| 1 | Determination of PCDD/Fs and dioxin-like PCBs in food and feed using gas chromatography-triple quadrupole mass spectrometry显示文摘A method was developed on a gas chromatograph coupled to a triple quadrupole mass spectrometer(GC-MS/MS) for trace level determination of polychlorinated dibenzo-p-dioxins/dibenzofurans(PCDD/Fs) and dioxin-like polychlorinated biphenyls(DL-PCBs) in food and feed. The results demonstrated good sensitivity and repeatability for PCDD/Fs and DL-PCBs at an extremely low level(10 pg mL^(-1) for 2,3,7,8-TCDD/F), as well as wide linear response of over 3 or 4 orders of magnitude in concentration ranges; 0.5–200 ng mL^(-1) for PeCDD/F and 0.2–2000 ng mL^(-1) for DL-PCBs. The method detection limits for PCDD/Fs and DL-PCBs were in the range from 0.018–0.17 pg g^(-1) to 0.13–0.36 pg g^(-1), respectively. The performance of the GC-MS/MS for food and feed sample analysis showed high precision and accuracy compared to the high resolution gas chromatograph/high resolution mass spectrometer. | 2 | Application of isotopic and hydro-geochemical methods in identifying sources of mine inrushing water显示文摘Isotopic and hydro-geochemical surveys were carried out to identify the source of mine inrushing water at the #73003 face in the Laohutai Mine. Based on the analysis of isotopes and hydro-chemical features of surface water, groundwater from different levels and the inrushing water, a special relationship between water at the #73003 face and cretaceous water has been found. The results show that the isotopic and hydro-chemical features of the inrushing water are completely different from those of other groundwater bodies, except for the cretaceous water. The isotopic and hydrochemical characteristics of cretaceous water are similar to the inrushing water of the #73003 face, which aided with obtaining the evidence for the possible source of the inrushing water at the #73003 face. The isotope calculations show that the inrushing water at the #73003 face is a mixture of cretaceous water and Quaternary water. Water from the cretaceous conglomerate is the main source, accounting for 67% of the inrushing water, while the Quaternary water accounts for 33%. | 3 | Atmospheric levels and distribution of Dechlorane Plus in an E-waste dismantling region of East China显示文摘Atmospheric concentrations of Dechlorane Plus(DP)were investigated in Taizhou,an electronic-waste(E-waste)dismantling region in East China.Passive air samplers with polyurethane foam(PUF)disks were deployed every three months during the sampling period of September 2009-August 2010.The total DP(syn-and anti-DP)concentrations in air ranged from not detected to 277 pg/m^3,with a mean concentration of 53.9 pg/m^3.A generally declining trend of DP levels was found from the E-waste dismantling region to the peripheral areas.The median values of total DP concentrations in autumn,winter,spring and summer were 52.2,28.8,39.7 and 30.1 pg/m^3,respectively.The seasonal variations of DP concentrations were mainly associated with the intensity of E-waste dismantling activities and meteorological conditions.The mean value of anti-DP fractional abundance(f_(anti))was 0.74±0.08,which was consistent with those in the commercial DP products.This study confirmed a significant emission source related to the distribution of atmospheric DP in the E-waste dismantling area and supplied information for the seasonal variation of DP in the atmosphere. | Huizhong Sun Yingming Li Pu Wang Shucheng Zheng Julius Matsiko Dou Wang Weiwei Zhang Yanfen Hao Qinghua Zhang Guibin Jiang | 2017 | Science China Chemistry2017,60,2: | 2 |
| 4 | Colorimetric determination of nanomolar concentrations of silicate in natural waters after liquid-liquid extraction using methyl isobutyl ketone显示文摘A sensitive solvent extraction method for the determination of nonamolar concentrations of silicate in natural waters is developed. According to the traditional aqueous silicate method,silicomolybdenum blue formed by the reaction between silicate and ammonium molydate and reduced by metol-sulfite reagent is extracted by methyl isobutyl ketone. The absorbance can be enhanced substantially up to 10-folds. The detection limit of silicate is 8 nmol/dm3,which is one tenth smaller than the traditional method,with the precision of 4.0% at a silicate level of 50 nmol/dm3 and 3.2% at a silicate level of 6 μmol/dm3. Comparing the calibration curves in the distilled water and seawater,it can be seen that the salt effect also exists in the extraction method. However,the salt effect is a linear function of the salinity and can be corrected by simple calibration. The proposed method is successfully applied to the determination of silicate in natural waters. | 5 | Nicotiana benthamiana LRR-RLP Nb EIX2 mediates the perception of an EIX-like protein from Verticillium dahliae显示文摘Verticillium wilt diseases caused by the soil-borne fungus Verticillium dahliae result in devastating yield losses in many economically important crops annually. Here, we identified a novel ethyleneinducing xylanase(EIX)-like protein, VdEIX3, from V. dahliae, which exhibits immunity-inducing activity in Nicotiana benthamiana. In vitro-purified VdEIX3 can induce strong oxidative burst, activate the expression of defense-related genes, and increase resistance against oomycete and fungal pathogens in N. benthamiana. VdEIX3 orthologs of other Verticillium pathogens also induce cell death in N. benthamiana, which form a new type of EIX protein family that is distinct from the known EIX proteins. A leucine-rich repeat receptor-like protein, NbEIX2, regulates the perception of VdEIX3 in N. benthamiana. | 6 | A G-type lectin receptor-like kinase regulates the perception of oomycete apoplastic expansin-like proteins显示文摘Phytophthora capsici is one of the most harmful pathogens in agriculture, which threatens the safe production of multiple crops and causes serious economic losses worldwide. Here, we identified a P. capsici expansin-like protein, Pc EXLX1, by liquid chromatography-tandem mass spectrometry from Nicotiana benthamiana apoplastic fluid infected with P. capsici. Clustered regularly interspaced short palindromic repeats/crispr associated protein9(CRISPR/Cas9)-mediated Pc EXLX1 knockout mutants exhibited significantly enhanced virulence,while the overexpression of Pc EXLX1 impaired the virulence. Prokaryotically expressed Pc EXLX1 activated multiple plant immune responses, which were BRI1-associated kinase 1(BAK1)-and suppressor of BIR1-1(SOBIR1)-dependent. Furthermore, overexpression of Pc EXLX1 homologs in N. benthamiana could also increase plant resistance to P. capsici. A G-type lectin receptor-like kinase from N. benthamiana, expansin-regulating kinase 1(ERK1), was shown to regulate the perception of Pc EXLX1 and positively mediate the plant resistance to P. capsici. | 7 | Monitoring the binding of metal cations and histones to DNA in real time using fluorescence assays显示文摘The binding of cations (Na +,K +,Mg 2+,Ca 2+,Mn 2+) and histones to DNA can be studied using fluorescence assays.Here,we measured the fluorescence intensity and fluorescence anisotropy of DNA and DNA-histone complexes in the presence of cations.We demonstrate that when different cations are added into a DNA solution,the fluorescence intensities of the stained DNA are reduced by different amounts.Compared with divalent cations,monovalent cations had a weaker effect on fluorescence intensity and fluorescence anisotropy.Divalent (Mn 2+,Mg 2+,Ca 2+) cations markedly enhanced the fluorescence anisotropy of DNA.The binding modes of monovalent and divalent cations to DNA may be different.Divalent cations can change the structure of DNA molecules,or promote the assembly of DNA strands.The addition of histones causes DNA condensation,which mostly occurs during the first few seconds.Cation binding to DNA is abrupt,and is much faster than that of histones. | LIU YuYing WANG PengYe DOU ShuoXing ZHANG WeiWei WANG XueJin SANG HongYi | 2011 | Chinese Science Bulletin2011,56,11: | 0 |
| 8 | Study of the interaction of DNA and histones by spin-stretching and droplet evaporation显示文摘Molecular combing is a powerful and simple method for aligning DNA molecules onto a surface. Using this technique combined with fluorescence microscopy, DNA-histone complexes are stretched on a hydrophobic polymethyl methacrylate (PMMA) surface and observed directly. We have developed a new method to stretch single DNA-histone complexes, termed spin-stretching. The results show that the histones markedly enhance DNA binding to the PMMA surface. DNA winds around the histones and therefore decreases in length. The number of histones that bind to each DNA molecule is found to correlate with the histone concentration. The combed DNA-histone complexes are found to depend on two factors: the binding force on the surface and the centrifugal force at its local position.
