| 1 | Exosomes and their role in the micro-/macro-environment: acomprehensive review显示文摘The importance of extracellular vesicles(EVs) in cell-cell communication has long been recognized due to their ability to transfer important cellular cargoes such as DNA, mRNA, miRNAs, and proteins to target cells. Compelling evidence supports the role of EVs in the horizontal transfer of cellular material which has the potential to influence normal cellular physiology and promote various disease states. Of the different types of EVs, exosomes have garnered much attention in the past decade due to their abundance in various biological fluids and ability to affect multiple organ systems. The main focus of this review will be on cancer and how cancer-derived exosomes are important mediators of metastasis, angiogenesis, immune modulation, and the tumor macro-/microenvironment. We will also discuss exosomes as potential biomarkers for cancers due to their abundance in biological fluids, ease of uptake, and cellular content. | 2 | Leaf Photosynthetic Activity and Antioxidant Defense Associated with Sub1 QTL in Rice Subjected to Submergence and Subsequent Re-aeration显示文摘The influence of submergence on photosynthesis and antioxidant capacities in rice varieties Swarna and Swarna-Sub1 with or without Sub1 QTL were evaluated under control,simulated complete submergence and subsequent re-aeration.The leaf photosynthetic rate and stomatal conductance decreased in the both varieties during the progression of submergence as compared to the control plants,but significant varietal differences were observed after 1 d of submergence.Submergence also altered the PSII activity,as reflected in a decrease in the values of Fo,Fm and Fv/Fm and degradation of chlorophyll,more in Swarna than in Swarna-Sub1.During early submergence period,the activities of superoxide dismutase(SOD),ascorbate peroxidase(APX),dehydroascorbate reductase(DHAR),glutathione reductase(GR) against reactive oxygen species were increased in the both varieties.However,with the progress of submergence period(after 7 d),the activities of SOD,catalase(CAT),APX,guaiacol peroxidase(GPX),GR and DHAR declined,more in Swarna than in Swarna-Sub1.During re-aeration,Swarna-Sub1 showed significant increase of above antioxidant enzymes but not in Swarna.Swarna-Sub1 improves photosynthetic activity,showing higher photosynthetic rate compared to Swarna under submergence and subsequent re-aeration because of less degradation of chlorophyll,higher stomatal conductance,and efficient PSII activity along with better antioxidant protection from oxidative damage. | Debabrata PANDA Ramani Kumar SARKAR | 2012 | Rice science2012,19,2: | 5 |
| 3 | Effect of Silver Nanoparticles on Growth of Eukaryotic Green Algae显示文摘Silver nanoparticles,endowed with powerful antimicrobial property,are the most widely used nanomaterial in consumer products,with associated risk of their easy access to environment and freshwater ecosystems by surface runoff.Although toxic effects of nanosilver on bacterial,fungal and mammalian cells have been documented,its impact on algal growth remains unknown.Pithophora oedogonia and Chara vulgaris are predominant members of photosynthetic eukaryotic algae,which form major component of global aquatic ecosystem.Here we report for the first time that nanosilver has significant adverse effects on growth and morphology of these filamentous green algae in a dose-dependent manner.Exposure of algal thalli to increasing concentrations of silver nanoparticles resulted in progressive depletion in algal chlorophyll content,chromosome instability and mitotic disturbance,associated with morphological malformations in algal filaments.SEM micrographs revealed dramatic alterations in cell wall in nanoparticle-treated algae,characterized with cell wall rupture and degradation in Pithophora.Although these observations underscore severe deleterious effects of nanosilver on aquatic environment,the information can also be exploited as a bioengineering strategy to control unwanted and persistent growth of noxious algal weeds that clog the municipal water supply and water channels and produce fouling of water bodies. | Anjali Dash Anand P.Singh Bansh R.Chaudhary Sunil K.Singh Debabrata Dash | 2012 | Nano-Micro Letters2012,4,3: | 5 |

| 11 | Cell-free derivatives from mesenchymal stem cells are effective in wound therapy显示文摘AIM:To compare the efficacy of cell-free derivatives from Bone marrow derived human mesenchymal stem cells(hMSCs) in wound therapy.METHODS:hMSCs have been shown to play an important role in wound therapy.The present study sought to compare efficacy of hMSCs and cell-free derivatives of hMSCs,which may be clinically more relevant as they are easier to prepare,formulate and transport.hMSCs were isolated from human bone marrow and cultured.Multi lineage differentiation of hMSCs was performed to confirm their identity.The ability of hMSCs to migrate was evaluated using in vitro and in vivo migration assays.Cell lysates and conditioned medium concentrate was prepared from hMSCs(see Methods for details).Wounds were induced in mice and wound areas were measure before and after cell and cell-free derivative treatment.RNA and proteins were extracted from the skin and cytokine levels were measured.RESULTS:Co-culture of hMSCs with keratinocytes resulted in increased expression of CXCL-12(SDF1) and ENA78(CXCL-5) in the conditioned media indicating that the hMSCs can respond to signals from keratinocytes.Accelerated wound closure was observed when hMSCs were injected near the site of excisional wounds in athymic as well as NOD/SCID mice.Interestingly,cell-free lysates prepared from hMSCs were also effective in inducing accelerated wound closure and increased expression of SDF1 and CXCL-5 at the wound bed.Additionally,concentrated media from hMSCs as well as an emulsion containing lysates prepared from hMSCs was also found to be more effective in rapid re-epithelialization than fibroblasts or vehicle-alone control.Use of cell-free derivatives may help replace expensive wound care approaches including use of growth factors,epidermal/dermal substitutes,synthetic membranes,cytokines,and matrix components,and most importantly avoid transmission of pathogens from human and animal products.CONCLUSION:These results encourage development of derivatives of hMSCs for wound care and re-epithelialization applications. | Pravin J Mishra Prasun J Mishra Debabrata Banerjee | 2012 | World Journal of Stem Cells2012,4,5: | 3 |
| 12 | Leaf Traits and Antioxidant Defense for Drought Tolerance During Early Growth Stage in Some Popular Traditional Rice Landraces from Koraput, India显示文摘Three popular traditional rice landraces, namely Kalajeera, Machakanta and Haladichudi, from Koraput, India were used to analyse the leaf traits and antioxidant defence for drought tolerance. When rice plants were exposed to different levels of drought stress by varying concentrations of polyethylene glycol(PEG) 6000, seed germination and growth parameters were significantly declined in all the rice landraces compared to the control. Drought stress also altered the leaf phenotypic traits based on chlorophyll fluorescence parameters and chlorophyll index, with more significant differences in susceptible variety IR64 than in traditional landraces. Furthermore, activities of antioxidative enzymes and proline and protein contents overtly increased under drought stress. The traditional rice landraces showed higher relative ratios for different parameters compared to the susceptible variety IR64. | 17 | Assessment of Variation in Morpho-Physiological Traits and Genetic Diversity in Relation to Submergence Tolerance of Five Indigenous Lowland Rice Landraces显示文摘The present study evaluated submergence responses in 88 lowland indigenous rice(Oryza sativa L.) landraces from Koraput, India, to identify submergence-tolerant rice genotypes. In pot experiments, variations in survival rate, shoot elongation, relative growth index, dry matter, chlorophyll, soluble sugar and starch contents were evaluated in two consecutive years under well-drained and completely submerged conditions. Principal component analysis showed that the first three axes contributed 96.820% of the total variation among the landraces, indicating wide variation between genotypes. Major traits such as survival rate, relative growth index, soluble sugar and starch contents appeared to be important determinants of phenotypic diversity among the landraces. Phenotypic coefficient of variance was higher than genotypic coefficient of variance for all the traits and all showed high heritability(90.38%–99.54%). Five rice landraces(Samudrabali, Basnamundi, Gadaba, Surudaka and Dokarakuji) were the most tolerant to submergence. When submerged for up to 14d, Samudrabali, Basnamundi and Godoba were notable for having greater survival rates than a standard submergence tolerant variety FR13 A, and also notable for elongating more vigorously and accumulating more biomass. These three landraces may therefore be especially useful in lowland rice growing areas that are affected by both moderate stagnant water and flash flooding.
