3Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 5Real-world study of surgical treatment of pancreatic cancer in China: annual report of China Pancreas Data Center (2016-2020)显示文摘Objective: In 2015, the Chinese Pancreatic Association of the Chinese Society of Surgery of the Chinese Medical Association launched a national multicenter online system for registration of surgical treatment of pancreatic cancer in China, called China Pancreas Data Center (CPDC). With continued effort, the CPDC has developed over time. Herein, we report the general results of the CPDC from January 2016 to January 2020 to present the real-world situation of surgical treatment of pancreatic cancer in China.Methods: The data of the CPDC from January 2016 to January 2020 were retrieved and analyzed in this real-world study, including the data on patient demographics, comorbidities, diagnostic modalities, neoadjuvant treatment, surgical procedures, postoperative complications and treatment, pathological examinations, postoperative adjuvant treatment, survival, and risk factors.Results: A total of 13,595 cases from 70 centers in 28 provinces were retrieved for analysis. This study reported the largest cohort of patients who underwent surgical treatment for pancreatic cancer in China to date. More cases were derived from the Eastern regions, among which Shanghai, Beijing, and Zhejiang ranked in the top three. The peak age of the patients ranged from 60 to 69 years. The ratio of males to females was 1.5:1. Overall, 64.3% of the tumors were located in the head and neck of the pancreas, and 35.7% in the body and tail of the pancreas. Of the patients, 23.0% underwent positron-emission tomography-computed tomography, 21.6% underwent endoscopic ultrasound, and 4.8% underwent preoperative biopsy. Two percent of the patients underwent neoadjuvant treatment, while 68.9% underwent R0 surgical resection (margin free of tumor cells). Of the latter, 78.6% of the operations were open procedures, 12.6% were laparoscopic procedures, 2.9% were robotic procedures, and 3.7% were converted to open procedures. The in-hospital mortality rate after surgery was 0.4%. The incidence of grade 2 and grade 3 postoperative pancreatic fistulas was 25.5% and 2.5%, respectively. The incidence of complications based on the Clavien-Dindo classification was 17.9% of grade II, 4.3% of grade IIIa, 1% of grade IIIb, and 0.6% of grade IV. Of the patients, 28.9% underwent postoperative adjuvant chemotherapy. The 1-year, 2-year, and 3-year overall survival of these patients were 77%, 51%, and 38%, respectively. In the 8542 patients who underwent R0 resection, the 1-year, 2-year, and 3-year overall survival and disease-free survival were 77% , 54%, and 43%, and 68%, 49%, and 41%, respectively. The factors related to the prognosis of these patients were also identified after uni-and multi-variate analyses.Conclusion: The surgical quality, safety, and long-term survival of the patients in CPDC are similar to those of international high-volume pancreatic centers. 6Metformin inhibits pancreatic cancer metastasis caused by SMAD4 deficiency and consequent HNF4G upregulation显示文摘Pancreatic ductal adenocarcinoma(PDAC)has poor prognosis due to limited therapeutic options.This study examines the roles of genome-wide association study identified PDAC-associated genes as therapeutic targets.We have identified HNF4G gene whose silencing most effectively repressed PDAC cell invasiveness.HNF4G overexpression is induced by the deficiency of transcriptional factor and tumor suppressor SMAD4.Increased HNF4G are correlated with SMAD4 deficiency in PDAC tumor samples and associated with metastasis and poor survival time in xenograft animal model and in patients with PDAC(log-rank P=0.036;HR=1.60,95%CI=1.03–2.47).We have found that Metformin suppresses HNF4G activity via AMPK-mediated phosphorylation-coupled ubiquitination degradation and inhibits in vitro invasion and in vivo metastasis of PDAC cells with SMAD4 deficiency.Furthermore,Metformin treatment significantly improve clinical outcomes and survival in patients with SMAD4-deficient PDAC(log-rank P=0.022;HR=0.31,95%CI=0.14–0.68)but not in patients with SMAD4-normal PDAC.Pathway analysis shows that HNF4G may act in PDAC through the cell-cell junction pathway.These results indicate that SMAD4 deficiency-induced overexpression of HNF4G plays a critical oncogenic role in PDAC progression and metastasis but may form a druggable target for Metformin treatment.Chengcheng Wang Taiping Zhang Quan Liao Menghua Dai Junchao Guo Xinyu Yang Wen Tan Dongxin Lin Chen Wu Yupei Zhao 2021Protein & Cell2021,12,2:5
7Pancreatic head cancer: Open or minimally invasive pancreaticoduodenectomy?显示文摘Pancreatic head cancer still represents an insurmountable barrier for patients and pancreatic surgeons.Pancreaticoduodenectomy(PD)continues to be the operative standard of care and potentially curative procedure for pancreatic head cancer.Despite the rapid development of minimally invasive techniques,whether the efficacy of minimally invasive pancreaticoduodenectomy(MIPD)is noninferior or superior to open pancreaticoduodenectomy(OPD)remains unclear.In this review,we summarized the history of OPD and MIPD and the latest staging and classification information for pancreatic head cancer as well as the proposed recommendations for MIPD indications for patients with pancreatic head cancer.By reviewing the MIPD-vs.OPD-related literature,we found that MIPD shows noninferiority or superiority to OPD in terms of safety,feasibility,enhanced recovery after surgery(ERAS)and several short-term and long-term outcomes.In addition,we analyzed and summarized the different MIPD outcomes in the USA,Europe and China.Certain debates over MIPD have continued,however,selection bias,the large number of low-volume centers,the steep MIPD learning curve,high conversion rate and administration of neoadjuvant therapy may limit the application of MIPD for pancreatic head cancer.Mengyu Feng Zhe Cao Zhiwei Sun Taiping Zhang Yupei Zhao 2019Chinese Journal of Cancer Research2019,31,6:4

9Advance in herpes simplex viruses for cancer therapy显示文摘Oncolytic virotherapy is an attractive approach that uses live viruses to selectively kill cancer cells. Oncolytic viruses can be genetically engineered to induce cell lyses through virus replication and cytotoxic protein expression. Herpes simplex virus (HSV) has become one of the most widely clinically used oncolytic agent. Various types of HSV have been studied in basic or clinical research. Combining oncolytic virotherapy with chemotherapy or radiotherapy generally produces synergic action with unclear molecular mechanisms. Arming HSV with therapeutic transgenes is a promising strategy and can be used to complement conventional therapies. As an efficient gene delivery system, HSV has been successfully used to deliver various immuno-modulatory molecules. 10Mechanisms of obesity-and diabetes mellitus-related pancreatic carcinogenesis:a comprehensive and systematic显示文摘Research on obesity-and diabetes mellitus(DM)-related carcinogenesis has expanded exponentially since these two diseases were recognized as important risk factors for cancers.The growing interest in this area is prominently actuated by the increasing obesity and DM prevalence,which is partially responsible for the slight but constant increase in pancreatic cancer(PC)occurrence.PC is a highly lethal malignancy characterized by its insidious symptoms,delayed diagnosis,and devastating prognosis.The intricate process of obesity and DM promoting pancreatic carcinogenesis involves their local impact on the pancreas and concurrent wholebody systemic changes that are suitable for cancer initiation.The main mechanisms involved in this process include the excessive accumulation of various nutrients and metabolites promoting carcinogenesis directly while also aggravating mutagenic and carcinogenic metabolic disorders by affecting multiple pathways.Detrimental alterations in gastrointestinal and sex hormone levels and microbiome dysfunction further compromise immunometabolic regulation and contribute to the establishment of an immunosuppressive tumor microenvironment(TME)for carcinogenesis,which can be exacerbated by several crucial pathophysiological processes and TME components,such as autophagy,endoplasmic reticulum stress,oxidative stress,epithelialmesenchymal transition,and exosome secretion.This review provides a comprehensive and critical analysis of the immunometabolic mechanisms of obesity-and DM-related pancreatic carcinogenesis and dissects how metabolic disorders impair anticancer immunity and influence pathophysiological processes to favor cancer initiation.Rexiati Ruze Jianlu Song Xinpeng Yin Yuan Chen Ruiyuan Xu Chengcheng Wang Yupei Zhao 2023Signal Transduction and Targeted Therapy2023,8,4:2

12Versatile.design and synthesis of mesoporous sulfonic acid catalysts显示文摘Mesoporous sulfonic acid catalysts(MSAC) are widely used in acid-catalyzed reactions, including biomass conversions with plenty of polar solvents and precursors. The catalytic efficiency of MSAC is greatly affected by the microenvironment around the sulfonic acid sites. In this review, the progress on modification of microenvironment of MSAC is reviewed over the past decade. Hydrophobic modification allows MSAC prevent the adhesion of water molecules onto sulfonic acid sites, to abate the risk of reduced acid strength and catalytic efficiency. In comparison, hydrophilic properties can bring positive effect on acidcatalyzed reactions with the aid of hydrophilic interaction between polar functional groups on MSAC and hydrophilic groups of specific substrates. Amphiphilic MSAC with tunable wettability for specific substrates and solvents tend to improve the efficiency in certain reactions with mixed solvents or reactants of different polarity, especially for biphasic systems of immiscible liquids. Furthermore, much attention has been attracted on modification of surface to simulate the microenvironment of homogeneous solvents and enzyme biocatalysts in recent research. 13Selective potassium uptake via biocompatible zeolite–polymer hybrid microbeads as promising binders for hyperkalemia显示文摘Patients with chronic kidney disease are at high risk of hyperkalemia that is associated with various lifethreatening complications.Treatments primarily rely on orally administered potassium binding agents,along with low curative effects and various side effects.Herein,direct serum potassium uptake was realized via zeolite–heparin-mimicking-polymer hybrid microbeads.The preparation process involved the synthesis of the heparin-mimicking polymer via the in situ cross-linking polymerization of acrylic acid and N-vinylpyrrolidone in polyethersulfone solution,the fabrication of microbeads via zeolite-mixing,electro-spraying and phase-inversion,and the subsequent aqueous-phase modifications based on ion-exchange and metal-leaching.An ultra-high(about 88%)amount of zeolite could be incorporated and well locked inside the polymer matrix.Potassium uptake capability was verified in water,normal saline and human serum,showing high selectivity and fast adsorption.The microbeads exhibited satisfying blood compatibility,negligible hemolysis ratio,prolonged clotting time,inhibited contact activation,and enhanced antifouling property toward serum proteins and cells.The proposed approach toward zeolite–heparin-mimicking-polymer hybrid microbeads provided a cheap,efficient and safe treatment protocol of hyperkalemia for the high-risk patients.Zhoujun Wang Wei Sun Zhiwei Wei Jianxu Bao Xin Song Yupei Li Haifeng Ji Jue Zhang Chao He Baihai Su Weifeng Zhao Changsheng Zhao 2021Bioactive Materials2021,6,2:2
14Increased COMT expression in pancreatic cancer and correlation with clinicopathologic parameters显示文摘Catechol-O-methyl transferase (COMT) is an enzyme involved in estrogen metabolism. Proteomic and immunoproteomic screens suggested COMT might be an immunogenic membrane antigen in human pancreatic cancer. The aim of this study was to investigate the dynamic expression of COMT in pancreatic ductal adenocarcinoma (PDAC) and noncancerous pancreatic tissue, and to determine the relationship between COMT expression and clinicopathologic parameters. COMT expression was analyzed by reverse transcription-polymerase chain reaction (RT-PCR) and western blot in five pancreatic cell lines and five pairs of PDAC and noncancerous pancreatic tissue. Immunohistochemistry was used to evaluate COMT expression in tissue microarrays and 20 cases of paraffin-embedded clinical specimens. The results indicated that COMT expression was detected in AsPC-1, BxPC-3, MIA PaCa-2, Capan-1 and SW1990 pancreatic cell lines, and in five pairs of PDAC and noncancerous pancreatic tissue, at the mRNA and protein levels. Immunohistochemistry analysis revealed that COMT expression was significantly higher in PDAC than in nonmalignant pancreatic tissue. High expression of COMT significantly correlated to early T stages. 15Analysis of clinical characteristics and treatment of pancreatic cystic tumors显示文摘Objective: To summarize experience in the diagnosis and treatment of pancreatic cystic neoplasms.Methods: This is a retrospective study of 207 patients who were diagnosed with pancreatic cystic tumors at Peking Union Medical College Hospital between Jan 2009 and Mar 2014.Clinical data, such as clinical manifestations, radiological and pathological images and surgical recordings, were collected.Results: Of the 207 included patients, females accounted for 76.81%, and the mean patient age was 52.04 years.Malignancy was more common in older patients who presented with marasmus and jaundice.Other risk factors included solid components in the tumor, a large tumor size, and elevated levels of tumor markers.Surgical treatment was required when a malignant tumor was suspected.The operation approach was selected based on the location, size and characteristics of the tumor.The position of the tumor relative to the pancreatic duct also played a significant role.Conclusions: No specific symptoms were observed for the patients with pancreatic cystic tumors.Imaging played an important role in making a differential diagnosis.Furthermore, surgical treatment should be proposed for patients with significant symptoms and potentially malignant tumors.The tumor resection rate is high, suggestive of good prognosis.Lei You Jianchun Xiao Zhe Cao Wanying Zhang Quan Liao Menghua Dai Taiping Zhang Yupei Zhao 2016Chinese Journal of Cancer Research2016,28,5:1
