| 3 | Preconcentration and Separation of Trace Copper in Water Samples with Nanometer-Size TiO_2 Colloid and Determination by FAAS显示文摘A new absorbent of nanometer-size TiO2 colloid for Cu(Ⅱ) was studied in this work. The adsorption rate could reach above 99% when the pH values were at the range of 5-6. The adsorption balance time, adsorption capacities, and the eluent were investigated. A novel method of trace Cu(Ⅱ) preconcentration and separation with nanometer-size titanium dioxide colloid and determination by flame atomic absorption spectrometry (FAAS) was advanced. The detection limit (3σ) of the method was 1.15 μg· L^(-1) , and the relative standard deviation (R.S.D) was 1.53% (n=6). | 4 | Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors. | Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang | 2022 | Animal Models and Experimental Medicine2022,5,1: | 3 |
| 5 | On island landscape pattern of forests in Helan Mountain and its cause of formation显示文摘Based on the spatial information techniques such as RS, GIS, and GPS, the forest landscape patterns in the Helan Mountain, western China, were studied. The Landsat 5 TM data were used to classify the forest landscapes through RS digital cartography, and then, the landscape characteristics and landscape pattern were analyzed quantificationally. Furthermore, through spatial data collection and spatial analysis of the main disturbances in this area, the cause of landscape formation was studied. The results showed that the total 1177 forest landscape patches could be classified into 21 landscape types, and the forest landscape in the Helan Mountain was island pattern, which was encircled by deserta as matrix. The values of landscape diversity index and landscape fragmentation index were 2.61 and 0.43, respectively. In this area, the landscape pattern was clearly formed and continuously changed in response to geological processes, climate, activities of organisms, forest fire, desertification, human activities and so on. This landscape pattern had an obviously negative effect on the stability and ecosystem services of forests. | 6 | Optimization of post-classification processing of high-resolution satellite image:A case study显示文摘The application of remote sensing monitoring techniques plays a crucial role in evaluating and governing the vast amount of ecological construction projects in China. However, extracting information of ecological engineering target through high-resolution satellite image is arduous due to the unique topography and complicated spatial pattern on the Loess Plateau of China. As a result, enhancing classification accuracy is a huge challenge to high-resolution image processing techniques. Image processing techniques have a definitive effect on image properties and the selection of different parameters may change the final classification accuracy during post-classification processing. The common method of eliminating noise and smoothing image is majority filtering. However, the filter function may modify the original classified image and the final accuracy. The aim of this study is to develop an efficient and accurate post-processing technique for acquiring information of soil and water conservation engineering, on the Loess Plateau of China, using SPOT image with 2.5 rn resolution. We argue that it is vital to optimize satellite image filtering parameters for special areas and purposes, which focus on monitoring ecological construction projects. We want to know how image filtering influences final classified results and which filtering kernel is optimum. The study design used a series of window sizes to filter the original classified image, and then assess the accuracy of each output map and image quality. We measured the relationship between filtering window size and classification accuracy, and optimized the post-processing techniques of SPOT5satellite images. | 8 | Efficient production of pronuclear embryos in breeding and nonbreeding season for generating transgenic sheep overexpressing TLR4显示文摘Background: Brucella is a zoonotic Gram-negative pathogen that causes abortion and infertility in ruminants and humans. TLR4 is the receptor for LPS which can recognize Brucella and initiate antigen-presenting cell activities that affect both innate and adaptive immunity. Consequently, transgenic sheep over-expressing TLR4 are an suitable model to investigate the effects of TLR4 on preventing Brucellosis. In this study, we generated transgenic sheep overexpressing TLR4 and aimed to evaluate the effects of different seasons(breeding and non-breeding season) on superovulation and the imported exogenous gene on growth.Results: In total of 43 donor ewes and 166 recipient ewes in breeding season, 37 donor ewes and 144 recipient ewes in non-breeding season were selected for super-ovulation and injected embryo transfer to generate transgenic sheep.Our results indicated the no. of embryos recovered of donors and the rate of pronuclear embryos did not show any significant difference between breeding and non-breeding seasons(P > 0.05). The positive rate of exogenous TLR4 tested were 21.21 % and 22.58 % in breeding and non-breeding season by Southern blot. The expression level of TLR4 in the transgenic sheep was 1.5 times higher than in the non-transgenic group(P < 0.05). The lambs overexpressing TLR4 had similar growth performance with non-transgenic lambs, and the blood physiological parameters of transgenic and non-transgenic were both in the normal range and did not show any difference.Conclusions: Here we establish an efficient platform for the production of transgenic sheep by the microinjection of pronuclear embryos during the whole year.
