1Persistence and clearance of viral RNA in 2019 novel coronavirus disease rehabilitation patients显示文摘Background:A patient’s infectivity is determined by the presence of the virus in different body fluids,secretions,and excreta.The persistence and clearance of viral RNA from different specimens of patients with 2019 novel coronavirus disease(COVID-19)remain unclear.This study analyzed the clearance time and factors influencing 2019 novel coronavirus(2019-nCoV)RNA in different samples from patients with COVID-19,providing further evidence to improve the management of patients during convalescence.Methods:The clinical data and laboratory test results of convalescent patients with COVID-19 who were admitted to from January 20,2020 to February 10,2020 were collected retrospectively.The reverse transcription polymerase chain reaction(RT-PCR)results for patients’oropharyngeal swab,stool,urine,and serum samples were collected and analyzed.Convalescent patients refer to recovered non-febrile patients without respiratory symptoms who had two successive(minimum 24 h sampling interval)negative RT-PCR results for viral RNA from oropharyngeal swabs.The effects of cluster of differentiation 4(CD4)+T lymphocytes,inflammatory indicators,and glucocorticoid treatment on viral nucleic acid clearance were analyzed.Results:In the 292 confirmed cases,66 patients recovered after treatment and were included in our study.In total,28(42.4%)women and 38 men(57.6%)with a median age of 44.0(34.0-62.0)years were analyzed.After in-hospital treatment,patients’inflammatory indicators decreased with improved clinical condition.The median time from the onset of symptoms to first negative RT-PCR results for oropharyngeal swabs in convalescent patients was 9.5(6.0-11.0)days.By February 10,2020,11 convalescent patients(16.7%)still tested positive for viral RNA from stool specimens and the other 55 patients’stool specimens were negative for 2019-nCoV following a median duration of 11.0(9.0-16.0)days after symptom onset.Among these 55 patients,43 had a longer duration until stool specimens were negative for viral RNA than for throat swabs,with a median delay of 2.0(1.0-4.0)days.Results for only four(6.9%)urine samples were positive for viral nucleic acid out of 58 cases;viral RNA was still present in three patients’urine specimens after throat swabs were negative.Using a multiple linear regression model(F=2.669,P=0.044,and adjusted R2=0.122),the analysis showed that the CD4+T lymphocyte count may help predict the duration of viral RNA detection in patients’stools(t=-2.699,P=0.010).The duration of viral RNA detection from oropharyngeal swabs and fecal samples in the glucocorticoid treatment group was longer than that in the non-glucocorticoid treatment group(15 days vs.8.0 days,respectively;t=2.550,P=0.013)and the duration of viral RNA detection in fecal samples in the glucocorticoid treatment group was longer than that in the non-glucocorticoid treatment group(20 days vs.11 days,respectively;t=4.631,P<0.001).There was no statistically significant difference in inflammatory indicators between patients with positive fecal viral RNA test results and those with negative results(P>0.05).Conclusions:In brief,as the clearance of viral RNA in patients’stools was delayed compared to that in oropharyngeal swabs,it is important to identify viral RNA in feces during convalescence.Because of the delayed clearance of viral RNA in the glucocorticoid treatment group,glucocorticoids are not recommended in the treatment of COVID-19,especially for mild disease.The duration of RNA detection may relate to host cell immunity.Yun Ling Shui-Bao Xu Yi-Xiao Lin Di Tian Zhao-Qin Zhu Fa-Hui Dai Fan Wu Zhi-Gang Song Wei Huang Jun Chen Bi-Jie Hu Sheng Wang En-Qiang Mao Lei Zhu Wen-Hong Zhang Hong-Zhou Lu 2020Chinese Medical Journal2020,,9:58
2Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. 4Species clarification of the prize medicinal Ganoderma mushroom“Lingzhi”显示文摘“Lingzhi”is a mushroom that has been renowned in China for more than 2,000 years because of its claimed medicinal properties plus its symbolic fortune.“Lingzhi”has high economic value mostly as a dietary supplement in the modern market especially in East Asia,and its medicinal functions have become a hot study topic.For over a century,the highly prized medicinal fungus,known as“Lingzhi”in East Asia,has been assigned to Ganoderma lucidum,a species originally described from Europe.Molecular studies in recent years have revealed that the commercially cultivated‘G.lucidum’(“Lingzhi”)in East Asia is a different species from the true G.lucidum.The present study aims to clarify the species identity of“Lingzhi”based on morphological studies and analysis of rDNA nuc-ITS sequences,and additional gene fragments of mt-SSU,RPB1,RPB2,and TEF1-αof“Lingzhi”were provided.All Ganoderma species that mostly resemble“Lingzhi”in phylogeny and/or morphology were included for analysis.We propose a new species G.lingzhi for“Lingzhi”,which has an East Asia distribution.The most striking characteristics which differentiate G.lingzhi from G.lucidum are the presence of melanoid bands in the context,a yellow pore surface and thick dissepiments(80–120μm)at maturity.G.curtisii is most closely related to G.lingzhi in phylogeny and is from North America.Ganoderma flexipes,G.multipileum,G.sichuanense,G.tropicum and‘G.tsugae’,are also closely related with G.lingzhi and are reported from China.These species are compared and discussed.‘Ganoderma tsuage’reported from China is determined as conspecific with G.lucidum,hence the distribution of G.lucidum extends from Europe to northeastern China.Yun Cao Sheng-Hua Wu Yu-Cheng Dai 2012Fungal Diversity2012,,5:25

7Sirtuin 3 deficiency exacerbates diabetic cardiomyopathy via necroptosis enhancement and NLRP3 activation显示文摘Sirtuin 3(SIRT3)is a potential therapeutic target for cardiovascular,metabolic,and other aging-related diseases.In this study,we investigated the role of SIRT3 in diabetic cardiomyopathy(DCM).Mice were injected with streptozotocin(STZ,60 mg/kg,ip)to induce diabetes mellitus.Our proteomics analysis revealed that SIRT3 expression in the myocardium of diabetic mice was lower than that of control mice,as subsequently confirmed by real-time PCR and Western blotting.To explore the role of SIRT3 in DCM,SIRT3-knockout mice and 129S1/SvImJ wild-type mice were injected with STZ.We found that diabetic mice with SIRT3 deficiency exhibited aggravated cardiac dysfunction,increased lactate dehydrogenase(LDH)level in the serum,decreased adenosine triphosphate(ATP)level in the myocardium,exacerbated myocardial injury,and promoted myocardial reactive oxygen species(ROS)accumulation.Neonatal rat cardiomyocytes were transfected with SIRT3 siRNA,then exposed to high glucose(HG,25.5 mM).We found that downregulation of SIRT3 further increased LDH release,decreased ATP level,suppressed the mitochondrial membrane potential,and elevated oxidative stress in HG-treated cardiomyocytes.SIRT3 deficiency further raised expression of necroptosis-related proteins including receptor-interacting protein kinase 1(RIPK1),RIPK3,and cleaved caspase 3,and upregulated the expression of inflammation-related proteins including NLR family pyrin domain-containing protein 3(NLRP3),caspase 1 p20,and interleukin-1βboth in vitro and in vivo.Collectively,SIRT3 deficiency aggravated hyperglycemia-induced mitochondrial damage,increased ROS accumulation,promoted necroptosis,possibly activated the NLRP3 inflammasome,and ultimately exacerbated DCM in the mice.These results suggest that SIRT3 can be a molecular intervention target for the prevention and treatment of DCM.Shu Song Yue Ding Guo-liang Dai Yue Zhang Meng-ting Xu Jie-ru Shen Ting-ting Chen Yun Chen Guo-liang Meng 2021Acta Pharmacologica Sinica2021,42,2:14

9Esophageal Helicobacter pylori colonization aggravates esophageal injury caused by reflux显示文摘AIM: To investigate esophageal Helicobacter pylori(H. pylori) colonization on esophageal injury caused by reflux and the related mechanisms. METHODS: An esophagitis model, with acid and bile reflux, was surgically produced in male rats. The rats were randomly divided into either:(1) an esophagogastroduodenal anastomosis(EGDA) group;(2) an EGDA with H. pylori infection group;(3) a pseudo-operation with H. pylori infection group; or(4) a pseudooperation group. All rats were kept for 36 wk. Based on the location of H. pylori colonization, the EGDA rats with H. pylori infection were subdivided into those with concomitant esophageal H. pylori colonization or those with only gastric H. pylori colonization. The esophageal injuries were evaluated grossly and microscopically. The expressions of CDX2 and MUC2 were determined by real-time polymerase chain reaction(RT-PCR) and immunohistochemistry. Ki-67 antigen expression was determined by immunohistochemistry. The m RNA levels of cyclin D1, c-Myc, Bax and Bcl-2 were determined by RT-PCR. Cell apoptosis was evaluated using the Td Tmediated d UTP nick-end labeling method.RESULTS: Esophagitis, Barrett's esophagus(BE), and esophageal adenocarcinoma(EAC) developed in rats that underwent EGDA. When comparing rats with EGDA and concomitant esophageal H. pylori colonization to EGDA-only rats, the severity of injury(87.9 ± 5.2 vs 77.2 ± 8.6, macroscopically, 92.5 ± 8.0 vs 83.8 ± 5.5, microscopically, both P < 0.05) and the incidences of BE(80.0% vs 33.3%, P = 0.055) and EAC(60.0% vs 11.1%, P < 0.05) were increased. These increases were associated with upregulation of CDX2 and MUC2 m RNA(10.1 ± 5.4 vs 3.0 ± 2.9, 8.4 ± 4.6 vs 2.0 ± 3.2, respectively, P s < 0.01) and protein(8.1 ± 2.3 vs 3.3 ± 3.1, 7.3 ± 4.0 vs 1.8 ± 2.7, respectively, all P < 0.05). The expression of Ki-67(8.9 ± 0.7 vs 6.0 ± 1.7, P < 0.01) and the presence of apoptotic cells(8.3 ± 1.1 vs 5.3 ± 1.7, P < 0.01) were also increased significantly in rats with EGDA and concomitant esophageal H. pylori colonization compared with rats with EGDA only. The m RNA levels of cyclin D1(5.8 ± 1.9 vs 3.4 ± 1.3, P < 0.01), c-Myc(6.4 ± 1.7 vs 3.7 ± 1.2, P < 0.01), and Bax(8.6 ± 1.6 vs 5.1 ± 1.3, P < 0.01) were significantly increased, whereas the m RNA level of Bcl-2(0.6 ± 0.3 vs 0.8 ± 0.3, P < 0.01) was significantly reduced in rats with EGDA and concomitant esophageal H. pylori colonization compared with rats with EGDA only.CONCLUSION: Esophageal H. pylori colonization increases esophagitis severity, and facilitates the development of BE and EAC with the augmentation of cell proliferation and apoptosis in esophageal mucosa.Yun-Xiang Chu Wei-Hong Wang Yun Dai Gui-Gen Teng Shu-Jun Wang 2014World Journal of Gastroenterology2014,20,42:12

16Novel Ru - K/Carbon Nanotubes Catalyst for Ammonia Synthesis显示文摘A novel ammonia synthesis catalyst, potassium-promoted ruthenium supported on carbon nanotubes, was developed. It was found that the Ru-K/carbon nanotubes catalyst had higher activity for ammonia synthesis (20.85 ml NH3/h/g-cat ) than the Ru-K/fullerenes (13.3 ml NH3/h/g-cat ) at atmospheric pressure and 623 K. The catalyst had activity even at 473 K. and had the highest activity (23.46 ml NH3/h/g-cat ) at 643 K. 17A cyclodextrin-based nanoformulation achieves co-delivery of ginsenoside Rg3 and quercetin for chemo-immunotherapy in colorectal cancer显示文摘The immune checkpoint blockade therapy has profoundly revolutionized the field of cancer immunotherapy. However, despite great promise for a variety of cancers, the efficacy of immune checkpoint inhibitors is still low in colorectal cancer(CRC). This is mainly due to the immunosuppressive feature of the tumor microenvironment(TME). Emerging evidence reveals that certain chemotherapeutic drugs induce immunogenic cell death(ICD), demonstrating great potential for remodeling the immunosuppressive TME. In this study, the potential of ginsenoside Rg3(Rg3) as an ICD inducer against CRC cells was confirmed using in vitro and in vivo experimental approaches. The ICD efficacy of Rg3 could be significantly enhanced by quercetin(QTN) that elicited reactive oxygen species(ROS). To amelioratein vivo delivery barriers associated with chemotherapeutic drugs, a folate(FA)-targeted polyethylene glycol(PEG)-modified amphiphilic cyclodextrin nanoparticle(NP) was developed for co-encapsulation of Rg3 and QTN. The resultant nanoformulation(CD-PEG-FA.Rg3.QTN) significantly prolonged blood circulation and enhanced tumor targeting in an orthotopic CRC mouse model, resulting in the conversion of immunosuppressive TME. Furthermore, the CD-PEG-FA.Rg3.QTN achieved significantly longer survival of animals in combination with Anti-PD-L1. 18Effect of a transverse plasma jet on a shock wave induced by a ramp显示文摘We conducted experiments in a wind tunnel with Mach number 2 to explore the evolution of a transverse plasma jet and its modification effect on a shock wave induced by a ramp with an angle of 24°. The transverse plasma jet was created by arc discharge in a small cylindrical cavity with a 2 mm diameter orifice. Three group tests with different actuator arrangements in the spanwise or streamwise direction upstream from the ramp were respectively studied to compare their disturbances to the shock wave. As shown by a time-resolved schlieren system, an unsteady motion of the shock wave by actuation was found: the shock wave was significantly modified by the plasma jet with an upstream motion and a reduced angle. Compared to spanwise actuation, a more intensive impact was obtained with two or three streamwise actuators working together. From shock wave structures, the control effect of the plasma jet on the shock motion based on a thermal effect, a potential cause of shock modification, was discussed. Furthermore, we performed a numerical simulation by using the Improved Delayed Detached Eddy Simulation(IDDES) method to simulate the evolution of the transverse plasma jet plume produced by two streamwise actuators. The results show that flow structures are similar to those identified in schlieren images. 19Translational Research in Oncology Research & Development and Its Impact on Early Development in China: report of the 5th Annual Meeting of the US Chinese Anti-Cancer Association (USCACA) at 2013 AACR Annual Meeting显示文摘In April 2013, the US Chinese Anti-Cancer Association (USCACA) held its 5th annual meeting in conjunction with the American Association for Cancer Research (AACR) 2013 Annual Meeting in Washington DC. The USCACA executive committee reported activities and programs and highlighted the partnership and collaboration between USCACA and other major organizations. The key initiatives and programs of USCACA included 1) USCACA-TIGM Esophageal Cancer Program that funds translational research of esophageal cancer prevention and treatment at the Xinxiang Medical University in Henan province, China; 2) the USCACA-NFCR-AFCR Scholarship Program, which has supported 10 young outstanding Chinese cancer researchers and will award 4 fellowships at the Guangzhou International Symposium on Oncology in November this year; 3) USCACA-Hengrui Training Program for Early Phase Clinical Research, which has supported the training of a Chinese scholar at two major cancer centers in the US; and 4) USCACA has continued its partnership with the Chinese Journal of Cancer, which has reached significant international impact.Lingjie Guan Yun Dai Roger Luo 2013Chinese Journal of Cancer2013,32,7:6
20Association between Metabolic Syndrome and Carotid Atherosclerosis: A Cross-sectional Study in Northern China显示文摘Objective Increased carotid artery intima-media thickness(CIMT)and carotid plaque as manifestations of carotid atherosclerosis have been used as markers of cardiovascular disease(CVD).The components of metabolic syndrome(Met S)are linked to CVD,but the association between Met S and CVD is controversial.Methods A total of 8,933 Chinese adults aged 40 years or older from 2010 to 2014 were selected from the Jidong and Kailuan communities.Met S was defined by the International Diabetes Federation criteria.CIMT and carotid plaque were measured using color Doppler ultrasound.Logistic regression models were used to assess the association of Met S with carotid plaque and CIMT.Results Met S was found among 3,461(3,461/8,933)participants.The odds ratio and 95%confidence internal(CI)for carotid plaques in participants with Met S was 1.16(1.03-1.30).The risk of carotid plaques increased with the number of Met S components.The average CIMT was higher in participants with Met S(β=0.020,95%CI,0.014-0.027)and in participants with more Met S components.Conclusion Individuals with Met S are at an increased risk for carotid atherosclerosis compared to those without MetS.ZHOU Ping An ZHANG Chen Huan CHEN Yan Ru LI Dong SONG Dai Yu LIU Hua Min ZHOU Ming Yue SONG Guo Shun CHEN Sheng Yun 2019Biomedical and Environmental Sciences2019,32,12:6
