Effects of thymol and carvacrol supplementation on intestinal integrity and immune responses of broiler chickens challenged with Clostridium perfringens显示文摘Background: Necrotic enteritis caused by Clostridium perfringens infection leads to serious economic losses in the global poultry production. In the present study, we investigated the protective effects of essential oils(EO, which contained 25 % thymol and 25 % carvacrol as active components) supplementation on growth performance, gut lesions, intestinal morphology, and immune responses of the broiler chickens infected with C. perfringens. A total of448 1-day-old male broiler chicks were allocated into eight treatment groups following a 4 × 2 factorial arrangement with four dietary EO dosages(0, 60, 120, or 240 mg/kg) and two infection status(with or without C. perfringens challenge from d 14 to 20).Results: The challenge did not impair the growth performance of birds, but induced gut lesions and increased crypt depth in the ileum(P ≤ 0.05). It also down-regulated the claudin-1 and occludin m RNA expression(P ≤ 0.05),up-regulated the m RNA expression of interleukin-1β(P ≤ 0.05), tended to increase the toll-like receptor(TLR)2 m RNA expression(P < 0.10) in the ileum, and enhanced the mucosal secretory Ig A production(P ≤ 0.05). In the challenged birds, dietary EO supplementation linearly alleviated the gut lesions and improved the ratio of vil us height to crypt depth(P ≤ 0.05), and the supplementation of 120 and 240 mg/kg EO increased the serum antibody titers against Newcastle disease virus(P ≤ 0.05). Regardless of challenge, the EO supplementation showed a tendency to linearly elevate the feed conversion efficiency between 14 and 28 d of age as well as the occludin m RNA expression(P < 0.10), and linearly inhibited the m RNA expression of TLR2 and tumor necrotic factor-α in the ileum(P ≤ 0.05).Conclusions: The dietary supplementation of EO could alleviate the intestinal injury by improving intestinal integrity and modulating immune responses in the C. perfringens-challenged broiler chickens. | Encun Du Weiwei Wang Liping Gan Zhui Li Shuangshuang Guo Yuming Guo | 2016 | Journal of Animal Science and Biotechnology2016,7,4: | 43
In vitro antibacterial activity of thymol and carvacrol and their effects on broiler chickens challenged with Clostridium perfringens显示文摘Background:In the post-antibiotic era, essential oils(EO) are promising alternatives to growth-promoting antibiotics. The aim of the present study was to investigate the antibacterial activities of an EO product and its components thymol and carvacrol in vitro, and the efficacy of EO to control Clostridium perfringens challenge in broiler chickens. Results:The in vitro minimum inhibitory concentration assay showed strong antibacterial activity of the EO product, thymol, and carvacrol against pathogenic Escherichia coli,C.perfringens, and Salmonella strains, and weak activity towards beneficial Lactobacillus strains.Besides,an additive effect was observed between thymol and carvacrol. The in vivo study was carried out with 448 male broiler chicks following a 4 × 2 factorial arrangement to test the effects of EO supplementation(0,60,120,or 240 mg/kg EO in wheat-based diet),pathogen challenge(with or without oral gavage of C perfringens from day 14 to day 20) and their interactions. Each treatment consisted of eight replicate pens(seven birds/pen).The challenge led to macroscopic gut lesions, and resulted in a significant increase in ileal populations of C.perfringens and Escherichia subgroup(P< 0.05) on day 21.Dietary EO supplementation did not influence C perfringens numbers, but linearly alleviated intestinal lesions on day 21 and 28(P=0.010 and 0.036,respectively),and decreased Escherichia populations in ileum with increased EO dosages(P=0.027 and 0.071 for day 21 and 28,respectively).For caecum,EO quadratically influenced Lactobacillus populations on day 21(p=0.002),and linearly decreased the numbers of total bacteria and Escherichia on day 28(P=0.026 and 0.060,respectively).Mean thymol and carvacrol concentrations in the small intestine were 0.21 and 0.20 μg/g in intestinal digesta(wet weight),respectively, for birds fed 60 mg/kg EO,and 0.80 and 0.71 μg/g,respectively,for birds fed 240 mg/kg EO.Conclusions:These results indicated that dietary EO supplementation could affect intestinal microbiota and alleviate intestinal lesions in broilers, which may contribute in controlling C.perfringens infection in broiler chickens. | Encun Du Liping Gan Zhui Li Weiwei Wang Dan Liu Yuming Guo | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 14

Restorative effect and mechanism of mecobalamin on sciatic nerve crush injury in mice显示文摘Mecobalamin,a form of vitamin B12 containing a central metal element(cobalt),is one of the most important mediators of nervous system function. In the clinic,it is often used to accelerate recovery of peripheral nerves, but its molecular mechanism remains unclear. In the present study,we performed sciatic nerve crush injury in mice, followed by daily intraperitoneal administration of mecobalamin(65 μg/kg or 130 μg/kg) or saline(negative control).Walking track analysis, histomorphological examination, and quantitative real-time PCR showed that mecobalamin significantly improved functional recovery of the sciatic nerve, thickened the myelin sheath in myelinated nerve fibers, and increased the cross-sectional area of target muscle cells.Furthermore, mecobalamin upregulated m RNA expression of growth associated protein 43 in nerve tissue ipsilateral to the injury, and of neurotrophic factors(nerve growth factor,brain-derived nerve growth factor and ciliary neurotrophic factor) in the L4–6 dorsal root ganglia. Our findings indicate that the molecular mechanism underlying the therapeutic effect of mecobalamin after sciatic nerve injury involves the upregulation of multiple neurotrophic factor genes. | Lin Gan Minquan Qian Keqin Shi Gang Chen Yanglin Gu Wei Du Guoxing Zhu | 2014 | Neural Regeneration Research2014,9,22: | 11
Chinese Herbal Medicine Combined with Entecavir for HBeAg Positive Chronic Hepatitis B:Study Protocol for a Multi-Center,Double-Blind Randomized-Controlled Trial显示文摘Background: The domestic prevalence of chronic hepatitis B(CHB) in China is 7.18% in 2006, imposing great societal healthcare burdens. Nucleot(s)ide analogues(NUCs) anti-hepatitis B virus(HBV) therapies are widely applied despite the relatively low rate of seroconversion and high risk of drug-resistant mutation. More effective treatments for CHB deserve further explorations. Combined therapy of NUCs plus Chinese herbal medicine(CHM) is widely accepted in China, which is recognized as a prospective alternative approach. The study was primarily designed to confirm the hypothesis that Tiaogan-Yipi Granule(调肝益脾颗粒, TGYP) or Tiaogan-Jianpi-Jiedu Granule(调肝健脾解毒颗粒, TGJPJD) plus entecavir tablet(ETV) was superior over ETV monotherapy in enhancing HBeA g loss rate. Methods: The study was a nationwide, large-scale, multi-center, double-blind, randomized, placebo-control ed trial with a designed duration of 108 weeks. A total of 16 hospitals and 596 eligible Chinese HBeA g positive CHB patients were enrolled from November 2012 to September 2013 and randomly allocated into 2 groups in 1:1 ratio via central randomization system: experimental group(EG) and control group(CG). Subjects in EG received CM formulae(TGYP or TGJPJD, 50 g per dose, twice daily) plus ETV tablet(or ETV placebo) 0.5 mg per day in the first 24 weeks(stage 1), and CHM granule plus ETV tablet(0.5 mg per day) from week 25 to 108(stage 2). Subjects in CG received CHM Granule placebo plus ETV tablet(0.5 mg per day) for 108 weeks throughout the trial. The assessments of primary outcomes(HBV serum markers and HBV-DNA) were conducted by a third-party College of American Pathologists(CAP) qualified laboratory. Adverse effects were observed in the hospitals of recruitment. Discussion: The study was designed to compare the curative effect of CM plus ETV and ETV monotherapy in respect of HBe Ag loss, which is recognized by the European Association for the Study of the Liver as 'a valuable endpoint'. We believe this trial could provide a reliable status for patients' 'journey' towards durable responses after treatment discontinuation. The trial was registered before recruitment on Chinese Clinical trial registry(No. ChiC TR-TRC-12002784, Version 1.0, 2015/12/23). | YE Yong-an LI Xiao-ke ZHOU Da-qiao CHI Xiao-ling LI Qin WANG Li LU Bing-jiu MAO De-wen WU Qi-kai WANG Xian-bo ZHANG Ming-xiang XUE Jing-dong LI Yong LU Wei GUO Jian-chun JIANG Feng ZHANG Xin-wei DU Hong-bo YANG Xian-zhao GUO Hui GAN Da-nan LI Zhi-guo | 2018 | Chinese Journal of Integrative Medicine2018,24,9: | 10

Costunolide alleviates HKSA-induced acute lung injury via inhibition of macrophage activation显示文摘Staphylococcus aureus (S. aureus) infection leads to a severe inflammatory response and causes acute lung injury (ALI), eventually threatening human life. Therefore, it is of importance to find an agent to inhibit infiammation and reduce ALI. Here, we found that costunolide, a sesquiterpene lactone, displays anti-infiammatory effects and ameliorates heat-killed S. aureus (HKSA)-induced pneumonia. Costunolide treatment attenuated HKSA-induced murine ALI in which pulmonary neutrophil infltration was inhibited, lung edema was decreased, and the production of pro-inflammatory cytokines was significantly reduced. In addition, costunolide dose-dependently inhibited the generation of IL-6, TNF-α, IL-1β, and keratinocyte-derived cytokine (KC), as well as the expression of iNOS, in HKSA-induced macrophages. Furthermore, costunolide attenuated the phosphorylation of p38 MAPK and cAMP response element-binding protein (CREB). Collectively, our findings suggested that costunolide is a promising agent for alleviating bacterial-induced ALI via the inhibition of the MAPK signaling pathways. | Yun-tian Chen Yao Du Bo Zhao Li-xing Gan Kai-kai Yu Lei Sun Jian Wang Feng Qian | 2019 | Acta Pharmacologica Sinica2019,40,8: | 7
Effects of arbuscular mycorrhizal fungi on the drought resistance of the mining area repair plant Sainfoin显示文摘To solve the problem of lack of water in arid areas and barren soil in the process of ecological restoration, the growth and drought resistance of Sainfoin(Onobrychis viciifolia Scop.) was studied by inoculation of AMF(arbuscular mycorrhizal fungi), under drought stress. The results show that the inoculation of AMF can significantly improve the growth and mycorrhizal infection rate of Sainfoin and accelerate the absorption of water and nutrients by the roots compared with uninoculated plants. On the 40 th day under drought stress, the relative water content, total nitrogen and total phosphorus content of the leaves of inoculated plants increased by 7.27%, 4.21% and 2.40%, respectively, the osmotic adjustment property in plant cells was improved, the protective enzyme system was improved and membrane lipid peroxidation was reduced. The inoculation of AMF can slow down the damage from drought, improve the drought resistance of Sainfoin, and will be of great significance to the ecologic restoration in arid areas. | Kong Jing Pei Zongping Du Min Sun Gan Zhang Xin | 2014 | International Journal of Mining Science and Technology2014,24,4: | 6
Group VB transition metal dichalcogenides for oxygen reduction reaction and strain-enhanced activity governed by p-orbital electrons of chalcogen显示文摘Developi ng alter native oxyge n reducti on reactio n (ORR) catalysts to replace precious Pt-based metals with abundant materials is the key challe nge of commercial application of fuel cells. Owing to their various compositi ons and tun able electronic properties, transition metal dichalcogenides (TMDs) have the great potential to realize high-efficiency catalysts for ORR. Here, various 3R-phase dichalcogenides of group VB and VIB transition metals (MX2, M = Nb, Ta, Mo, W;X = S, Se, Te) are investigated for ORR catalysts by using density functional theory calculations. The computed over-potentials of group VB TMDs are much less than those of group VIB TMDs. For group VB TMDs, a volcano-type plot of ORR catalytic activity is established on the adsorption energies of *OH, and NbS2 and TaTe2 exhibit best ORR activity with an oveepotential of 0.54 V. To achieve even better activity, strain engineering is performed to tune ORR catalytic activity, and the minimum over-potential of 0.43 V can be realized. We further dem on strate that the shift of p orbital center of surface chalcoge n elements under strain is responsible for tuning the catalytic activity of TMDs. | Shuyang Zhao Ke Wang Xiaolong Zou Lin Gan Hongda Du Chengjun Xu Feiyu Kang Wenhui Duan Jia Li | 2019 | Nano Research2019,12,4: | 4
Synergistic refining mechanism of Mg-3%Al alloy refining by carbon inoculation combining with Ca addition显示文摘Mg-3%Al alloy was refined by carbon inoculation combining with 0.2%Ca addition.High grain refining efficiency was obtained and the synergistic refining mechanism was deeply discussed in the present study.Al-C-O particles, actually Al4C3 particles, were formed in the carbon-inoculated Mg-3%A1 alloy acting as nuclei forα-Mg grains.Ca addition had no obvious effect on size distribution of the nucleating particles.Ca segregation was proved on Al4C3 particles, which should reduce the interfacial energy of nuclei/Mg.The constitutional undercooling in front of nucleus/liquid was increased from 0.12℃to 0.15℃induced by 0.2%Ca addition. The synergistic grain refining efficiency can be attributed to the higher constitutional undercooling and lower the interface energy of nucleus/Mg induced by Ca addition.More nucleating particles with small size could be activated acting as potent nuclei ofα-Mg grains.Consequently,Mg-3%Al alloy could be effectively refined due to the synergistic effect induced by carbon inoculation combining with Ca. | Chengbo Li Shuqing Yang Jun Du HengBin Liao Gan Luo | 2020 | Journal of Magnesium and Alloys2020,8,4: | 3

Analytical algorithm for longitudinal deformation profile of a deep tunnel显示文摘To investigate the longitudinal deformation profile(LDP)of a deep tunnel in non-hydrostatic condition,an analytical model is proposed in our study. In this model, the problem is considered as a superposition of two partial models, and the displacement field of the second partial model is the same as that of the concerned problem. Therefore, the problem can be solved by a model with simple boundary conditions. We obtain the solutions for the stress and displacement fields of an infinite body caused by arbitrary surface tractions on the boundary of the coming tunnel(zone inside the tunnel before excavation)by integrating the extended Kelvin solution over the boundary. The obtained stress solution is used to solve the specific surface tractions, which can satisfy the boundary conditions of the second partial model,on the boundary of the coming tunnel in an infinite body. Then, the specific surface tractions are substituted into the obtained displacement solution to solve the displacement on the wall and face of the tunnel. Therefore, the LDP can also be calculated. The proposed solution is verified by both numerical simulation and the LDP functions recommended by other researchers. The major advantage of our analytical model is that it can consider the effects of the axial and horizontal lateral pressure coefficients. It is revealed that the horizontal lateral pressure coefficient majorly affects the LDP behind the tunnel face, while the axial lateral pressure coefficient dominates the LDP ahead of the tunnel face.Furthermore, the deformation characteristics of the LDPs ahead of the face and the unexcavated core are investigated. The axial displacements of the excavation face can be used to predict the crown displacements ahead of the face. | Qian Fang Gan Wang Fucai Yu Jianming Du | 2021 | Journal of Rock Mechanics and Geotechnical Engineering2021,13,4: | 2
Transferrin plays a central role in coagulation balance by interacting with clotting factors显示文摘Coagulation balance is maintained through fine-tuned interactions among clotting factors, whose physiological concentrations vary substantially. In particular, the concentrations of coagulation proteases(pM to nM)are much lower than their natural inactivator antithrombin(AT,〜3μM),suggesting the existence of other coordinators. In the current study,we found that transferrin(norm plasma concentration〜40 μM)interacts with fibrinogen, thrombin, factor Xlla(FXlla),and AT with different affinity to maintain coagulation balance.Normally, transferrin is sequestered by binding with fibrinogen(normal plasma concentration〜10 μM)at a molar ratio of 4:1.In atherosclerosis, abnormally up-regulated transferrin interacts with and potentiates thrombin/FXlla and blocks ATs inactivation effect on coagulation proteases by binding to AT,thus inducing hypercoagulability. In the mouse model, transferrin overexpression aggravated atherosclerosis, whereas transferrin inhibition via shRNA knockdown or treatment with antitransferrin antibody or designed peptides interfering with transferrin-thrombin/FXlla interactions alleviated atherosclerosis.Collectively, these findings identify that transferrin is an important clotting regulator and an adjuster in the maintenance of coagulation balance and modifies the coagulation cascade. | Xiaopeng Tang Zhiye Zhang Mingqian Fang Yajun Han Gan Wang Sheng Wang Min Xue Yaxiong Li Li Zhang Jian Wu Biqing Yang James Mwangi Qiumin Lu Xiaoping Du Ren Lai | 2020 | Cell Research2020,30,2: | 2

Hierarchically mesoporous carbon spheres coated with a single atomic Fe-N-C layer for balancing activity and mass transfer in fuel cells显示文摘Novel cost-effective fuel cells have become more attractive due to the demands for rare and expensive platinum-group metal(PGM)catalysts for mitigating the sluggish kinetics of the oxygen reduction reaction(ORR).The high-cost PGM catalyst in fuel cells can be replaced by earth-abundant transition-metalbased catalysts, that is,an Fe-N-C catalyst, which is considered one of the most promising alternatives. However, the performance of the Fe-N-C catalyst is hindered by the low catalytic activity and poor stability, which is caused by insufficient active sites and the lack of optimization of the triple-phase interface for mass transportation.Herein,a novel Fe–N–C catalyst consisting of mono-dispersed hierarchically mesoporous carbon sphere cores and single Fe atom-dispersed functional shells are presented. The synergistic effect between highly dispersed Fe-active sites and well-organized porous structures yields the combination of high ORR activity and high mass transfer performance. The half-wave potential of the catalyst in 0.1M H_(2)SO_(4) is 0.82 V versus reversible hydrogen electrode, and the peak power density is 812 mW·cm^(−2) in H_(2)–O_(2) fuel cells.Furthermore,it shows superior methanol tolerance, which is almost immune to methanol poisoning and generates up to 162 mW·cm^(−2) power density in direct methanol fuel cells. | Chengyong Shu Qiang Tan Chengwei Deng Wei Du Zhuofan Gan Yan Liu Chao Fan Hui Jin Wei Tang Xiao-dong Yang Xiaohua Yang Yuping Wu | 2022 | Carbon Energy2022,4,1: | 2
| 18 | Silica nanocone array as a template for fabricating a plasmon induced hot electron photodetector显示文摘Plasmon induced hot electrons have attracted a great deal of interest as a novel route for photodetection and lightenergy harvesting. Herein, we report a hot electron photodetector in which a large array of nanocones deposited sequentially with aluminum, titanium dioxide, and gold films can be integrated functionally with nanophotonics and microelectronics. The device exhibits a strong photoelectric response at around 620 nm with a responsivity of 180 μA/W under short-circuit conditions with a significant increase under 1 V reverse bias to 360 μA/W. The increase in responsivity and a red shift in the peak value with increasing bias voltage indicate that the bias causes an increase in the hot electron tunneling effect.
