Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram. Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. This stratification of patients according to their PNI status may provide a basis for individualized auxiliary treatment. | Yanqi Huang Lan He Di Dong Caiyun Yang Cuishan Liang Xin Chen Zelan Ma Xiaomei Huang Su Yao Changhong Liang Jie Tian Zaiyi Liu | 2018 | Chinese Journal of Cancer Research2018,30,1: | 23
Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis. However, little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins, including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types, and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels. The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies. Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry, providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations. | Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu | 2022 | Cell Research2022,32,1: | 8
Activation of β-catenin signaling in aggrecan-expressing cells in temporomandibular joint causes osteoarthritis-like defects显示文摘β-Catenin plays a critical role in cartilage formation and development. To further understand the role of β-catenin in osteoarthritis(OA) development in temporomandibular joint(TMJ), we have generated β-catenin conditional activation mice(β-cat(ex3)^(Agc1CreER))by breeding Agc1-CreER mice with β-catenin^(flox(ex3)/+)mice. Results of histologic analysis showed the progressive TMJ defects in 3-and 6-month-old β-cat(ex3)^(Agc1CreER)mice(tamoxifen induction was performed at 2 weeks of age), including decreased chondrocyte numbers in the superficial layer associated with less Alcian blue staining, increased numbers of hypertrophic chondrocytes in deep layers, and rough articular surface. Compared to the TMJ phenotype of β-cat(ex3)^(Col2CreER)mice, β-cat(ex3)^(Agc1CreER)mice showed much severe morphological defects in the superficial layer of TMJ. This may reflect that Agc1-CreER mice could efficiently target cells in the superficial layer of TMJ. Results of immunostaining showed significantly increased expression of MMP13, Col-X, Adamts4,and Adamts5 in TMJ of β-cat(ex3)^(Agc1CreER)mice. Results of proliferating cell nuclear antigen(PCNA), Ki67, and terminal deoxinucleotidyl transferase-mediated d UTP-fluorescein nick end labeling(TUNEL) staining further demonstrated that cell proliferation was decreased and cell apoptosis was increased in condylar cartilage of β-cat(ex3)^(Agc1CreER)mice. Our findings indicate that abnormal upregulation of β-catenin in TMJ leads to defects assembling to OA-like phenotype, further demonstrating that β-catenin plays a critical role in TMJ pathogenesis. | Tianqian Hui Yachuan Zhou Tingyu Wang Jun Li Shanxing Zhang Lifan Liao Jianhong Gu Ling Ye Lan Zhao Di Chen | 2018 | International Journal of Oral Science2018,10,2: | 6
Molecular mechanisms of dietary restriction in aging—insights from Caenorhabditis elegans research显示文摘Dietary restriction(DR) is one of the most robust environmental manipulations that not only extend life span but also delay the onset of age-related diseases in almost every species examined. Caenorhabditis elegans plays an important role in aging studies due to its simple life cycle, easy genetic manipulations and highly conserved genome. Recent studies have demonstrated that the beneficial effects of DR are mediated by the highly conserved transcription factors and signaling pathways in C. elegans. Here we review recent progress in the methodology and molecular mechanisms of DR using C. elegans as a model, as well as prospects for future research. | LAN JianFeng ZHANG Xuan CHEN Di | 2015 | Science China(Life Sciences)2015,58,4: | 6
Electrical and thermal transport behaviours of high-entropy perovskite thermoelectric oxides显示文摘Oxide-based ceramics could be promising thermoelectric materials because of their thermal and chemical stability at high temperature. However, their mediocre electrical conductivity or high thermal conductivity is still a challenge for the use in commercial devices. Here,we report significantly suppressed thermal conductivity in SrTiO_(3)-based thermoelectric ceramics via high-entropy strategy for the first time, and optimized electrical conductivity by defect engineering. In high-entropy(Ca_(0.2)Sr_(0.2)Ba_(0.2)Pb_(0.2)La_(0.2))TiO_(3)bulks,the minimum thermal conductivity can be 1.17 W/(m·K)at 923 K,which should be ascribed to the large lattice distortion and the huge mass fluctuation effect. The power factor can reach about 295μW/(m·K^(2))by inducing oxygen vacancies. Finally, the ZT value of 0.2 can be realized at 873 K in this bulk sample. This approach proposed a new concept of high entropy into thermoelectric oxides, which could be generalized for designing high-performance thermoelectric oxides with low thermal conductivity. | Yunpeng Zheng Mingchu Zou Wenyu Zhang Di Yi Jinle Lan Ce-Wen Nan Yuan-Hua Lin | 2021 | Journal of Advanced Ceramics2021,10,2: | 5
Dendrocalamus latiflorus and its component rutin exhibit glucose-lowering activities by inhibiting hepatic glucose production via AKT activation显示文摘The potential medicinal value of Ma bamboo(Dendrocalamus latiflorus), one of the most popular and economically important bamboo species in China, has been underestimated. In the present study, we found that D. latiflorus leaf extract(DLE) reduced fasting blood glucose levels, body weight, and low-density lipoprotein cholesterol with low liver toxicity in db/db mice. In addition, gene expression profiling was performed and pathway enrichment analysis showed that DLE affected metabolic pathways.Importantly, DLE activated the AKT signaling pathway and reduced glucose production by downregulating glucose-6-phosphatase(G6 PC) and phosphoenolpyruvate carboxykinase 1(PCK1) expression. Moreover, network pharmacology analysis identified rutin as an active component in DLE through targeting insulin growth factor 1 receptor(IGF1 R), an upstream signaling transducer of AKT. Due to its hypoglycemic effects and low toxicity, DLE may be considered an adjuvant treatment option for type 2 diabetes patients. | Kun Luo Wenting Huang Liansheng Qiao Xiaoling Zhang Di Yan Zhiyu Ning Chengmei Ma Honglei Dang Dong Wang Hongyan Guo Lan Xie Jing Cheng | 2022 | Acta Pharmaceutica Sinica B2022,12,5: | 4
Photonuclear production of nuclear isomers using bremsstrahlung induced by laser-wakefield electrons显示文摘In this study,we theoretically investigate the feasibility of using laser-wakefield accelerated(LWFA)electrons for the photonuclear measurement of nuclear isomers according to the characteristics of the electrons obtained from LWFA experiments conducted at the Compact Laser-Plasma Accelerator(CLAPA)laboratory. The experiments at the CLAPA show that a stable electron beam with an energy of 78-135 MeV and a charge of 300-600 pC can be obtained. The bremsstrahlung spectra were simulated using Geant4,which suggests that a bremsstrahlung source with a peak intensity of 10^(19) photons/s can be generated.Theoretical calculations of isomer production cross sections from the photonuclear reactions on six target nuclei,^(197)Au,^(180)Hf,^(159)Tb,^(115)In,^(103)Rh,and ^(90)Zr,were performed and compared with the available experimental data in EXFOR,which suggest that further experiments are required for a series of photonuclear reaction channels.Flux-averaged cross sections and isomer ratios(IR)resulting from such bremsstrahlung sources are theoretically deduced. The results suggest that IR measurements can be used to constrain nuclear components, such asγstrength function and optical model potential. In addition, the detection of the decay characteristics was evaluated with Geant4 simulations. The use of the LWFA electron beam and its bremsstrahlung for photonuclear studies involving nuclear isomers is anticipated. | Hao-Yang Lan Di Wu Jia-Xin Liu Jian-Yao Zhang Huan-Gang Lu Jian-Feng Lv Xue-Zhi Wu Wen Luo Xue-Qing Yan | 2023 | Nuclear Science and Techniques2023,34,5: | 3

Effect of simulated thermomechanical processing on transformation behavior and microstructure of 82B steel显示文摘The effects of loop-laying temperature and austenite deformation on the phase(ransformation behavior during continuous cooling, microstructure, and pearlite interlaminar spaci ng in 82B steels were investigated.Static and dynamic conti nu ous cooling transformation(CCT)diagrams were measured with a Gleeble-3500 thermal simulator, and the mechanisms governing changes in the initial temperature, initial time, and duration of the phase transformation zone were also analyzed and discussed. The results show that CCT diagram shifted to the bottom right, the initial temperature of the phase transition decreased, the initial time of the phase transition increased, the duration of the phase transition increased, and the lamellar spacing of pearlite was finer as the loop-layi ng temperature in creased. The initial phase transition time decreased, and the phase transition duration first reduced, then increased, and finally decreased in the static condition and in the dynamic condition at 850℃as the cooling rate increased.Meanwhile, the phase transition duration continuously decreased in the dynamic condition at 900℃.At a given loop-laying temperature, the lamellar spacing in pearlite was finer due to austenite deformation compared with the undeformed case.Compared with the results shown in the dynamic CCT diagram, the corresponding phase diagrams of the static CCT diagram slightly shifted to the bottom right. Moreover, there was a clear lin ear relationship between the reciprocal of the lamellar spaci ng in pearlite and the average undercooling degree in the phase transformation zone. | He Wei Yin-li Chen Lan Su Di Tang | 2019 | Journal of Iron and Steel Research(International)2019,26,1: | 2
Heterostructured CoFe@N-doped carbon porous polyhedron for efficient microwave absorption显示文摘The synthesis of wide bandwidth, thin thickness, and high performance microwave absorbing materials has become a hot topic of current research.Metal-organic frameworks with heterojunctions and porous structures are considered as suitable candidates to meet these characteristics.Herein, heterogeneous CoFe@N-doped porous carbon polyhedron composites were successfully synthesized via Fe^(2+)to replace Co in zeolite imidazole frame-67.The dielectric properties of composites were enhanced by the replacement of Fe^(2+),and the synergistic effect of dielectric loss and magnetic loss was realized. The petal-like lamellar structure increases the travel of electromagnetic(EM)waves, and the formation of porous structures improves impedance matching.Specifically,a reflection loss of−67.30 dB was obtained at a thickness of 2.88 mm,and an ultrabroad wide effective absorption bandwidth of 8.40 GHz was obtained, covering most of the X-band(8–12 GHz)and the whole Ku-band(12–18 GHz).The radar cross section(RCS)reduction value can reach 29.4 dB·m^(2),which means that the radar detector has a smaller probability of detecting targets. This work describes the unique advantages of metal ion replacement metal-organic frameworks derived materials in structural design, impedance matching, and performance adjustment, and provides a new reference for the field of electromagnetic wave absorption. | Dan Wu Yiqun Wang Shuanglin Deng Di Lan Zhongning Xiang Qinchuan He | 2023 | Nano Research2023,16,2: | 2
Calibration of CR-39 solid-state track detectors for study of laser-driven nuclear reactions显示文摘It is of particular interest to investigate nuclear fusion reactions generated by high-intensity lasers in plasma environments that are similar to real astrophysical conditions. We have experimentally investigated2H(d,p)3H,one of the most crucial reactions in big bang nucleosynthesis models,at the Shenguang-Ⅱlaser facility. In this work,we present a new calibration of CR-39 solidstate track detectors, which are widely employed as the main diagnostics in this type of fusion reaction experiment. We measure the dependence of the track diameter on the proton energy. It is found that the track diameters of protons with different energies are likely to be identical. We propose that in this case, the energy of the reaction products can be obtained by considering both the diameters and gray levels of these tracks. The present results would be very helpful for analyzing the2 H(d,p)3H reaction products recorded with the same batch of CR-39 solid-state track detectors. | Yang-Fan He Xiao-Feng Xi Shi-Lun Guo Bing Guo Chuang-Ye He Fu-Long Liu Di Wu Ji-Hong Wei Wan-Sha Yang Luo-Huan Wang Dong-Hai Zhang Meng-Lin Qiu Guang-Fu Wang Chao-Yang Li Xiao-Fei Lan | 2020 | Nuclear Science and Techniques2020,31,4: | 2
Characterization of Porcine Matrix Metalloproteinase 23 (pMMP-23) Gene and Its Association with Litter Size Traits显示文摘The matrix metalloproteinase 23 (MMP-23), which might play a role in ovulation in mammals, was one of the promising candidate genes for litter size traits in pigs. In the present research, partial sequence of porcine MMP-23 (pMMP-23) gene, including exons 2-8 (GenBank: EU360790), was obtained. Real-time PCR analysis revealed that pMMP-23 gene was highly expressed in ovary. PCR-Sau3A I-RFLP and PCR-Acc II-RFLP assay were established to detect a C/T mutation in exon 3 (EU360790: g. 269C>T) and an A/G mutation in exon 4 (EU360790: g. 505A>G), respectively. Association study for these two SNPs with litter size was assessed in three independent populations (Minzhu, Landrace I and Landrace II). Statistical analysis demonstrated that for second and subsequent litters, TT sows produced more TNB than CC pigs in Landrace breed (P<0.05) at g. 269C>T locus, and the additive effect was significant (P<0.05); GG sows produced more TNB and NBA than AA pigs in Minzhu (P<0.01) and Landrace breeds (P<0.05) at g. 505A>G locus, and the additive effect was significant (P<0.01 or P<0.05). Our study suggested that the pMMP-23 gene might be a novel candidate gene for litter size traits, and g. 505A>G locus might be a useful molecular marker for marker assisted selection (MAS). | NIU Bu-yue LAN Xiao-ming YAN Xiao-hong DI Sheng-wei WANG Yang LI Hai-tao XIONG Yuan- zhu WANG Xi-biao | 2013 | Journal of Integrative Agriculture2013,12,1: | 2
Curcumin alleviates TGF-β1-induced fibrosis in NRK-49F cells via suppression of miR-21 expression, and regulation of the TGF-β1/smad3 signaling pathway显示文摘Objective:Overexpression of microRNA-21(miR-21)has been well-recognized during fibrosis in a wide range of tissue types through regulation of fibrogenesis via diverse pathways.Curcumin exhibits antifibrotic effects in various organs. We and others have previously demonstrated that curcumin has positive effects on the protection against fibrosis. However, the effect of curcumin on miR-21 expression has not been reported. Methods:In this study, transforming growth factor-b1(TGF-b1)-stimulated rat renal fibroblast cells(NRK-49F)were transfected with miR-21 inhibitors, then the expression of Smad3,a-smooth muscle actin(a-SMA),type I collagen(COL1A1),and type III collagen(COL3A1)was determined using quantitative real-time-PCR(qRT-PCR)and western blot analysis. The effect of curcumin on production of the extracellular matrix was evaluated using immunofluorescence,qRT-PCR,and western blot analysis.Results:TGF-b1 stimulation upregulated the expression of miR-21 and induced fibrogenesis in NRK-49F cells.Transfection with miR-21 inhibitors selectively decreased smad3 activity, and noticeably reduced the expression of a-SMA,COL1A1,and COL3A1.Curcumin treatment significantly inhibited the expressions of smad3,a-SMA,COL1A1,COL3A1,as well as miR-21 expression in NRK-49F cells in a dosedependent manner. Conclusion:Curcumin exerted its anti-fibrotic effects by targeting the TGF-b1/smad3 signaling pathway and suppressing miR-21 expression, thereby provides novel insight in the protective effects of curcumin against fibrosis in various organs. | Lan Zhang Yi Liu Xiting Wang Cheng Li Mengmeng Shen Liming Di Shuangshuang He Yu Li | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 2

Mechanism of Learning and Memory Impairment in Rats Exposed to Arsenic and/or Fluoride Based on Microbiome and Metabolome显示文摘Objective Arsenic(As) and fluoride(F) are two of the most common elements contaminating groundwater resources. A growing number of studies have found that As and F can cause neurotoxicity in infants and children, leading to cognitive, learning, and memory impairments. However, early biomarkers of learning and memory impairment induced by As and/or F remain unclear. In the present study, the mechanisms by which As and/or F cause learning memory impairment are explored at the multi-omics level(microbiome and metabolome). Methods We stablished an SD rats model exposed to arsenic and/or fluoride from intrauterine to adult period. Results Arsenic and/fluoride exposed groups showed reduced neurobehavioral performance and lesions in the hippocampal CA1 region. 16S rRNA gene sequencing revealed that As and/or F exposure significantly altered the composition and diversity of the gut microbiome, featuring the Lachnospiraceae_NK4A136_group, Ruminococcus_1, Prevotellaceae_NK3B31_group, [Eubacterium]_xylanophilum_group. Metabolome analysis showed that As and/or F-induced learning and memory impairment may be related to tryptophan, lipoic acid, glutamate, gamma-aminobutyric acidergic(GABAergic) synapse, and arachidonic acid(AA) metabolism. The gut microbiota, metabolites, and learning memory indicators were significantly correlated. Conclusion Learning memory impairment triggered by As and/or F exposure may be mediated by different gut microbes and their associated metabolites. | ZHANG Xiao Li YU Sheng Nan QU Ruo Di ZHAO Qiu Yi PAN Wei Zhe CHEN Xu Shen ZHANG Qian LIU Yan LI Jia GAO Yi LYU Yi YAN Xiao Yan LI Ben REN Xue Feng QIU Yu Lan | 2023 | Biomedical and Environmental Sciences2023,36,3: | 2
Stretchable and self-healable hydrogel artificial skin显示文摘Hydrogels have emerged as promising materials for the construction of skin-like mechanical sensors. The common design of hydrogel-based artificial skin requires a dielectric sandwiched between two hydrogel layers for capacitive sensing. However, such a planar configuration limits the sensitivity, stretchability and self-healing properties. Here,we report the design of single-layer composite hydrogels with bulk capacitive junctions as mechanical sensors. We engineer dielectric peptide-coated graphene(PCG)to serve as homogenously dispersed electric double layers in hydrogels.Any mechanical motions that alter the microscopic distributions of PCG in the hydrogels can significantly change the overall capacitance. We use peptide self-assembly to render strong yet dynamic interfacial interactions between the hydrogel network and graphene. The resulting hydrogels can be stretched up to 77 times their original length and self-heal in a few minutes. The devices can effectively sense strain and pressure in both air and aqueous environments, providing tremendous opportunities for next-generation iontronics. | Bin Xue Hui Sheng Yongqiang Li Lan Li Weishuai Di Zhengyu Xu Linjie Ma Xin Wang Haoting Jiang Meng Qin Zhibo Yan Qing Jiang Jun-Ming Liu Wei Wang Yi Cao | 2022 | National Science Review2022,9,7: | 2
