1Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
2Van der Waals epitaxy of ultrathin crystalline PbTe nanosheets with high near-infrared photoelectric response显示文摘Lead telluride(PbTe)is one of the reliable candidates for infrared(IR)optoelectronics with optimum band-gap as well as excellent photoelectric properties.Great interests had been paid on the growth and device applications with PbTe for the development of high-performance IR photodetectors especially those working in the near-infrared regime.Although a great deal of effort had been made to prepare PbTe nanostructures for miniaturized detectors,it is difficult to synthesize high-quality two-dimensional(2D)PbTe crystals due to its rock-salt non-layered structure.Herein,a facile strategy for controllable synthesis of ultrathin crystalline PbTe nanosheets by van der Waals epitaxy is reported.With an optimized growth temperature,which determines the morphology transit from triangular pyramid islands to regular square 2D planars,PbTe nanosheets in lateral size of tens of microns with thickness down to~7 nm are achieved.Meanwhile,ultrasensitive near-infrared detectors(NIRDs)based on the as-grown 2D PbTe nanosheets have been demonstrated with an ultrahigh responsivity exceeding 3,847 A/W at the wavelength of 1,550 nm under room temperature.Our approach demonstrates that 2D PbTe nanosheets have great latent capacity of developing high-performance miniaturized IR optoelectronic devices.Xinxin Zhao Qing Yin Hao Huang Qiang Yu Bo Liu Jie Yang Zhuo Dong Zhenjiang Shen Benpeng Zhu Lei Liao Kai Zhang 2021Nano Research2021,14,6:3
3Super-plasticity of Zr_(64.80)Cu_(14.85)Ni_(10.35)Al_(10) bulk metallic glass at room temperature显示文摘Generally,bulk metallic glasses(BMGs)exhibit a very limited plastic deformation under a compression load at room temperature,often less than 2% before fracturing.In this letter,through an appropriate choice of BMGs' composition,an amorphous rod of Zr64.80Cu14.85Ni10.35Al10 with a diameter of 2 mm was prepared by using copper mold suction casting.X-ray diffraction and differential scanning calorimetry were utilized to determine its structure and thermal stability,and the uniaxial compression test was adopted to study its plastic deformation behavior at room temperature simultaneously.The results showed that the glass transition temperature and onset temperature of the exothermic reaction of the amorphous rod were 646 and 750 K,respectively,and its micro-hardness was 594.7 Hv.During com-pression,when the engineering strain and engineering stress arrived at 9.05% and 1732 MPa,respec-tively,i.e.,the true strain and true stress reached 9.42% and 1560 MPa,respectively,the amorphous rod started to yield.After yielding,with the increase of load,the strain increased and the glass rod ulti-mately were compressed into flake-like form.Although the maximum engineering strain was larger than 70%,i.e.,the maximum true strain exceeded by 120%,the amorphous specimen was not fractured,indicating that it has super-plasticity at room temperature.Through the appropriate choice of compo-sition and optimization of the technological process,flexible BMG with super-plasticity at room tem-perature could be produced.TAO PingJun YANG YuanZheng BAI XiaoJun XIE ZhiWei CHEN XianCao DONG ZhenJiang Wen JianGuo 2008Chinese Science Bulletin2008,53,3:3

5Android Apps:Static Analysis Based on Permission Classification显示文摘Android has a strict permission management mechanism. Any applications that try to run on the Android system need to obtain permission. In this paper, we propose an efficient method of detecting malicious applications in the Android system. First, hundreds of permissions are classified into different groups. The application programming interfaces (APIs) associated with permissions that can interact with the outside environment are called sink functions. The APIs associated with other permissions are called taint functions. e construct association tables for block variables and function variables of each application. 10MBGM: A Graph-Mining Tool Based on MapReduce and BSP显示文摘This paper proposes an analytical mining tool for big graph data based on Map Reduce and bulk synchronous parallel(BSP) com-puting model. The tool is named Mapreduce and BSP based Graph-mining tool(MBGM). The core of this mining system are foursets of parallel graph-mining algorithms programmed in the BSP parallel model and one set of data extraction-transformation-load-ing(ETL) algorithms implemented in Map Reduce. To invoke these algorithm sets, we designed a workflow engine which optimizedfor cloud computing. Finally, a well-designed data management function enables users to view, delete and input data in the Ha-doop distributed file system(HDFS).
