| 2 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |

| 4 | The Reference Genome Sequence of Scutellaria baicalensis Provides Insights into the Evolution of Wogonin Biosynthesis显示文摘Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,'Huang Qin,' are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. | 5 | Study on Relationship between Microstructure of Active Phase and HDS Performance of Sulfided Ni-Mo Catalysts: Effect of Metal Loading显示文摘Six Ni-Mo catalysts with different metal contents were prepared and characterized by N2 adsorption and X-ray diffractometry. The active phase microstructure of these catalysts was examined by the Raman spectroscopy, temperatureprogrammed reduction(TPR), X-ray photoelectron spectroscopy, and high-resolution transmission electron microscopy. Hydrodesulfurization(HDS) activity of catalyst samples were analyzed in a flow fixed-bed microreactor. The sulfidation degree of Mo and the length of the MoS2 slab slightly increased with the amount of metal loaded following sulfidation. This small change is attributed to polymolybdate species observed in all the oxidized catalysts. Weak metal-support interactions, as determined by the TPR technique, increased the NiSx sulfidation phase and MoS2 slab stacking. The HDS activity of the catalyst samples increased with the number of active sites. For high metal loading catalysts, their HDS activity was nearly identical because the sulfur atoms cannot easily approach active sites. | 7 | Changes in vitreous VEGF, bFGF and fibrosis in proliferative diabetic retinopathy after intravitreal bevacizumab显示文摘AIM: To evaluate the relationship between intravitreal bevacizumab(IVB) treatment and the levels of vitreous vascular endothelial growth factor(VEGF), basic fibroblast growth factor(b FGF) and vitreous-retina surface fibrosis in patients with proliferative diabetic retinopathy(PDR).·METHODS: This study was a prospective, open-label,controlled, randomized clinical trial. Sixty-eight eyes of PDR patients(n =53) and macular hole patients(n =15)were enrolled in this study. Thirty-four eyes of the PDR patients received IVB before vitrectomy. Twenty-three of the 34 PDR patients received IVB treatment 5d before vitrectomy(subgroup a), and 11 of the 34 PDR patients received IVB treatment greater than 2wk prior to vitrectomy(subgroup b). Nineteen of the PDR patients did not receive IVB treatment at any time prior to vitrectomy. The levels of b FGF and VEGF in vitreous samples were measured using enzyme-linked immunosorbent assay(ELISA) and the degree of vitreoretinal fibrosis was characterized using clinical data and data obtained intra-operatively.·RESULTS: In PDR patients, VEGF and b FGF levels were significantly increased compared to non-PDR(control) subject's eyes(P <0.01). In PDR patients,vitreous VEGF levels were significantly decreased following IVB treatment compared to PDR patients that did not receive IVB treatment(P <0.01). The degree of vitreoretinal fibrosis was significantly increased in subgroup b compared to subgroup a(P <0.05) and to patients that did not receive IVB(P <0.05). Vitreous b FGF levels were significantly greater in subgroup b than subgroup a(P <0.01) or in patients who did not receive IVB treatment(P <0.05). A Spearman's rank correlationtest indicated that higher levels of vitreous b FGF, but not VEGF, correlated with the degree of vitreoretinal fibrosis.·CONCLUSION: We found that b FGF levels increase in PDR patient's vitreous after IVB treatment longer than two weeks prior to vitrectomy and correlated with the degree of fibrosis after IVB treatment. | 10 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. | 11 | Old canalicular laceration repair:a retrospective study of the curative effects and prognostic factors显示文摘AIM: To investigate the epidemiology and surgical outcomes of old canalicular laceration and analyze the variables impacting on the prognosis of reparation. METHODS: A retrospective review of all old canalicular laceration repairs from Jan. 1, 2008 to Dec. 30, 2015 was performed. Analyzed data included demographics, mechanisms of injury, the time from injury to repair, causes for delayed repair, old associated injuries, the types of surgery, and the effects of repair using canaliculus anastomosis combined with bicanalicular stent intubation. RESULTS: Totally 148 patients with old canalicular laceration received surgical repair and were enrolled. The mean age at presentation was 32.52 years old(ranged from 3 to 63 years old). The 110 patients(74.32%) were male and 127 patients(85.81%) were adults(≥18 years old). The old upper, lower, and bicanalicular lacerations were found in 5(3.38%), 39(26.35%), and 104 patients(70.27%), respectively. The mechanism of old injury was primarily due to motor vehicle accidents(n=53, 35.81%). The mean time from injury to repair was 43.61mo(ranged from 1 to 360mo). Associated old ocular and orbit injuries were found in 65 patients(43.92%), and chronic dacryocystitis in 18 patients(12.16%). The main cause of delayed repair was that doctors or patients didn't pay attention to the canalicular laceration because of the concurrent severe injuries(n=71, 47.97%). Totally 136 patients(91.89%) with old canalicular laceration underwent canaliculus anastomosis combined with bicanalicular stent intubation. In all of them, 20 patients(13.51%) were combined with dacryocystorhinostomy. In these cases, 132 patients(97.06%) attained anatomic success, 121 patients(88.97%) reported no epiphora(functional success), 11 patients(8.09%) reported significant epiphora anesis(functional improvement), and 4(2.94%) reported no significant anesis(functional failure). Rates of anatomic success and functional success were significantly correlated with different canaliculus involved. | 12 | One-stitch anastomosis through the skin with bicanalicular intubation:a modified approach for repair of bicanalicular laceration显示文摘AIM:To evaluate the efficacy and safety of one-stitch anastomosis through the skin with bicanalicular silicone tube intubation in repairing of bicanalicular laceration.METHODS:The clinical data of 15 consecutive patients with both superior and inferior canalicular laceration in one eye who underwent surgical repair using one-stitch anastomosis through the skin and bicanalicular stent were retrospective studied.All the operations were performed under surgical microscope,5-0 silk sutures were used and were with bicanalicular silicone tube(diameter was 8mm) intubation,for one lacerated canaliculi one-stitch anastomosis through the skin.The stents were left in place for 3 months postoperatively and then removed.The follow-up period was 3-36 months(average 14 months). RESULTS:In 15 patients,13 patients were cured entirely,1 patient was meliorated,1 patient with no effects.All patients had got good recovery of eyelid laceration with no traumatic deformity in eyelid and canthus.Complication was seen in one case,for not followed the doctor's guidance to come back to hospital to had the suture removed on the 7thday after operation,when he came at the 15thday,the inferior canalicular wall and eyelid skin were corroded by the suture caused 2mm wound,and the inside silicone tube was exposed,a promptly repair with 10-0 nylon suture was done,the wound healed in a week.There were no early tube protrusions and punctal slits in the patients.CONCLUSION:One-stitch anastomosis through the skin with bicanalicular silicone tube intubation is a good method in repair of bicanalicular laceration in one eye,the cut ends can be anastomosed directly,and with excellent cosmetic results,it is acceptable for the patients.For there is no suture remained in the wound permanently,so there is no suture-related granuloma which may cause obstruction or stenosis of canaliculi.It is simple,economical,effective and safe. | Hai Tao Peng Wang Cui Han Jian Zhang Fang Bai Zhao-Yan He | 2013 | International Journal of Ophthalmology(English edition)2013,6,5: | 14 |

| 14 | MicroRNA-218 is upregulated in gastric cancer after cytoreductive surgery and hyperthermic intraperitoneal chemotherapy and increases chemosensitivity to cisplatin显示文摘AIM: To investigate the molecular mechanisms of miRNA in advanced gastric cancers(AGCs) before and after cytoreductive surgery(CRS) + hyperthermic intraperitoneal chemotherapy(HIPEC). METHODS: A miRNA microarray containing human mature and precursor miRNA sequences was used to compare expression profiles in serum samples of 5 patientswith AGC before and after CRS + HIPEC. The upregulation of miR-218 was confirmed by real-time reverse transcription polymerase chain reaction and its expression was analyzed in SGC7901 gastric cancer cells. RESULTS: miRNA microarray chip analysis found that the level of miR-218 expression was upregulated more than 8 fold after CRS + HIPEC. | 17 | Lira-YOLO: a lightweight model for ship detection in radar images显示文摘For the detection of marine ship objects in radar images, large-scale networks based on deep learning are difficult to be deployed on existing radar-equipped devices. This paper proposes a lightweight convolutional neural network, LiraNet, which combines the idea of dense connections, residual connections and group convolution, including stem blocks and extractor modules.The designed stem block uses a series of small convolutions to extract the input image features, and the extractor network adopts the designed two-way dense connection module, which further reduces the network operation complexity. Mounting LiraNet on the object detection framework Darknet, this paper proposes Lira-you only look once(Lira-YOLO), a lightweight model for ship detection in radar images, which can easily be deployed on the mobile devices. Lira-YOLO's prediction module uses a two-layer YOLO prediction layer and adds a residual module for better feature delivery. At the same time, in order to fully verify the performance of the model, mini-RD, a lightweight distance Doppler domain radar images dataset, is constructed. Experiments show that the network complexity of Lira-YOLO is low, being only 2.980 Bflops, and the parameter quantity is smaller, which is only 4.3 MB. The mean average precision(mAP) indicators on the mini-RD and SAR ship detection dataset(SSDD) reach 83.21% and 85.46%, respectively,which is comparable to the tiny-YOLOv3. Typically, males vocalize in choruses during one or more periods in a twenty-four-hour cycle, which varies, however, among species. Nevertheless, the causal factors influencing circadian variations of calling patterns in anuran species are not clear. In this study, male chorus vocalizations were monitored in the Emei music frog(Babina daunchina) for 17 consecutive days during the breeding season, while its habitat air temperature and relative humidity in the course of experiments were measured as well. The results revealed that the circadian calling patterns were characterized by two periods of peak vocalization, which were observed from 0500 h to 0700 h and from 1300 h to 2000 h, while the lowest activity period was found from 2100 h to 2200 h. Both calls/h and notes/h were positively correlated with air temperature and negatively with relative humidity. | 20 | A Brassica miRNA Regulates Plant Growth and Immunity through Distinct Modes of Action显示文摘In plants,high disease resistance often results in a reduction of yield.Therefore,breeding crops with balanced yield and disease resistance has become a major challenge.Recently,microRNA(miRNA)-mediated R gene turnover has been shown to be a protective mechanism used by plants to prevent autoimmunity in the absence of pathogens.However,whether these miRNAs play a role in plant growth and how miRNA-mediated R gene turnover responds to pathogen infection have rarely been explored.Here,we found that a Brassica miRNA,miR1885,targets both an immune receptor gene and a development-related gene for negative regulation through distinct modes of action.MiR1885 directly silences the TIR-NBS-LRR class of R gene BraTNL1 but represses the expression of the photosynthesis-related gene BraCP24 by targeting the Trans-Acting Silencing(TAS)gene BraTIR1 for trans-acting small interfering RNAs(tasiRNAs)-mediated silencing.We found that,under natural conditions,miR1885 was kept at low levels to maintain normal development and basal immunity but peaked during the floral transition to promote flowering.Interestingly,upon Turnip mosaic virus(TuMV)infection,miR1885-dependent trans-acting silencing of BraCP24 was enhanced to speed up the floral transition,whereas miR1885-mediated R gene turnover was overcome by TuMV-induced BraTNL1 expression,reflecting precise regulation of the arms race between plants and pathogens.Collectively,our results demonstrate that a single Brassica miRNA dynamically regulates both innate immunity and plant growth and responds to viral infection,revealing that Brassica plants have developed a sophisticated mechanism in modulating the interplay between growth,immunity,and pathogen infection. | Chen Cui Jing-Jing Wang Jian-Hua Zhao Yuan-Yuan Fang Xiang-Feng He Hui-Shan Guo Cheng-Guo Duan | 2020 | Molecular Plant2020,13,2: | 11 |
