| 4 | Gut microbiota mediates the absorption of FLZ,a new drug for Parkinson’s disease treatment显示文摘The gut microbiota plays an important role in regulating the pharmacokinetics and pharmacodynamics of many drugs.FLZ,a novel squamosamide derivative,has been shown to have neuroprotective effects on experimental Parkinson’s disease(PD)models.FLZ is under phase I clinical trial now,while the underlying mechanisms contributing to the absorption of FLZ are still not fully elucidated.Due to the main metabolite of FLZ was abundant in feces but rare in urine and bile of mice,we focused on the gut microbiota to address how FLZ was metabolized and absorbed.In vitro studies revealed that FLZ could be exclusively metabolized to its major metabolite M1 by the lanosterol 14 alpha-demethylase(CYP51)in the gut microbiota,but was almost not metabolized by any other metabolism-related organs,such as liver,kidney,and small intestine.M1 was quickly absorbed into the blood and then remethylated to FLZ by catechol O-methyltransferase(COMT).Notably,dysbacteriosis reduced the therapeutic efficacy of FLZ on the PD mouse model by inhibiting its absorption.The results show that the gut microbiota mediate the absorption of FLZ through a FLZ-M1-FLZ circulation.Our research elucidates the vital role of the gut microbiota in the absorption of FLZ and provides a theoretical basis for clinical pharmacokinetic studies and clinical application of FLZ in the treatment of PD. | Junmei Shang Shurong Ma Caixia Zang Xiuqi Bao Yan Wang Dan Zhang | 2021 | Acta Pharmaceutica Sinica B2021,11,5: | 3 |

| 15 | Financial decision‑making behaviors of Ethnic Tibetan Households based on mental accounting显示文摘Ethnic Tibetans(ETs)typically reside in the remote plateaus of China and possess strong cultural and spiritual values.Their financial decision-making is influenced by economic and physical factors,unique culture,social norms,and psychological motivators.We conducted an in-person survey of 480 randomly selected ET households across four provinces in rural China.The survey data was analyzed using three different econometric models—probit,ordered probit,and ranked ordered logit—to examine the choice of borrowing from formal or informal credit sources,the number of sources borrowed from,and repayment priority.Our findings indicate that mental accounting plays a significant role in the financial decision-making process of ET households.Additionally,we find that the informal credit source is strongly associated with the financial decisions of ET households.The majority of loans from formal financial institutions are used to meet daily needs,as opposed to purchasing productive inputs.Our results also suggest that strong social relationships and religious beliefs prevent households from defaulting,and that loans from formal financial sources receive repayment priority.China would benefit from promoting inclusive finance and encouraging the adoption of improved agricultural practices to support the prosperity of ET and other minority communities. | DunGang Zang Krishna P.Paudel Yan Liu Dan Liu Yating He | 2023 | Financial Innovation2023,9,1: | 0 |
| 16 | The F5 gene predicts poor prognosis of patients with gastric cancer by promoting cell migration identified using a weighted gene co-expression network analysis显示文摘Distal gastric cancer(DGC)is a subgroup of gastric cancer(GC),which has different molecular characteristics from proximal gastric cancer(PGC).These differences result in different overall survival(OS)rates;however,data pertaining to the survival rate in PGC or DGC are contradictory.This suggests that the location of GC is not the unique cause of the different survival rates,while the molecular characteristics might be more important factors determining the prognosis of DGC.Therefore,the aim of this study was to discover key prognostic factors in DGC using bioinformatic methods and to explore the potential molecular mechanism.The Cancer Genome Atlas(TCGA)public database was employed to screen data relating to DGC,and we conducted a weighted gene co-expression network analysis(WGCNA)on DGC patient samples to establish co-expression modules.High-weight genes(hub genes)in a dominant color module were identified.In vitro experiments and gene set enrichment analyses(GSEA)were carried out to elucidate the potential molecular mechanism.In this study,139 DGC samples were enrolled to perform a co-expression analysis.According to the correlation between gene modules and clinical characteristics,the royal blue module related to stage M of DGC was screened,and a survival analysis was conducted to show that highcoagulation-factor V(F5)expression was related to the short OS of patients with GC.In vitro experiments confirmed that F5 could promote the migration of GC cells.GSEA suggested that F5 might have affected the prognosis of GC by modulating the activities of the Wnt and/or the TGF-βsignaling pathways.Our results indicated that high F5 expression predicts poor prognosis of patients with DGC,and it functions probably by promoting cell migration through the Wnt and/or the TGF-βsignaling pathways. | MENGYI TANG BOWEN YANG CHUANG ZHANG CHAOXU ZHANG DAN ZANG LIBAO GONG YUNPENG LIU ZHI LI XIUJUAN QU | 2021 | BIOCELL2021,45,4: | 0 |

| 20 | FUS-mediated HypEVs: Neuroprotective effects against ischemic stroke显示文摘Few studies have investigated the properties and protein composition of small extracellular vesicles (sEVs) derived from neurons under hypoxic conditions. Presently, the extent of the involvement of these plentiful sEVs in the onset and progression of ischemic stroke remains an unresolved question. Our study systematically identified the characteristics of sEVs derived from neurons under hypoxic conditions (HypEVs) by physical characterization, sEV absorption, proteomics and transcriptomics analysis. The effects of HypEVs on neurites, cell survival, and neuron structure were assessed in vitro and in vivo by neural complexity tests, magnetic resonance imaging (MRI), Golgi staining, and Western blotting of synaptic plasticity-related proteins and apoptotic proteins. Knockdown of Fused in Sarcoma (FUS) small interfering RNA (siRNA) was used to validate FUS-mediated HypEV neuroprotection and mitochondrial mRNA release. Hypoxia promoted the secretion of sEVs, and HypEVs were more easily taken up and utilized by recipient cells. The MRI results illustrated that the cerebral infarction volume was reduced by 45% with the application of HypEVs, in comparison to the non- HypEV treatment group. Mechanistically, the FUS protein is necessary for the uptake and neuroprotection of HypEVs against ischemic stroke as well as carrying a large amount of mitochondrial mRNA in HypEVs. However, FUS knockdown attenuated the neuroprotective rescue capabilities of HypEVs.
