| 3 | Inositol Pyrophosphate lnsP8 Acts as an Intracellular Phosphate Signal in Arabidopsis显示文摘The maintenance of cellular phosphate(Pi)homeostasis is of great importance in living organisms.The SPX domain-containing protein 1(SPX1)proteins from both Arabidopsis and rice have been proposed to act as sensors of Pi status.The molecular signal indicating the cellular Pi status and regulating Pi homeostasis in plants,however,remains to be identified,as Pi itself does not bind to the SPX domain.Here,we report the identification of the inositol pyrophosphate lnsP8 as a signaling molecule that regulates Pi homeostasis in Arabidopsis.Polyacrylamide gel electrophoresis profiling of InsPs revealed that lnsP8 level positively correlates with cellular Pi concentration.We demonstrated that the homologs of diphosphoinositol pentaki-sphosphate kinase(PPIP5K),VIH1 and VIH2,function redundantly to synthesize lnsP8,and that the vih1 vih2 double mutant overaccumulates Pi.SPX1 directly interacts with PHR1,the central regulator of Pi starvation responses,to inhibit its function under Pi-replete conditions.However,this interaction is compromised in the vih1 vih2 double mutant,resulting in the constitutive induction of Pi starvation-induced genes,indicating that plant cells cannot sense cellular Pi status without lnsP8.Furthermore,we showed that lnsP8 could directly bind to the SPX domain of SPX1 and is essential for the interaction between SPX1 and PHR1.Collectively,our study suggests that lnsP8 is the intracellular Pi signaling molecule serving as the ligand of SPX1 for controlling Pi homeostasis in plants. | Jinsong Dong Guojie Ma Liqian Sui Mengwei Wei Viswanathan Satheesh Ruyue Zhang Shenghong Ge Jinkai Li Tong-En Zhang Christopher Wittwer Henning JJessen Huiming Zhang Guo-Yong An Dai-Yin Chao Dong Liu Mingguang Lei | 2019 | Molecular Plant2019,12,11: | 8 |

| 6 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 8 | The telomere-to-telomere genome of Fragaria vesca reveals the genomic evolution of Fragaria and the origin of cultivated octoploid strawberry显示文摘Fragaria vesca,commonly known as wild or woodland strawberry,is the most widely distributed diploid Fragaria species and is native to Europe and Asia.Because of its small plant size,low heterozygosity,and relative ease of genetic transformation,F.vesca has been a model plant for fruit research since the publication of its Illumina-based genome in 2011.However,its genomic contribution to octoploid cultivated strawberry remains a long-standing question.Here,we de novo assembled and annotated a telomere-to-telomere,gap-free genome of F.vesca‘Hawaii 4’,with all seven chromosomes assembled into single contigs,providing the highest completeness and assembly quality to date.The gap-free genome is 220785082 bp in length and encodes 36173 protein-coding gene models,including 1153 newly annotated genes.All 14 telomeres and seven centromeres were annotated within the seven chromosomes.Among the three previously recognized wild diploid strawberry ancestors,F.vesca,F.iinumae,and F.viridis,phylogenomic analysis showed that F.vesca and F.viridis are the ancestors of the cultivated octoploid strawberry F.×ananassa,and F.vesca is its closest relative.Three subgenomes of F.×ananassa belong to the F.vesca group,and one is sister to F.viridis.We anticipate that this high-quality,telomere-to-telomere,gap-free F.vesca genome,combined with our phylogenomic inference of the origin of cultivated strawberry,will provide insight into the genomic evolution of Fragaria and facilitate strawberry genetics and molecular breeding. | Yuhan Zhou Jinsong Xiong Ziqiang Shu Chao Dong Tingting Gu Pengchuan Sun Shuang He Mian Jiang Zhiqiang Xia Jiayu Xue Wasi Ullah Khan Fei Chen Zong-Ming Cheng | 2023 | Horticulture Research2023,10,4: | 4 |

| 12 | Effect of Nao Yikang on choline acetyltransferase and caspase-3 brain expression in a rat model of Alzheimer's disease显示文摘BACKGROUND: The main components of the traditional Chinese medicine compound Nao Yikanghave been shown to possibly alleviate neural damage.OBJECTIVE: To observe the effects of Nao Yikang on expression of choline acetyltransferase(ChAT) and caspase-3 in the rat brains of an experimental Alzheimer's disease (AD) model, and toinvestigate the mechanisms of potential neuroprotective effects.DESIGN, TIME AND SETTING: A randomized, controlled experiment was performed at theDepartment of Pathophysiology, Medical School of Nantong University between November 2006and December 2007.MATERIALS: The main active components of Nao Yikang were as follows: prepared polygonummultiflorum, Rhizoma anemarrhenae, and Rhizoma acori tatarinowii. Nao Yikang granules wereprepared by Nantong Hospital of Traditional Chinese Medicine. Ibotenic acid (IBO) was purchasedfrom Sigma-Aldrich, USA, ChAT goat anti-rat antibody from Chemicon, USA, and cleaved caspase-3rabbit anti-rat (Asp175) (5A1) antibody from Cell Signaling, USA.METHODS: A total of 60 male, Sprague Dawley rats (2 months old) were randomly assigned to 6groups: sham-surgery, model, Nao Yikang 1.73, 3.45, 6.90 g/kg per day, and piracetam, with 10 ratsin each group. Bilateral infusions of 5 μg IBO into the nucleus basalis of Meynert were performedwith Hamilton syringe and stereotaxic apparatus for AD model establishment. For the sham-surgerygroup, rats received 1 μL saline in the identical stereotaxic position. From the second day, NaoYikang groups were administrated 1.73, 3.45, and 6.90 g/kg per day Nao Yikang, respectively, whilethe piracetam group received 0.04 g/mL piracetam, the model group received 0.5% sodiumcarboxymethyl cellulose, and the sham-surgery group received normal saline. Rats wereintragastrically administered 1 mL/100 g daily for 28 consecutive days.MAIN OUTCOME MEASURES: Following treatment of the various solutions for 28 days, Westernblot was utilized to observe ChAT expression in the frontal cortex of AD rats, andimmunohistochemistry was applied to quantify caspase-3-positive cells in the frontal cortex.RESULTS: ChAT protein expression significantly decreased in the model group (P < 0.01), howevercaspase-3 expression was significantly elevated (P < 0.01) compared with the sham-surgery group.Compared with the model group, ChAT protein expression increased in the Nao Yikang 1.73 g/kgper day, 3.45 g/kg per day, 6.90 g/kg per day groups, and the piracetam group (P < 0.05 or P < 0.01)and the number of caspase-3-positive cells decreased in the Nao Yikang 3.45 g/kg per day and6.90 g/kg per day groups (P < 0.01). | 13 | Colonization of endophyte Acremonium sp. D212 in Panax notoginseng and rice mediated by auxin and jasmonic acid显示文摘Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms under-lying colonization of Acremonium spp. remain unclear. In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng, enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin bio-synthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid (IAA) and jasmonic acid (JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. noto-ginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate (MeJA) (2–15μmol/L) and 1-naphthalenacetic acid (NAA) (10–20μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and miR393b-overexpressing lines, and the colonization was res-cued by MeJA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. | 14 | Assessment of the risk of infectious aerosols leaking to the environment from BSL-3 laboratory HEPA air filtration systems using model bacterial aerosols显示文摘To assess the risk of infectious bacterial aerosols leaking to the environment, the filtration efficiency of a biosafety level 3(BSL-3) laboratory high-efficiency particulate(HEPA) filter was investigated using the aerosolized bacteria Serratia marcescens. The aerosol size was measured using an Andersen sampler. Eight first stage HEPA filters(numbered 1-8) were distributed in contaminated labs and exhausts from each0f the first stage HEPA filters were aggregated and filtered through one second stage HEPA filter before being released to the environment. In total, 8 first-stage and 1 second-stage HEPA filters from the BSL-3 air purification system were analyzed. No S. marcescens was detected in first stage filters 1,2,4,5,7 and 8 and the second stage HEPA filter. The filtration efficiencies against aerosolized S. marcescens were >99.9999%.First stage filter numbers 3 and 6 had filtration efficiencies of 99.9825% and 99.9906%, respectively. When filter number 3 was replaced by a new filter and the bracket for filter number 6 was sealed, no aerosolized S. marcescens was detected in the filtered air.
