| 1 | Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood.In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux,and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners.Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux,leading to an increase in intracellular total cholesterol,free cholesterol,and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression,resulting in a decrease in cholesterol efflux from macrophages.The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor.On the contrary,the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection.The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice,which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly,aortic lipid deposition and plaque area were exacerbated,and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone.These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages,leading to foam cell formation and AS development. | Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong | 2019 | Acta Biochimica et Biophysica Sinica2019,51,5: | 4 |

| 3 | Orderly coexistence and accumulation models of conventional and unconventional hydrocarbons in Lower Permian Fengcheng Formation,Mahu sag,Junggar Basin显示文摘By using the latest geological,seismic,drilling and logging data,this article studies the basic conditions for the formation of the total petroleum system and the orderly coexisting characteristics and accumulation models of conventional&unconventional reservoirs in the Lower Permian Fengcheng Formation in the Junggar Basin.Controlled by thermal evolution,hydrocarbon generation and expulsion process of the high-quality source rocks in alkaline lake as well as the characteristics of multi-type reservoirs(conglomerate,sandstone,dolomite and shale),conventional structure-lithologic reservoirs and tight oil and shale oil reservoirs controlled by source-reservoir structure have been formed.On the plane,mature conventional reservoirs,medium-high mature tight oil,and medium-high mature shale oil reservoirs coexist orderly from the slope area around Mahu sag to the sag.Based on the orderly coexisting characteristics of conventional and unconventional reservoirs in the Fengcheng Formation,it is clear that oil and gas in the Fengcheng Formation accumulate continuously over a large area in three accumulation models:integrated source-reservoir,source-reservoir in close contact,and separated source-reservoir model.The three accumulation models differ in relationship between source-reservoir structure,reservoir lithology and spatial distribution,hydrocarbon migration,oil and gas type.It is pointed out that the conventional&unconventional oil and gas should be explored and developed as a whole to achieve an overall breakthrough of the total petroleum system.This study is expected to enrich the geological theory of oil and gas enrichment in continental basins and to provide an analogy for exploration and research in other hydrocarbon-rich sags. | ZHI Dongming TANG Yong HE Wenjun GUO Xuguang ZHENG Menglin HUANG Liliang | 2021 | Petroleum Exploration and Development2021,48,1: | 2 |
| 4 | SSD Real-Time Illegal Parking Detection Based on Contextual Information Transmission显示文摘With the improvement of the national economic level,the number of vehicles is still increasing year by year.According to the statistics of National Bureau of Statics,the number is approximately up to 327 million in China by the end of 2018,which makes urban traffic pressure continues to rise so that the negative impact of urban traffic order is growing.Illegal parking-the common problem in the field of transportation security is urgent to be solved and traditional methods to address it are mainly based on ground loop and manual supervision,which may miss detection and cost much manpower.Due to the rapidly developing deep learning sweeping the world in recent years,object detection methods relying on background segmentation cannot meet the requirements of complex and various scenes on speed and precision.Thus,an improved Single Shot MultiBox Detector(SSD)based on deep learning is proposed in our study,we introduce attention mechanism by spatial transformer module which gives neural networks the ability to actively spatially transform feature maps and add contextual information transmission in specified layer.Finally,we found out the best connection layer in the detection model by repeated experiments especially for small objects and increased the precision by 1.5%than the baseline SSD without extra training cost.Meanwhile,we designed an illegal parking vehicle detection method by the improved SSD,reaching a high precision up to 97.3%and achieving a speed of 40FPS,superior to most of vehicle detection methods,will make contributions to relieving the negative impact of illegal parking. | Huanrong Tang Aoming Peng Dongming Zhang Tianming Liu Jianquan Ouyang | 2020 | Computers, Materials & Continua2020,,1: | 2 |
| 5 | Deep learning-based evaluation of factor of safety with confidence interval for tunnel deformation in spatially variable soil显示文摘The random finite difference method(RFDM) is a popular approach to quantitatively evaluate the influence of inherent spatial variability of soil on the deformation of embedded tunnels.However,the high computational cost is an ongoing challenge for its application in complex scenarios.To address this limitation,a deep learning-based method for efficient prediction of tunnel deformation in spatially variable soil is proposed.The proposed method uses one-dimensional convolutional neural network(CNN) to identify the pattern between random field input and factor of safety of tunnel deformation output.The mean squared error and correlation coefficient of the CNN model applied to the newly untrained dataset was less than 0.02 and larger than 0.96,respectively.It means that the trained CNN model can replace RFDM analysis for Monte Carlo simulations with a small but sufficient number of random field samples(about 40 samples for each case in this study).It is well known that the machine learning or deep learning model has a common limitation that the confidence of predicted result is unknown and only a deterministic outcome is given.This calls for an approach to gauge the model’s confidence interval.It is achieved by applying dropout to all layers of the original model to retrain the model and using the dropout technique when performing inference.The excellent agreement between the CNN model prediction and the RFDM calculated results demonstrated that the proposed deep learning-based method has potential for tunnel performance analysis in spatially variable soils. | Jinzhang Zhang Kok Kwang Phoon Dongming Zhang Hongwei Huang Chong Tang | 2021 | Journal of Rock Mechanics and Geotechnical Engineering2021,13,6: | 2 |

The lower assemblage in the southern margin of the Junggar Basin has favorable geological conditions for forming large Petroleum fields, including:(1) Multiple sets of source rocks, of which the Jurassic and Permian are the main source rocks, with a large source kitchen.(2) Multiple sets of effective reservoirs,namely Cretaceous Qingshuihe Formation, Jurassic Toutunhe Formation and the Khalza Formation etc.(3) Regional thick mudstone caprock of Cretaceous Tugulu Group, generally with abnormally high pressure and good sealing ability.(4) Giant structural traps and litho-stratigraphic traps are developed. The northern slope also has the conditions for large-scale litho-stratigraphic traps.(5) Static elements such as source rocks, reservoirs and caprocks are well matched, and the dynamic evolution is suitable for large oil and gas accumulation. | 12 | Yanshanian–Himalayan geodynamic transformation of the northwestern Junggar Basin, southwestern Central Asian Orogenic Belt (CAOB), and its significance for petroleum accumulation显示文摘The northwestern Junggar Basin in the southwestern Central Asian Orogenic Belt is a typical petroliferous basin.The widely distributed reservoirs in Jurassic–Cretaceous strata indicate that the region records Yanshanian–Himalayan tectonic activity,which affected the accumulation and distribution of petroleum.The mechanism of this effect,however,has not been fully explored.To fill the knowledge gap,we studied the structural geology and geochemistry of the well-exposed Wuerhe bitumen deposit.Our results indi-cate that deformation and hydrocarbon accumulation in the northwestern Junggar Basin during the Yanshanian–Himalayan geodynamic transformation involved two main stages.During the Yanshanian orogeny,a high-angle extensional fault system formed in Jurassic–Cretaceous strata at intermediate to shallow depths owing to dextral shear deformation in the orogenic belt.This fault system connected at depth with the Permian–Triassic oil–gas system,resulting in oil ascending to form fault-controlled reservoirs(e.g.,a veined bitumen deposit).During the Himalayan orogeny,this fault system was deacti-vated owing to sinistral shear caused by far-field stress related to uplift of the Tibetan Plateau.This and the reservoir densification caused by cementation formed favorable hydrocarbon preservation and accu-mulation conditions.Therefore,the secondary oil reservoirs that formed during the Yanshanian–Himalayan tectonic transformation and the primary oil reservoirs that formed during Hercynian–Indosinian orogenies form a total and complex petroleum system comprising conventional and uncon-ventional petroleum reservoirs.This might be a common feature of oil–gas accumulation in the Central Asian Orogenic Belt and highlights the potential for petroleum exploration at intermediate–shal-low depths. | Dongming Zhi Jingkun Zhang Tao Wu Anbin Wu Yong Tang Yin Liu Jian Cao | 2023 | Geoscience Frontiers2023,14,4: | 0 |

| 14 | Establishment of an indirect immunofluorescence assay for the detection of African swine fever virus antibodies显示文摘African swine fever(ASF)continues to cause enormous economic loss to the global pig industry.Since there is no safe and effective vaccine,accurate and timely diagnosis of ASF is essential to implement control measures.Indirect immunofluorescence assay(IFA)is a gold standard serological method recommended by the World Organization for Animal Health(WOAH).In this study,we used primary fetal kidney cells to establish a wild boar cell line(BK2258)that supported the efficient replication of ASF virus(ASFV)SD/DY-I/21 and showed visible cytopathic effect(CPE).Moreover,using BK2258,we established a sensitive and specific IFA for ASFV antibody detection.To standardize and evaluate the performance of this assay,we used serum samples from pigs infected with the low virulent genotype I SD/DY-I/21 and genotype II HLJ/HRB1/20,and immunized with the vaccine candidate HLJ/18-7GD,field samples,and negative serum samples.The IFA reacted with the ASFV-positive sera and displayed bright fluorescence foci.There was no non-specific green fluorescence due to cellular senescence or other cell damage-causing factors.Compared to a commercial indirect enzyme-linked immunosorbent assay(iELISA),ASFV antibodies were detected 1–4 days earlier using our IFA.The detection limits of the IFA and iELISA for the same ASFV-antibody positive serum samples were 1:25,600 and 1:6,400,respectively,indicating that the IFA is more sensitive than iELISA.The newly established IFA was highly specific and did not cross-react with sera positive for six other important porcine pathogens(i.e.,Classical swine fever virus(CSFV),Porcine reproductive and respiratory syndrome virus(PRRSV),Porcme circovirus type 2(PCV2),Pseudorabies virus(PRV),Foot-and-Mouth disease virus type O(FMDV/O),and Porcine epidemic diarrhea virus(PEDV)).This study thus provides a sensitive,specific,and reliable detection method that is suitable for the serological diagnosis of ASF. | Wan Wang Zhenjiang Zhang Weldu Tesfagaber Jiwen Zhang Fang Li Encheng Sun Lijie Tang Zhigao Bu Yuanmao Zhu Dongming Zhao | 2024 | Journal of Integrative Agriculture2024,23,1: | 0 |
| 15 | Preliminary Study of the Characteristics of Several Glossy Cabbage(Brassica oleracea var. capitata L.) Mutants显示文摘To determine the characteristics and potential practical applications of glossy cabbage(Brassica oleracea var. capitata L.) mutants, five different glossy mutants were studied. The amount of epicuticular wax covering the mutant leaves was only approximately 30% that of the wild-type(WT) leaves. The wax crystals of WT plants were columnar and linear, while they were granular and rod-shaped in the mutants. Additionally, in WT cabbage, the primary wax components were alkanes, alcohols, fatty acids, ketones, and aldehydes. There was a significant decrease in the abundance of alkanes and ketones in the wax of the mutants. The glossy-green trait of the mutants may be the result of an inhibited alkane-forming pathway. Higher rates of chlorophyll leaching and water loss demonstrate that the mutant leaves were more permeable and sensitive to drought stress than the WT leaves. Growth curve results indicated that the growth rate of mutant-1 and mutant-3 was slower than that of the corresponding WT cabbage, resulting in shorter plants. However, the growth rate of mutant-2 was not influenced by the lack of coating wax. An investigation of the agronomic traits and heterosis of the glossy cabbage mutants indicated that all five mutants had glossy-green leaves, which was a favorable characteristic. The F1 plants derived from crosses involving mutant-2 exhibited obvious heterosis, suggesting the observed glossy-green trait is controlled by a dominant gene. | 16 | Co-evolution of paleo-environment and bio-precursors in a Permian alkaline lake,Mahu mega-oil province,Junggar Basin:Implications for oil sources显示文摘The co-evolution of paleo-environment and bio-precursors in alkaline lakes is of significance to understand the extreme environment system and associated enigmatic hydrocarbon potential.Here we used organic petrology and biomarker geochemistry to investigate the bio-precursors in a Permian alkaline paleo-lake in the Mahu mega-oil province within about a hundred miles of the Mahu Sag,Junggar Basin,China,and its effect on oil generation and accumulation.In general,the bioprecursors in the alkaline lacustrine source rocks of the Fengcheng Formation were mainly bacteria and algae,with a low abundance of higher plants.Therefore,these source rocks were mainly prone to oil generation.Two distinctive hydrocarbongenerating bio-precursors—Dunaliella-like algae and cyanobacteria—were identified.In addition to fossil evidence for these bio-precursors,the former results in a high C_(28)/C_(29)sterane ratio andβ-carotane abundance,and the latter results in the formation of medium-chain monomethyl alkanes in terms of biomarkers.The nature of the bio-precursors varied with the sedimentary paleo-environment,and was controlled by the salinity and stratification of the lake.Dunaliella-type source rocks were deposited in the central area of the alkaline lake,and cyanobacteria-type source rocks were formed around the lake margins.The crude oils in different parts of the Mahu mega-oil province within about a hundred miles have different sources.The bio-precursors in the saline lacustrine source rocks were jointly controlled by the age and water salinity of the source rocks.The physiological synthesis of carotenoids and sterols by haloalkaliphilic green algae may have affected the evolution of ancient green algae. | Liuwen XIA Jian CAO Lizeng BIAN Wenxuan HU Tingting WANG Dongming ZHI Yong TANG Erting LI | 2022 | Science China Earth Sciences2022,65,3: | 0 |
