1HBB-deficient Macaca fascicularis monkey presents with human β-thalassemia显示文摘Dear Editor,β-Thalassemia is a common severe genetic disease caused by mutations in HBB and affects approximately 1.5% of the global population (Origa, 2017). In southern China, the carrier rate of β-thalassemia is as high as 6.43%, creating a high socio-economic burden (Xiong et al., 2010). In adult humans, there are three types of hemoglobin: HbA1 (~97%), HbA2 (~2%) and HbF (~1%). HbA1 (α2β2) is composed of two a-globin and two β-globi n sub units en coded by HBA and HBB, respectively;HbF (α2β2)is made up of two α-globin subunits and two β-globin sub units en coded by HBG. Mutations in the coding region or regulatory region of HBB are involved in β-thalassemia pathogenesis. Except for some rare dominant mutations, most HBB mutations are recessive (Origa, 2017). Depending on the mutation type, the β-globin level will either be reduced or completely depleted, resulting in α-globin accumulation and precipitation. These α-globin precipitates lead to red blood cell death, resulting in anemia and tissue damage, and even death in thalassemia major patients. Blood transfusions can help slow disease progression but lead to iron overload, ultimately resulting in iron toxicity. Bone marrow transfer is the only cure in the clinic and is available only to a small percentage of patients with human leukocyte antigervmatched donors. Recently, gene therapy and gene editing therapy have shown great promise in curing β-thalassemia (Glaser et al., 2015;Thompson et al., 2018). However, no appropriate animal models are available for evaluating the safety and efficacy of such advanced therapeutic strategies in vivo.β-thalassemia mice are the sole animal model available for research. However, substantial differences have been reported between the types and expressi on patter ns of human and mouse globins (McColl and Vadolas, 2016). Moreover, mice contain no fetal globin gene equivalent, and homozygous mutations of HBB in mouse for early models of β-thalassemia major or Cooley anemia are all embryonic lethal (Huo et al., 2009). Recently, significant phenotype and physiology differences have been reported between SIRT6- null mice and the non-human primate model (Zhang et al., 2018). 4Claw-shaped flexible and low-impedance conductive polymer electrodes for EEG recordings: Anemone dry electrode显示文摘Electroencephalogram(EEG) signals provide important brain information in healthcare settings. Wet electrodes have several limitations, including their time-consuming preparation and no long-term acquisition, and dry electrodes have become a promising candidate. However, dry electrodes face several challenges related to signal quality, comfort, conductivity and cost.This study aims to develop and evaluate a novel dry electrode for EEG recordings. The electrode material properties are characterized by electrochemical, mechanical force, and skin measurements. The electrode performance is evaluated by comparing the impedance, signal-to-noise ratio(SNR), and EEG signal features of the proposed electrode with those of a wet electrode and two commercial dry electrodes. The dry electrode design is based on the biological structure of the sea anemone and demonstrates an improved scalp fit and reduced signal noise and motion artifacts. The electrode materials include advanced conducting polymers combined with thermoplastic elastomers(TPEs) and carbon nanotubes(CNTs), which exhibit good electrical conductivity, mechanical properties, and safety. A low-cost injection molding fabrication method is proposed. The dry electrodes show a scalp contact impedance of 7 kΩ at 20 Hz, which is lower than that of wet and commercial dry electrodes.Resting-state EEG and event-related potential signals collected by the anemone dry electrode achieved more than 90% similarity with signals acquired by wet electrodes. Thus, a low-cost, comfortable anemone dry electrode that exhibits excellent EEG recording performance is presented. 8Nonlinearity-enabled higher-order exceptional singularities with ultra-enhanced signal-to-noise ratio显示文摘Higher-order exceptional points(HOEPs)with extraordinary responsivity are expected to exhibit a vastly improved performance in detection-related applications.However,over the past few years,such an approach has been questioned due to several potential drawbacks,including the stringent parameter re quirements,fundamental resolution limits and noise.Here,exploring the conse quence of nonlinear gain saturation in exceptional singularities of non-Hermitian systems,we offer a feasible scheme to overcome all the above difficulties.We provide a simple and intuitive example by demonstrating with both theory and circuit experiments an'exceptional nexus'('EX'),a HOEP with an ultra-enhanced signal-to-noise ratio(SNR),in only two coupled resonators with the aid of nonlinear gain.The tedious parameter tuning in a six-dimensional hyper-dimensional space is reduced to two dimensions.The feedback mechanism of nonlinear saturable gain can give a solution to the ongoing debate on the SNR of EPs in other linear systems.Our findings advance the fundamental understanding of the peculiar topology of nonlinear non-Hermitian systems,significantly reduce the practical difficulty in EP sensing and possibly open new avenues for applications.Kai Bai Liang Fang Tian-Rui Liu Jia-Zheng Li Duanduan Wan Meng Xiao 2023National Science Review2023,10,7:0

12Effects of tissue heterogeneity on trabecular micromechanics examined by microCT-based finite element analysis and digital volume correlation显示文摘Trabecular bone is natural material with heterogeneous tissue properties.The effect of tissue heterogeneity on the micromechanical behavior of trabecular bone is commonly evaluated by microCT-based finite element(microFE)analysis.Results from prior work remain inconclusive and lack of experimental validation.To address these issues,we combined microFE analysis with mechanical testing and microCT-based digital volume correlation(DVC),as a validation for the microFE approach.Porcine trabecular specimens were tested in compression as sequential microCT scans were taken.DVC was performed to extract“realistic”boundary conditions that were applied to microFE models,and to measure microstructural deformation and strain of the trabecular specimens.Heterogeneous and homogeneous microFE models of each trabecular specimen were created and compared with the experimentally measured microstructural displacement and strains.Results showed strong correlations between DVC-measured and microFE-predicted trabecular displacement and strain fields(R^(2)>0.9,p<0.05),regardless of heterogeneous or homogeneous material assignments.The heterogeneous and homogeneous models predicted similar magnitudes for maximum or minimum principal strains(R^(2)=1,p<0.05).However,incorporation of tissue heterogeneity decreased more than 16.5%in the overall stress level of the trabecular tissues.Regardless,very strong correlations were found between the heterogeneous and homogeneous model-predicted principal strains or stresses.These results together suggest that tissue heterogeneity may have little effect on microFE modeling of typical elastic displacement and strains in the trabecular bone,suggesting that homogeneous material models might be sufficient to predict general trabecular micromechanics.Jizhi Fu Haoye Meng Changhao Zhang Youjun Liu Duanduan Chen Aiyuan Wang Russell P.Main Haisheng Yang 2021Medicine in Novel Technology and Devices2021,,3:0
13Determination of large diameter bored pile's effective length based on Mindlin's solution显示文摘The calculation equation of large diameter bored pile's effective length is connected with its distribution of pile shaft resistance.Thus,there is a great difference between the calculation results under the different distributions of pile shaft resistance.Primarily,this paper summarizes the conceptualized mode of pile shaft resistance under the circumstance that the soil surrounding the piles presents different layer distributions.Secondly,based on Mindlin's displacement solution and in consideration of the effect of pile diameter,the calculation equation is optimized with the assumption that the pile shaft resistance has a parabolic distribution.The influencing factors are analyzed according to the calculation result of effective pile length.Finally,combined with an engineering example,the calculation equation deduced in this paper is analyzed and verified.The result shows that both the Poisson ratio of soil and pile diameter have impacted the effective pile length.Compared with the Poisson ratio of soil,the effect of pile diameter is more significant.If the pile diameter remains the same,the effect of the Poisson ratio of soil to the effective pile length decreases as the ratio of pile elastic modulus and soil share modulus increases.If the Poisson ratio of soil remains the same,the effect of the pile diameter to the effective pile length increases as the ratio of pile elastic modulus and soil share modulus increases.Thus the optimized calculation result of pile effective length under the consideration of pile diameter effect is more close to the actual situation of engineering and reasonably practicable.Zhijun Zhou Duanduan Wang Lipeng Zhang Weisi Ma 2015Journal of Traffic and Transportation Engineering(English Edition)2015,2,6:0
14Research advances in gene therapy for Parkinson's disease显示文摘During the long period of time when people have been seeking for an effective therapeutic method for PD, the gene therapy has shown greater and greater advantages over other methods. It can be performed mainly in two ways: ex vivo and in vivo. With the former, TH gene as well as some neurotrophic factor genes (such as GDNF, BNDF genes) can be invited to some cell lines or primary cells thus forming engineered cells and then implanting them into brain. While with the latter, viral vectors including HSV-1, Ad, AAV that can be utilized to construct recombinant viruses, or non-virus vectors can be used to delived DNA into brain directly. 15Generation of 100 nJ pulse,1 W average power at 2μm from an intermode beating mode-locked all-fiber laser显示文摘We report on the investigation of intermode beating mode-locked(IBML)pulse generation in a simple all-fiber Tm^3+-doped double clad fiber laser(TDFL).This IBML TDFL is implemented by matching longitudinal-mode frequency between 793 nm laser and TDFL without extra mode locker.The central wavelength of 1983 nm,the fundamental pulse frequency of 9.6 MHz and the signal-to-noise ratio(SNR)of>50 dB are achieved in this IBML TDFL.With laser cavity optimization,the IBML TDFL can finally generate an average output power of 1.03 W with corresponding pulse energy of 107 nJ.These results can provide an easily accessible way to develop compact large-energy,highpower TDFLs.Jiaji Zhang Duanduan Wu Ruwei Zhao Rongping Wang Shixun Dai 2019High Power Laser Science and Engineering2019,7,4:0
