucic acid content QTL where BnA8.FAE1 is located.Seven independent QTLs for seed oil content(qOC) were detected around the two seed erucic acid content QTLs(qEA) across 39.20 cM on linkage group A8.Two of the qOCs co-localized with the two qEAs,respectively,and were detected in a single environment.The other five qOCs were detected in 10 of 11 environments independent of qEAs.Alleles from Tapidor in all the QTLs at the 0-39.20 cM region contributed negative effects to either erucic acid content or oil content in seeds.Parallel,geno-typing showed that on 5 of the 7 QTLs regions,Tapidor alleles had the same genotypes with that in 'Liho',the original low seed erucic acid content source.Through rounds of crossbreeding with oil-cropped cultivars and intensive selection for multi generations,Tapidor still had the inferior alleles for low seed oil content from 'Liho',the forage rape.This showed a strong linkage drag of low seed oil content,which was controlled by the five qEA-independent qOCs,with low seed erucic acid content.Ninety cultivars of B.napus from 8 countries were used to analyze the genetic drag with 9 molecular markers located in the QTL confidence intervals(24.04 cM) on linkage group A8.It was noticed that more than 46% of the cultivars with low seed erucic acid content trait remained the genotype of low seed oil content at least in one locus.Backcross and marker-assisted selection could break the genetic drag between the low oil content and erucic acid in seeds in the process for breeding modern high seed oil content canola cultivars. | Zhengying Cao Fang Tian Nian Wang Congcong Jiang Bing Lin Wei Xia Jiaqin Shi Yah Long Chunyu Zhang Jinling Meng | 2010 | Journal of Genetics and Genomics2010,37,4: | 2 |
| 4 | Endothelial Notch activation promotes neutrophil transmigration via downregulating endomucin to aggravate hepatic ischemia/reperfusion injury显示文摘Inflammatory leukocytes infiltration is orchestrated by mechanisms involving chemokines,selectins,addressins and other adhesion molecules derived from endothelial cells(ECs),but how they respond to inflammatory cues and coordinate leukocyte transmigration remain elusive.In this study,using hepatic ischemia/reperfusion injury(HIRI)as a model,we identified that endothelial Notch activation was rapidly and dynamically induced in liver sinusoidal endothelial cells(LSECs)in acute inflammation.In mice with EC-specific Notch activation(NICeCA),HIRI induced exacerbated liver damage.Consistently,endothelial Notch activation enhanced neutrophil infiltration and tumor necrosis factor(TNF)-αexpression in HIRI.Transcriptome analysis and further qRT-PCR as well as immunofluorescence indicated that endomucin(EMCN),a negative regulator of leukocyte adhesion,was downregulated in LSECs from NICeCA mice.EMCN was downregulated during HIRI in wild-type mice and in vitro cultured ECs insulted by hypoxia/re-oxygenation injury.Notch activation in ECs led to increased neutrophil adhesion and transendothelial migration,which was abrogated by EMCN overexpression in vitro.In mice deficient of RBPj,the integrative transcription factor of canonical Notch signaling,although overwhelming sinusoidal malformation aggravated HIRI,the expression of EMCN was upregulated;and pharmaceutical Notch blockade in vitro also upregulated EMCN and inhibited transendothelial migration of neutrophils.The Notch activation-exaggerated HIRI was compromised by blocking LFA-1,which mediated leukocyte adherence by associating with EMCN.Therefore,endothelial Notch signaling controls neutrophil transmigration via EMCN to modulate acute inflammation in HIRI. | Peiran Zhang Kangyi Yue Xinli Liu Xianchun Yan Ziyan Yang Juanli Duan Congcong Xia Xinyuan Xu Mei Zhang Liang Liang Lin Wang Hua Han | 2020 | Science China(Life Sciences)2020,63,3: | 1 |

| 8 | In situ Observation of Li Deposition-Induced Cracking in Garnet Solid Electrolytes显示文摘Lithium(Li)penetration through solid electrolytes(SEs)induces short circuits in Li solid-state batteries(SSBs),which is a critical issue that hinders the development of high energy density SSBs.While cracking in ceramic SEs has been often shown to accompany Li penetration,the interplay between Li deposition and cracking remains elusive.Here,we constructed a mesoscale SSB inside a focused ion beam-scanning electron microscope(FIB-SEM)for in situ observation of Li deposition-induced cracking in SEs at nanometer resolution.Our results revealed that Li propagated predominantly along transgranular cracks in a garnet Li_(6.4)La_(3)Zr_(1.4)Ta_(0.6)O_(12)(LLZTO).Cracks appeared to initiate from the interior of LLZTO beneath the electrode surface and then propagated by curving toward the LLZTO surface.The resulting bowl-shaped cracks resemble those from hydraulic fracture caused by high fluid pressure on the surface of internal cracks,suggesting that the Li deposition-induced pressure is the major driving force of crack initiation and propagation.The high pressure generated by Li deposition is further supported by in situ observation of the flow of filled Li between the crack flanks,causing crack widening and propagation.This work unveils the dynamic interplay between Li deposition and cracking in SEs and provides insight into the mitigation of Li dendrite penetration in SSBs. | Jun Zhao Yongfu Tang Qiushi Dai Congcong Du Yin Zhang Dingchuan Xue Tianwu Chen Jingzhao Chen Bo Wang Jingming Yao Ning Zhao Yanshuai Li Shuman Xia Xiangxin Guo Stephen J.Harris Liqiang Zhang Sulin Zhang Ting Zhu Jianyu Huang | 2022 | Energy & Environmental Materials2022,5,2: | 0 |
| 9 | Kunming mouse strain is less susceptible to elastase-induced abdominal aortic aneurysms显示文摘Background:Porcine pancreatic elastase(PPE)is successfully used to induce abdominal aortic aneurysm(AAA)in mice.However,differences between mouse strains in susceptibility to PPE induction have been reported.Kunming mouse is one of the most frequently used strains in China but whether it is suitable for induction of AAA by PPE application remains unclear.Methods:PPE infusion(1.5 units/ml)in temporary controlled aorta was performed to induce AAAs in both C57BL/6J and Kunming mice.Phosphatebuffered saline(PBS)application was used as vehicle control.The aorta diameters of all mice were measured at days 0 and 14 after surgery to evaluate the AAA formation.Results:After 14 days of PPE or PBS infusion,all mice were sacrificed and aorta tissues were collected for histological staining analysis.At the 14th day after infusion,PPE successfully induced aortic dilation in Kunming mice and typical AAA in C57BL/6J mice.The aorta diameter increased by 0.23 mm in Kunming mice after PPE infusion,while it was 0.72 mm in the C57BL/6J strain.PPE induced mild elastin degradation,smooth muscle cell(SMC)depletion and mural leucocyte infiltration in Kunming mice,but in PPE-sensitive C57BL/6J mice,it induced total loss of SMCs,elastin disappearance and diffused infiltrated leucocytes in aortic aneurysmal segments.The effects of PPE in inducing angiogenesis and upregulating matrix metalloproteinase 2 and 9 expression in Kunming mice were also weaker than that in C57BL/6J mice.Conclusion:At the reported dose of PPE,Kunming mouse is not as susceptible to AAA formation as C57BL/6J mice.The failure of PPE to induce AAA formation in Kunming mice may be associated to its inability to boost a strong inflammatory response. | Haole Liu Kangli Tian Congcong Xia Panpan Wei Boyu Xu Weilai Fu Yankui Li Yafeng Li Liang Bai Rong Wang Weirong Wang Baohui Xu Enqi Liu Sihai Zhao | 2022 | Animal Models and Experimental Medicine2022,5,1: | 0 |
