| 2 | A Cloud-edge Cooperative Dispatching Method for Distribution Networks Considering Photovoltaic Generation Uncertainty显示文摘With the increasing penetration of renewable energy generation,uncertainty and randomness pose great challenges for optimal dispatching in distribution networks.We propose a cloud-edge cooperative dispatching(CECD)method to exploit the new opportunities offered by Internet of Things(IoT)technology.To alleviate the huge pressure on the modeling and computing of large-scale distribution system,the method deploys edge nodes in small-scale transformer areas in which robust optimization subproblem models are introduced to address the photovoltaic(PV)uncertainty.Considering the limited communication and computing capabilities of the edge nodes,the cloud center in the distribution automation system(DAS)establishes a utility grid master problem model that enforces the consistency between the solution at each edge node with the utility grid based on the alternating direction method of multipliers(ADMM).Furthermore,the voltage constraint derived from the linear power flow equations is adopted for enhancing the operation security of the distribution network.We perform a cloud-edge system simulation of the proposed CECD method and demonstrate a dispatching application.The case study is carried out on a modified 33-node system to verify the remarkable performance of the proposed model and method. | Lu Shen Xiaobo Dou Huan Long Chen Li Ji Zhou Kang Chen | 2021 | Journal of Modern Power Systems and Clean Energy2021,9,5: | 4 |
| 3 | Three-Dimensional Seismic Response in Complex Site Conditions: A New Approach Based on an Auxiliary-Model Method显示文摘In this paper, an auxiliary-model method is proposed for calculating equivalent input seismic loads in research of ground motions. This method can be used to investigate the local effect of 3 D complex sites subjected to obliquely incident SV and P waves. Using this method, we build a fictitious auxiliary model along the normal direction of the boundary of the area of interest, with the model’s localized geological features remaining the same along a vector normal to this boundary. This model is divided into five independent auxiliary models, which are then dynamically analyzed to obtain the equivalent input seismic loads. Unlike traditional methods, in this new technique, the mechanical behavior of the auxiliary model can be nonlinear, and its geometry can be arbitrary. In addition, a detailed description of the steps to calculate the equivalent input seismic loads is given. Numerical examples of incident plane-wave propagation at uniform sites with local features validate the effectiveness of this method. | 4 | Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. | 5 | A new method for optimal FTU placement in distribution network under consideration of power service reliability显示文摘Modern distribution network with high penetration of intermittent renewable sources and the so-called prosumers requires more reliable distribution automation(DA) system for safe operation and control. The design of optimal feeder terminal units(FTU)placement is critical and economical for the effective DA application. Previously proposed solutions of optimal FTU placement aiming to ensure the accuracy of state estimation(SE), typically include the following two main shortcomings: 1) only to obtain the optimal FTU placement in quantity, and the analysis of FTU location is not considered yet; 2) few consider the uncertainty of intermittent power injections in the analysis of state estimation. In this paper, a modified methodology of FTU placement is proposed not only aiming to ensure the accuracy of state estimation with the minimum number of meters, but also finding those specific FTU locations to guarantee the power service reliability. Moreover, the uncertainty models of those intermittent power injections are also considered by using probability density function(PDF). The resultant optimization problem is addressed by using the covariance matrix adaptation evolution strategy(CMA-ES). | 8 | Engineering crop Phytophthora resistance by targeting pathogen-derived PI3P for enhanced catabolism显示文摘Phytophthora pathogens lead to numerous economically damaging plant diseases worldwide,including potato late blight caused by P.infestans and soybean root rot caused by P.sojae.Our previous work showed that Phytophthora pathogens may generate abundant phosphatidylinositol 3-phosphate(PI3P)to promote infection via direct association with RxLR effectors.Here,we designed a disease control strategy for metabolizing pathogen-derived PI3P by expressing secreted Arabidopsis thaliana phosphatidylinositol-4-phosphate 5-kinase 1(AtPIP5K1),which can phosphorylate PI3P to PI(3,4)P2.We fused AtPIP5K1 with the soybean PR1a signal peptide(SP-PIP5K1)to enable its secretion into the plant apoplast.Transgenic soybean and potato plants expressing SP-PIP5K1 showed substantially enhanced resistance to various P.sojae and P.infestans isolates,respectively.SP-PIP5K1 significantly reduced PI3P accumulation during P.sojae and soybean interaction.Knockout or inhibition of PI3 kinases(PI3Ks)in P.sojae compromised the resistance mediated by SP-PIP5K1,indicating that SP-PIP5K1 action requires a supply of pathogen-derived PI3P.Furthermore,we revealed that SP-PIP5K1 can interfere with the action of P.sojae mediated by the RxLR effector Avr1k.This novel disease control strategy has the potential to confer durable broad-spectrum Phytophthora resistance in plants through a clear mechanism in which catabolism of PI3P interferes with RxLR effector actions. | Kun Yang Qiang Yan Yi Wang Wenyi Zhu Xiaodan Wang Xiaobo Li Hao Peng Yang Zhou Maofeng Jing Daolong Dou | 2023 | Plant Communications2023,4,2: | 0 |
| 9 | Sensing of cytoplasmic chromatin by cGAS activates innate immune response in SARS-CoV-2 infection显示文摘The global coronavirus disease 2019(COVID-19)pandemic is caused by severe acute respiratory syndrome coronavirus 2(SARSCoV-2),a positive-sense RNA virus.How the host immune system senses and responds to SARS-CoV-2 infection remain largely unresolved.Here,we report that SARS-CoV-2 infection activates the innate immune response through the cytosolic DNA sensing cGAS-STING pathway.SARS-CoV-2 infection induces the cellular level of 2′3′-cGAMP associated with STING activation.cGAS recognizes chromatin DNA shuttled from the nucleus as a result of cell-to-cell fusion upon SARS-CoV-2 infection.We further demonstrate that the expression of spike protein from SARS-CoV-2 and ACE2 from host cells is sufficient to trigger cytoplasmic chromatin upon cell fusion.Furthermore,cytoplasmic chromatin-cGAS-STING pathway,but not MAVS-mediated viral RNA sensing pathway,contributes to interferon and pro-inflammatory gene expression upon cell fusion.Finally,we show that cGAS is required for host antiviral responses against SARS-CoV-2,and a STING-activating compound potently inhibits viral replication.Together,our study reported a previously unappreciated mechanism by which the host innate immune system responds to SARS-CoV-2 infection,mediated by cytoplasmic chromatin from the infected cells.Targeting the cytoplasmic chromatin-cGAS-STING pathway may offer novel therapeutic opportunities in treating COVID-19.In addition,these findings extend our knowledge in host defense against viral infection by showing that host cells’self-nucleic acids can be employed as a“danger signal”to alarm the immune system. | Zhuo Zhou Xinyi Zhang Xiaobo Lei Xia Xiao Tao Jiao Ruiyi Ma Xiaojing Dong Qi Jiang Wenjing Wang Yujin Shi Tian Zheng Jian Rao Zichun Xiang Lili Ren Tao Deng Zhengfan Jiang Zhixun Dou Wensheng Wei Jianwei Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 0 |
