1DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR.Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen 2020Acta Biochimica et Biophysica Sinica2020,52,11:5
2Seasonal variations in the plant diet of the Chinese Monal revealed by fecal DNA metabarcoding analysis显示文摘The Chinese Monal(Lophophorus thuysii)is an alpine-obligate galliform species of global conservation priority.It has been listed as a first class protected wildlife species in China,requiring conservation actions during the 14 th Five-Year Plan period.However,the diet composition of Chinese Monal and its seasonal variations have rarely been studied,constraining the effective conservation of the species.Here,we investigated the plant diet composition of the Chinese Monal and its seasonal variations using a DNA metabarcoding approach on fecal samples.We collected 190 fecal samples of the Chinese Monals from the central Qionglai Mountains located in China,and analyzed the plant diet of this species using a DNA metabarcoding approach.Taxonomic profiling of higher plants in the fecal samples was performed using the second internal transcribed spacer(ITS2)amplicon.Downstream analyses,including rarefaction curves,nonmetric multidimensional scaling(NMDS)and permutational multivariate analysis of variance(PERMANOVA),were used to explore the seasonal variations in diet composition.The Chinese Monal foraged a wide range of plant recipes composed of 35 families and 83 genera throughout the year,with Brassicaceae,Apiaceae,and Poaceae as the dominant families,and Cardamine as the dominant genus.The species consumed plants from 62 genera from 28 families during the breeding season(n=81)and 66 genera from 31 families during the non-breeding season(n=109).Further,the plant diet composition significantly varied between the breeding and non-breeding seasons,especially for the frequency of occurrence and relative read abundances at genus level.Our study analyzed the plant diet of the Chinese Monal at a high resolution for the first time,and the results revealed that the seasonal variations in its plant diet composition was adapted to plant phenology and foraging strategy.Fritillaria species,a previously confirmed important food resource for the Chinese Monal,were not detected in any fecal samples,potentially due to overharvesting of Fritillaria bulbs for Traditional Chinese Medicine.Therefore,we highly recommend further restriction of herb gathering in Chinese Monal habitats to facilitate the conservation of this endangered species.Altogether,our study enriches essential ecological information for the Chinese Monal and also provides insights into conservation management for this endangered species.Bin Wang Yun Li Guiquan Zhang Jian Yang Cao Deng Haoyu Hu Long Zhang Xiaoqin Xu Caiquan Zhou 2022Avian Research2022,13,2:2
3Nuclear translocation of OsMFT1 that is impeded by OsFTIP1 promotes drought toleranee in rice显示文摘Drought is the leading environmental threat affecting crop productivity,and plants have evolved a series of mechanisms to adapt to drought stress.The FT-interacting proteins(FTIPs)and phosphatidylethanola mine.binding proteins(PEBPs)play key roles in developmental processes,whereas their roles in the regulation of stress response are still largely unknown.Here,we report that OsFTIP1 negatively regulates drought response in rice.We showed that OsFTIP1 interacts with rice MOTHER OF FT AND TFL1(OsMFT1),a PEBP that promotes rice tolerance to drought treatment.Further studies discovered that OsMFT1 interacts with two key drought-related transcription factors,OsbZIP66 and OsMYB26,regulating their binding capacity on drought-related genes and thereby enhancing drought toleranee in rice.Interestingly,we found that OsFTIP1 impedes the nucleocytoplasmic translocation of OsMFT1,implying that dynamic modulation of drought-responsive genes by the OsMFT1-OsMYB26 and OsMFT1-OsbZIP66 complexes is integral to OsFTIP1-modulated nuclear accumulation of OsMFT1.Our findings also suggest that OsMFT1 might act as a hitherto unknown nucleocytoplasmic trafficking signal that regulates drought tolerance in rice in response to environmental signals.Ying Chen Jun Shen Liang Zhangi Haoyue Qi Lijia Yang Huanyu Wang Jiaxuan Wang Yuexing Wang Hao Du Zeng Tao Ting Zhao Pingchuan Deng Qingyao Shu Qian Qian Hao Yu Shiyong Song 2021Molecular Plant2021,14,8:2
4Design of a clustered data-driven array processor for computer vision显示文摘Computer vision(CV)is widely expected to be the next big thing in emerging applications.So many heterogeneous architectures for computer vision emerge.However,plenty of data need to be transferred between different structures for heterogeneous architecture.The long data transfer delay becomes the mainly problem to limit the processing speed for computer vision applications.For reducing data transfer delay and fasting computer vision applications,a clustered data-driven array processor is proposed.A three-level pipelining processing element is designed which supports two-buffer data flow interface and 8 bits,16 bits,32 bits subtext parallel computation.At the same time,for accelerating transcendental function computation,a four-way shared pipelining transcendental function accelerator is designed,which is based on Y-intercept adjusted piecewise linear segment algorithm.A distributed shared memory structure based on unified addressing is also employed.To verify efficiency of architecture,some image processing algorithms are implemented on proposed architecture.Simultaneously the proposed architecture has been implemented on Xilinx ZC 706 development board.The same circuitry has been synthesized using SMIC 130 nm CMOS technology.The circuitry is able to run at 100 MHz.Area is 26.58 mm2.山蕊 Deng Junyong Jiang Lin Zhu Yun Wu Haoyue He Feilong 2020High Technology Letters2020,26,4:2
5Parallel design of convolutional neural networks for remote sensing images object recognition based on data-driven array processor显示文摘Object recognition in very high-resolution remote sensing images is a basic problem in the field of aerial and satellite image analysis.With the development of sensor technology and aerospace remote sensing technology,the quality and quantity of remote sensing images are improved.Traditional recognition methods have a certain limitation in describing higher-level features,but object recognition method based on convolutional neural network(CNN)can not only deal with large scale images,but also train features automatically with high efficiency.It is mainly used on object recognition for remote sensing images.In this paper,an AlexNet CNN model is trained using 2100 remote sensing images,and correction rate can reach 97.6%after 2000 iterations.Then based on trained model,a parallel design of CNN for remote sensing images object recognition based on data-driven array processor(DDAP)is proposed.The consuming cycles are counted.Simultaneously,the proposed architecture is realized on Xilinx V6 development board,and synthesized based on SMIC 130 nm complementary metal oxid semiconductor(CMOS)technology.The experimental results show that the proposed architecture has a certain degree of parallelism to achieve the purpose of accelerating calculations.Shan Rui Jiang Lin Deng Junyong Cui Pengfei Zhang Yuting Wu Haoyue Xie Xiaoyan 2020The Journal of China Universities of Posts and Telecommunications2020,27,6:1

8Acclimation of CH_(4)emissions from paddy soil to atmospheric CO_(2)enrichment in a growth chamber experiment显示文摘Elevated levels of atmospheric CO_(2)(eCO_(2))promote rice growth and increase methane(CH_(4))emissions from rice paddies,because increased input of plant photosynthate to soil stimulates methanogenic archae.However,temporal trends in the effects of eCO_(2)on rice growth and CH_(4)emissions are still unclear.To investigate changes in the effects of eCO_(2)over time,we conducted a two-season pot experiment in a walk-in growth chamber.Positive effects of eCO_(2)on rice leaf photosynthetic rate,biomass,and grain yield were similar between growing seasons.However,the effects of eCO_(2)on CH_(4) emissions decreased over time.Elevated CO_(2)increased CH_(4)emissions by 48%-101%in the first growing season,but only by 28%-30%in the second growing season.We also identified the microbial process underlying the acclimation of CH4 emissions to atmospheric CO_(2)enrichment:eCO_(2)stimulated the abundance of methanotrophs more strongly in soils that had been previously exposed to eCO_(2)than in soils that had not been.These results emphasize the need for long-term eCO_(2)experiments for accurate predictions of terrestrial feedbacks.Haoyu Qian Yaguo Jin Jin Chen Shan Huang Yunlong Liu Jun Zhang Aixing Deng Jianwen Zou Genxing Pan Yanfeng Ding Yu Jiang Kees Jan van Groenigen Weijian Zhang 2022The Crop Journal2022,10,1:1
9LOOKNoC: a low-loss, non-blocking, scalable passive optical interconnect network-on-chip architecture显示文摘Low-loss, non-blocking, scalable passive optical interconnect network on-chip(LOOKNoC) structure was proposed based on 2×2 optical exchange switches, using wavelength division multiplexing(WDM)technology to expand to 8×8, 16×16, 32×32, 64×64 passive optical interconnection networks, which can achieve non-blocking communication. The experimental results show that based on the 16×16 optical interconnection network structure, the number of microring resonators(MRs) in LOOKNoC was reduced by 90.9%, 90.9%, 20.0% and 75.0% compared with the generic wavelength-routed optical router(GWOR), λ-router, topology and CrossBar structure. 10Eco-geographical Regionalization of China:An Approach Using the Rough Set Method显示文摘Eco-geographical regionalization involves dividing land into regions by considering both intra-regional consistency and interregional disparity and is based on the pattern of differentiation of eco-geographical elements.Owing to the complexity of the land surface,and the limitation of data and appropriate methods,regions in China have hitherto been mapped manually,meaning that the process of mapping was non-repeatable.To make the regionalization technique repeatable,this study aimed to extract and quantify the expert knowledge of regionalization using an automated method.The rough set method was adopted to extract rules of regionalization based on the existing eco-geographical regionalization map of China,as well as its corresponding meteorological and geological datasets.Then,the rules for regionalization were obtained hierarchically for each natural domain,each temperature zone,and each humidity region.Owing to differences in zonal differentiation,the rule extraction sequence for the eastern monsoon zone and Tibetan Alpine zone was temperature zone first followed by humidity region,with the reverse order being applied for the northwest arid/semi-arid zone.Results show that the extracted indicators were similar to those of the existing(expert-produced)regionalization scheme but more comprehensive.The primary indicator for defining temperature zones was the≥10℃ growing season,and the secondary indicators were the January and July mean temperatures.The primary and secondary indicators for identifying humid regions were aridity index and precipitation,respectively.Eco-geographical regions were mapped over China using these rules and the gridded indicators.Both the temperature zones and humidity regions mapped by the rules show≥85%consistency with the existing regionalization,which is higher than values for mapping by the commonly used simplified method that uses the classification of one indicator.This study demonstrates that the proposed rough set method can establish eco-geographical regionalization that is quantitative and repeatable and able to dynamically updated.DENG Haoyu WU Shaohong YIN Yunhe GAO Jiangbo ZHAO Dongsheng 2022Chinese Geographical Science2022,32,1:0
11Design and Fabrication of Reflective Phase Shifter for Two-Dimensional Terahertz Beam-Scanning Reflectarray显示文摘A reflective phase shifter is proposed to realize the two-dimensional beam-scanning reflectarray.The reflectarray is composed of double-dipole resonant elements in which the phase shift is implemented by applying a driving voltage to the liquid crystal(LC). A 30 ×30 phase shifter sample is fabricated on a quartz substrate with a 480 μm thickness and a 4 cm × 4 cm area.According to the experimental results, 0 to 360° phase shift can be achieved within the bandwidth of 5 GHz. In order to accomplish two-dimensional beam-scanning and reduce the negative impact of driving lines on the reflectarray, a new LC driving method is developed. To verify the accuracy of the proposed approach, three samples of different driving line numbers are designed using the above-mentioned phase shifter, which all have achieved a0-360° phase shift in the bandwidth of 4 GHz. 12Upregulation of DRG protein TMEM100 facilitates dry skin-induced pruritus by enhancing TRPA1 channel function显示文摘The dry skin tortures numerous patients with severe itch.The transient receptor potential cation channel V member 1(TRPV1)and A member 1(TRPA1)are two essential receptors for peripheral neural coding of itch sensory,mediating histaminergic and nonhistaminergic itch separately.In the dorsal root ganglion,transmembrane protein 100(TMEM100)is structurally related to both TRPV1 and TRPA1 receptors,but the exact role of TMEM100 in itch sensory coding is still unknown.Here,in this study,we find that TMEM100+DRG neurons account for the majority of activated neurons in an acetone-ether-water(AEW)-induced dry skin itch model,and some TMEM100+DRG neurons are colocalized with both TRPA1 and the chloroquine-related Mrgpr itch receptor family.Both the expression and function of TRPA1 channels,but not TRPV1 channels,are upregulated in the AEW model,and specific DRGTmem100 gene knockdown alleviates AEW-induced itch and rescues the expression and functional changes of TRPA1.Our results strongly suggest that TMEM100 protein in DRG is the main facilitating factor for dry skin-related chronic itch,and specific suppression of TMEM100 in DRG could be a novel effective treatment strategy for patients who suffer from dry skin-induced itch.Chao Pan Yingfu Jiao Dexu Kong Haoyue Deng Saihong Xu Dan Tang Wen Yin Po Gao Weifeng Yu Yinghui Fan Daxiang Wen 2023Acta Biochimica et Biophysica Sinica2023,55,3:0
13Intensified risk to ecosystem productivity under climate change in the arid/humid transition zone in northern China显示文摘Assessing the climate change risk faced by the ecosystems in the arid/humid transition zone(AHTZ)in northern China holds scientific significance to climate change adaptation.We simulated the net primary productivity(NPP)for four representative concentration pathways(RCPs)using an improved Lund-Potsdam-Jena model.Then a method was established based on the NPP to identify the climate change risk level.From the midterm period(2041–2070)to the long-term period(2071–2099),the risks indicated by the negative anomaly and the downward trend of the NPP gradually extended and increased.The higher the scenario emissions,the more serious the risk.In particular,under the RCP8.5 scenario,during 2071–2099,the total risk area would be 81.85%,that of the high-risk area would reach 54.71%.In this high-risk area,the NPP anomaly would reach–96.00±46.95 gC·m-2·a-1,and the rate of change of the NPP would reach–3.56±3.40 gC·m-2·a-1.The eastern plain of the AHTZ and the eastern grasslands of Inner Mongolia are expected to become the main risk concentration areas.Our results indicated that the management of future climate change risks requires the consideration of the synergistic effects of warming and intensified drying on the ecosystem.YIN Yunhe DENG Haoyu MA Danyang WU Shaohong 2021Journal of Geographical Sciences2021,31,9:0
14Targeting TRPV1-mediated autophagy attenuates nitrogen mustard-induced dermal toxicity显示文摘Nitrogen mustard(NM)causes severe vesicating skin injury,which lacks effective targeted therapies.The major limitation is that the specific mechanism of NM-induced skin injury is not well understood.Recently,autophagy has been found to play important roles in physical and chemical exposure-caused cutaneous injuries.However,whether autophagy contributes to NM-induced dermal toxicity is unclear.Herein,we initially confrmed that NM dose-dependently caused cell death and induced autophagy in keratinocytes.Suppression of autophagy by 3-methyladenine,chloroquine,and baflomycin A1 or ATG5 siRNA attenuated NM-induced keratinocyte cell death.Furthermore,NM increased transient receptor potential vanilloid 1(TRPV1)expression,intracellular Ca2+content,and the activities of Car+/calmodulin-dependent kinase kinaseβ(CaMKKB),AMP-activated protein kinase(AMPK),unc-51-like kinase 1(ULK1),and mammalian target of rapamycin(mTOR).NM-induced autophagy in keratinocytes was abolished by treatment with inhibitors of TRPV1(capsazepine),CaMKKβ(STO-609),AMPK(compound C),and ULK1(SBI-0206965)as well as TRPV1,CaMKKB,and AMPK siRNA transfection.In addition,an mTOR inhibitor(rapamycin)had no signifcant effect on NM-stimulated autophagy or cell death of keratinocytes.Finally,the results of the in vivo experiment in NM-treated skin tissues were consistent with the findings of the in vitro experiment.In conclusion,NM-caused dermal toxicity by overactivating autophagy partially through the activation of TRPV1-Ca2+-CaMKKB-AMPK-ULK1 signaling pathway.These results suggest that blocking TRPV1-dependent autophagy could be a potential treatment strategy for NM-caused cutaneous injury.Mingliang Chen Xunhu Dong Haoyue Deng Feng Ye Yuanpeng Zhao Jin Cheng Guorong Dan Jiqing Zhao Yan Sai Xiuwu Bian Zhongmin Zou 2021Signal Transduction and Targeted Therapy2021,6,2:0
15Dietary cysteine drives body fat loss via FMRFamide signaling in Drosophila and mouse显示文摘Obesity imposes a global health threat and calls for safe and effective therapeutic options.Here,we found that protein-rich diet significantly reduced body fat storage in fruit flies,which was largely attributed to dietary cysteine intake.Mechanistically,dietary cysteine increased the production of a neuropeptide FMRFamide(FMRFa).Enhanced FMRFa activity simultaneously promoted energy expenditure and suppressed food intake through its cognate receptor(FMRFaR),both contributing to the fat loss effect.In the fat body,FMRFa signaling promoted lipolysis by increasing PKA and lipase activity.In sweet-sensing gustatory neurons,FMRFa signaling suppressed appetitive perception and hence food intake.We also demonstrated that dietary cysteine worked in a similar way in mice via neuropeptide FF(NPFF)signaling,a mammalian RFamide peptide.In addition,dietary cysteine or FMRFa/NPFF administration provided protective effect against metabolic stress in flies and mice without behavioral abnormalities.Therefore,our study reveals a novel target for the development of safe and effective therapies against obesity and related metabolic diseases.Tingting Song Wusa Qin Zeliang Lai Haoyu Li Daihan Li Baojia Wang Wuquan Deng Tingzhang Wang Liming Wang Rui Huang 2023Cell Research2023,33,6:0
16Reinforcement learning-based missile terminal guidance of maneuvering targets with decoys显示文摘In this paper,a missile terminal guidance law based on a new Deep Deterministic Policy Gradient(DDPG)algorithm is proposed to intercept a maneuvering target equipped with an infrared decoy.First,to deal with the issue that the missile cannot accurately distinguish the target from the decoy,the energy center method is employed to obtain the equivalent energy center(called virtual target)of the target and decoy,and the model for the missile and the virtual decoy is established.Then,an improved DDPG algorithm is proposed based on a trusted-search strategy,which significantly increases the train efficiency of the previous DDPG algorithm.Furthermore,combining the established model,the network obtained by the improved DDPG algorithm and the reward function,an intelligent missile terminal guidance scheme is proposed.Specifically,a heuristic reward function is designed for training and learning in combat scenarios.Finally,the effectiveness and robustness of the proposed guidance law are verified by Monte Carlo tests,and the simulation results obtained by the proposed scheme and other methods are compared to further demonstrate its superior performance.Tianbo DENG Hao HUANG Yangwang FANG Jie YAN Haoyu CHENG 2023Chinese Journal of Aeronautics2023,36,12:0
