1Genome-Wide Prediction of Highly Specific Guide RNA Spacers for CRISPR-Cas9-Mediated Genome Editing in Model Plants and Major Crops显示文摘Dear Editor, RNA-guided genome editing (RGE) using the Streptococcus pyogenes CRISPR-Cas9 system (Jinek et al.,2012;Cong et al.,2013;Mali et al.,2013b) is emerging as a simple and highly efficient tool for genome editing in many organisms.The Cas9 nuclease can be programmed by dual or single guide RNA (gRNA) to cut target DNA at specific sites,thereby introducing precise mutations by error-prone non-homologousend-joining repairing or by incorporating foreign DNAs via homologous recombination between target site and donor DNA.The gRNA-Cas9 complex recognizes targets based on the complementarity between one strand of targeted DNA (referred as protospacer) and the 5'-end leading sequence of gRNA (referred to as gRNA spacer) that is approximately 20 base pairs (bp) long (Figure 1A).Besides gRNA-DNA pairing,a protospacer-adjacent motif (PAM) following the paired region in the DNA is also required for Cas9 cleavage.Recent studies reveal that Cas9 could cut the PAM-containing DNA sites that imperfectly match gRNA spacer sequences,resulting in genome editing at undesired positions.Kabin Xie Jianwei Zhang Yinong Yang 2014Molecular Plant2014,7,5:35
2Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
3Chemical characterization of size-resolved aerosols in four seasons and hazy days in the megacity Beijing of China显示文摘Size-resolved aerosol samples were collected by MOUDI in four seasons in 2007 in Beijing. The PM10 and PM1.8mass concentrations were 166.0 ± 120.5 and 91.6 ± 69.7 μg/m^3, respectively,throughout the measurement, with seasonal variation: nearly two times higher in autumn than in summer and spring. Serious fine particle pollution occurred in winter with the PM1.8/PM10 ratio of 0.63, which was higher than other seasons. The size distribution of PM showed obvious seasonal and diurnal variation, with a smaller fine mode peak in spring and in the daytime. OM(organic matter = 1.6 × OC(organic carbon)) and SIA(secondary inorganic aerosol) were major components of fine particles, while OM, SIA and Ca_2+were major components in coarse particles. Moreover, secondary components, mainly SOA(secondary organic aerosol) and SIA,accounted for 46%–96% of each size bin in fine particles, which meant that secondary pollution existed all year. Sulfates and nitrates, primarily in the form of(NH_4)_2SO_4, NH_4NO_3, Ca SO_4, Na_2SO_4 and K_2SO_4, calculated by the model ISORROPIA II, were major components of the solid phase in fine particles. The PM concentration and size distribution were similar in the four seasons on non-haze days, while large differences occurred on haze days, which indicated seasonal variation of PM concentration and size distribution were dominated by haze days. 5Single-cell transcriptomic analysis defines the interplay between tumor cells,viral infection,and the microenvironment in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an Epstein-Barr virus(EBV)-associated malignancy with a complex tumor ecosystem.How the interplay between tumor cells,EBV,and the microenvironment contributes to NPC progression and immune evasion remains unclear.Here we performed single-cell RNA sequencing on^104,000 cells from 19 EBV^+NPCs and 7 nonmalignant nasopharyngeal biopsies,simultaneously profiling the transcriptomes of malignant cells,EBV,stromal and immune cells.Overall,we identified global upregulation of interferon responses in the multicellular ecosystem of NPC.Notably,an epithelial–immune dual feature of malignant cells was discovered and associated with poor prognosis.Functional experiments revealed that tumor cells with this dual feature exhibited a higher capacity for tumorigenesis.Further characterization of the cellular components of the tumor microenvironment(TME)and their interactions with tumor cells revealed that the dual feature of tumor cells was positively correlated with the expression of co-inhibitory receptors on CD8^+tumor-infiltrating T cells.In addition,tumor cells with the dual feature were found to repress IFN-γproduction by T cells,demonstrating their capacity for immune suppression.Our results provide new insights into the multicellular ecosystem of NPC and offer important clinical implications.Shanzhao Jin Ruoyan Li Ming-Yuan Chen Chao Yu Lin-Quan Tang Yan-Min Liu Jiang-Ping Li Yi-Na Liu Yi-Ling Luo Yifan Zhao Yu Zhang Tian-Liang Xia Shang-Xin Liu Qi Liu Guan-Nan Wang Rui You Jing-Yun Peng Jiang Li Feng Han Jianwei Wang Qiu-Yan Chen Li Zhang Hai-Qiang Mai Benjamin E.Gewurz Bo Zhao Lawrence S.Young Qian Zhong Fan Bai Mu-Sheng Zeng 2020Cell Research2020,30,11:16
6ERA modal identification method for hydraulic structures based on order determination and noise reduction of singular entropy显示文摘Modal parameter has been considered as one of the indices in assessing the working state of hydraulic structures. The method for obtaining the modal parameter indices is crucial for health monitoring of hydraulic structures. Traditionally, model parameters are measured from the prototype dynamic test under the excitation of special equipment. The preparation is time-consuming and the method is difficult to carry out. We propose a method in this paper to denoise, reconstruct, determine the order and identify modal parameters based on singular entropy (SE) of eigenmatrix. It can be used to solve the problem of system eigenmatrix order determination, and to identify the modal order and the characteristics of structures under the working state. Finally, this method is applied to identify the operational modal parameters of Ertan arch dam under different flood discharging states and vibration frequencies of the powerhouse with a two-row placed unit in Lijiaxia hydropower station during the unit halting process. 8Addition of hydrogen peroxide for the simultaneous control of bromate and odor during advanced drinking water treatment using ozone显示文摘Complete removal of the characteristic septic/swampy odor from Huangpu River source water could only be achieved under an ozone dose as high as 4.0 mg/L in an ozone-biological activated carbon(O3-BAC) process, which would lead to the production of high concentrations of carcinogenic bromate due to the high bromide content. This study investigated the possibility of simultaneous control of bromate and the septic/swampy odor by adding H2O2 prior to the O3-BAC process for the treatment of Huangpu River water. H2O2 addition could reduce the bromate concentration effectively at an H2O2/O3(g/g) ratio of 0.5 or higher. At the same time, the septic/swampy odor removal was enhanced by the addition of H2O2, although optimization of the H2O2/O3 ratio was required for each ozone dose. At an ozone dose of 2.0 mg/L, the odor was removed completely at an H2O2/O3 ratio of 0.5. 9A new bone repair scaffold combined with chitosan/ hydroxyapatite and sustained releasing icariin显示文摘Icariin, a plant-derived flavonol glycoside, has been proved as an osteoinductive agent for bone tissue engineering. A new bone repair scaffold was generated by thorough mixing of icariin and chitosan/ hydroxyapatite (icariin-CS/HA) using freeze-drying technigue. Characteristics of morphology, mechanical properties, biocompatibility, drug release behavior and bone repair abilities in vivo were evaluated. The results show that drug loading process of icariin did not affect physical structure of CS/HA composite significantly but decreased mechanical properies of CS/HA composite, which happened with a high dosage; icariin-CS/HA had favorable cell compatibility and promoted osteogenic differentiation of hBMSCs; the controlled release of icariin was satisfactory and the release retained after 90 d in vitro. In addition, icariin-CS/HA scaffolds had favorable osteoconduction and osteoinduction in vivo, and could fill bone defect sites and stimulate newborn bone tissues formation at early stage. 13Characterization of disinfection byproduct formation potential in 13 source waters in China显示文摘The formation potential of four trihalomethanes (THMFP) and seven haloacetic acids (HAA 7 FP) in 13 source waters taken from four major water basin areas in China was evaluated using the simulated distribution system (SDS) chlorination method.The specific ultraviolet absorbance (SUVA 254:the ratio of UV 254 to dissolved organic carbon (DOC)),which ranged between 0.9 and 5.0 L/(mg·m),showed that the organic compounds in different source waters exhibited different reactivities with chlorine.The HAA 7 FP of source waters ranged from 20 to 448 μg/L and the THMFP ranged from 29 to 259 μg/L.The HAA 7 FP concentrations were higher than the THMFP concentrations in all but one of the samples.Therefore,the risks of haloacetic acids (HAAs) should be of concern in some source waters.TCM (chloroform) and BDCM (bromodichloromethane) were the major THM constituents,while TCAA (trichloroacetic acid) and DCAA (dichloroacetic acid) were the major HAA species.Br-THM (brominated THM species) were much higher than BrHAA (brominated HAA species),and the formation of Br-DBP (Br-THM and Br-HAA) should be of concern when the bromide concentration is over 100 μg/L.Junzhi Zhang Jianwei Yu Wei An Juan Liu Yongjing Wang Youjun Chen Jia Tai Min Yang 2011Journal of Environmental Sciences2011,23,2:13
14Soil respiration in tropical seasonal rain forest in Xishuangbanna, SW China显示文摘With the static opaque chamber and gas chromatography technique, from January 2003 to January 2004 soil respiration was investigated in a tropical seasonal rain forest in Xishuangbanna, SW China. In this study three treatments were applied, each with three replicates: A (bare soil), B (soil+litter), and C (soil+litter+seedling). The results showed that soil respiration varied seasonally, low from December 2003 to February 2004, and high from June to July 2004. The annual average values of CO2 efflux from soil respiration differed among the treatments at 1% level, with the rank of C (14642 mgCO2· m-2. h-1)>B (12807 mgCO2· m-2. h-1)>A (9532 mgCO2· m-2. h-1). Diurnal variation in soil respiration was not apparent due to little diurnal temperate change in Xishuangbanna. There was a parabola relationship between soil respiration and soil moisture at 1% level. Soil respiration rates were higher when soil moisture ranged from 35% to 45%. There was an exponential relationship between soil respiration and soil temperature (at a depth of 5cm in mineral soil) at 1% level. The calculated Q1o values in this study,ranging from 2.03 to 2.36, were very near to those of tropical soil reported. 15On-board measurement of particle numbers and their size distribution from a light-duty diesel vehicle:Influences of VSP and altitude显示文摘In this study,the particle size-resolved distribution from a China-3 certificated light-duty diesel vehicle was measured by using a portable emission measurement system(PEMS).In order to examine the influences of vehicle specific power(VSP) and high-altitude operation,measurements were conducted at 8 constant speeds,which ranged from 10 to 80 km/hr at10 km/hr intervals,and two different high altitudes,namely 2200 and 3200 m.The results demonstrated that the numbers of particles in all size ranges decreased significantly as VSP increased when the test vehicle was running at lower speeds(< 20 km/hr),while at a moderate speed(between 30 and 60 km/hr),the particle number was statistically insensitive to increase VSP.Under high-speed cruising conditions,the numbers of ultrafine particles and PM2.5were insensitive to changes in VSP,but the numbers of nanoparticles and PM10 surged considerably.An increase in the operational altitude of the test vehicle resulted in increased particle number emissions at low and high driving speeds;however,particle numbers obtained at moderate speeds decreased as altitude rose.When the test vehicle was running at moderate speeds,particle numbers measured at the two altitudes were very close,except for comparatively higher number concentrations of nanoparticles measured at 2200 m.Jia Liu Yunshan Ge Xin Wang Lijun Hao Jianwei Tan Zihang Peng Chuanzhen Zhang Huiming Gong Ying Huang 2017Journal of Environmental Sciences2017,29,7:12
16Sox2 enhances the tumorigenicity and chemoresistance of cancer stem-like cells derived from gastric cancer显示文摘Gastric cancer stem-like cells(GCSCs) have been identified to possess the ability of self-renewal and tumor initi-ation.However,the mechanisms involved remain largely unknown.Here,we isolated and characterized the GCSCs by side population(SP) sorting procedure and cultured sphere cells(SC) from human gastric cancer cell lines SGC-7901,BGC-823,MGC-803,HGC-27 and MKN-28.The sorting and culture assay revealed that SP cells proliferated in an asymmetric division manner.In addition,SP cells exhibited a higher potential of spheroid colony formation and greater drug resistance than non-SP cells(NSP).Moreover,the SC were found with enhanced capabilities of drug resistance in vitro and tumorigenicity in vivo.Sox2 mRNA and protein was highly and significantly overex-pressed in the SP cells and SC.Importantly,downregulation of Sox2 with siRNA obviously reduced spheroid colony formation and doxorubicin efflux,as well as increased apoptosis rate in sphere cells in vitro and suppressed tumori-genicity in vivo.These results suggest that both SP cells and cultured SC enrich with GCSCs and that Sox2 plays a pivotal role in sustaining stem cell properties and might be a potential target for gastric cancer therapy.Tian Tian Yajie Zhang Shouyu Wang Jianwei Zhou Shan Xu 2012The Journal of Biomedical Research2012,26,5:11

18Lattice-Based Double-Authentication-Preventing Ring Signature for Security and Privacy in Vehicular Ad-Hoc Networks显示文摘Amidst the rapid development of the Internet of Things (loT), Vehicular Ad-Hoc NETwork (VANET), a typical loT application, are bringing an ever-larger number of intelligent and convenient services to the daily lives of individuals. However, there remain challenges for VANETs in preserving privacy and security. In this paper, we propose the first lattice-based Double-Authentication-Preventing Ring Signature (DAPRS) and adopt it to propose a novel privacy-preserving authentication scheme for VANETs, offering the potential for security against quantum computers. The new construction is proven secure against chosen message attacks. Our scheme is more efficient than other ring signature in terms of the time cost of the message signing phase and verification phase, and also in terms of signature length. 20Treatment of multiple sclerosis by transplantation of neural stem cells derived from induced pluripotent stem cells显示文摘Multiple sclerosis(MS) is an autoimmune disease of the central nervous system(CNS), with focal T lymphocytic infiltration and damage of myelin and axons. The underlying mechanism of pathogenesis remains unclear and there are currently no effective treatments. The development of neural stem cell(NSC) transplantation provides a promising strategy to treat neurodegenerative disease. However, the limited availability of NSCs prevents their application in neural disease therapy. In this study, we generated NSCs from induced pluripotent stem cells(iPSCs) and transplanted these cells into mice with experimental autoimmune encephalomyelitis(EAE), a model of MS. The results showed that transplantation of iPSC-derived NSCs dramatically reduced T cell infiltration and ameliorated white matter damage in the treated EAE mice. Correspondingly, the disease symptom score was greatly decreased, and motor ability was dramatically rescued in the iPSC-NSC-treated EAE mice, indicating the effectiveness of using iPSC-NSCs to treat MS.
