2Quantification of microcystin-producing and non-microcystin producing Microcystis populations during the 2009 and 2010 blooms in Lake Taihu using quantitative real-time PCR显示文摘Lake Taihu,a large,shallow hypertrophic freshwater lake in eastern China,has experienced lake-wide toxic cyanobacterial blooms annually during summer season in the past decades.Spatial changes in the abundance of hepatotoxin microcystin-producing and nonmicrocystin producing Microcystis populations were investigated in the lake in August of 2009 and 2010.To monitor the densities of the total Microcystis population and the potential microcystin-producing subpopulation,we used a quantitative real-time PCR assay targeting the phycocyanin intergenic spacer(PC-IGS) and the microcystin synthetase gene(mcyD),respectively.On the basis of quantification by real-time PCR analysis,the abundance of potential toxic Microcystis genotypes and the ratio of the mcyD subpopulation to the total Microcystis varied significantly,from 4.08×104 to 5.22×107 copies/mL,from 5.7% to 65.8%,respectively.Correlation analysis showed a strong positive relationship between chlorophyll-a,toxic Microcystis and total Microcystis;the abundance of toxic Microcystis correlated positively with total phosphorus and ortho-phosphate concentrations,but negatively with TN:TP ratio and nitrate concentrations.Meanwhile the proportion of potential toxic genotypes within Microcystis population showed positive correlation with total phosphorus and ortho-phosphate concentrations.Our data suggest that increased phosphorus loading may be a significant factor promoting the occurrence of toxic Microcystis bloom in Lake Taihu.Daming Li Fanxiang Kong Xiaoli Shi Linlin Ye Yang Yu Zhen Yang 2012Journal of Environmental Sciences2012,24,2:5
3IL1RN promotes osteoblastic differentiation via interacting with ITGB3 in osteoporosis显示文摘The occurrence and progress of osteoporosis(OP)are partially caused by impaired osteoblast differentiation.Interleukin-I receptor antagonist(IL1RN)is an immune modulatory molecule that commonly functions by means of competing the binding site of IL-1R with IL-1.Although it was recently reported that IL1RN is involved in osteoblast differentiation,the role of IL1RN in osteogenesis remains unclear.In this work,we first investigated the expression pattern of IL1RN in ovariectomy mice and in vitro osteogenic induction of MC3T3-E1 and C3H10T1/2 cells.To verify the exact role of IL1RN in osteoblast differentiation,we established IL1RN-downregulated/upregulated cell lines.The results indicated that IL1RN was constantly expressed in MC3T3-E1 and C3H10T1/2 cells.Interestingly,an increase of IL1RN expression in osteoblasts occurred when osteoblasts were cultured in osteogenic medium(OM).As expected,silencing of IL1RN attenuated the osteogenic effect of OM,while IL1RN overexpression increased the osteogenic staining and promoted the expression of osteogenic markers,including alkaline phosphatase,osterix,and osteocalcin.In addition to evaluating the function of IL1RN in osteoblasts,we also investigated the molecular mechanism of the role of IL1RN in osteoblasts.We found that IL1RN interacts with integrinβ3 to activateβ-catenin signaling,which finally regulates osteoblast differentiation.Taken together,this study provides the framework that IL1RN,as a novel regulator of osteogenesis,may be a potential therapeutic target for the treatment of OP.Zehua Zou Ruixuan Liu Yiwen Wang Yufei Xing Zuowei Shi Kaifu Wang Daming Dong 2021Acta Biochimica et Biophysica Sinica2021,53,3:3

5Robust facial landmark detection and tracking across poses and expressions for in-the-wild monocular video显示文摘We present a novel approach for automatically detecting and tracking facial landmarks acrossposesandexpressionsfromin-the-wild monocular video data,e.g.,You Tube videos and smartphone recordings.Our method does not require any calibration or manual adjustment for new individual input videos or actors.Firstly,we propose a method of robust 2D facial landmark detection across poses,by combining shape-face canonical-correlation analysis with a global supervised descent method.Since 2D regression-based methods are sensitive to unstable initialization,and the temporal and spatial coherence of videos is ignored,we utilize a coarse-todense 3D facial expression reconstruction method to refine the 2D landmarks.On one side,we employ an in-the-wild method to extract the coarse reconstruction result and its corresponding texture using the detected sparse facial landmarks,followed by robust pose,expression,and identity estimation.On the other side,to obtain dense reconstruction results,we give a face tracking flow method that corrects coarse reconstruction results and tracks weakly textured areas;this is used to iteratively update the coarse face model.Finally,a dense reconstruction result is estimated after it converges.Extensive experiments on a variety of video sequences recorded by ourselves or downloaded from You Tube show the results of facial landmark detection and tracking under various lighting conditions,for various head poses and facial expressions.The overall performance and a comparison with state-of-art methods demonstrate the robustness and effectiveness of our method.Shuang Liu Yongqiang Zhang Xiaosong Yang Daming Shi Jian J.Zhang 2017Computational Visual Media2017,3,1:2

8Identification of weathered troughs in granites for tunneling at Daya Bay reactor-neutrino experiment site显示文摘Weathered troughs are frequently encountered in granites.They can cause problems to tunneling in the rocks and have to be properly addressed.The structures or spatial shapes of weathered troughs were seldom clarified in the past.In this paper,four weathered troughs are identified by means of geophysical exploration,core drilling,logging,and in-situ stress measurement at Daya Bay reactor-neutrino experiment site in Shenzhen,China.The weathered troughs are exposed on the ground or partially covered by grass and soils,which will threaten the safety of horizontal tunneling at a shallow depth.High electrical resistivity(HER) method is adopted for its feasibility and practicality,in combination with field geological observation,ultrasonic televiewer in boreholes and in-situ stress measurement.By comparing the HER values of completely decomposed to fresh rocks,it is indicated that the HER values of weathered troughs in natural state are 0.4-100 ·m,and the in-situ stress is abnormally lower than those at upper and lower layers.Field investigations show that the depths of the four weathered troughs are 30-182 m,with bottom elevation over 10 m.The volume of each weathered trough is mostly over 1×10 6 m 3 in inverted conic form.The weathered troughs often occur in various kinds of landforms,such as ridges,gullies or gently dipping dish-like depression areas.Faults and boundaries of different granitic plutons as well as joints govern the formation,locations and strikes of these troughs under development.Yanjun Shang Yongyue Shi Weijun Jin Daming Lin Fengbo Wu Wei Zhang 2012Journal of Rock Mechanics and Geotechnical Engineering2012,4,2:1

11Science with the 2.5-meter Wide Field Survey Telescope(WFST)显示文摘The Wide Field Survey Telescope(WFST) is a dedicated photometric surveying facility being built jointly by University of Science and Technology of China(USTC) and the Purple Mountain Observatory(PMO). It is equipped with a 2.5-meter diameter primary mirror, an active optics system, and a mosaic CCD camera with 0.73 gigapixels on the primary focal plane for highquality image capture over a 6.5-square-degree field of view. The installation of WFST near the summit of Saishiteng mountain in the Lenghu region is scheduled in summer of 2023, and the operation is planned to start three months later. WFST will scan the northern sky in four optical bands(u, g, r and i) at cadences from hourly/daily in the deep high-cadence survey(DHS) program, to semi-weekly in the wide field survey(WFS) program. During a photometric night, a nominal 30 s exposure in the WFS program will reach a depth of 22.27, 23.32, 22.84, and 22.31(AB magnitudes) in these four bands, respectively, allowing for the detection of a tremendous amount of transients in the low-z universe and a systematic investigation of the variability of Galactic and extragalactic objects. In the DHS program, intranight 90 s exposures as deep as 23(u) and 24 mag(g), in combination with target of opportunity follow-ups, will provide a unique opportunity to explore energetic transients in demand for high sensitivities, including the electromagnetic counterparts of gravitational wave events, supernovae within a few hours of their explosions,tidal disruption events and fast, luminous optical transients even beyond redshift of unity. In addition, the final 6-year co-added images, anticipated to reach g■25.8 mag in WFS or 1.5 mags deeper in DHS, will be of fundamental importance to general Galactic and extragalactic science. The highly uniform legacy surveys of WFST will serve as an indispensable complement to those of the Vera C. 16Fish Assemblage Responses to a Low-head Dam Removal in the Lancang River显示文摘Dam removal is becoming an effective approach for aquatic biodiversity restoration in damming river in order to balance the aquatic ecosystem conservation with large-scale cascade damming. However, the effects of dam removal on fish communities in Asian mountainous rivers, which are dominated by Cypriniformes fishes, are still not well known. To determine whether dam removal on a mountainous river benefit restoration of fish diversity, we investigated the response of fish assemblage to dam removal using a before-after-control-impact design in two tributaries of the Lancang River(dam removal river: the Jidu River, and control river: the Fengdian River). Fish surveys were conducted one year prior to dam removal(2012) and three years(2013–2015) following dam removal. We observed rapidly and notably spatio-temporal changes in fish biodiversity metrics and assemblage structure, occurring in the Jidu River within the first year after dam removal. Overall, fish species richness, density and Shannon-Wiener diversity all increased immediately in above-and below-dam sites, and maintained a stable level in subsequent years, compared to unchanged situation in the control river. All sites in the Jidu River experienced shifts in fish composition after dam removal, with the greatest temporal changes occurred in sites below-and above-the former dam, resulting in a temporal homogenization tendency in the dam removed river.
