1A Strigolactone Biosynthesis Gene Contributed to the Green Revolution in Rice显示文摘Plant architecture is a complex agronomic trait and a major factor of crop yield,which is affected by several important hormones.Strigolactones(SLs)are identified as a new class hormoneinhibiting branching in many plant species and have been shown to be involved in various developmental processes.Genetical and chemical modulation of the SL pathway is recognized as a promising approach to modify plant architecture.However,whether and how the genes involved in the SL pathway could be utilized in breeding still remain elusive.Here,we demonstrate that a partial loss-of-function allele of the SL biosynthesis gene,HIGH TILLERING AND DWARF 1/DWARF17(HTD1/D17),which encodes CAROTENOID CLEAVAGE DIOXYGENASE 7(CCD7),increases tiller number and improves grain yield in rice.We found that the HTD1 gene had been widely utilized and co-selected with Semidwarf 1(SD1),both contributing to the improvement of plant architecture in modern rice varieties since the Green Revolution in the 1960s.Understanding how phytohormone pathway genes regulate plant architecture and how they have been utilized and selected in breeding will lay the foundation for developing the rational approaches toward improving crop yield.Yuexing Wang Lianguang Shang Hong Yu Longjun Zeng Jiang Hu Shen Ni Yuchun Rao Sanfeng Li Jinfang Chu Xiangbing Meng Lei Wang Ping Hu Jijun Yan Shujing Kang Minghao Qu Hai Lin Tao Wang Quan Wang Xingming Hu Hongqi Chen Bing Wang Zhenyu Gao Longbiao Guo Dali Zeng Xudong Zhu Guosheng Xiong Jiayang Li Qian Qian 2020Molecular Plant2020,13,6:23
2Extraction kinetics of neodymium from chloride medium using HEH/EHP saponified with magnesium bicarbonate solution显示文摘Magnesium bicarbonate solution is considered as an environmentally friendly extractant saponification agent for the solvent extraction of rare earth elements due to its advantage of minimum water pollution.In order to reveal the extraction regularity, optimize production-process and guide the use of this new extraction system, the extraction of Nd(Ⅲ) in chloride medium with HEH/EHP saponified by magnesium bicarbonate solution was investigated with the self-designed constant interfacial area cell. Besides, the effects of stirring rate, temperature, specific interfacial area and concentration of Mg-HEH/EHP on the extraction rate of Nd(Ⅲ) were systematically investigated. Results show that, the rate of extraction is governed by both diffusion and chemical reaction, and the extraction reaction takes place at the interface. The apparent activation energy of the extraction reaction is 16.88 kJ/mol. The corresponding rate equation is deduced. 7Toward atomistic models of intact severe acute respiratory syndrome coronavirus 2 via Martini coarsegrained molecular dynamics simulations显示文摘The causative pathogen of coronavirus disease 2019(COVID-19),severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is an enveloped virus assembled by a lipid envelope and multiple structural proteins.In this study,by integrating experimental data,structural modeling,as well as coarse-grained and all-atom molecular dynamics simulations,we constructed multiscale models of SARS-CoV-2.Our 500-ns coarse-grained simulation of the intact virion allowed us to investigate the dynamic behavior of the membrane-embedded proteins and the surrounding lipid molecules in situ.Our results indicated that the membrane-embedded proteins are highly dynamic,and certain types of lipids exhibit various binding preferences to specific sites of the membrane-embedded proteins.The equilibrated virion model was transformed into atomic resolution,which provided a 3D structure for scientific demonstration and can serve as a framework forfuture exascale allatom molecular dynamics(MD)simulations.A short all-atom molecular dynamics simulation of 255 ps was conducted as a preliminary test for largescale simulationsofthis complexsystem.Dali Wang Jiaxuan Li Lei Wang Yipeng Cao Bo Kang Xiangfei Meng Sai Li Chen Song 2023Quantitative Biology2023,11,4:0
8A de novo missense mutation in MPP2 confers an increased risk of Vogt–Koyanagi–Harada disease as shown by trio-based whole-exome sequencing显示文摘Vogt–Koyanagi–Harada(VKH)disease is a leading cause of blindness in young and middle-aged people.However,the etiology of VKH disease remains unclear.Here,we performed the first trio-based whole-exome sequencing study,which enrolled 25 VKH patients and 50 controls,followed by a study of 2081 VKH patients from a Han Chinese population to uncover detrimental mutations.A total of 15 de novo mutations in VKH patients were identified,with one of the most important being the membrane palmitoylated protein 2(MPP2)p.K315N(MPP2-N315)mutation.The MPP2-N315 mutation was highly deleterious according to bioinformatic predictions.Additionally,this mutation appears rare,being absent from the 1000 Genome Project and Genome Aggregation Database,and it is highly conserved in 10 species,including humans and mice.Subsequent studies showed that pathological phenotypes and retinal vascular leakage were aggravated in MPP2-N315 mutation knock-in or MPP2-N315 adeno-associated virus-treated mice with experimental autoimmune uveitis(EAU).In vitro,we used clustered regularly interspaced short palindromic repeats(CRISPR‒Cas9)gene editing technology to delete intrinsic MPP2 before overexpressing wild-type MPP2 or MPP2-N315.Levels of cytokines,such as IL-1β,IL-17E,and vascular endothelial growth factor A,were increased,and barrier function was destroyed in the MPP2-N315 mutant ARPE19 cells.Mechanistically,the MPP2-N315 mutation had a stronger ability to directly bind to ANXA2 than MPP2-K315,as shown by LC‒MS/MS and Co-IP,and resulted in activation of the ERK3/IL-17E pathway.Overall,our results demonstrated that the MPP2-K315N mutation may increase susceptibility to VKH disease.Xianyang Liu Jiayu Meng Xingyun Liao Yusen Liu Qian Zhou Zongren Xu Shuming Yin Qingfeng Cao Guannan Su Siyuan He Wanqian Li Xiaotang Wang Guoqing Wang Dali Li Peizeng Yang Shengping Hou 2023Cellular & Molecular Immunology2023,20,11:0
9The Sm core protein SmEb regulates salt stress responses through maintaining proper splicing of RCD1 pre-mRNA in Arabidopsis显示文摘Salt stress adversely impacts crop production.Several spliceosome components have been implicated in regulating salt stress responses in plants,however,the underlying molecular basis is still unclear.Here we report that the spliceosomal core protein SmEb is essential to salt tolerance in Arabidopsis.Transcriptome analysis showed that SmEb modulates alternative splicing of hundreds of pre-mRNAs in plant response to salt stress.Further study revealed that SmEb is crucial in maintaining proper ratio of two RCD1 splicing variants(RCD1.1/RCD1.2)important for salt stress response.In addition,RCD1.1 but not RCD1.2 is able to interact with the stress regulators and attenuates saltsensitivity by decreasing salt-induced cell death in smeb-1 mutant.Together,our findings uncovered the essential role of SmEb in the regulation of alternative pre-mRNA splicing in salt stress response.Yechun Hong Yang Gao Jia Pang Huazhong Shi Tingting Li Huiying Meng Dali Kong Yunjuan Chen Jian-Kang Zhu Zhen Wang 2023Journal of Integrative Plant Biology2023,65,6:0
