| 1 | mTOR signaling in skeletal development and disease显示文摘The mammalian/mechanistic target of rapamycin(m TOR) is a serine/threonine protein kinase that integrates inputs from nutrients and growth factors to control many fundamental cellular processes through two distinct protein complexes m TORC1 and m TORC2.Recent mouse genetic studies have established that m TOR pathways play important roles in regulating multiple aspects of skeletal development and homeostasis. In addition, m TORC1 has emerged as a common effector mediating the bone anabolic effect of Igf1,Wnt and Bmp. Dysregulation of m TORC1 could contribute to various skeletal diseases including osteoarthritis and osteoporosis.Here we review the current understanding of m TOR signaling in skeletal development and bone homeostasis, as well as in the maintenance of articular cartilage. | 3 | Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery. | Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang | 2020 | Light(Science & Applications)2020,9,1: | 6 |
| 4 | Soil Heavy Metal Pollution and Health Risk Assessment of Abandoned Land in Coal Mining Areas in Xingren County显示文摘Total contents of heavy metals( Pb,Cr,Cd,Hg,As,Cu,Zn,and Ni) in 68 soil samples were determined to evaluate the contamination level of soil from five coal mine areas in Xingren County,Guizhou province.The pollution levels of heavy metals were characterized by anthropogenic influence multiple,and the pollution of heavy metal in soil was evaluated using geo-accumulation index.Based on the health risk models recommended by the U.S.Environmental Protection Agency( US EPA),the health risks of heavy metals in the soils were assessed.The results showed that the abandoned land in the five coal mining areas were contaminated by heavy metals,and the order of pollution levels of eight heavy metals was As>Pb>Cu>Hg>Ni>Cr>Cd>Zn.The soils suffered moderate-heavy pollution from As and Pb,and the contents of As and Pb in the soils were 13.16 and 4.76 times higher than the background values of Guizhou Province,followed by Cu,Hg and Ni,while soil pollution from Cr,Cd and Zn was mild.The order of non-carcinogenic risks of the heavy metals in the soils was As>Pb>Cr>Ni>Cu>Hg>Cd>Zn,and the non-carcinogenic risk value of As was higher than that its threshold value 1,which indicated that As had a high potential health risk to adults and children.The order of carcinogen risks of the four heavy metals was As>Cd>Cr>Ni,and the carcinogenic risk value of As was higher than its threshold value,indicating that As had a high carcinogenic risk to adults and children.The carcinogenic risk values of other three heavy metals(Cd,Cr and Ni)were lower than the threshold value,so they had no carcinogenic risk to human health.Moreover,children were more sensitively affected by heavy metals from the abandoned land.Therefore,the health of children in such areas should be paid more attention to.The HI value of the eight heavy metals to children was seven times higher than that to adults.The contribution rate of HQ_(Asto) HI was about 88%,and the contribution rate of CR_(As) to TCR was about 98%,so As was the most crucial factor influencing non-carcinogenic and carcinogenic risk in the abandoned soils. | Qin Fanxin Pang Wenpin Liu Wenzheng | 2015 | Meteorological and Environmental Research2015,6,11: | 6 |

| 6 | Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19. | Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 4 |
| 7 | Unintended targeting of Dmp1-Cre reveals a critical role for Bmpr1a signaling in the gastrointestinal mesenchyme of adult mice显示文摘Cre/lox P technology has been widely used to study cell type-specific functions of genes. Proper interpretation of such data critically depends on a clear understanding of the tissue specificity of Cre expression. The Dmp1-Cre mouse, expressing Cre from a 14-kb DNA fragment of the mouse Dmp1 gene, has become a common tool for studying gene function in osteocytes, but the presumed cell specificity is yet to be fully established. By using the Ai9 reporter line that expresses a red fluorescent protein upon Cre recombination, we find that in2-month-old mice, Dmp1-Cre targets not only osteocytes within the bone matrix but also osteoblasts on the bone surface and preosteoblasts at the metaphyseal chondro-osseous junction. In the bone marrow, Cre activity is evident in certain stromal cells adjacent to the blood vessels, but not in adipocytes. Outside the skeleton, Dmp1-Cre marks not only the skeletal muscle fibers, certain cells in the cerebellum and the hindbrain but also gastric and intestinal mesenchymal cells that express Pdgfra. | 8 | Inducible expression of Wnt7b promotes bone formation in aged mice and enhances fracture healing显示文摘There remain unmet clinical needs for safe and effective bone anabolic therapies to treat aging-related osteoporosis and to improve fracture healing in cases of nonunion or delayed union. Wnt signaling has emerged as a promising target pathway for developing novel bone anabolic drugs. Although neutralizing antibodies against the Wnt antagonist sclerostin have been tested,Wnt ligands themselves have not been fully explored as a potential therapy. Previous work has demonstrated Wnt7b as an endogenous ligand upregulated during osteoblast differentiation, and that Wnt7b overexpression potently stimulates bone accrual in the mouse. The earlier studies however did not address whether Wnt7b could promote bone formation when specifically applied to aged or fractured bones. Here we have developed a doxycycline-inducible strategy where Wnt7b is temporally induced in the bones of aged mice or during fracture healing. We report that forced expression of Wnt7b for 1 month starting at 15 months of age greatly stimulated trabecular and endosteal bone formation, resulting in a marked increase in bone mass. We further tested the effect of Wnt7b on bone healing in a murine closed femur fracture model. Induced expression of Wnt7b at the onset of fracture did not affect the initial cartilage formation but promoted mineralization of the subsequent bone callus. | 9 | Differential involvement of Wnt signaling in Bmp regulation of cancellous versus periosteal bone growth显示文摘Bone morphogenetic proteins(Bmp) are well-known to induce bone formation following chondrogenesis,but the direct role of Bmp signaling in the osteoblast lineage is not completely understood. We have recently shown that deletion of the receptor Bmpr1 a in the osteoblast lineage with Dmp1-Cre reduces osteoblast activity in general but stimulates proliferation of preosteoblasts specifically in the cancellous bone region,resulting in diminished periosteal bone growth juxtaposed with excessive cancellous bone formation.Because expression of sclerostin(SOST), a secreted Wnt antagonist, is notably reduced in the Bmpr1 adeficient osteocytes, we have genetically tested the hypothesis that increased Wnt signaling might mediate the increase in cancellous bone formation in response to Bmpr1 a deletion. Forced expression of human SOST from a Dmp1 promoter fragment partially rescues preosteoblast hyperproliferation and cancellous bone overgrowth in the Bmpr1 a mutant mice, demonstrating functional interaction between Bmp and Wnt signaling in the cancellous bone compartment. To test whether increased Wnt signaling can compensate for the defect in periosteal growth caused by Bmpr1 a deletion, we have generated compound mutants harboring a hyperactive mutation(A214 V) in the Wnt receptor Lrp5. However, the mutant Lrp5 does not restore periosteal bone growth in the Bmpr1 a-deficient mice. | 12 | Insights into the floatability between spodumene and albite from crystal chemistry standpoint显示文摘Spodumene is a silicate mineral rich in lithium.However,the gangue mineral,albite has similar crystal chemical structure and composition to spodumene.In this work,the density functional theory(DFT)calculation was taken to research the floatability from the perspective of crystal chemistry.And contact angle test and reagent adsorption test were used to support the simulation results.In addition,the weighted total density of broken bonds is proposed for the first time to judge the generation probability of cleavage surface.The DFT calculation results display that the Na and Al sites on the albite surface are the adsorption sites of water molecules.While the water molecules only bond with Al atom on the surface of spodumene and each Al site can adsorb only one water molecule.Thus,the wetting effect of water molecules on the albite surface is stronger than that of spodumene.This is consistent with the result of contact angle.In sodium oleate system,the oleic acid anion is adsorbed on the spodumene surface in form of a multicomponent ring while the albite is a single ring.Theoretically,oleic acid anion can be strongly adsorbed on the surface of spodumene and albite under vacuum.The adsorption strength of spodumene is higher than that of albite.However,on hydrated surface,the adsorption strength of oleic acid anion on mineral surface is greatly reduced due to the interaction between water molecules and metal site on mineral surface.Thus,spodumene and albite are hard to float without external activated ions in sodium oleate system. | Hepeng Zhou Fanxin Xie Yongbing Zhang Boyuan Zhang Siqi Yang Xianping Luo | 2022 | International Journal of Mining Science and Technology2022,32,6: | 1 |
