1Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy,efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade,thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade.These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC.Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin 2021Signal Transduction and Targeted Therapy2021,6,12:3

3Lactulose mediates suppression of dextran sulfate sodium-induced colon inflammation显示文摘Objective:Assessment of digestion colonic flora lactulose can be induced to produce hydrogen and prove lactulose with a potential anti-inflammatory effects. Methods:We established a mouse model of human ulcerative colitis (UC) by supplying mice with water containing dextran sulfate sodium (DSS). At 7-day, DSS-induced pathogenic outcomes, including loss of body weight, increase of colitis score, pathogenic shortening of colon length, elevated level of TNF-α, IL-1β, malondialdehyde (MDA) and myeloperoxidase (MPO) deficiency in colon lesions, were significantly suppressed by lactulose administration and intraperitoneally injected hydrogen water. Results:After eliminating colon microflora by antibiotics, the anti-inflammatory effects of lactulose were significantly compromised. Histological analysis also revealed that the DSS mediated colonic tissue destruction accompanied by neutrophils infiltration was remarkably suppressed by lactulose and hydrogen water. 4Genome-wide identification and expression analysis of NIN-like Protein(NLP)genes reveals their potential roles in the response to nitrate signaling in watermelon显示文摘To balance the relationship between high yield and low nitrogen supply,the nitrogen utilization efficiency of watermelon needs to be improved urgently.Nodule inception-like Protein(NLP)transcription factors play a key node role in nitrate response and growth and development of plant,however,comprehensive analysis of the NLP gene family in watermelon is unclear.This study explored the functional classification,evolutionary characteristics,and expression profile of the ClNLP gene family.Three ClNLPs were categorized into three groups according to their gene structure and phylogeny.All of them contained the conserved RWP-RK and PB1 domains.Evolutionary analysis of ClNLPs revealed that ClNLP1 and ClNLP3 underwent strong purified selection.In addition,cis-acting elements related to plant hormones and abiotic stresses were present in the ClNLP promoter.According to tissue-specific analysis ClNLP was widely expressed in roots,stems,leaves,flowers and fruits,and ClNLP1 was significantly induced in the roots of different nitrogen utilization varieties under different nitrate nitrogen supply.The SRTING functional protein association network suggested that ClNLP1 is associated with most genes,such as NRT1.1,NRT2.1,NIA1,and NIR1,and the dual-luciferase reporter assay found that ClNLP1 positively regulates the expression of ClNRT2.1.We speculated that ClNLP1 might play a central role in regulating the response of watermelon to nitrate nitrogen.Gaopeng Yuan Dexi Sun Yifan Wang Guolin An Weihua Li Wenjing Si Junpu Liu Yingchun Zhu 2022Horticultural Plant Journal2022,8,5:2

13Energy partitioning and evapotranspiration in a black locust plantation on the Yellow River Delta,China显示文摘Plantations of woody tree species play a crucial role in ecological security along coastal zones.Understanding energy partitioning and evapotranspiration can reveal land–atmosphere interaction processes.We investigated energy fluxes,evapotranspiration,and their related biophysical factors using eddy covariance techniques in a black locust(Robinia pseudoacacia L.)plantation in 2016,2018,and 2019 on the Yellow River Delta.Downward longwave radiation offsets 84–85%of upward longwave radiation;upward shortwave radiation accounted for 12–13%of downward shortwave radiation.The ratio of net radiation to downward radiation was 18–19%over the three years.During the growing season,latent heat flux was the largest component of net radiation;during the dormant season,the sensible heat flux was the dominant component of net radiation.The seasonal variation in daily evapotranspiration was mainly controlled by net radiation,air temperature,vapor pressure deficit,and leaf area index.Black locust phenology influenced daily evapotranspiration variations,and evapotranspiration was greater under sea winds than under land winds because soil water content at 10-cm depth was greater under sea winds during the day.Seasonal patterns of daily evaporative fraction,Bowen ratio,crop coefficient,Priestley–Taylor coefficient,surface conductance,and decoupling coefficient were mainly controlled by leaf area index.The threshold value of daily surface conductance was approximately 8 mm sover the plantation.Xiang Gao Zhenyu Du Qingshan Yang Jinsong Zhang Yongtao Li Xiaojie Wang Fengxue Gu Weiping Hao Zekun Yang Dexi Liu Jianmin Chu 2022Journal of Forestry Research2022,33,4:1

16Occurrence Regularity and Control of Thrips on Leek显示文摘[Objective] The paper was to clear the species and occurrence regularity of thrips severely occurred in leek field, and to study the main control methods. [Methods] The species of thrips on leek were identified in Sichuan Province, and the damage symptoms, occurrence regularity and control methods of thrips in leek field were studied. [Result] The morphological characteristic of thrips on leek was Frankliniella occidentalis(Pergande). ITS molecular identification results showed that the amplified fragment was 520 bp, which shared the similarity of 99.81% with the base se-quence of F. occidentalis. F. occidentalis concentrated at the height of 80 cm from the ground; in sunny days, the occurrence frequency of F. occidentalis was relatively high during 15:00 and 17:00; rainwater had obvious lethal effect on F. occidentalis, and the lethal rate was 29.30%. Field trial showed that 20% dinotefuran SG and 40% acetamiprid GR had good control effect against F. occidentalis. The control effect of 20% dinotefuran SG 1 500 times dilution was the best, up to 92.56%. 18Watermelon domestication was shaped by stepwise selection and regulation of the metabolome显示文摘Although crop domestication has greatly aided human civilization,the sequential domestication and regulation of most quality traits remain poorly understood.Here,we report the stepwise selection and regulation of major fruit quality traits that occurred during watermelon evolution.The levels of fruit cucurbitacins and flavonoids were negatively selected during speciation,whereas sugar and carotenoid contents were positively selected during domestication.Interestingly,fruit malic acid and citric acid showed the opposite selection trends during the improvement.We identified a novel gene cluster(CGC1,cucurbitacin gene cluster on chromosome 1)containing both regulatory and structural genes involved in cucurbitacin biosynthesis,which revealed a cascade of transcriptional regulation operating mechanisms.In the CGC1,an allele caused a single nucleotide change in Cl ERF1 binding sites(GCC-box)in the promoter of Cl Bh1,which resulted in reduced expression of Cl Bh1 and inhibition of cucurbitacin synthesis in cultivated watermelon.Functional analysis revealed that a rare insertion of 244 amino acids,which arose in C.amarus and became fixed in sweet watermelon,in Cl OSC(oxidosqualene cyclase)was critical for the negative selection of cucurbitacins during watermelon evolution.This research provides an important resource for metabolomics-assisted breeding in watermelon and for exploring metabolic pathway regulation mechanisms.Pingli Yuan Congping Xu Nan He Xuqiang Lu Xingping Zhang Jianli Shang Hongju Zhu Chengsheng Gong Hanhui Kuang Tang Tang Yong Xu Shuangwu Ma Dexi Sun Weiqin Zhang Muhammad J.Umer Jian Shi Alisdair R.Fernie Wenge Liu Jie Luo 2023Science China(Life Sciences)2023,66,3:0
19Correlation study of relative parameters of intracranial pressure and prognosis of patient with craniocerebral injury显示文摘Objective:To analyze the correlation of the relative parameters of intracranial pressure to the prognosis in patients with craniocerebral injury.Methods:The clinical data of 80 patients with closed craniocerebral injury were retrospectively analyzed,and all of these patients underwent conventional examinations of arterial blood pressure and intracranial pressure.Neumatic DCR system was used to monitor relative parameters of intracranial pressure from patients.According to the score of Glasgow outcome scale(GOS)upon discharge,they were divided into favorable prognosis group(GOSⅢ-Ⅴ,n=46)and unfavorable prognosis group(GOSⅠ-Ⅱ,n=34).The relative parameters of intracranial pressure of two groups were compared so as to analyze the correlation of the prognosis in patients to ICP-related parameters.Results:Pressure reactivity index(PRx)and intracranial pressure(ICP)of favorable prognosis group were significantly higher than those of unfavorable prognosis group(t=12.27,t=5.22,p<0.05).Meanwhile,cerebral perfusion pressure(CPP)and ICP-ABP wave amplitude correlation(IAAC)of favorable prognosis group were significantly lower than those of unfavorable prognosis group(t=14.54,t=14.78,p<0.05).The average age,gender,duration of admission to neurosurgical intensive care unit(NICU)and GCS(Glasgow coma scale)score on admission of the two groups were not statistically significant.Conclusions:The prognosis and ICP-related parameters(such as PRx,ICP,CPP,etc.)in patients with craniocerebral injury are risk factors for the prognosis effect.Therefore,to monitor the above-mentioned indicators has an important clinical value for assessing the prognosis of craniocerebral injury.Laixing Liu Yixin Deng Fuquan Liu Lei Chen Dexi Li Xiangyi Wang 2019Discussion of Clinical Cases2019,6,4:0
20Utilization and Inheritance of Watermelon Resistance to Fusarium oxysporum f.sp.niveum Introduced from Bottle Gourd显示文摘Sugar Baby susceptible to Fusarium oxysporum f. sp. niveum was crossed with high resistant male parents D3 - 1 and D3 - 2, respectively. F1 hybrids showed high resistance. The segregation ratios of resistance to susceptibility of F2 and of BC1 hybrid population from Sugar Baby tallied with 3: 1 and 1: 1, respectively. The results indicated that the resistance to Fusarium wilt was a kind of dominant inheritance controlled by mono- gene or mono- segment DNA. Furthermore, 5 hybrid combinations with fine character were bred.
